Changes in Protein Synthesis Induced by Chilling in Tomato Chloroplasts

  • To find out the effect of low temperature on the regulation of tomato chloroplast genes, the optimization of the system in chloroplast protein synthesis and the identification of the changes in chloroplast protein synthesis induced by chilling were studied. Incorporation reaction occurred rapidly at the first 30 minutes and was constantly maintained after 60 minutes. A broad optimal temperature on protein synthesis was found around 20 to $30^{\circ}C$. No difference was shown in the chloroplast protein synthesis under high light intensity (1600 ${\mu}E/m^2/s$) as well as under low light intensity (400 ${\mu}E/m^2/s$) even darkness. $K^+$, $Mg^{++}$ and ATP at an optimal concentration act as an activator, while DTT, chloramphenicol, cycloheximide, $Ca^{++}$ and inorganic phosphate act as an inhibitor in the chloroplast protein synthesis. Synthesis of 15, 55 and 60 kd chloroplast encoded stromal proteins and 18, 24, 33 and 55 kd chloroplast encoded thylakoid membrane proteins were reduced by chilling, while 17 kd chloroplast encoded stromal protein and 16 kd chloroplast encoded thylakoid membrane protein was induced by chilling. It was expected that the 55 kd stromal protein would be the large subunit of rubisco and the 33 kd thylakoid membrane protein would be the D1 protein which was drastically reduced by chilling.

중금속이 Cyanobacterium synechocystis sp.PCC 6803의 성장과 단백질 합성에 미치는 영향 (Effects of Heavy Metals on Growth and Protein Synthesis in Cyanobacterium synechocystis sp. PCC 6803)

  • The changes of growth and protein synthesis pattern by aluminum (Al), cadmium (Cd), zinc (Zn) treatments were studied in Cyanobacterium synechocystis sp. PCC 6803. When exposed to Al from 5ppm to 3oppm, synechocystis grows normally. But more than that retard the growth of algae notably. The 0.05ppm Cd additions had no effect on the growth of algae. 0.1, 0.2, and 0.5ppm Cd inhibited growth. Under 1 and 2ppm Cd stress, growth was greatly diminished. Zn had dual effects. The growth of algae in media containing 5ppm was stimulated. As concentration increases more than l5ppm, growth inbition increases. Under 25ppm Zn stress, growth was greatly diminished. According to logistic theory, r and K values of each heavy metal-treated groups were estimated. Correlation analysis of r and K values with metal concentration shows that there is negative correlation between K and concentration in Cd and Zn treatments. Critical concentration which shows lethal or sublethal effect was estimated by t-test of each r and K value. The cells cultured in 10, 20, 30, 40 and 5oppm of Al, 1 and 2ppm of Cd, and 10, 15, 20, 25 and 30ppm of Zn for 4 days was used for protein analysis. Analysis of protein synthesis with SDS-PACE showed alterations of protein synthesis pattern. The synthesis of protein about 220kD increased markedly. In this study, it showed that resistance mechanism against Al, Cd, and Zn is K selection and that metal stress induced the change of protein synthesis in Cyanobacterium synechocystis sp. The Inhibitory Effects of Ahnjeonbaekho-tang on FRTL-5 Cell Proliferation and Thyroxine Synthesis

  • Objective : Graves' disease, the most common cause of hyperthyroidism, is an autoimmune disorder associated with autoantibodies to the TSH receptor. The clinical features of Graves' disease are goiter and hypermetabolic symptoms induced by excessive hormones. Antithyroid drug therapy is the first-line treatment for Graves' disease in Korea, Japan and European countries. Yet in spite of a long period and high-dose of treatment, it is hard to achieve remission because of adverse effects, frequent recurrence and resistance to antithyroid drugs. Recently, it has been reported that the abnormal thyroid hormone and clinical symptoms of Graves' disease were reduced by Ahnjeonbaekho-tang (AJBHT). Methods : To investigate the effectiveness and action mechanism of AJBHT, we studied the influence of AJBHT on FRTL-5 thyroid cell proliferation, DNA synthesis and expression of T4, TSH, cAMP, Tg and TPO mRNA. Results : AJBHT significantly inhibited the FRTL-5 cell proliferation, DNA synthesis, T4 synthesis, cAMP production and the expression of Tg mRNA in comparison with control and MMI. Conclusions : These results suggest that AJBHT may inhibit the cell proliferation and DNA synthesis by regulating the cAMP, and suppress the T4 synthesis by modulating Tg mRNA expression and cAMP synthesis, and that it may be useful agent for treating the goiter and hormone abnormality of Graves' disease.

Glucosamine Hydrochloride and N-Acetylglucosamine Influence the Response of Bovine Chondrocytes to TGF-β3 and IGF in Monolayer and Three-Dimensional Tissue Culture

  • BACKGROUND: Glucosamine hydrochloride (GlcN HCl) has been shown to inhibit cell growth and matrix synthesis, but not with N-acetyl-glucosamine (GlcNAc) supplementation. This effect might be related to an inhibition of critical growth factors (GF), or to a different metabolization of the two glucosamine derivatives. The aim of the present study was to evaluate the synergy between GlcN HCl, GlcNAc, and GF on proliferation and cartilage matrix synthesis. METHOD: Bovine chondrocytes were cultivated in monolayers for 48 h and in three-dimensional (3D) chitosan scaffolds for 30 days in perfusion bioreactors. Serum-free (SF) medium was supplemented with either growth factors (GF) $TGF-{\beta}$ ($5ng\;mL^{-1}$) and IGF-I ($10ng\;mL^{-1}$), GlcN HCl or GlcNAc at 1mM each or both. Six groups were compared according to medium supplementation: (a) SF control; (b) SF + GlcN HCl; (c) SF + GlcNAc; (d) SF + GF; (e) SF + GF + GlcN HCl; and (f) SF + GF + GlcNAc. Cell proliferation, proteoglycan, collagen I (COL1), and collagen II (COL2) synthesis were evaluated. RESULTS: The two glucosamines showed opposite effects in monolayer culture: GlcN HCl significantly reduced proliferation and GlcNAc significantly augmented cellular metabolism. In the 30 days 3D culture, the GlcN HCl added to GF stimulated cell proliferation more than when compared to GF only, but the proteoglycan synthesis was smaller than GF. However, GlcNAc added to GF improved the cell proliferation and proteoglycan synthesis more than when compared to GF and GF/GlcN HCl. The synthesis of COL1 and COL2 was observed in all groups containing GF. CONCLUSION: GlcN HCl and GlcNAc increased cell growth and stimulated COL2 synthesis in long-time 3D culture. However, only GlcNAc added to GF improved proteoglycan synthesis.

분산형 시스템을 적용한 음성합성에 관한 연구 (A Study on Speech Synthesizer Using Distributed System)

  • 최근 광대역 무선 통신망의 보급과 소형 저장매체의 대용량화로 인하여 이동형 단말기가 주목 받고 있다. 이로 인해 이동형 단말기에 문자정보를 청취할 수 있도록 문자를 음성으로 변환해 주는 TTS(Text-to-Speech) 기능이 추가되고 있다. 사용자의 요구사항은 고음질의 음성합성이지만 고음질의 음성합성은 많은 계산량이 필요하기 때문에 낮은 성능의 이동형 단말기에 는 적합하지 않다. 본 논문에서 제안하는 분산형 음성합성기 (DTTS)는 고음질 음성합성이 가능한 코퍼스 기반 음성합성 시스템을 서버와 단말기로 나누어 구성한다. 서버 음성합성 시스템은 단말기에서 전송된 텍스트를 데이터베이스 검색 후 음성파형 연결정보를 생성하여 단말기로 전송하고, 단말기 음성합성 시스템은 서버 음성합성 시스템에서 생성된 음성파형 연결정보와 단말기에 존재하는 데이터베이스를 이용하여 간단한 연산으로 고음질 합성음을 생성할 수 있는 시스템이다. 제안하는 분산형 합성기는 단말기에서의 계산량을 줄여 저가의 CPU 사용, 전력소모의 감소, 효율적인 유지보수를 할 수 있도록 하는 장점이 있다.
