In virucidal efficacy testing, the chemical inactivation cannot be determined for all viruses due to the difficulties or the inability to culture sufficiently or the risk of exposure to the viruses. Therefore, disinfectants against these viruses could be evaluated by different methods and surrogate viruses are used as alternative. In this study we developed a method for efficacy testing of veterinary disinfectants using one of the candidate surrogate viruses, bacteriophage MS2, as part of the research on the selection of surrogate viruses for efficiency of efficacy testing of veterinary disinfectants. This method is based on the Animal and Plant Quarantine Agency (APQA) guidelines for efficacy testing of veterinary disinfectants. Bacteriophage and disinfectant are reacted in suspension in accordance with the APQA guidelines and then a newly established double agar layer method is applied for the efficacy test. The double agar layer method is summarized as follows: 1) The bottom agar with 1.5% agar is boiled and cooled before poured into petri dishes at volume of 20 mL, and dried under biological safety cabinet. 2) The top agar with 0.7% agar is boiled and kept at 50℃ before E. coli culture was seeded. 3) The serially diluted bacteriophage MS2-disinfectant mixtures 0.05 mL and E. coli host 0.01 mL (OD600 0.2~0.3) are mixed with 5 mL of top agar and incubate them at 50℃ for 5 min for reaction. 4) The resulting mixture is poured over top of a bottom agar plate and rocked sufficiently to ensure that the top agar covers the entire surface of the bottom agar. 5) The double agar layer is then placed under biological safety cabinet to allow the agar layer to solidify and subsequently incubated at 37℃ for 24 hr. 6) Following incubation, the plates may be inspected for plaques and record results.

In case of analyzing PAHs (EPA 16 compounds) in oil-contaminated soils, the lump of peaks may occur because of the aliphatic and polar compounds in oil. This phenomenon is due to the lower accuracy of the analysis. To solve this problem, evaluation of application of silicagel-alumina multi-layer fraction was performed using standard substances and oil-contaminated soils. As a result of application of silicagel-alumina multi-layer fraction cleanup method using standard substances, recovery rates of surrogate standards (5 compounds including Naphthalene-d8) were 83~100% and those of target standards were 75~129%. These were to meet the target values (60~130%) in this study. When used 4% water-silicagel column analyze PAHs in oil-contaminated soils, Some problems were generated for quantitative analysis of PAHs; concentration of PAHs was underestimated due to an upward baseline of internal standard (recovery rate: less than 60%) and overestimated by the lump of peaks which were not purified (the biggest recovery rate: more than 400%). On the other hand, in case of silicagel-alumina multi-layer fraction cleanup method, recovery rate of surrogate standards were 61~101.6%. Therefore this cleanup method was considered a valid method to improve accuracy of analysis of PAHs in oil-contaminated soils.
In this study, a fluorescent silica nano particle is used as the surrogate for challenging test of membrane surface integrity. The particles are functionalized by a fluorescent dying agent so that as an ultraviolet light is imposed a bright fluorescent image from the particles can be taken. If a membrane surface is damaged and has a compromised part larger than the size of surrogate the fluorescent particles would pass through and contained in the permeate. An operator can directly notice whether the membrane surface is damaged or not by detecting a fluorescent image taken from the permeate. Additionally, the size of compromised part is estimated through analysing the fluorescent image in which we surmise the mass of particles included in the permeate by calculating an average RGB value of the image. The pilot scale experiments showed that this method could be applied successfully to determine if a membrane surface had a damaged parts regardless of the test condition. In the testing on the actual damaged area of $4.712mm^2$, the lowest error of estimating the damaged area was -1.32% with the surrogate concentration of 80 mg/L, flux of $40L/m^2/hr$ for 25 minutes of detection. A further study is still going on to increase the lowest detection limit and thus decrease the error of estimation.

The damage tolerance analysis is required to guarantee the structural safety and the reliability for aircraft components. The damage tolerance method, which evaluate the life considering the initial crack, considers a fatigue design model of the aircraft main structure. The fatigue crack growth life should be calculated in damage tolerance analysis and the inspection time to define the replacement cycle. In this paper, the damage tolerance analysis is performed for a turbine wheel which has complex geometry. The equation of the stress intensity factor for complex geometry is hard to know, so that they are usually processed by finite element analysis which takes long time. To solve this problem, the stress intensity factors at specified crack are obtained by the FEA and the crack growth life is evaluated using the surrogate model which is generated by the regression analysis of the FEA data. From the results, the efficiency of the crack growth life calculation and the damage tolerance analysis could be increased by taking the surrogate model.
Computer modeling is essential to evaluate possible design of suspension for a railway vehicles. By creating a simulation, the engineers are able to assess the feasibility of a given design and change the design factors to get a better design. But if one wishes to perform complex analysis on the simulation, such as railway vehicle dynamic, the computational time can become overwhelming. Therefore, many researchers have turned to surrogate modeling. A surrogate model is essentially a regression performed on a data sampling of the simulation. In the most general sense, metamodels(surrogate model) take the form $y(x)=f(x)+{\varepsilon}$, where y(x) is the true simulation output, f(x) is the metamodel output, and $\varepsilon$ is the error between the two. In this paper, a second order polynomial equation is partially used as a metamodel to represent the forty-six dynamic performances for high speed train. The number of factors as design variables of the metamodel is twenty-nine, which are composed the dynamic characteristics of suspension. This metamodel is used to search the optimum values of suspension characteristics which minimize the dynamic responses for high speed train. This optimization is a multi-objective problem which have many design variables. This paper shows that the response surface model which is made through the design of analysis of computer experiments method is very efficient to solve this complex optimization problem.

An optimization procedure for the design of a ventilation axial-flow fan is presented in this paper. Flow analyses of the preliminary fan are performed by solving three-dimensional Reynolds-averaged Navier-Stokes equations via a finite-volume solver with the shear-stress transport turbulence model as a turbulence closure. Three variables, the hub-to-tip ratio and the stagger angles at the mid and tip spans, are selected for the optimization. The Latin-hypercube sampling method as a design-of-experiments technique is used to generate twenty-five design points within the design space. and the weighted average surrogate models, WTA1, WTA2, and WTA3, are applied for find optimal designs. The results show that the efficiency is considerably enhanced.
Understanding the fundamental characteristics of supersonic hydrogen jets is important for the optimization of combustion in hydrogen engines. Previous studies have used helium as a surrogate gas to characterize the hydrogen jet characteristics due to potential explosion risks of hydrogen. It was based on the similarity of hydrogen and helium jet structures in supersonic conditions that has been confirmed using hole-type injectors and large-cone-angle pintle-type injectors. However, the validity of using helium as a surrogate gas has not been examined for recent small-cone-angle pintle-type injectors applied to direct-injection hydrogen engines, which form a supersonic hollow cone near the nozzle and experience the jet collapse downstream. Differences in the physical properties of hydrogen and helium could alter the jet development characteristics that need to be investigated and understood. This study compares supersonic jet structures of hydrogen and helium injected by a small-cone-angle (50°) pintle-type hydrogen injector and discusses their differences and related mechanisms. Jet penetration length and dispersion angle are measured using the Schlieren imaging method under engine-like injection conditions. As a result, the penetration length of hydrogen and helium jets showed a slight difference of less than 5%, and the dispersion angle showed a maximum of 10% difference according to the injection condition.
While job stress evaluations are reported in the recent surveys upon the nuclear power plants(NPPs), any significant advance in the types of questionnaires is not currently found. There are limitations to their usefulness as analytic tools for the management of safety resources in NPPs. Data mining(DM) has emerged as one of the key features for data computing and analysis to conduct a survey analysis. There are still limitations to its capability such as dimensionality associated with many survey questions and quality of information. Even though some survey methods may have significant advantages, often these methods do not provide enough evidence of causal relationships and the statistical inferences among a large number of input factors and responses. In order to address these limitations on the data computing and analysis capabilities, we propose an advanced procedure of survey analysis incorporating the DM method into a statistical analysis. The DM method can reduce dimensionality of risk factors, but DM method may not discuss the robustness of solutions, either by considering data preprocesses for outliers and missing values, or by considering uncontrollable noise factors. We propose three steps to address these limitations. The first step shows data mining with response surface method(RSM), to deal with specific situations by creating a new method called response surface data mining(RSDM). The second step follows the RSDM with detailed statistical relationships between the risk factors and the response of interest, and shows the demonstration the proposed RSDM can effectively find significant physical, psycho-social, and environmental risk factors by reducing the dimensionality with the process providing detailed statistical inferences. The final step suggest a robust stress management system which effectively manage job stress of the workers in NPPs as a part of a safety resource management using the surrogate variable concept.
Norovirus has become the most common cause of human gastroenteritis in developed countries. Detection procedures of foodborne viruses from foods require several steps. The concentration step using polyethylene glycol (PEG) is time-consuming and the detection efficiency of reverse transcription-polymerase chain reaction (RT-PCR) is affected by inhibitors from food components. In this study, a rapid detection method based on buoyant density centrifugation was developed to replace the time-consuming chloroform-polyethylene glycol-Tris Tween method. Feline calicivirus that belongs to the family Caliciviridae was used as a surrogate model for norovirus. After artificial inoculation of feline calcivirus (FCV) to oyster and lettuce, 830 ${\mu}L$ of homogenized sample suspension was layered on the top of 670 ${\mu}L$ 20% percoll and centrifuged. Then RNA extraction step was proceeded with the supernatant. By varying several physical conditions, the detection limits were lowered to $2.4{\times}10^2$ PFU per 1 g in oyster and $2.4{\times}10^0$ PFU per 1 g in lettuce. The protocol obtained in this study could be used to develop new detection method for norovirus in foods.
The studies for the derivatization of 3-monochloropropane-1,2-diol (3-MCPD) were performed mainly as acylation with HFBI (heptafluorobutyrylimidazole), alkylation with PBA (phenylboric acid) and silylation with BSTFA (N,O-bis[trimethylsilyl]trifluoroacetamide). Also silylation with MTBSTFA(N-methyl-N-[tert.-butyldimethylsilyl] trifluoroacetamide) and acylation with MBTFA (N-Methyl-bis[trifluoro-acetamide]) were also considered. Except the TBDMS derivative of 3-MCPD, all the derivatives were detected well. The derivatives of 3-MCPD with HFBI, PBA and BSTFA showed below 10 ${\mu}g/kg$ which was sensitive enough to satisfy Korea maximum residue limit 0.3 mg/kg. Among the tested adsorbents, Extrelut20 and Florisil were evaluated as the proper adsorbents to eliminate the soy sauce matrix for 3-MCPD. Ethyl acetate was the most efficient eluent with good recovery rate. The desired surrogate compound and internal standard were 1,2-butanediol and 1,2-dibromo-3-chloropropane, respectively. The limit of detection for PB-MCPD and TMS-MCPD were 10.16 and 7.06 ${\mu}g/kg$ on GC/MSD, respectively. HFB-MCPD derivative showed the lowest detection limits 2.98 and 5.32 ${\mu}g/kg$ by GC/ECD and GC/MSD, respectively.
