Allelism and Molecular Marker Tests for Genic Male Sterility in Paprika Cultivars (파프리카 시판 품종에 대한 유전자적 웅성불임성의 대립성 및 분자표지의 유용성 검정)

  • Paprika (Capsicum annuum L.), a colored bell-type sweet pepper, is one of the most important money making vegetable crops in Korea. The cultivation area, total production, and exports of paprika are gradually getting increased, but the paprika cultivars used in Korea are all imported. It was well-known that the genic male sterility (GMS) is the main way to produce paprika hybrid seeds. However, it is little known that how many and what kinds of ms genes are used for breeding of paprika $F_1$ varieties. In this study, eight paprika cultivars ('Special', 'Debla', 'Plenty', 'Fiero', 'Boogie', 'Fiesta', 'Derby', and 'Minibell'), popularly cultivated in Korea and three different genic male sterile lines ('GMSP', 'GMS3', and 'GMSK') were used. For allelism test among the $F_1$ cultivars, half diallel crosses were performed. The result demonstrated that the most of the GMS in paprika cultivars except for 'Minibell' were same allele. To identify which GMS gene(s) were used for paprika $F_1$ cultivars, top crosses between previously known GMS lines and the $F_1$ cultivars were performed. As a result, we found that the $ms_k$ and the $ms_p$ genes were alleles for the GMS of 'Minibell' and for the other cultivars, respectively. We also confirmed that the GMS gene identification using GMSK-CAPS marker linked to the $ms_k$ gene and the PmsM1-CAPS marker linked to the $ms_p$ gene in $F_2$ progenies of 'Minibell' and 'Fiesta' and 'Derby' cultivars, respectively. In addition, we developed the PmsM2-CAPS marker for 'Plenty', 'Fiero', and 'Boogie' cultivars. We expect that these markers will be very useful for breeding new maternal (male sterile) line of paprika.

Resistance of Commercial Tomato Cultivars to Meloidogyne arenaria and M. incognita (시판 토마토품종의 고구마 뿌리혹선충과 땅콩 뿌리혹선충에 대한 저항성)

  • Root-knot nematodes (Meloidogyne spp.) are among the main pathogens of greenhouse crops worldwide. Plant resistance is currently the method of choice for controlling these pests. To select resistant tomato against two common species of root-knot nematodes, M. incognita and M. arenaria, 36 commercial tomato (Lycopersicon esculentum Mill.) cultivars were screened. Seventeen tomato cultivars were resistant to both root-knot nematodes: six in cherry tomato, 'Tenten', 'Cadillac', 'Cutti', 'Sweet', 'Ppotto', 'Lycopin-9', eight in globe tomato, 'Lovely 240', 'Dotaerang Dia', 'Cupirang', 'Dotaerang Master', 'Super Dotaerang', 'Dotaerang Season', 'Miroku', 'Hoyong', and three in root stock, 'Special', 'Fighting', and 'Magnet'.

Schisandra Fructus Butanol Fraction Reduces Serum Total Cholesterol and Triglyceride Levels in Hyperlipidemic Mice (오미자 부탄올 분획물이 고지혈증이 유도된 생쥐의 지질대사 및 간조직 유전자 발현에 미치는 영향)

  • The berries of Schisandra chinensis (Schisandra fructus) are given the name Omiza in Korean (五味子), which translates as "five flavor fruit" because they possess all five basic flavors in Korean traditional herbal medicine: salty, sweet, sour, pungent (spicy), and bitter. It is used as a remedy for many ailments: to resist infections, increase skin health, and cure insomnia, coughing, and thirst. This study was designed to investigate the effects of Schisandra fructus butanol fraction (SFB) on serum lipid levels in hyperlipidemic mice. In this experiment, effects on total cholesterol, HDL-cholesterol, and triglyceride in serum were measured. In our results, SFB did not affect weight gain in hyperlipidemic mice. Oral administration of SFB lowered levels of total cholesterol and triglyceride, which were elevated by induction of hyperlipidemia. Finally, administration of SFB regulated changes in gene expression which were related to cell growth and differentiation.

Anti-obesity Effect of Vegetable Formula on Obese Mice in High Fat Diet (고지방식이 비만 마우스에 대한 야채 조성물의 항비만 효과)

  • This study was conducted to investigate the cholesterol and Triglyceride (TG) lowering and anti-obesity effects of water extract of vegetable formula (onion 25%, carrot 20%, Cabbage 20%, Sweet Pumpkins 20%, Chinese plum 5%, turmeric 5%, morus leaves 5%, SLS) in mice fed high fat-diet. ICR mice were divided into 3 groups; a normal diet group (ND), a high-fat diet group (HFD), a high-fat diet and SLS with 300 mg/kg treated group (HFD+SLS). Body fat gain was increased by high-fat diet and HFD+SLS group showed a definite weight loss. The total cholesterol level in the HFD+SLS group was 328.4 mg/dl which was 27.4% lower than that in the HFD group and the cholesterol-lowering effect of SLS was confirmed. The HFD + SLS group showed 118.1 mg/dl and the triglyceride level in the serum was decreased by 88.7% compared to the HFD group and SLS significantly decreased blood triglyceride levels. RT-PCR showed that the expression of PPAR-${\gamma}$ and the target gene SCD-1 was inhibited in a dose-dependent manner in the SLS-treated group. These results suggest that the SLS water extract may have a cholesterol and triglyceride-lowering effect and inhibit the expression of PPAR-${\gamma}$ and SCD-1 to have an anti-obesity effect.

  • Tamarind (Tamarindus indica L.) is one of the multipurpose tree species distributed in the tropical and sub-tropical climates. It is an important fruit yielding tree that supports the livelihood and has high social and cultural values for rural communities. The vegetative, reproductive, qualitative, and quantitative traits of tamarind vary widely. Characterization of phenotypic and genetic structure is essential for the selection of suitable accessions for sustainable cultivation and conservation. This study aimedto examine the genetic relationship among the collected accessions of sweet, red, and sour tamarind by using Random Amplified Polymorphic DNA (RAPD) primers. Nine accessions were collected from germplasm gene banks and subjected to marker analysis. Fifteen highly polymorphic primers generated a total of 169 fragments, out of which 138 bands were polymorphic. The polymorphic information content of RAPD markers varied from 0.10 to 0.44, and the Jaccard's similarity coefficient values ranged from 0.37 to 0.70. The genetic clustering showed a sizable genetic variation in the tamarind accessions at the molecular level. The molecular and biochemical variations in the selected accessions are very important for developing varieties with high sugar, anthocyanin, and acidity traits in the ongoing tamarind improvement program.

Studies on Resistance of Sweet potato Varieties to Black Rot(Ceratocystis fimbriata E.) (고구마 품종의 흑반병 저항성에 관한 연구)

  • To find out highly resistant gene source to black rot fungus in sweetpotato varieties, a screening test was carried out with inoculated tubers and sprouts in 1976 and 1977. Fifteen varieties out of 165 and 54 varieties out of 161 were highly resistant to black rot in the tuber and sprout tests. respectively. The sprout test showed bigger variation from year to year compared to tuber test. Varieties highly resistant to black rot in both tuber and sprout tests were Suweon # 59, #81, #90, Norin #17, #23. Chilship-il cho, Sachun Jong #36, Hamkao, and Kandee.

Some Aspects of High Lysine Maize Breeding using Opaque-2 Gene (Opaque-2 인자를 이용한 고라이신 옥수수의 육종)

  • Several field and sweet corn varietiea from several sources were crossed with a variety carrying the opaque-2 gene to determine the phenotypic interactions in the breeding of high lysine maize. Although opaque-2 lines showed lower protein content than the corresponding normal varieties, there was no correlation between the protein levels of the two types. opaque-2 maize contained more lysine, but no relationship was found between the protein content and the lysine content of either normal or opaque-2 types, suggesting that high lysine corn using the opaque-2 gene may be developed independently from the protein content. The F2 segregation ratios for normal and opaque-2, 100-kernel weights, percentage seed set, opaque-2 phenotype, disease susceptibility, and the relationship between protein and lysine content of normal and opaque-2 were investigated. The determinations and observations were made on the F2, F3, and BC1 Lysine content was determined by the ion exchangeresin combined with paper chromatography method. Most crosses segregated in a 1-opaque-2 : 3-normal ratio as expected. Opaque-2 segregates were lighter than the normal type and smaller in size. A mottled phenotype of opaque-2 maize observed in the Philippines yellow endersperm. In some varieties opaque-2 maize was very susceptible to the ear and kernel rot disease. No. 5(female) and opaque-2(male). Selectlon of a double mutant of waxy and opaque-2 by using the iodine technique and electric lamp was discussed. opaque-2 and floruy-2 were not allels. Different percentage of seed set were observed in the segregation of aewx crossed with opaque-2. An unusual gametophytie relationship was involved in a cross between Glutinous.

Evaluation of Early Generation of Crosses for Incorporation of Resistance to Bacterial Spot into Sweet Pepper (감미종(甘味種) 고추에 더뎅이병(病) 저항성(抵抗性)을 도입(導入)하기 위한 교잡(交雜) 초기세대(初期世代) 검정(檢定))

  • A leading sweet pepper cultivar, Keystone Resistant Giant #3, was crossed with a line with resistance to Xanthomonas campestris pv. vesicatoria, PI271322, for incorporation of the resistance and to study the inheritance of resistance to the disease. Seedlings of parents, $F_1$, $F_2$ and backcross populations of a cross between Keystone Resistant Giant #3 and PI271322 were inoculated with race 1 and race 3 of Xanthamonas campestris pv. vesicatoria by infiltrating bacterial suspension into abaxial side of leaves. PI271322 was carrying a gene ($Bs_3$) for hypersensitive resistance to race 1 of X. c. pv. vesicatoria and also a component of non-hypersensitive resistance to race 3 inheriting in a quantitative mode. Correlation coefficient between disease indices to race 1 and race 3 of non-hypersensitive plants in $F_2$ population was highly significant, thus indicating that a component of non-hypersensitive general resistance in PI271322 acted consistently to both races. Individual plants with resistance to both race 1 and race 3 of X. c. pv. vesicatoria were selected and a breeding program for incorporation of the resistance is continued.

Anti-inflammatory Activity of Stevia rebaudiana in LPS-induced RAW 264.7 Cells

  • Stevia rebaudiana (SR) is an herb used traditionally as a sweetener in Paraguay and Brazil, whose use is spreading to other countries, such as Japan, Korea and China. In addition to its low calorie sweet taste, SR appears to have other beneficial properties, such as hypotensive capabilities and inflammation reduction. To identify the bioactive natural constituents exerting anti-inflammatory activities, we examined the EtOAc fraction of SR. In the inflammatory mediator inhibitory assay from lipopolysaccharide (LPS)-activated macrophages, the EtOAc fraction significantly, and dose dependently, inhibited the enhanced production of nitric oxide (NO) and inducible nitric oxide synthase (iNOS) expression. We also found that treatment of cells with the EtOAc fraction significantly inhibited LPS-stimulated nuclear factor-${\kappa}B$ (NF-${\kappa}B$) reporter gene expression. Such inhibition of NF-${\kappa}B$ was closely associated with the inhibition of interleukin-6 (IL-6) and the monocyte chemoattractant protein-1 (MCP-1). Therefore, we suggest that SR has the potential for development as a functional food for the treatment of immune diseases, such as rheumatoid arthritis and lupus.

Quality Characteristics and Antioxidant Activity of Syrup Added with Maca (Lepidium meyenii) Extract (마카 추출액 첨가에 따른 시럽의 품질특성과 항산화성)

  • This study was conducted to develop a functional syrup by using maca extract. Five different levels (0%, 7%, 14%, 21%, 28%) of maca extract was added and the physicochemical properties were investigated. The pH of control syrup was lower than that of syrup added with maca extract. Viscosity increased with increasing amount of maca extract. Hunter L value decreased while b value increased as maca extract level increased. Results of consumer acceptance test revealed that no significant differences in flavor, sweet taste and overall acceptability were observed among samples. Therefore, it is suggested that maca extract can be incorporated into syrup up to 28% without depressing the quality. Total polyphenol content and DPPH radical, superoxide anion radical and hydroxyl radical scavenging tests were conducted in view of estimating the functionality of maca syrup. Results showed that incorporation of maca extract into syrup exhibited higher polyphenol content and DPPH radical and superoxide anion radical scavenging activity than control syrup (p<0.05).
