담배세균성마름병[립고병(立枯病)]에 대한 담배품종의 저항성 검정법 (Screening Procedure of Tobacco Cultivars for Resistant to Bacterial Wilt Caused by Ralstonia solanacearum)

  • Bacterial wilt caused by Ralstonia solanacearum has become a severe problem on tobacco in Korea. No effective single control measure is available at present time. One of the most potential way for controlling the bacterial wilt on tobacco is growing tobacco cultivars resistant to the bacterial wilt. In this study, optimal conditions for screening tobacco cultivars resistant to the bacterial wilt were examined to provide reproducible and efficient methods in growth chamber testing and field experiments for evaluating plant disease resistance. For this, already-known inoculation methods, inoculum densities, and incubation temperature, and plant growth stages at the time of inoculation were compared using tobacco cultivars resistant (Nicotiana tabacum cv, NC95), moderately resistant (N. tabacum cv. SPG70), and susceptible (N. tabacum BY4) to the bacterial disease. It was determined that root-dipping of tobacco seedlings at six true leaf stage into the bacterial suspension with inoculum level of $10^8$ colony-forming units (CFU)/ml for 20 min before transplanting was simple and most efficient in testing for resistance to the bacterial wilt of tobacco caused by R. solanacearum, for which disease incidences and severities were examined at 2 weeks of plant growth after inoculation at $20{\sim}25^{\circ}C$ in a growth chamber. These experimental conditions could discriminate one tobacco cultivar from the others by disease severity better than any other experimental conditions. In field testing, the optimum time for examining the disease occurrence was late June through early July. These results can be applied to establishing a technical manual for the screening of resistant tobacco cultivars against the bacterial wilt caused by R. solanacearum.

Beach-chair lateral traction position using a lateral decubitus distracter in shoulder arthroscopy

  • The beach-chair traction position is designed to allow the use of traction while allowing the surgeon to orient the shoulder in an upright position and convert to an open procedure, if necessary. The patient is placed in the beach-chair position under general anesthesia. A three-point shoulder holder (Arthrex, Naples, Florida) is attached to the rail of the operating table on the same side as the surgeon, whereas it is placed on the side opposite the surgeon in the lateral decubitus position. A shoulder traction and rotation sleeve (Arthrex) are affixed to the arm following the manufacturer's instructions. Positioning the thumb toward the closed side of the sleeve ensures a field for the anterior portion of the rotator cuff and prevents the tendency of the suspension apparatus to place the arm in internal rotation. The arm is maintained in 30 to 40 degree abduction and 30 to 40 degree flexion by controlling the length and height of the bar and the location of the universal clamp. The universal clamp allows multiple planes of adjustment to control abduction and forward movement of the arm. The sleeve is attached to the longitudinal traction cable using a sterile hook, and a lateral strap is secured around the proximal portion of the sleeve to the overhead traction cable to ensure a field for glenohumeral reconstruction. The use of a lateral strap permits ideal shoulder positioning for improved access to the anterior and inferior glenohumeral joint. The lateral strap can be released or removed to widen the subacromial space during subacromial decompression or rotator cuff repair. A 10-lb weight is attached to the longitudinal traction cable for an average-sized person.

초음파 처리 조건에 따른 집락형 유해남조류 Microcystis 세포수 변화 연구 (Colonial Cyanobacteria, Microcystis Cell Density Variations using Ultrasonic Treatment)

  • It is difficult to count colonial cyanobacteria Microcystis cells since the thickness of colonies is constrained by amorphous mucilage, making it impossible to estimate the number of cells. Disaggregation of Microcystis colonies into single cell is needed to improve the accuracy and precision of cell density estimation of naturally collected samples. Uultrasonic treatment method is commonly used owing to the simplicity and immediacy of the procedure. However, amplitude, frequency, and duration of ultrasonic treatment also cause cell loss during the experiment. Optimal ultrasonic treatment has not been standardized yet. Therefore, the objective of this study was to investigate optimal ultrasonic treatment by analyzing cell density and colony numbers. We collected colonial Microcystis from Changnyeong-Haman weir area in Nakdong River during harmful algal boom period from September to October in 2017. Ultrasonic treatment method was applied to disrupt colonies into single cells to enumerate cell density. Among treatment conditions, results from continuously treated for 100 seconds were found to be the optimum to reduce colonies to a suspension of single cell without cell losses under high and low density of Microcystis cells. Lugol iodine fixed cells followed by sonication showed less negative impact of cell damage within the optimal treatment time (100 seconds). Furthermore, disaggregated cells treated by sonication enables microscopic observation more easily since gas vacuoles were collapsed to facilitate sedimentation of cells under the counting chamber for quantitative enumeration of buoyant Microcystis cells.

  • 복합재료 중 가장 많이 사용되는 탄소계 나노복합재의 성능 향상에 대한 연구가 수행되었다. 나노 사이즈의 탄소나노튜브(CNT)와 마이크로 사이즈의 그레파이트(Graphite) 입자를 일정한 형태로 배열하여 나노복합재를 제작하였다. 입자를 배열하기 위하여 전기장을 활용하였다. 일정한 방향 혹은 형태로 배열된 입자는 에폭시 기지 속에서 섬유강화 복합재 처럼 일정한 방향으로 강성의 향상을 나타낸다. 복합재의 구조적 강성이나 물리적 특성은 강화 입자의 배열상태에 따라 매우 달라지게 된다. 본 연구에서는 특히 탄소나노튜브와 그레파이트 강화입자의 전기장 배열에 의해서 만들어진 나노복합재의 강성 및 방사능 차폐특성을 규명하였다. 전기장에 의한 입자의 배열로 만들어진 탄소계 복합재는 구조적 성능뿐만 아니라 물리적인 방사능 차폐에서도 훌륭한 특성을 나타내었다.

적혈구를 이용한 Daunorubicin의 배송시스템 (Delivery System of Daunorubicin by Red Blood Cells)

  • Drug delivery system by the use of red blood cells was established to sustain the release of drugs in the circulatory system by the intravenous injection. The entrapment method by the preswelling technique was re-examined and evaluated for searching the new entrapping conditions without hemolysis. The addition of 4 volume of $0.6{\times}\;hank's$ balanced salt solution (HBSS) into 1 volume of 50% red blood cells suspension did not induce the hemolysis and change the hematocrit level in this experimental condition (within 15 min). Most of daunorubicin could be entrapped into red blood cells within 15 min. While the intracellular adenosine triphosphate (ATP) level followed by the entrapment was reduced to 86% of normal ATP level, the membrane fluidity and the shape factor of red blood cells were not altered. The release rate of daunorubicin from red blood cells was affected by the hemolysis under this condition. To maintain the intracellular ATP in red blood cells, the new reaction buffer was made With the addition of ATP and sodium pyruvate during the entrapment procedure because the hemolysis during the release test would reflect the loss of intracellular ATP that might result in the decrease of the viability in vivo. The addition of ATP raised the intracellular ATP level, which protect the hemolysis during the release test.

Indica 벼의 원형질체들로부터 식물체 재분화 (Plant Regeneration from Protoplasts of Indica Rice)

  • Indica 벼의 원형질체들로 부터 효과적인 식물체 재분화 방법을 개발하였다. 이 방법은 배형성 진탕 세포 배양체로 부터 나출된 원형질체들을 feeder cell들이 agarose에 embedded된 배지 표면에 놓인 filter membrane 위에 배양하는 것이다. Feeder cell로서 Lolium multiflorum 세포배양체를 사용했을 때가 Oryza ridleyi를 사용했을 때보다 효과적이었고, 원형질체 평탄효율은 세포 배양체 age에 따라 달랐지만 최고로 0.68% 까지 증가되었다. Carbohydrate source로서 maltose를 사용하거나 maltose와 sucrose를 1:1로 조합했을 때가 sucrose 단독으로 사용했을 때보다 식물체 재분화율이 증가되었고, 고농도의 agarose를 이용하여 원형질체로부터 유도된 캘러스를 dehydration시켰을 때 또한 재분화율이 괄목하게 증가되었다. 식물체 재분화율은 control 캘러스로부터 3.1∼30.6% 였지만 dehydration처리한 캘러스로부터는 30.7∼70.7%까지 증가되었다. 원형질체로 부터 유도된 식물체들은 형태적으로 정상이며 개화했다.

  • 광범위 제초제인 Bast $a^{(R)}$에 대해 저항성을 가지는 형질전환체 벼를 개발하였다. Bar유전자를 함유하고 있는 플라스미드 pCaMV35S::Bar를 embryogenic 현탁 배양체로부터 분리한 벼의 원형질체에 도입하였다. Phosphinotricin에 대해 저항성을 가지는 형질전환체 식물체들이 재분화되었고, 이들을 15 mg/l phosphinotricin이 함유한 배지에서 다시 선별하였다. 형질전환체 벼에서 bar유전자의 삽입과 발현을 Southern과 Northern blot분석으로 확인하였고, 또한 $R_1$ 형질전환 식물체들을 PAT 활성 assay로 재차 유전자 발현을 확인하였다. Bar 유전자는 다음 세대인 $R_1$ 식물체에서 3:1 멘델 유전 양상을 나타내었고, 형질전환체 $R_1$$R_2$ 식물체들은 fieild에서 살포되는 제초제 양만큼 Bast $a^{(R)}$ 를 살포했을 때 제초제 저항성을 나타내었다..

방사선에 의한 EL4 마우스 백혈병세포 및 정상 마우스 비장 임파구 DNA strand breaks의 측정 (Radiation-induced DNA strand breaks in EL4 cells and mouse spleen lymphocytes)

  • The filter elution technique was used to assay $^{60}Co$ $\gamma$ ray-induced DNA strand breaks(SB) in EL4 mouse leukemia cell and mouse spleen lymphocyte. The lymphocytes were stimulated with lipopolysaccharide (LPS, $20{\mu}g/ml$) to label $[^3H]$ thymidine. EL4 cells and lymphocytes in suspension were exposed at $0^{\circ}C$ to 0Gy, 1Gy, 5Gy, 10Gy or l5Gy for DNA single strand breaks(SSB) assay and 0Gy, 25Gy, 50Gy, 75Gy or 100Gy for DNA double strand breaks(DSB) assay of $^{60}Co$ radiation and elution procedure was performed at pH12.1 and 9.6. The number of DNA strand breaks increased with increasing doses of r rays. The strand scission factor(SSF) was estimated in each experiment (eluted volume 21ml). The slope of SSB EL4 cells was $0.01301{\pm}0.00096Gy^{-1}$ (n=5), the slope of SSB for lymphocytes was $0.01097{\pm}0.00091Gy^{-1}$ (n=5) and the slope of DSB for lymphocytes was $0.001707{\pm}0.0000573Gy^{-1}$ (n=5). Thus EL4 cells were more sensitive to induction of DNA SSB by ionizing radiation than lymphocytes (p<0.005). The ratio of slope of dose-response relationship (SSF versus dose) of lymphocytes DNA SSB as compared with the slope of DNA DSB was 6.4.

남북 투자분쟁해결의 법적쟁점에 관한 고찰 (A Study on the Legal Issues of Inter-Korean Investment Disputes Settlement System)

  • The resumption of economic cooperation between South and the North Korea will be a new growth engine for our economy. Many Korean companies are preparing to invest in North Korea in accordance with the progress of inter-Korean relations. However, there are many risks inherent in inter-Korean economic cooperation, as experienced in previous cases. Specifically, one should be prepared for unfair measures such as the expropriation of investment assets of South Korean enterprises by North Korea authorities. Therefore, it is essential to review the protection measures of investment in North Korea and to review the investment dispute settlement system. The South and the North have an agreement to establish the inter-Korean Commercial Arbitration Committee to resolve the disputes that may arise if one party's investments are lost due to inappropriate or unfair measures due to the other party's authority. However, the Investment Agreement, which governs the Inter-Korean Commercial Arbitration Committee, contains a number of declarative statements that are somewhat ineffective. Even today, nearly 20 years after the adoption of the Agreement, the specific detailed procedures have shown no real progress, such as in the enactment of arbitration rules. Therefore, at present, it is difficult to expect a system that can effectively address the damage of our corporations which have invested in North Korea. When the assets freeze after the suspension of Kumgang tourism and the closure of the Kaeseung Industrial Complex by North Korea, the activation of the inter-Korean Commercial Arbitration Committee is the most important prerequisite for economic cooperation with North Korea. For this purpose, the resolution of disputes through the Inter-Korean Commercial Arbitration Committee has to be made more concrete, with the effectiveness of the dispute settlement system enhanced by means of various efforts.

Geotechnical engineering behavior of biopolymer-treated soft marine soil

  • Soft marine soil has high fine-grained soil content and in-situ water content. Thus, it has low shear strength and bearing capacity and is susceptible to a large settlement, which leads to difficulties with coastal infrastructure construction. Therefore, strength improvement and settlement control are essential considerations for construction on soft marine soil deposits. Biopolymers show their potential for improving soil stability, which can reduce the environmental drawbacks of conventional soil treatment. This study used two biopolymers, an anionic xanthan gum biopolymer and a cationic ${\varepsilon}-polylysine$ biopolymer, as representatives to enhance the geotechnical engineering properties of soft marine soil. Effects of the biopolymers on marine soil were analyzed through a series of experiments considering the Atterberg limits, shear strength at a constant water content, compressive strength in a dry condition, laboratory consolidation, and sedimentation. Xanthan gum treatment affects the Atterberg limits, shear strength, and compressive strength by interparticle bonding and the formation of a viscous hydrogel. However, xanthan gum delays the consolidation procedure and increases the compressibility of soils. While ${\varepsilon}-polylysine$ treatment does not affect compressive strength, it shows potential for coagulating soil particles in a suspension state. ${\varepsilon}-Polylysine$ forms bridges between soil particles, showing an increase in settling velocity and final sediment density. The results of this study show various potential applications of biopolymers. Xanthan gum biopolymer was identified as a soil strengthening material, while ${\varepsilon}-polylysine$ biopolymer can be applied as a soil-coagulating material.
