The feasibility of stimulating in situ aerobic cometabolic activity of indigenous microorganisms was investigated in a trichloroethylene (TCE)-contaminated aquifer. A series of single-well natural drift tests (SWNDTs) was conducted by injecting site groundwater amended with a bromide tracer and combinations of toluene, oxygen, nitrate, ethylene and TCE into an existing monitoring well and by sampling the same well over time. Three field tests, Push-pull Transport Test, Drift Biostimulation Test, and Drift Surrogate Activity Test, were performed in sequence. Initial rate of toluene degradation was much faster than the rate of bromide dilution resulting from natural groundwater drift, indicating stimulation of indigenous toluene-oxidizing microorganisms. Transformation of ethylene, a surrogate probing overall activity of TCE transformation, was also observed, and its transformation results in the production of ethylene oxide, suggesting that some tolueneoxidizing microorganisms stimulated may express a orthomonooxygenase enzyme. Also in situ transformation of TCE was confirmed by greater retardation of TCE than bromide after the stimulation of toluene-oxidizing microorganisms. These results indicate that, in this environment, toluene and oxygen additions stimulated the growth and aerobic cometabolic activity of indigenous microorganisms expressing orthomonooxygenase enzymes. The simple, low-cost field test method presented in this study provides an effective method for conducting rapid field assessments and pilot testing of aerobic cometabolism, which has previously hindered application of this technology to groundwater remediation.
This paper proposes a sampling-based sensitivity method for dealing with electromagnetic coupled design problems effectively. The black-box modeling technique is basically applied to obtain an optimum regardless of how strong the electromagnetic, thermal and structural analyses are coupled with each other. To achieve this, Kriging surrogate models are produced in a hyper-cubic local window with the center of a current design point. Then design sensitivity values are extracted from the differentiation of basis functions which consist of the models. The proposed method falls under a hybrid optimization method which takes advantages of the sampling-based and the sensitivity-based methods. Owing to the aforementioned feature, the method can be applied even to electromagnetic problems of which the material properties are strongly coupled with thermal or structural outputs. To examine the accuracy and validity of the proposed method, a strongly nonlinear mathematical example and a coil design problem for local induction heating are tested.

The building energy performance indicator, called Energy Performance Index (EPI), has been used for the past decades in South Korea. It has a list of design variables assigned with weighting factors (a, b). Unfortunately, the current EPI method is not performance-based but very close to a prescriptive rating. With this in mind, this study aims to propose a new performance-based EPI method. For this purpose, a global sensitivity analysis method, Sobol, is employed. The Sobol method is suitable for complex nonlinear models and can decompose all the output variance due to every input. The Sobol sensitivity index of each variable is defined as 0 to 1 (0 to 100%), and the sum of all sensitivity indices is equal to 1 (100%). In this study, an office building was modeled using EnergyPlus and then the Latin Hypercube Sampling (LHS) was conducted to generate a surrogate model to EnergyPlus. The sensitivity index was suggested to replace weight (a) in the existing EPI. In addition, the discrete weight (b) in the existing EPI was replaced by a set of continuous regression functions. Due to the introduction of the sensitivity index and the continuous regression functions, the new proposed approach can provide far more accurate outcome than the existing EPI (R2: 0.83 vs. R2: 0.01 for cooling, R2: 0.66 vs. R2: 0.01 for total energy). The new proposed approach proves to be more rational, objective and performance-based than the existing EPI method.
While the assessment of mean flow field is very important to characterize the hydrodynamic aspect of the flow regime in river, the conventional methodologies have required very time-consuming efforts and cost to obtain the mean flow field. The paper provides an efficient technique to quickly assess mean flow field by developing and applying spatial averaging method utilizing repeatedly surveyed acoustic Doppler current profiler(ADCP)'s cross-sectional measurements. ADCP has been widely used in measuring the detailed velocity and discharge in the last two decades. In order to validate the proposed spatial averaging method, the averaged velocity filed using the spatial averaging was compared with the bench-mark data computed by the time-averaging of the consistent fix-point ADCP measurement, which has been known as a valid but a bit inefficient way to obtain mean velocity field. The comparison showed a good agreement between two methods, which indicates that the spatial averaging method is able to be used as a surrogate way to assess the mean flow field. Bed shear stress distribution, which is a derived hydrodynamic quantity from the mean velocity field, was additionally computed by using both spatial and time-averaging methods, and they were compared each other so as to validate the spatial averaging method. This comparison also gave a good agreement. Therefore, such comparisons proved the validity of the spatial averaging to quickly assess mean flow field. The mean velocity field and its derived riverine quantities can be actively used for characterizing the flow dynamics as well as potentially applicable for validating numerical simulations.
Foodborne illness caused by Noroviruses (NVs) is increasing rapidly in Korea. This study developed an effective detection protocol for NVs found in contaminated oysters and lettuce through an investigation using the major steps of virus particle separation, concentration and RT-PCR. As a surrogate model for NVs, the cultivable feline calicivirus (FCV) that belongs to the same Caliciviridae family was used. Instead of using a time-consuming ultracentrifugation method, efficient methods based on solvent extraction and PEG precipitation procedure were applied. Direct homogenization of a 25g sample of whole oyster and lettuce in 175mL PBS provided the simplicity that would be needed in the actual field of food product examination. The overnight PEG precipitation step at $4^{\circ}C$ was reduced to 3 h by placing the reaction tube in ice and by adjusting the PEG concentrations. The application of the use of chloroform and 0.2 ${\mu}m$ syringe filtration together showed a better detection efficiency than the use of chloroform alone in removing PCR inhibitors for both oyster and lettuce samples. Also, dilution of the extracted RNA solution before PCR provided increased sensitivity. The improved detection protocol developed in this study could be efficiently applied to detect FCV and most likely NVs from oysters and lettuce.

In order to characterize the mixing process of confluence for understanding the impacts of a river on the other river, it has been crucial to analyze the spatial mixing patterns for main streams depending on various inflow conditions of tributaries. However, most conventional studies have mostly relied upon hydraulic or water quality numerical models for understanding mixing pattern analysis of confluences, due to the difficulties to acquire a wide spatial range of in-situ data for characterizing mixing process. In this study, backscatters (or SNR) measured from ADCPs were particularly used to track sediment mixing assuming that it could be a surrogate to estimate the suspended sediment concentration. Raw backscatter data were corrected by considering the beam spreading and absorption by water. Also, an optical Laser diffraction instrument (LISST) was used to verify the method of acoustic backscatter and to collect the particle size distribution of main stream and tributary. In addition, image-based spatial distributions of sediment mixture in the confluence were monitored in various flow conditions by using an unmanned aerial vehicle (UAV), which were compared with the spatial distribution of acoustic backscatter. As results, we found that when acoustic backscatter by ADCPs were well processed, they could be proper indicators to identify the spatial patterns of the three-dimensional mixing process between two rivers. For this study, flow and sediment mixing characteristics were investigated in the confluence between Nakdong and Nam river.

Laidback fan shaped film-cooling hole is formulated numerically and optimized with the help of three-dimensional numerical analysis, surrogate methods, and the multi-objective evolutionary algorithm. As Pareto optimal front produces a set of optimal solutions, the trends of objective functions with design variables are predicted by hybrid multi-objective evolutionary algorithm. The problem is defined by four geometric design variables, the injection angle of the hole, the lateral expansion angle of the diffuser, the forward expansion angle of the hole, and the ratio of the length to the diameter of the hole, to maximize the film-cooling effectiveness compromising with the aerodynamic loss. The objective function values are numerically evaluated through Reynolds- averaged Navier-Stokes analysis at the designs that are selected through the Latin hypercube sampling method. Using these numerical simulation results, the Response Surface Approximation model are constructed for each objective function and a hybrid multi-objective evolutionary algorithm is applied to obtain the Pareto optimal front. The clustered points from Pareto optimal front were evaluated by flow analysis. These designs give enhanced objective function values in comparison with the experimental designs.

Primordial germ cell (PGC) is the progenitor cell of the germ cell lineage and eventually give rise to gametes that are responsible for creating individual organisms via a fertilization process. This means that PGC is a unique cell that can be converted into individual fish. This advantage of PGCs would make it possible to develop various applications in the field of fish bioengineering. First, PGCs may make it easier to preserve the genetic resources of fish. Cryopreservation of fish eggs or embryos has not been successfully achieved so far. Therefore, the only possible method to preserve genetic resources of fishes is to raise fish as live individuals. If PGCs isolated from various fishes could be cryopresewed, these cells could be converted into live fishes via germ-line chimera production. This is particularly useful for preserving genetic materials of endangered species. Even if the species of interest were to become extinct, it could be recovered by the transplantation of cryopreserved PGCs into the embryos of a closely related species. Another application of this technology is in what could be termed "surrogate broodstock technology". (중략)

Consider a capacitated facility location problem in which the demands of customers are all equal and so are the capacities of facilities. Under the restriction that each customer's uniform demand be met by exactly one facility, the objective is to select a set of facilities to open, and to assign customer's demand to them so as to minimize the total cost which includes fixed costs of opening facilities as well as variable assignment costs. The problem is modelled as a pure zero-one program which may be viewed as a variant of well-known capacitated facility location problems. The purpose of this study is to develop efficient computational procedures for solving the pure zero-one facility location problems. Due to the special structure of our zero-one location problem with uniform demand, it can be converted to a location problem with the unimodular property. A Lagrangean relaxation algorithm is developed to solve the location problem. The algorithm is made efficient by employing a device which exploits the special structure of a surrogate constraint. The efficiency of the algorithm is analyzed through computational experiments with some test problems.
This study analyzes guidelines to select optimum number of grids to represent behavior of a given water system appropriately. The EFDC model was chosen as a 3-D hydrodynamic and water quality model and salt was chosen as a surrogate variable of pollutant. The model is applied to an artificial canal that receives salt water from coastal area and fresh water from a river from respective gate according to previously developed gate operation rule. Grids are subdivided in vertical and horizontal (longitudinal) directions, respectively until no significant changes are found in salinity concentrations. The optimum grid size was determined by comparing errors in average salt concentrations between a test grid systems against the most complicated grid system. MSE (mean squared error) and MAE (mean absolute error) are used to compare errors. The CFL (Courant-Friedrichs-Lewy) number was used to determine the optimum number of grid systems for the study site though it can be used when explicit numerical method is applied only. This study suggests errors seem acceptable when both MSE and MAE are less than unity approximately.
