Mulberry Low-Fat Ice Cream Supplemented with Synbiotic: Formulation, Phytochemical Composition, Nutritional Characteristics, and Sensory Properties

  • For this study, we designed and produced mulberry low-fat ice cream supplemented with synbiotics (MLF-ISS). The sensory characteristics and physical, chemical, and microbiological qualities of MLF-ISS were then determined. Mulberry juice inoculated with or without probiotic (Lactobacillus plantarum TISTR 926 and Saccharomyces boulardii CNCM I-745) was also tested at 37℃ for 24 h to determine probiotic growth rate, pH, total anthocyanin content (TAC), total phenolic content (TPC), and antioxidant activity (AA). Only the TAC of mulberry juice inoculated with S. boulardii CNCM I-745 increased considerably (p < 0.05) among these parameters. MLF-ISS was produced with varied mulberry fruit concentrations (0, 10, 20, 30, or 40%) (w/w). The MLF-ISS prepared with 30% mulberry fruit (w/w) (30-MLF-ISS) had a higher score in appearance, color, and sweetness (p < 0.05) when sensory qualities were measured using the 9-point hedonic scale method. In the CIE lab system (L*, a*, b*), the color values of 30-MLF-ISS were 27.80 ± 0.26, 12.99 ± 0.59, and 1.43 ± 0.05, respectively. The 30-MLF-ISS was also subjected to a proximate analysis. The melting rate of 30-MLF-ISS was 0.29 ± 0.03 g/min and the time it took for the first drop to fall was 37.00 ± 7.00 min. TAC, TPC, and AA of 30-MLF-ISS were observed to alter significantly (p < 0.05) during varied intervals of storage at - 18℃ (0, 30, and 60 days). The viability of probiotics in 30-MLF-ISS slightly decreased after storage at -18℃ for 8 weeks, but remained about 6 log CFU/g. During storage at -18℃ for 0 and 120 days, no pathogenic bacteria were detected in 30-MLF-ISS. These findings show that 30-MLF-ISS has nutritional and functional value, is free of foodborne pathogenic bacteria, is safe for consumers' health, and is suitable for application in the ice cream and related food industries.

Synbiotic Synthesis of Oligosaccharides During Milk Fermentation by Addition of Leuconostoc Starter and Sugars

  • Synthesis of oligosaccharides during milk fermentation was attempted by inoculating Leuconostoc citreum with Lactobacillus casei, Lb. delbrueckii subsp. bulgaricus, and Streptococcus thermophilus as starters. Dextransucrase of Ln. citreum worked as a catalyst for the transglycosylation reaction of sugars; sucrose was added as the glucose donor, and lactose or maltose acted as the acceptor compound for the reaction. When 4% sucrose was added in milk, glucosyl-lactose was synthesized (about 1%, w/v) after 1-2 days of fermentation at 15 or $25^{\circ}C$. Alternatively, when sucrose and maltose (2% each, w/v) were added, panose (about 1 %, w/v) and other isomaltooligosaccharides were made in a day at $15-35^{\circ}C$. Growth patterns of lactobacilli and streptococci starters were not affected by the coculture of leuconostoc starter, but the rate of acid synthesis was slightly slowed at every temperature. Addition of sugars in milk did not give any adverse effect on the lactate fermentation. Accordingly, the use of leuconostoc starter and addition of sugars in milk allowed the production of oligosaccharides-containing fermented milk, and application of this method will facilitate the extensive development of synbiotic lactate foods.

  • Disease associated with microorganisms are far from resolved by current therapeutics. One of effective approach to health maintenance and disease control is the use of dietary bacterial and carbohydrate supplements. This comprises use of probiotics and prebiotics. Probiotics mean the live microorganisms, which when administered in adequate amounts confer a health benefit on the host. Prebiotics mean a nondigestible food ingredient that beneficially affects the host by selectively stimulating the growth and/or activity of one or a limited number of bacteria that can Improve the host health. Especially, probiotics has the relation which is close with innate immunity and adaptive immunity. And probiotics has the clinical value with many disease like lactose intolerance, constipation, acute gastroenteritis, food hypersensitivity and allergy, atopic dermatitis, crohn's disease, rheumatoid arthritis, pelvic radiotherapy, intestinal inflammation and chemical exposure, colon cancer, inhibitory effect of Helicobacter pylori and lowering the level of cholesterol. We use jointly korean medicine and probiotics and it has the more therapeutic effect in the many disease.

Prevalence of canine heartworm infection among dogs in Kwangju and Chonnam area (광주와 전남지역 사육견의 개심장사상충(Dirofilaria immitis) 감염률 조사)

  • We have investigated the prevalence of canine heartworm (Dirofilaria immitis) infection among 100 mixed-breed dogs raised in Chonnam Province (Koheung, Posung, Changheung, and Hwasoon) and 100 dogs randomly selected from visiting dogs to the College of Veterinary Medicine, Chonnam National University for routine physical examination. Blood samples taken from dogs were examined for the presence of Dirofilaria immitis microfilaria by the modified Knott's test, and an antigen test which was based on the sandwich ELISA(DiroCHEK(equation omitted), Synbiotics, Inc., San Diego, California, USA) was used to detect the adult heartworm-specific antigen from serum samples. The results were then compared with those of the modified Knott's test. Five(2.5%) of the 200 examined dogs were microfilaria-positive, while 8 dogs(4%) were anti-gen-positive, which suggested that the antigen test was more sensitive than the microfilarial test in detecting heartworm infection. All dogs that were microfilaria-positive were also antigen-positive. The place of origin of the heartworm-positive dogs were of Naju (4/26, 15.4%), Koheung(3/32, 9.4%), and Hwasoon (1/19, 5.3%), while none of dogs raised in Kwangju, Posung, and Changheung areas were infected with the parasite. Although not significant, more infected dogs were observed in older ages, while the difference in infection rate between male (4/98, 4.1%) and female (4/102, 3.9%) was not recognized. Our results indicated that a relatively low infection rate was observed in dogs raised in Kwangju and Chonnam area compared to the infection rate reported previously in other areas of South Korea.

A New Method of Producing a Natural Antibacterial Peptide by Encapsulated Probiotics Internalized with Inulin Nanoparticles as Prebiotics

  • Synbiotics are a combination of probiotics and prebiotics, which lead to synergistic benefits in host welfare. Probiotics have been used as an alternative to antibiotics. Among the probiotics, Pediococcus acidilactici (PA) has shown excellent antimicrobial activity against Salmonella Gallinarum (SG) as a major poultry pathogen and has improved the production performances of animals. Inulin is widely used as a prebiotic for the improvement of animal health and growth. The main aim of this study was to investigate the antimicrobial activity of inulin nanoparticle (IN)-internalized PA encapsulated into alginate/chitosan/alginate (ACA) microcapsules (MCs) for future in vivo application. The prepared phthalyl INs (PINs) were characterized by DLS and FE-SEM. The contents of phthal groups in the PINs were estimated by $^1H-NMR$ measurement as 25.1 mol.-%. The sizes of the PINs measured by DLS were approximately 203 nm. Internalization into PA was confirmed by confocal microscopy and flow cytometry. The antimicrobial activity of PIN-internalized probiotics encapsulated into ACA MCs was measured by coculture antimicrobial assays on SG. PIN-internalized probiotics had a higher antimicrobial ability than that of ACA MCs loaded with PA/inulin or PA. Interestingly, when PINs were treated with PA and encapsulated into ACA MCs, as a natural antimicrobial peptide, pediocin was produced much more in the culture medium compared with other groups with inulin-loaded ACA MCs and PA encapsulated into ACA MCs.

Perinatal Nutrition, Post-Hatch Holding Time and In Ovo Feeding (부화 전후 영양, 부화 후 유지시간, 그리고 In Ovo Feeding)

  • The most important times during the development of young chicks are the days immediately prior to hatching and the days immediately after hatching, known as the perinatal period. A sufficient supply of nutrients during the perinatal period is a crucial during the late stage of embryonic development and the starvation period of the young chicks. The delayed post-hatch holding time can restrict the development of the gastrointestinal tract, reduce final body weight, impair muscle development, and change immunological capacities. These symptoms are deleterious to the development of young chicks. Therefore, the post-hatch holding time and its influence on the fitness of young chicks are major concerns to the poultry industry. The in ovo feeding is a practical technology for perinatal nutrition to optimize poultry production and for attenuating the stress experienced by fasting young chicks. This study will discuss in ovo feeding and its effect on the development of the chick embryo, the establishment of a healthy microbiota, and the improving immune response.

Prevalence-based Interpretation of Predictive Values of Diagnostic Tests: An Example for Detection of Canine Heartworm Infection (진단키트 검사결과에 대한 유병율 위주 해석: 개 심장사상충의 예)

  • The use of screening tests as part of a diagnostic work-up is common in domestic canine practice, but understanding of the diagnostic test characteristics and factors affecting diagnostic accuracy is not clear among clinicians. This article was aimed to provide clinicians with a better understanding on the selection of test kits and with a proper interpretation of test results using an example from heartworm(Dirofilaria immitis) studies. From the literatures, diagnostic accuracy varied depending on the kits: percent average sensitivity and specificity of ELISA antigen-detecting kits were DiroChek(Synbiotics, USA) 78.1 and 95.2, SNAP(IDEXX, USA) 66.3 and 98.1, and Solo Step(Heska, Switzerland) 69.5 and 97.5, respectively, while the values for three hematological methods(Modified Knott's, direct smear and capillary tube) ranged from 38.4 to 81.8% and from 96.9 to 100%, respectively. Furthermore, it was also reported that the prevalence of heartworm disease in domestic dog populations varied depending on the regions studied: 2.5-22.8% for microfilarial test and 2.2-66.3% by ELISA. The values of predictive values for positive(PPV) and negative(NPV) provide useful information to clinicians on the probability of heartworm infection, but the PPV and NPV are greatly dependent on the heartworm prevalence. This suggests that PPV or NPV values of a test should be interpreted carefully in different clinical settings. Practical methods on the interpretation taking into account heartworm prevalence were discussed.

White Rice Fermented by Clostridium butyricum IDCC 9207 as an Alternative to Antibiotic having Antibacterial and Immunostimulatory Activity (항생제 대체제로서 Clostridium butyricum IDCC 9207 백미 발효물의 항균작용과 장관 면역증진효과)

  • The antagonistic activities against animal entero-pathogenic bacteria were investigated with 444 natural substances fermented by various probiotics. A white rice product fermented (FWR) by Clostridium butyricum IDCC 9207 with a high growth inhibition of Salmonella typhimurium KCTC 2054 and Escherichia coli O157:H7 was selected. Also, a FWR was shown to suppress 8 among 21 pathogenic bacteria. In a mouse model with salmonella (${\times}10^9$ CFU/mouse) infection, 5 samples (200 ${\mu}{\ell}$/mouse/day) were fed to mice (n = 25) for 18 days. A fermented white rice containing C. butyricum IDCC 9207 (FWRCb9207) among 5 samples significantly inhibited the growth of salmonella, while in the control group (PBS, tetracycline) the number of salmonella increased. And the treatment with FWRCb9207 was found to increase the secretory immunoglobulin A (sIgA) level in the feces of salmonella-infected mice. The results obtained in this study suggest that a FWRCb9207 might be utilized as a feed additive in pigs and poultry diets.
