아그로박테리움 매개에 의한 형질전환 기술을 이용하여 국내에서 육성된 품종 'Sweet Yellow'로부터 유도된 체세포배 (배발생캘러스 포함)로 intron-GUS유전자가 전이된 식물체를 획득하기까지의 과정이 제시되었다. Intron-GUS 유전자를 포함하고 있는 Agrobacterium tumefaciens AgL1(O.D=0.7~1.6)에 30분 감염시켜 3일간 공동배양 한 후 $4^{\circ}C$에서 7일간의 저온처리를 거친 후 cefotaxim $250\;mg{\cdot}L^{-1}$ 첨가 체세포배발아 배지에 배양된 체세포배 (배발생캘러스 포함)들 대부분으로 유전자가 전이된 것을 GUS transient assay에 의해 확인하였다. Intron-GUS유전자가 전이된 체세포배 (배발생캘러스 포함)로부터 신초원기를 유도한 후 신초를 재분화시켰고, 재분화된 신초로부터 다신초가 형성되도록 하였다. 다신초로부터 신초의 일부를 떼어 GUS transient assay 분석을 실시하여 intron-GUS 유전자의 발현을 확인한 후 발근시켜 순화 후 온실로 옮겼다. GUS transient assay에 의해 확인된 유전자 발현율은 100%였다.
R2R3 유형의 단백질인 IbMYB1 전사인자는 고구마의 뿌리에서 안토시아닌 생합성 과정을 조절하는 중요한 단백질이다. 선행연구에서 IbMYB1 유전자의 발현 증가를 통한 안토시아닌의 합성 증가가 담배, 애기장대 및 고구마의 저장뿌리에서 증명된 바 있다. 본 연구에서는 괴경(저장줄기) 특이적 PATATIN 프로모터와 산화스트레스 유도성 SWPA2 프로모터의 조절하에서 안토시아닌을 고함유하는 IbMYB1 유전자 과발현 형질전환 감자를 개발하여 그 특성을 분석하였다. PAT-IbMYB1 형질전환 식물체들은 대조구 식물체 및 SWPA2-IbMYB1 식물체 보다 높은 안토시아닌 함량을 괴경에서 나타내었다. 본 연구의 결과로서, IbMYB1의 과발현은 형질전환 기술을 이용한 특정 조직 특이적 안토시아닌 생산에 매우 좋은 개발 기술이 될 것으로 생각되는 바이다.
2012년 국내 양앵두나무에서 발생하는 CGRMV를 조사하기 위해 화성, 평택, 경주, 김천, 대구, 영주 음성의 양앵두 재배과원 7개 지역에서 잎 시료 154점을 채집하였다. 채집한 시료에 대해 CGRMV 유전자 검정을 수행한 결과 6점의 시료에서 807 bp 크기의 PCR 증폭산물이 검출되었다. 이들 PCR 증폭산물은 클로닝과 유전자 염기서열 분석 결과 GeneBank에 등록된 외국의 CGRMV 분리주들과 88% 이상의 외피단백질 유전자 염기서열 상동성을 보였다. 국내 양앵두나무에서 분리된 분리주들, CGR-KO 1-6 간에는 98.8-99.8%의 염기서열 및 99.6-100%의 아미노산 서열 상동성을 나타내었다. 국내 CGRMV 분리주들은 외피단백질 유전자 계통도 분석에서 기존에 분류된 I, II, III 그룹 중 II 그룹에 속하였다. 또한 CGRMV가 감염된 국내 양앵두나무 이병주들은 ACLSV 등 10종 바이러스에 대해서도 RT-PCR을 수행한 결과 모든 시료에서 ACLSV가 검출되어 CGRMV와 ACLSV가 복합 감염된 것을 확인하였다.
Pepper (Capsicum spp.) is one of the main quality features of this crop because of its sense of pungency, which is due to the presence of capsaicinoids. This compound is synthesized as a secondary metabolite and found only in the placental tissue of spicy fruit (Suzuki et al., 1980). Stewart et al. (2005) concluded that Pun1 encodes for the acyltransferase AT3 and they demonstrated its involvement in capsaicinoids metabolism. It was analyzed that the capsaicinoids content and PUN1 genotype in pepper genetic resources which were selected with excellent phenotype in field evaluation. The number of pepper genetic resources analyzed was 135, and species were C. annuum, C. baccatum, C. chinense, C. frutescens. The content of capsaicinoid ranged from 0 mg/100g to 828 mg/100g. The content of 0 mg/100g was the sweet pepper type, the highest content is IT 158530, the capsaicinoid content of which was 828 mg/100g and species was C. annuum. PUN1 gene analysis showed 117 pungent, 5 hetero, and 13 non-pungent. PUN1 analysis showed that 5 out of 13 non-pungent accessions were detected with low levels of capsaicinoid.

The goal of this study was to evaluate the influence of various supplementary feeds on the chemical composition and production of bioactive substances in Protaetia brevitarsis larvae. The primary feed-oak-fermented sawdust-was supplemented with a variety of substances, including aloe, apple, banana, sweet persimmon (S. persimmon) and sweet pumpkin (S. pumpkin). Crude protein and fat content were the highest in the control and S. pumpkin group, respectively. Supplementary feeds increased the content of unsaturated fatty acids, except in the group receiving S. pumpkin, in which oleic acid was the most abundant (58.2%-64.5%). Free essential amino acids in larvae receiving supplementary aloe were higher compared with the control group except for Lys and His. Polyphenol and flavonoid contents and the antioxidant activities of ABTS and DPPH were higher in all treated groups compared with the control group. Although supplementary feeds led to a decreased crude protein content in the treated larvae when compared with the control group, these treatments generally improved the levels of unsaturated fatty acids and antioxidative activity. Therefore, we suggest that among the supplementary foods tested, aloe is a better resource for P. brevitarsis based on crude protein content, free amino acids and other bioactive compounds such as unsaturated fatty acids and antioxidants.
본 논문에서는 multiple array 안테나 링크 상에서 각 스테이션 간의 안테나 빔의 스위트 스폿을 변형된 유전자 알고리즘을 이용하여 찾고 유지하는 방법을 제안하였다. 제안한 방식은 각 스테이션에서 전송하는 데이터에 안테나의 정보를 같이 전송하며 범의 강도를 거리함수로 나타내고 그 거리함수의 곱을 적합도 함수로 이용하여 최대값이 되는 각도를 찾는 방식이다. 변형된 유전자 알고리즘 방식은 전처리 과정을 통하여 우수한 초기세대를 선택하는 방식으로 일반적인 유전자 알고리즘방식에서 랜덤하게 초기세대를 갖는 것과는 차별화가 된다. 통식 방식은 시분할 이중화 (TDD: Time Division Duplex) 방식으로 하여 전송하는 데이터에 안테나 정보를 같이 보낸다. 제안한 방식의 유용성을 확인하기 위하여 1:1, 1:2, 1:5 array 안테나의 세 가지 경우에 대하여 시뮬레이션 하였다. 염색체의 길이는 8bit, 16bit, split인 경우에 대하여 개체 수와 세대 수를 변화시켜 수렴 확률을 비교 검토하였다. 제안한 16bit split는 실제로는 8bit이지만 16bit와 유사한 좋은 수렴율을 보여주었다.
Mosquitoes are carriers of malaria and encephalitis. This study performed for eco-friendly control of mosquitos by using genus Acorus. Several solvents were used for the extraction of genus Acorus; water, ethanol, and methanol. Grinded leaves and roots were also included. Acorus extracts killed mosquito larvae and the ethanol extract showed the best result. Autoclaved Acorus water needed long time to kill mosquito larvae. $LT_{50}$ of 1 % Acorus calamus decoction was 13.6 hrs and 1 % autoclaved Acorus water was 53.6 hrs. $LT_{50}$ of 0.05% Acorus calamus rhizome powder was 28.5 hrs. $LT_{50}$ of 0.5% Acorus calamus leaf powder was 10.8 hrs. $LT_{50}$ of 0.1 % Acorus calamus decoction was 63.4 hrs and 0.1 % Acorus calamus ethanol extracts was 48.6 hrs and 0.1% Acorus calamus methanol extracts was 53.9 hrs. $LT_{50}$ of 0.4% Acorus gramineus decoction was 45.5 hrs, 0.4% ethanol extracts was 10.9 hrs, 0.4% methanol extracts was 10.2 hrs. $LT_{50}$ of ethanol extracts was shorter than other extracts. Acorus calamus rhizome powder could be used for the eco-friendly control of the mosquito larvae.
A reverse transcription polymerase chain reaction (RT-PCR) protocol was developed using two specific 22-mer primers located in coat protein gene of SPFMV. A 411 bp PCR-product was detected in virus infected plants as well as tissue culture raised sweet potato but not in healthy plants. For optimization of RT-PCR protocol, the optimum crude nucleic acid concentration, annealing temperature, primer concentration and numbers of PCR-cycle for maximum sensitivity and specificity were determined. The optimum condition for RT-PCR was as follows: RT-PCR reaction mixture was one-step mixture, containing 50 pmol of primer, 30 units of reverse transcriptase, 5 units of RNasin, and the crude nucleic acid extracts (200 ng). In RT-PCR, cDNA was synthesized at $42^{\circ}C$ for 45 min before a quick incubation on ice after pre-denaturation at $95^{\circ}C$ for 5 min. The PCR reaction was carried out for 40 cycles at $96^{\circ}C$ for 30 see, $63^{\circ}C$ for 30 sec, $72^{\circ}C$ for 1 min, and finally at $72^{\circ}C$ for 10 min. The viral origin of the amplified product was confirmed by sequencing, with the sequence obtained having $95-98\%$ homology with published sequence data for SPFMV. The benefits of this RT-PCR based detection of SPFMV would be simple, rapid and specific.
