Establishment of Culture System of a New Strain NT0423 of Bacillus thuringiensis (새로운 Bacillus thuringiensis NT0423 균주의 배양체계)

  • For efficient and economical production of Btrc,illus tlz~rr.ingi~r1~sstirsa in NT0423 as amicrobial-control agents, a new culture medium and culture condition were developed. It was founcl that in terms of the cell growth rate and development ofsporulation of B, thrri.il~girrl.sis strain NT0423 in all SW culture media were more efficient than those inGYS and in LB media. Total cell number in all SW media showed similar values, hut SW32 lnediilm wasthe most efficient in the development of spore, which amo~~ntetod 3.7 x 10XC FUImI. Also. at the pHranging frorn 6.2 to 7.3 in the mediiun~ no ad\:erse effect was not made in the culture of B. thur-ingicnsisstrain NT0423. The optimal volume (%) of SW32 mecliuni in a 5 1 fernientor was determined as 4 8\rolume of total niediuni. resulting ill growth (4.2 x 1OTCFUlrnl) of H. t1~~irir1,yirrz.ssit.vr ain NT0423.   • 이 연구는 디지털 네트워크 세상으로 인한 새로운 학습이론인 커넥티비즘을 알아보고 SW 교육이 나가야할 방향을 알아보았다. 커넥티비즘에서 학습은 지식이나 정보를 연결하는 과정으로 정의하고 학습자가 기존에 알고 있는 지식을 바탕으로 새로운 지식을 끊임없이 연결하거나 단절시키는 과정이 반복된다고 말한다. 커넥티비즘에 따른 SW 교육의 새로운 방향을 제시하면, 1)SW 학습 목표는 지식의 연결과정으로 다양성과 가변성을 내포하여 설정하고, 2)현재 정보 교과서 개발 체재의 변화와, 3)협업과 네트워킹이 강조되는 도구를 활용한 SW 교육 활동을 지향하며, 4)교육 비용 절감을 위한 적시학습(just-in-time learning) 지향 및 5)SW 교육 목적에 합리적 의사결정을 통한 연결 지식 배양 역량을 추가하고, 6)문제 해결학습 보다는 문제발견학습을 중시하도록 설정해야 한다.

  • 인공지능과 소프트웨어가 중요한 역할을 하는 시대가 되었고, 이를 학생들에게 교육하여 미래의 AI/SW 인재를 양성하는 것에 많은 관심이 집중되고 있다. 해외 주요국에서는 이러한 시대적 흐름에 맞추어 AI/SW 분야의 인재 양성을 위해 노력하고 있으며, 국내에서도 여러 부처에서 관련된 다양한 정책을 시행하고 있다. 본 논문에서는 SW교육 관련 소셜미디어와 언론 데이터를 수집하고 이를 분석하여 국내 AI/SW교육에 대한 시사점을 제시하려고 한다. 이를 위해 2014년부터 2018년까지 총 5개년도의 데이터를 수집하고, 네트워크 분석 방법을 활용하여 연도별 SW교육의 흐름, 주요 등장 키워드, 연관 검색어들을 파악하였다. 이를 활용하여 미래의 AI/SW 교육 정책 수립 및 개선을 위한 시사점을 모색해보고자 한다.

Production of Extracellular Lipid by Rhodotorula graminis SW 214 (Rhodotorula graminis SW 214의 세포외 지방질 생산에 관한 연구)

  • A lipid producing yeast was screened from leaves of Albabiscus and was identified as a Rhodotorula graminis SW 214. During the shakining incubation of 8 days at $25^{\circ}C$, the yeast produced extracellular lipids of 7.3g/l of the media. The relative concentration of carbon and nitrogen sources in the media influenced the extracellular lipid production greatly. When with nitrogen sources in the media were almost exhausted for growth of the yeast the sufficient carbon sources, the lipid production proceeded vigorously. Eight days of batch cultivation with 8% glucose, 2.5g/l of yeast extract, $KH_{2}PO_{4}(1g/l)\;MgSO_{4}\;(0.2g/l)$ and pH 6 gave maximum biomass and extracellular lipid production of 8.05g/l and 8.89g/l, respectively. The acid value, saponificatio value, the iodine value, ad the unsaponifiable matter of the extracellular lipids of Rhodotorula graminis SW 214 were 2.6, 534, 5.1 and 2.4, respectively. Lipid was constituted 75.2% triglyceride, 5.9% free fatty acid, 10.8% phospholipid, 4.9% esterified sterol and 3.3% free sterol. Major fatty acids found were 3-hydroxypentadecanoate, 3-hydroxyhexadecanate, trans-9-octadecanate, cis-9-hexadecanate (hydroxy palmitic), 15-methylhexadecanate (oleic), 18-methylno-nadecanate, octadecanate (stearic) and 3-hydroxytridecante.

Acinetobacter marinus sp. novo and Acinetobacter seohaensis sp. nov., Isolated from Sea Water of the Yellow Sea in Korea

  • Two Gram-negative, nonmotile, coccobacilli, SW-$3^T$ and SW-$100^T$, were isolated from sea water of the Yellow Sea in Korea. Strains SW-$3^T$ and SW-$100^T$ contained ubiquinone-9 (Q-9) as the predominant respiratory lipoquinone and $C_{18:1}\;{\omega}9c$ and $C_{16:0}$ as the major fatty acids. The DNA G+C contents of strains SW-$3^T$ and SW- $100^T$ were 44.1 mol% and 41.9 mol%, respectively. A neighbor-joining tree based on l6S rRNA gene sequences showed that the two isolates fell within the evolutionary radiation enclosed by the genus Acinetobacter. Strains SW-$3^T$ and SW-$100^T$ exhibited a l6S rRNA gene similarity value of 95.7% and a mean DNA-DNA relatedness level of 9.2%. Strain SW-$3^T$ exhibited l6S rRNA gene sequence similarity levels of 93.5-96.9% to the validly described Acinetobacter species and fifteen Acinetobacter genomic species. Strain SW-$100^T$ exhibited l6S rRNA gene sequence similarity levels of less than 97.0% to the other Acinetobacter species except Acinetobacter towneri DSM $14962^T$ (98.0% similarity). Strains SW-$3^T$ and SW-$100^T$ exhibited mean levels of DNA-DNA relatedness of 7.3-l6.7% to the type strains of some phylogenetically related Acinetobacter species. On the basis of phenotypic, phylogenetic, and genetic data, strains SW-$3^T$ and SW-$100^T$ were classified in the genus Acinetobacter as two distinct novel species, for which the names Acinetobacter marinus sp. novo (type strain SW-$3^T$=KCTC $12259^T$=DSM $16312^T$) and Acinetobacter seohaensis sp. novo (type strain SW-$100^T$=KCTC $12260^T$=DSM $16313^T$) are proposed, respectively.

Exploring perception and experience of non-majors about SW education using CQR (SW교육에 대한 대학 비전공자의 인식과 경험 탐색: CQR을 중심으로)

  • The purpose of this study is to explore non-major students' perception and experiences in college software education. For this, we analyzed the reflection journals of 36 non-major students in D University based on the Consensual Qualitative Research(CQR). As a result, there was not general core concept to all students nor a typical core concept that appeared to more than 50% students. However, various variable core concepts could be derived. Overall, 57 variable concepts were derived from experience in SW education and 7 variable concepts for perception of SW education. Based on this result, we found many of non-major students feel difficulty from unfamiliarity to SW education. Also, many students have satisfaction in their perception to SW education about personalized learning that their professor provided in the class. Lastly, we conclude that a methodology for SW education needs to have a careful operation strategy and interactive design. Although this study has not been able to elucidate general core concepts that appear to all learners, it has significant implication in terms of providing various implicit core concepts and suggestions for effective software education for non-major students.

The Development of Stuttering Therapy Device and Clinical Application Cases Using Breathing Control Prolonged Speech Method (호흡 조절식 연장기법을 이용한 말더듬치료 장치개발 및 적용사례 연구)

  • The purpose of this study was to develop a stuttering therapy device to aid in stutter therapy. The research method used for this study was as follows: First, the stuttering therapy device based on analysis of the prolonged speech method used at home and abroad was designed to achieve the goal of research. Second, the stuttering therapy device was to be developed to maintain a vocalization state, to use bio-feedback visualization, to have enough inspiration, to use Korean language in this device, and to use transfer and maintenance training in daily life. Third, the stuttering therapy device effectiveness was to be verified through use in clinical cases. The results of subjects receiving speech therapy and using the breathing control prolonged speech device and SI(stuttering Interview) evaluation programs for 3 months were as follows: For subject A, the stuttered word rate was reduced from 3.20 SW/M to 0.5 SW/M. For subject B, the stuttered word rate was reduced from 1.90 SW/M to 0.75 SW/M. For subject C, the stuttered word rate was reduced from 3.37 SW/M to 0.34 SW/M. For Subject D, the stuttered word rate was reduced from 0.51 SW/M to 0 SW/M. Follow-up evaluations verified the effectiveness of how the stuttering therapy device can reduce subjects' SW/M.

The Mediating Effect of Learning Flow on Affective Outcomes in Software Education Using Games (게임을 활용한 SW교육의 정의적 성과에 대한 학습몰입의 매개 효과)

  • As software transforms the structure of industry, it becomes a key measure in determining market competitiveness. Therefore, various educational efforts have been attempted in Korea to cultivate software professionals to secure software competitiveness. While previous studies had focused mainly on the cognitive effectiveness of software education, the authors tried to focus on affective perspectives. The authors, therefore, aimed to analyze the predictive power of the recognition of software importance and learning flow on affective outcomes, such as efficacy of computational thinking skills, and attitude toward, and satisfaction with, software education. The data were collected from 103 sixth grade students who participated in a software education. Results show that software importance and learning flow had significant predictive power on affective outcomes; Learning flow mediated the relationship between software importance and affective outcomes. This study provides practical implications for improving affective outcomes in the design and implementation of software education.

Comparison of the operation of SW gifted curriculum: Focusing on face-to-face and non-face-to-face classes (SW영재학급 교육과정 운영 비교 : 대면 및 비대면 수업방식 중심으로)

  • In order for SW education to be established in the era of non-face-to-face caused by COVID-19, research on the efficiency of SW education according to face-to-face and non-face classes is needed. Therefore, this study classified the operation status of the curriculum of 30 SW gifted classes nationwide in 2020 according to the class method(face-to-face, non-face, and blended). Subsequently, the results of class time and production per person were compared and analyzed through quantitative analysis. According to the study, the type of classes that performed the most classes compared to the planned number of hours was non-face-to-face(90.9%), followed by face-to-face(84.2%) and the least was blended(80.5%). The average number of products per student was the highest in the face-to-face class(0.504), while the blended class(0.421) and non-face-to-face class(0.42). Based on the results of this study, the non-face-to-face approach is advantageous in securing the number of hours, but various measures should be prepared to solve this problem because teachers and students find it difficult to guide the output.

Comparison of Grain and Milling Properties between Waxy and Non-waxy Wheat (찰성밀과 보통밀간의 종실 및 제분특성비교)

  • For the purpose to verify the grain and evaluate milling properties of Korean waxy wheat, com­parison analysis between waxy wheat lines and their respective maternal parents were performed. The waxy lines showed various grain yields of 4.76${\~}$5.79 t/ha depending on parentages, which were corresponding to $80{\~}96\%$ levels of their respective maternal parents. One thousand grain weights of waxy lines were also lighter than its respective part in its parentage by exhibiting 32.8${\~}$34.6 g compared to 32.9${\~}$45.2 g of their parents. Test weights of waxy lines and their parents were 720${\~}$798 g/l and 786${\~}$797 g/l, respectively. The proportions of the grains above 2.5 mm in width were higher in order of Keumgang, Olgeuru, Geuru, SW97134, Suwon 292, Woori, SW97105, and SW97110. Waxy lines exhibited low milling properties by showing the straight flour yields ranging from $61.8\%$ to $67.1\%$ compared with the yields of their parents ranging from $66.1\%$ to $72.5\%$; the waxy lines were significantly lower in first break flour (Bl) and first reduction flour (Rl) yields in the Buller test mill, while significantly higher in the yields of second and third reduction flour (R2 and R3) than the respective ones of their parent wheat.
