Heat Stress로 유발된 생쥐의 혈중 corticosterone 농도변화에 대한 사군자탕(四君子湯) 구성약물의 효능 (Effect of Each Constituent-Herb of Sagoonja-Tang on the Change of Corticosterone Level induced by Heat Stress in Mice)

  • This study aimed to investigate the effect of each constituent-herb of Sagoonja-Tang on the plasma corticosterone level in mice exposed to heat stress. Each formula(0.5g/kg, 1.0g/kg) was injected intragastrically to mice exposed to forced hot water swimming(42$^{\circ}C$, 3min) before measuring the change of plasma corticosterone level of mice. The results were as follows: 1. At 15 minutes after the heat stress for 3 minutes the most significant increase of corticosterone level was shown. 2. Ginseng group didn't show significant changes. 3. Atractylodis group showed significant decrease at 0.5g/kg dose(P<0.05). But it showed slightly increase at 1.0g/kg dose. 4. Poria group didn't show significant changes. 5. Glycyrrhizae group showed slightly increase, but insignificant. These data revealed that Atractylodis macrocephalae Rhizoma might have the anti-stress effect through the control of plasma corticosterone level in mice exposed to heat stress.

  • This study aimed to investigate the effects of each constituent-herb of Sagoonja- Tang on the plasma corticosterone level in mice exposed to cold stress. Sagoonja-Tang is composed of Ginseng Radix, Atractylodis Radix, Poria, and Glycyrrhizae Radix. Each constisuent-herb(0.5g/kg, 1.0g/kg) was administered intragastrically to mice 1hr prior to expose to forced cold water swimming($4{\circ}C$, 3min) before measuring the change of plasma corticosterone level of mice. The results were as follows: 1. At 30 minutes after the cold stress for 3 minutes the most significant increase of corticosterone level was shown. 2. Ginseng group(PGR group) didn't show any significant changes. 3. Atractylodis group(AR group) showed significant decrease at 0.5g/kg dose (P<0.01). But it didn't show significant changes at 1.0g/kg dose. 4. Poria group(P group) didn't show significant changes. But it showed slightly increase at 1.0g/kg dose. 5. Glycyrrhizae group(GR group) didn't show significant changes. These data revealed that Atractylodis macrocephalae Rhizoma might have the anti-stress effect through the control of plasma corticosterone level and the dose of herbs had a different effect on the plasma corticosterone level in mice exposed to cold stress.

Dehydroevodiamine.HCl Improves Stress-Induced Memory Impairments and Depression Like Behavior in Rats

  • Dehydroevodiamine HCl (DHED) has been reported to prevent memory impairment and neuronal cell loss in a rat model with cognitive disturbance. We investigated the effect of DHED on memory impairment and behavioral abnormality caused by stress. We demonstrated that DHED can improve stress-induced memory impairments and depression-like behaviors by using open-field test, Y-maze test and forced swimming test. DHED treatment significantly recovered the decreases in the levels of neural cell adhesion molecule (NCAM) proteins caused by stress and the decreases in cell viability. Our results suggested that DHED is a potential drug candidate for neuronal death, memory impairment and depression induced by stress.

Hot Water Extract of Leather Carp (Cyprinus carpio nudus) Improves Exercise Performance in Mice

  • The hot water extract of leather carp (Cyprinus carpio nudus) has been used as a nourishing tonic soup and as an aid for recovery from physical fatigue. In this study, we investigated the effect of leather carp extract on exercise performance in mice. Swimming endurance and forelimb grip strength were assessed following oral administration of the extract (once per day for 7 days) at a dose of $0.5 mg/10{\mu}L/g$ body weight. After 7 days, mice given the leather carp extract had significantly greater swimming endurance [$105{\pm}18s$ (P<0.05); 52% longer than day 0] and forelimb grip strength [$1.18{\pm}0.05$ Newton (P<0.01); 17% greater than day 0]. The extract increased muscle mass, but had little effect on body weight. Following the swimming exercise, blood glucose, glutathione peroxidase, and superoxide dismutase levels in extract-fed mice were significantly higher (145%, 131%, and 106%, respectively) than in the saline control group. Blood levels of high-density lipoprotein cholesterol were also significantly increased (128%) in mice given the extract compared to the controls. These results suggest that leather carp extract can improve physical exercise performance and prevent oxidative stress caused by exhaustive workouts.

지치 유래 naphthoquinone을 전처치한 생쥐에서 우울 및 불안 조절 효과 (The effect of pretreated Lithospermum erythrorhizon derived-naphthoquinone on anxiety, depression in mice)

  • 본 연구는 불안, 불면증, 우울증 또는 삶의 질에 대하여 지치(Lithospermum erythrorhizon) 유래 naphthoquinone, 즉 shikonin의 영향을 고찰하고, 확실히 밝혀지지 않은 메카니즘을 규명하고자한다. 우리는 지치의 주요성분인 naphthoquinone이 스트레스에 의한 수면장애, 강제수영에 의한 우울, 미로에 의해 유발된 불안현상 등의 조절에 영향을 미칠 것이라고 가설을 세웠다. 수컷 쥐를 이용하여 부동 혹은 수영시간, 수면시간, open arms으로 지속시간 및 진입빈도를 측정하고 기록하였다. Naphthoquinone(10, 30 & 100mg/kg)의 투여군에서 GABAA receptor의 활성으로 일어나는 barbiturate-유도 수면을 증가시켰다. 미로에서 open arms 상태로 지속되는 시간이 증가되고, 강제수영에서 부동시간이 줄었다. 결과적으로 naphthoquinone은 불안을 완화, 수면과 항우울경향이 있고, 불안, 불면과 우울증에서 효과적인 치료효과가 있었다.

Stress related activities of Sun-ginseng in SD Rats and ICR Mice

  • The main aim of this study was to investigate stress related activities of Sun-ginseng extract as a candidate for anti-stress-related functional supplement by comparing its effect to those of red ginseng, which is also known to alleviate stress. Normal group was not exposed to stress while the control group was exposed to stress. Rats were orally administered once a day with 200 mg red ginseng (RG) extract, 100 or 200 mg Sun-ginseng (SG) extract/kg body weight. Mice were given water containing 400 mg red ginseng extract, 200 or 400 mg SG/100 mL potable water. Rats were given supplements for 5 days without stress, and 5 days with restraint and electroshock stress. After final stress, stress-related behavioral changes of experimental animals were recorded and levels of blood corticosterone were measured. Mice were given supplements for 5 days through drinking water, and then fatigue related motor activity were recorded. SG-supplementation partially blocked stress effect on locomotion and elevated plus maze test in rats, and also partially blocked stress-induced behavioral changes such as freezing, burrowing, smelling, facewashing, grooming and rearing behavior in rats. SG-supplementation decreased blood corticosterone level which is increased by stress in rats. Effects of SG may not be modulated by GABAnergic nervous system. SG-supplementation prolonged swimming time and staying time on the wire and rotarod wheel in mice. These results suggest that SG partially protects living organisms from stress attack in some cases and thus has the potential to be used as a functional food to alleviate stress response.

Effects of Sumsu (Bufonis venenum) Pharmacopuncture Treatment on Depression in Mice

  • Objectives: The main objective of this study was to evaluate the anti-depressant effects of pharmacopuncture using sumsu (Bufonis venenum). Methods: Animals were divided into three groups (control, sham, and experimental), with eight mice per group. The sham and the experimental groups were exposed to 2 hours of immobilization stress daily for 14 days. They were also injected with normal saline (sham) or subjected to pharmacopuncture with sumsu at the acupoints HT7, SP6, and GV20 (experimental). The depression or anxiety-like behaviors of the mice in each group were evaluated 1 day after treatment. Results: There was no difference in locomotor activity between the groups during the open-field test; i.e., all groups had normal motor function. However, the open-field and the forced-swimming tests revealed that depression and anxiety-like behaviors were decreased significantly in the group treated with sumsu pharmacopuncture. Conclusion: Sumsu pharmacopuncture attenuated depressive or anxiety-like behavior in mice stressed with chronic immobilization. These results suggest that sumsu pharmacopuncture has therapeutic potential for treating neuropsychiatric disorders such as anxiety or depression disorder.

Anti-fatigue effect of fermented porcine placenta through the regulation of fatigue-associated inflammatory cytokines

  • Fatigue is a common complaint and affects the quality of life in modern people. Physical stress may induce activation of certain immune cells. Fermented porcine placenta (FPP) has been used to alleviate fatigue. Inflammatory cytokines are produced by physical stress and results in symptoms of fatigue. However, the role of FPP on fatigue-associated inflammatory cytokine production has not been elucidated yet. Thus, we estimated the anti-fatigue effect of FPP and its active components, leucine (Leu) and lysine (Lys) in activated RAW264.7 macrophages and forced swimming test (FST) fatigue animal model. Pretreatment with FPP, Leu, or Lys significantly inhibited the lipopolysaccharide (LPS)-induced tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 production without inducing cytotoxicity on LPS-stimulated RAW264.7 macrophages. FPP, Leu, or Lys inhibited the production of nitric oxide and downregulated the expression of inducible nitric oxide synthase on LPS-stimulated RAW264.7 macrophages. Furthermore, caspase-1 activities increased by LPS were significantly reduced by FPP, Leu, or Lys. In the FST, inflammatory cytokine levels of the mice administrated with FPP, Lys, and Leu were significantly reduced compared with the control group at 21 days. Collectively, these results show that anti-fatigue effect of FPP and its active components, Leu and Lys might be derived from the down-regulating of inflammatory mediators.

DEPRESSION: CELLULAR AND PHYSIOLOGICAL CONSEQUENCES OF STRESS (ANTIDEPRESSANT EFFECT OF SEROTONIN N-ACETYLTRANSFERASE INHIBITOR)

  • Melatonin is secreted during the hours of darkness and is thought to influence the circadian and seasonal timing of a variety of physiological processes. Serotonin N-acetyltransferase (AA-NAT) which is found to be expressed in pineal gland, retina, and various tissues, catalyses the conversion of serotonin to N-acetylserotonin and is known as the rate-limiting enzyme in the biosynthetic pathway of melatonin. The compounds that modulate the activity of AA-NAT can be used to treat serotonin-and melatonin-related diseases such as insomnia, depression and seasonal affective disorders (SAD). Several assay methods have been developed by which to measure AA-NAT activity. We have also developed a simple, rapid and sensitive AA-NAT assay method that takes advantage of differences in the organic solubilities between acetyl CoA and N-acetyltryptamine. We screened modulators of AA-NAT activity from the water extracts of the medicinal plants. We found MNP1005 which strongly inhibited the activity of AA-NAT ($IC_{50}$=2.2$\mu$M). Enzyme inhibitory kinetic studies revealed that MNP1005 exhibited a noncompetitive inhibition toward tryptamine. The antidepressant effect of MNP1005 was investigated on behavioral despair test so called forced swimming test (FST). MNP1005 significantly increased swimming behavior by reducing immobility with treatment of 10 mg/kg when compared to the vehicle-treated control group (P < 0.05). This suggests that MNP1005 possesses antidepressant activity. The influence of chronic MNP1005 treatment on the expression of brain-derived neurotrophic factor (BDNF) was examined by in situ hybridization and Northern blot. Chronic treatment of MNP1005 blocked the downregulation of BDNF mRNA in the frontal cortex and other cortex regions in response to restraint stress.
