Comparative Study of Antioxidant Activity of Apple and Pear Peel

  • Apple and pear are popular fruits consumed in Korea and are common fruit in daily diet. In order to compare the antioxidant activity of the apple and pear peels, total polyphenol contents, total flavonoid contents, $ABTS^+$ free radical scavenging activity, and DPPH free radical scavenging activity were measured from hot water, ethanol, and methanol extracts of the two fruit peels. The total polyphenol and flavonoid contents were highest in 95% methanol extracts of the apple peelsand 70% ethanol extract of the pear peels, respectively. Total polyphenol contents of the pear peels were higher than that of apple peels, and total flavonoid contents of the apple peels were higher than that of pear peels. The apple and pear peels had the highest $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity in 95% methanol extracts and 70% ethanol extracts, respectively. $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity of pear peels was higher than that of apple peels, and the DPPH free radical scavenging activity of apple and pear peels were detected in hot water, 95% methanol, and 70% ethanol extracts, respectively. Ascorbic acid, a synthetic antioxidant used as positive control, had significantly higher scavenging activity than the apple and pear peels. In conclusion, the apple and pear peelshave great potential as natural antioxidants. Therefore, above results should be considered to provide the possibility for the development of high functional antioxidants.

Antimicrobial and Antioxidative Effects of Isoflavonoids on Skin Microbial Inflammation Pathogens (Isoflavonoid의 피부염증균에 대한 항균 및 항산화효과)

  • Puerarin and daidzin have been isolated from Puerariae thunbergiana Benth. Structures were determined by spectroscopic methods. Compounds showed weak antimicrobial activity against S. mutans, S. epidermidis, S. aureus, and C. albicans (MIC, $800{\mu}g/ml$). However, these compounds were not antioxidative. Puerariae thunbergiana Benth. extracts against microorganisms were evaluated in terms of the minimum inhibitory concentrations (MIC). In general, C. albicans was stronger antimicrobial activity than the other microorganisms. The antioxidative activity of was observed in the etyl acetate extract ($IC_{50},\;119.87{\pm}0.16\;{\mu}g/ml$). The DPPH radical scavenging effect ($IC_{50},\;1,673.3{\pm}0.54\;{\mu}M$) of the puerarin was comparable with that of synthetic antioxidant, BHA $(IC_{50},\;88.39{\pm}1.1){\times}10^{3}{\mu}M$. These results suggest that puerarin and daidzin have a potential antimicrobial activity.

Effects of Culture Conditions on Mycelial Growth and Polysaccharide Production of Tricholoma matsutake in Bioreactor

  • This experiment was carried out to obtain the optimal liquid culture conditions for the mycelial growth and the polysaccharide production of Tricholoma matsutake. For the mycelial growth and polysaccharide production, the synthetic medium was optimized with containing glucose 40 g/L, yeast extract 30 g/L, $KH_2PO_4$ 1.5 g/L and $MgSO_4.7H_2O$ 1 g/L. The effects of agitation and aeration were investigated for the cell growth and the polysaccharide production in batch culture. The biomass and polysaccharide concentrations were 21.87 g/L at 150 rpm and 8.86 g/L at 300 rpm, respectively. And the biomass concentration and the polysaccharide production were 20.85 g/L at 0.5 vvm and 8.83 g/L at 1.5 vvm, respectively.

Charateristics of Akalophilic Microorganism Developed for Color Removal of Dye (염료의 색도 제거를 위해 개발된 호알칼리성 미생물의 특성)

  • An alkalophilic microorganism capable of degrading dyes was developed for the treatment of alkaline dye solution. This strain was identified as Pseudomonas species. Using this microorganism, biological treatment of dye was studied in Erlenmeyer flasks. The characteristics of this microorganism were observed under various incubating-condition such as temperature, pH, nitrogen source, and macronutrients concentration. The removal effciencies of Disperse Red 60 from synthetic wastewater were 33.5 ~ 36.9% at the range of $30{\sim}40^{\circ}C$, and they were 31.1 ~ 36.7% at the range of initial pH 8 ~ pH 10, respectively. The optimal culture medium was found to be 0.25%(w/v) yeast extract, 0.25%(w/v) polypeptone, 0.1%(w/v) $KH_2PO_4$, 0.2%(w/v) $MgSO_4{\cdot}7H_2O$, and 1.0%(w/v) $Na_2CO_3$. In treatment of various dyes using Erlenmeyer flasks, the removal effciencies of Disperse Blue 87, Disperse Yellow 64, Disperse Red 60, Acid Blue 193, Acid Red 138, and Direct Yellow 23 were found to be 76%, 71%, 58%, 93%, 94%, and 90% respectively after 24hrs reaction of alkalophilic strain Pseudomonas sp. YBE-12.

Insecticidal Effect of Neem Cake Extracts on Cabbage Pests, Aphis gossypii and Pluetella xylostella

  • In organic agriculture, choose of effective and cheap bio-pesticide is very important. The authors developed an insecticidal extract from neem cake, waste of neem oil from kernel, and applied as a bio-pesticide. Bio-pesticide neem cake extracts experiment on cabbage pest was carried out at Wonju Agricultural Technology and Extension Center from 11 March to 30 May 2003. There were six treatments with three replications, using completely randomized design. Treatments involved three and six sprays of synthetic pyrethroid pesticide cypermethrin 10 EC at the dilution rate of 2.2 mL $L^{-1}$ of distilled water and four, five and six sprays of bio-pesticide neem at the dilution rate of 13.3 mL $L^{-1}$ of distilled water, and untreated control. For each treatment, designated sprayings were done at 7 days interval. Pre-spray data showed that the plants in all the experimental plots were already infested with aphid (Aphis gossypii), and diamondback moth (Pluetella xylostella). The results indicated that all neem pesticide treatments were more effective in insecticidal activity than the untreated control and the chemical treatments in controlling aphids and diamondback moth. Among the three neem treatments, there were no significant differences between them.

Study on Bio-H2 Production from Synthetic Food Waste and Activated Sludge from Industrial Waste Water Processes using Dark-fermentation (산업공정의 폐수처리에서 발생된 폐활성슬러지 및 인공음식폐기물을 이용한 생물학적 수소생성에 관한 연구)

  • This study performed to extract operation factors of major organic wastes, which were food wastes and waste activated sludge generated in industries in order to use them as a substrate for bio-H2 production. According to the results of experimental analysis for hydrogen production capacity by various organic concentrations, the hydrogen production yield was the highest at 80 g/L, and the efficiency was improved by the pretreatment of waste activated sludge (acid treatment, alkali treatment). Hydrogen production efficiency was improved by mixing food wastes and waste activated sludge if waste activated sludge was below than 30%, however, it was decreased when it was more than 50%. The impacts of heavy metals on the hydrogen production shows that the inhibition level depends on the concentration of Cr, Zn, and Cu, Fe was able to enhance the hydrogen production.

Bioactive Phenolic Constituents from the Culms of Phyllostachys bambusoides

  • In our search for bioactive phenolics from plants, the culms of Phyllostachys bambusoides has been selected for investigation of anti-cariogenic and 1,1-diphenylpicrylhydrazyl (DPPH) radical scavenging agents based on the initial screening results. Fractionation process of n-hexane and $CHCl_3$ extracts afforded four phenolic constituents, ferulic acid (1), vanillin (2), coniferaldehyde (3), and coniferyl alcohol (4) as guided by their DPPH free radical scavenging activities. Additionally, activity-guided fractionation of EtOAc extract with anti-cariogenic activity has resulted in the isolation of coniferaldehyde (3), 2,6-dimethoxy-p-benzoquinone (5), p-methoxycinnamic acid (6), (${\pm}$)-balanophonin (7), and 6-methoxychromanone (8). The structures of 1 - 8 were determined by spectroscopic data interpretation, and also by comparison of their data with the published values. Phenolic compounds 1 - 4 exhibited similar DPPH radical scavenging activities compared with the synthetic antioxidant, butylated hydroxytoluene (BHT), and compounds 3 and 5 - 8 showed significant antibacterial activity against cariogenic oral streptococci, Streptococcus mutans and S. sobrinus.

  • In this paper, the design technique of the wide swath TOPS(Terrain Observation by Progressive Scan) imaging mode is introduced. The TOPS mode overcomes the scalloping limitations imposed by ScanSAR mode by steering the antenna pattern along track direction during the acquisition of a burst. This paper reports the operation concept of TOPS imaging and mode design result to extract the SAR operational parameters. Finally, several analyzed results such as IRF(Impulse Response Function), NESZ(Noise Equivalent Sigma Zero) and DTAR(Distributed Target Ambiguity Ratio) are presented.

Composition in Carotenoids of Rainbow Trout Fed with CLA and Ascidian Tunic Extracts (CLA와 멍게 껍질 추출물 급이에 따른 무지개송어 색소조성)

  • Astaxanthin is a valuable pigment source for many aquacultured species, including salmonoids, shrimp, sea bream, and ornamental species. Conjugated linoleic acid (CLA) and ascidian tunic extracts were mixed with the basal diet of rainbow trout to investigate their pigmentation effects. Synthetic Carophyll Pink and natural carotenoids that came from the tunic extracts were incorporated into muscle and skin tissues. The main carotenoids found in muscle after 8 weeks were canthaxanthin in CP12 (13.4%), and CP52 (17.2%), and astaxanthin in CP12 (58.5%), and CP52 (59.2%) in the Carophyll Pink group, while those in skin were canthaxanthin in CP14 (34.5%), and CP54 (29.2%), and astaxanthin in CP14 (32.0%), and CP54 (36.5%) in the ascidian tunic extract group. The total carotenoid content in skin (53.0-69.3 mg/kg) was greater than that in muscle (9.5-13.8 mg/kg).

Chinese Skullcap (Scutellaria baicalensis) inhibits inflammation and proliferation on benign prostatic hyperplasia in rats

  • Benign prostatic hyperplasia (BPH), which is the most common disorder in elderly men, involves androgenic hormone imbalance with chronic inflammation that causes imbalance between cell apoptosis and cell proliferation. As the root cause of the BPH remains unclear and synthetic drugs for treatment of BPH have undesirable side effects, the development of effective alternative medicines has been considered. Chinese Skullcap has been considered natural remedy to treat pyrexia, micturition disorder and inflammation. Although skullcap has effective properties on various diseases, the effects and molecular mechanism of Skullcap on BPH are not fully understood. Therefore, in this study, we evaluated the efficacy of Chinese Skullcap root extract (SRE) in testosterone-induced BPH rats. Compared with the untreated group, the SRE treatment group suppressed pathological alterations, such as prostate growth and increase in serum dihydrotestosterone and $5{\alpha}$-reductase levels. Furthermore, SRE significantly decreased the expression of androgen receptor and proliferating cell nuclear antigen. SRE also restored Bax/Bcl-2 balance. These effect of SRE was more prevalent than commercial $5{\alpha}$-reductase inhibitor, finasteride. Taken together, we propose that SRE suppresses abnormal androgen events in prostate tissue and inhibits the development of BPH by targeting inflammation- and apoptosis-related markers. These finding strengthens that SRE could be used as plant-based $5{\alpha}$-reductase inhibitory alternative.
