The cDNA of the touch-induced genes (TCH) of the sweet potato [Ipomoea batatas (L.) Lam.] has been cloned and analyzed. IbTCH1, which exists as at least two-copy genes in the genome of the sweet potato, encodes for 148-amino acid polypeptides, and harbors four conversed $Ca^{2+}-binding$ motif EF-hands. IbTCH1 was shown to be expressed in the flower, leaf, thick pigmented root, and particularly in the white fibrous root, but expressed only weakly in the petiole. IbTCH1 is upregulated upon exposure to environmental stresses, dehydration, and jasmonic acid. Furthermore, IbTCH1 is developmentally regulated in the leaf and root. These results strongly indicate that the gene performs functions in both plant development and in defense/stress-signaling pathways.
본 연구에서는 국내산 자색고구마(Ipomoea batatas L. Lam.) 추출물을 제조하고 이들 추출물에 대하여 항산화 활성을 측정하였다. 자색고구마 추출물은 70% 에탄올 추출물과 그 추출물로부터 에틸아세테이트 분획으로 제조하였다. 추출물 및 분획의 수율은 건조 분말 당 각각 39.2% 및 3.49%이었다. 이들 추출물/분획에 대한 항산화 활성을 확인하기 위해 자유 라디칼(1,1-phenyl-2-picrylhydrazyl, DPPH) 소거활성과 루미놀 발광법을 이용한 총항산화능 그리고 세포손상에 대한 보호효과를 측정하였다. 자유 라디칼 소거활성($FSC_{50}$)은 70%에탄올 추출물과 에틸아세테이트 분획이 각각 $90.16{\mu}g/mL$와 $7.69{\mu}g/mL$를 나타내었다. 비교 대조군으로 사용한 (+)-${\alpha}$-tocopherol의 라디칼 소거활성은 $8.98{\mu}g/mL$로, 에틸아세테이트 분획의 라디칼 소거활성이 보다 큼을 알 수 있었다. 루미놀 발광법을 이용한 70% 에탄올 추출물과 에틸아세테이트 분획의 총 항산화능($OSC_{50}$)은 각각 $5.75{\mu}g/mL$와 $1.92{\mu}g/mL$이었다. 비교 대조군으로 사용한 항산화 효능이 매우 우수한 L-ascorbic acid의 항산화능은 $1.50{\mu}g/mL$로 추출물의 에틸아세테이트 분획과 유사한 활성을 나타내었다. $^1O_2$로 유도된 사람 적혈구의 세포손상에 대한 보호 효과 실험에서 에틸아세테이트 분획은 농도 의존적($5{\sim}50{\mu}g/mL$)으로 세포보호 효과를 나타내었다. 에틸아세테이트 분획의 세포보호활성(${\tau}_{50}$)은 $5{\mu}g/mL$에서 45.6 min으로 모든 농도에서 비교 대조군인 (+)-${\alpha}$-tocopherol 보다 더 높은 세포보호 효과를 나타내었다. 이상의 결과들로부터 자색고구마의 에틸아세테이트 분획은 뛰어난 항산화능을 나타내며, 이는 피부노화 억제를 위한 항노화 기능성 화장품원료로 응용될 수 있는 가능성을 시사하였다.
Greenhouse and laboratory studies were conducted to determine the allelopathic potentials of extracts or residues from sweet potato (Ipomoea batatas L. (Lam). The extracts applied on filter paper in a Petri dish bioassay significantly inhibited root growth of alfalfa. Aqueous leachates at 40g dry tissue $\textrm{L}^{-1}$ (g $\textrm{L}^{-1}$) from leaves showed the highest inhibition against alfalfa, and followed by stems and roots. Alfalfa root growth was significantly inhibited by methanol extracts of the same plants as the concentration increased. The effect of residue incorporation into soil on seedling growth of com, soybean, barnyard grass and eclipta was examined in the greenhouse, and results showed that the leaf residues at 200g $\textrm{kg}^{-1}$ by plant parts inhibited shoot dry and root dry weights of test plants by 60-80%. By means of HPLC, causative allelopathic substances present in plant parts of sweet potato "Sinyulmi" were identified as coumarin, trans-cinnamic acid, o-coumaric acid, p-coumaric acid, and chlorogenic acid. Total content of these compounds for leaves extracts were detected as the greatest amount in EtOAc fraction, especially trans-cinnamic acid was the greatest component. These results suggest that sweet potato plants have herbicidal potentials, and that their activities exhibit differently depending on plant parts.ant parts.
This study investigated the antioxidant characteristics of sweet potato according to different plant parts and drying methods. The sweet potato plant parts were divided into root tubers, stems, stalks, leaves, and tips, and the drying methods were freeze-drying and hot air drying. Total polyphenol and flavonoid contents and radical scavenging activity of the sweet potato plant parts were significantly different depending on the plant parts and drying methods. The total polyphenol content of freeze-dried sweet potato leaves and tips were 52.76 and 46.19 mg chlorogenic acid equivalents/g sample, and the total flavonoid contents were 222.47 and 214.12 mg quercetin equivalents/g sample, respectively, and decreased with hot air drying. DPPH radical scavenging activity was higher in freeze-drying than hot air drying and was significantly different depending on the plant parts. The ABTS radical scavenging activity of freeze-dried sweet potato leaves and tips were 43.48 and 44.68 mg Trolox equivalents/g sample, respectively, and decreased with hot air drying. Therefore, additional studies on the functionality of using by-products from sweet potato cultivation are needed.

The sweet potato (Ipomoea batatas [L.] Lam.) is the sixth-most important crop in the world following rice, wheat, potato, maize, and cassava. Four varieties ('Beniharuka', 'Annobeni', 'Pungwonmi', 'Hogammi') and their Japanese cultivars are broadly distributed in South Korea. In the Korean marketplace, sweet potatoes are classified by color and shape, not by variety, making it necessary to differentiate varieties for uniform production and consumption. In this study, molecular markers were developed to distinguish the four varieties of sweet potato using SNPs and genotyping-by-sequencing (GBS) analysis via a tetra-primer amplification refractory mutation system (ARMS)-PCR. The results revealed that three variety-specific fragments (164 bp and 241 bp of SNP 04-27457768 and 292 bp of SNP 03-16195623) were amplified in the 'Beniharuka', 'Pungwonmi', and 'Annobeni' sweet potato varieties. There were instances where some varieties produced three bands within the gel electrophoresis, indicating heterozygosity at the given SNPs loci. DNA sequencing analysis also confirmed the results of electrophoresis at the SNPs loci. Overall, these molecular markers would provide a useful, rapid, and, simple evaluation method for the Korean sweet potato marketplace, where the mixing of varieties is a serious issue.
재배방법을 달리하여 재배한 고구마의 품질특성과 화학성분 및 항산화활성의 변화를 분석하여 고구마 친환경재배에 적합한 녹비작물 설정의 기초자료로 활용하고자 하였다. 녹비작물 재배 후 후작으로 재배한 고구마에 대한 당도 및 색도 등의 품질특성과 호화특성을 분석한 결과 전작물에 따라 유의적으로 차이를 보이는 것으로 나타났다. 수확된 고구마를 과피와 과육으로 분리하여 수분(62.5-80.6 g/100 g), 단백질(2.6-3.9 g/100 g) 등의 일반성분과 칼륨, 칼슘, 마그네슘, 나트륨 등의 무기성분 함량을 분석한 결과 전작물에 따라 유의적으로 차이를 보였다. 재배방법을 달리하여 재배한 고구마를 과피와 과육으로 구분하여 메탄올로 추출하여 총 폴리페놀, 플라보노이드 함량을 분석한 결과 과육보다 과피 추출물이 유의적으로 높은 함량을 보였다(p<0.05). 가장 높은 함량을 보인 처리구는 관행재배 방법으로 재배한 고구마(SP1)로서 과피 추출물에서 각각 9.38 mg GAE/g sample 및 5.57 mg CE/g sample으로 유의적으로 가장 높았다(p<0.05). DPPH 및 ABTS radical 소거활성 또한 항산화성분과 마찬가지로 과육보다 과피 추출물이 유의적으로 높은 활성을 보였으며(p<0.05), 관행재배 방법으로 재배한 고구마(SP1) 과피 추출물이 각각 776.38 및 708.03mg TE/100 g sample로 유의적으로 가장 높은 활성을 보였고, 헤어리베치 후작으로 재배한 고구마(SP2)가 각각 715.20 및 708.58mg TE/100 g sample로 비교적 높은 활성을 보였다. 이상의 결과에서 관행재배 방법으로 재배한 고구마(SP1)가 높은 항산화성을 보였으며, 헤어리베치 후작으로 재배한 고구마(SP2)가 그 다음의 항산화성을 보여 다른 작물에 비해 유리할 것으로 판단된다.
고구마 현탁배양세포로부터 POD 고생산세포주로 선발한 SP-47세포주를 사용하여 POD생산성을 향상시키기 위하여 식물생장조절제 및 탄소원의 종류와 농도, 세포접종량 등의 배양조건을 적정화하였다. 30 g/L Surcrose, 1 mg/L 2,4-D가 첨가된 LS배지 50mL을 함유한 30 mL Erlenmeyer flask에 세포생중량 1 g을 접종하여 $25^{\circ}C$ 암소에서 100 rpm으로 진탕배양하였을 때 세포생장은 배양 후 15일에 절정에 달하였으나 단위세포당 POD 활성(unit/g dry cell wt)은 배양 25일에 약 6,800으로 이는 실생 서양겨자무뿌리의 것보다 약 30배 높았다. 배양시기별 단위세포당 단백질 함량은 계대배양후 약간 높았다가 감소한 후, 배양 25일까지는 거의 일정한 값을 유지하다가 계속 배양함에 따라 감소하였으나 POD 비활성(unit/mg protein)은 배양후 12일부터 배양말기 (40일)까지 계속하여 증가하였다. 배양시기별 POD 동위효소의 패턴은 배양시기에 관계없이 거의 일정하였으나 배양 25일이후 산성의 주요 동위효소들의 활성이 약간 증가하였다. 이러한 결과로부터 고구마 세포배양의 POD는 세포생장 및 계대배양과 배지고갈로 인한 배양스트레스와 밀접한 관련 이 있을 것으로 간주된다. 본 연구에서 확립한 SP-47세포주는 하나의 동위효소가 강하게 발현되어 높은 활성을 보임으로써 새로운 POD 대량생산 시스템에 활용될 수 있을 것으로 기대된다.
This study examined the quality characteristics of Makgeolli to investigate the availability of Korean sweet potato cultivars as alcoholic beverages. The following sweet potato varieties were used: Gogunmi, Daeyumi, Shingunmi, Shinyulmi, Shincheonmi, Jinyulmi, Jinhongmi, Jeungmi and Pungwonmi; their alcohol contents ranged from 12.20% to 14.20%, with the lowest value in Makgeolli made with Jeungmi and the highest value in Makgeolli made with Jinhongmi. The DPPH radical scavenging activity was in the range of 37.51~77.02%, with the lowest value in Makgeolli made with rice (control) and the highest value in Makgeolli made with Gogunmi. As a result of analyzing the aroma component of sweet potato Makgeolli, 27 kinds of aroma components were detected, and six kinds of alcohols, 13 kinds of esters, four kinds of acids, and four kinds of other compounds were found. Regarding the number of aroma compounds, Makgeolli made with Shinyulmi showed the lowest number with 14 kinds, while Makgeolli made with Pungwonmi showed the highest number with 27 kinds.
Extrafloral nectaries have been shown in many studies to promote mutualistic interactions between plants and insects(usually ants) that visit the glands. The insects gain sugars, water and amino acids secreted by the extrafloral nectaries and benefit the plants by reducing the damage caused by plant's inseet herbivores. Little is known about the occurrence of extrafloral nectaries in plants growing in Asia. To learn about the occurrence of extrafloral nectary bearing plants in Korea, living plants and herbarium material were examined for the glands. In addition, the cover of plants with extrafloral nectaries and the proportion of woody plants with extrafloral nectaries were measured in three forest communities on Kangwha Island. 131 species of plants belonging to 53 genera and 30 families were found to have extrafloral nectaries. These 131 species comprise about 4.0% of Korea's flora, a highet percentage of extrafloral nectary bearing plants than occurs in the studied areas of North America. Extrafloral nectary bearing plants occupied 7, 23 and 55% of the covers and comprised 15, 21 and 15% of the woody plants in the three different forests, a significant level of occurrence. Many important Korean crop plants were found to have extrafloral nectaries including : sesame (Sesamum indicum L.), squash (Cucurbita moschata Duchesne), sweet potato (Ipomoea batatas Lam), persimmon (Diospyros kaki Thunb.) cotton (Gossypium indicum Lam.), mung bean (Phaseolus radiatus L.), red bean (Phaseolus angularis W.F.), peach (Prunus persica (L) Batsch.), plum (Prunus salicina Lindl.). Many of these cultivated and wild plants may receive protection by ants and other beneficial insects that visit their extrafloral nectaries.
