Bovine piroplasmosis caused by Theileria sergenti results in economic loss in the dairy industry. The majority of calves infected with T. sergenti in Korea harbor mixed populations with Buffeli, Chitose and Ikeda types. The T. sergenti types of the infected calves were examined to evaluate the effects of diminazene aceturate on their infection. To confirm the type of the T. sergenti infection, the allele-specific PCR was performed with the erythrocyte specimen from the 5 naturally infected calves. The dfferent allele-specific genes encoding the p32, p33 and 34, the immunodominant piroplasmin surface proteins, were amplified using the 3 sets of the oligonucleotide primers by PCR. The calves were treated with diminazene aceturate at the dose of 2mg/kg once intravenously and monitored for 12 months at one month intervals by the allele-specific PCR. Diminazene aceturate did not effect the Ikeda type infection. But diminazene aceturate effected the Chitose and Buffeli type infection reducing T. sergenti parasitemia. It is postulated that diminazene aceturate may effect the infection of the Chitose and Buffeli types, but not that of Ikeda type.
In total, 131 Escherichia coli isolates from surface seawater of the Gomso Bay, of Korea, were analyzed for their susceptibility to 22 different antimicrobials and for genes associated with antimicrobial resistance and virulence. According to the disk diffusion susceptibility test, the resistance to tetracycline was most prevalent (33.6%), followed by that to ampicillin (22.1%), ticarcillin (22.1%), and trimethoprim (16.8%). More than 46.6% of the isolates were resistant to at least one antimicrobial, and 22.9% were resistant to three or more classes of antimicrobials; these were consequently defined as multidrug resistant. We further found that 29 ampicillin-resistant isolates possessed genes encoding TEM-type (93.1%) and SHV-type (6.9%) ${\beta}$-lactamases. Among the 44 tetracycline-resistant isolates, tetA and tetC were found in 35 (79.5%) and 19 (43.2%), respectively, whereas tetB was detected in only three isolates (6.8%). With regard to virulence genes, merely 0.8% (n = 1) and 2.3% (n = 3) of the isolates were positive for the enteroaggregative E. coli-associated plasmid (pCVD432) gene and the enteropathogenic E. coli-specific attaching and effacing (eae) gene, respectively. Overall, these results not only provide novel insight into the necessity for seawater sanitation in Gomso Bay, but they help reduce the risk of contamination of antimicrobial-resistant bacteria.
The rice blast disease caused by of Magnaporthe oryzae is one of the most destructive diseases of rice. By the microarray analysis, we profiled expression changes of genes during conidiation and found out many putative genes that are up-regulated. Among those, we first selected MGG_06399 encoding a dual-specificity tyrosine-regulated protein kinase (DYRK), homologous to YAK1 in yeast. To investigate functional roles of MoYAK1, We made ${\Delta}Moyak1$ mutants by homology dependent gene replacement. The deletion mutant showed a remarkable reduction in conidiation and produced abnormally shaped conidia smaller than those of wild type. The conidia form ${\Delta}Moyak1$ were able to develop a germ tube, but failed to form apppressoria on a hydrophobic coverslip. The ${\Delta}Moyak1$ formed appressria on a hydrophobic cover slip when exogenous cAMP was induced, but the appressoria shape was abnormal. The ${\Delta}Moyak1$ also formed appressoria abberent in shape on onion epidermis and rice sheaths and failed to penetrate the surface of the plants. These data indicate that MoYAK1 is associated with cAMP/PKA pathway and important for conidiation, appressorial formation and pathogenic development in Magnaporthe oryzae. Detailed characterization of MoYAK1 will be presented.
In this study, we demonstrate that the biological unnatural amino acid incorporation method can be utilized in vivo to synthesize an alkyne-terminated telechelic protein, Synthesis of terminally-functionalized polymers such as telechelic polymers is recognized to be important, since they can be employed usefully in many areas of biology and material science, such as drug delivery, colloidal dispersion, surface modification, and formation of polymer network. The introduction of alkyne groups into polymeric material is particularly interesting since the alkyne group can be a linker to combine other materials using click chemistry. To synthesize the telechelic recombinant protein, we attempted to incorporate the L-homopropargylglycine into the recombinant GroES fragment by expressing the recombinant gene encoding Met at the codons for both N- and C-terminals of the protein in the Met auxotrophic E. coli via Hpg supplementation. The Hpg incorporation rate was investigated and the incorporation was confirmed by MALDI-TOF analysis of the telcchelic recombinant protein.

Streptococcus pneumoniae (the pneumococcus) causes a major upper respiratory tract infection often leading to severe illness and death in the elderly. Thus, it is important to induce safe and effective mucosal immunity against this pathogen in order to prevent pnuemocaccal infection. However, this is a very difficult task to elicit protective mucosal IgA antibody responses in older individuals. A combind nasal adjuvant consisting of a plasmid encoding the Flt3 ligand cDNA (pFL) and CpG oligonucleotide (CpG ODN) successfully enhanced S. pneumoniae-specific mucosal immunity in aged mice. In particular, a pneumococcal surface protein A-based nasal vaccine given with pFL and CpG ODN induced complete protection from S. pneumoniae infection. These results show that nasal delivery of a combined DNA adjuvant offers an attractive potential for protection against the pneumococcus in the elderly.
In this report, we describe the high-yield secretory expression of the recombinant human anti-HBsAg Fab fragment from Pichia pastoris that was achieved by co-integration of the genes encoding the heavy and light chains (both under the control of alcohol oxidase promoter) into the genome of the yeast cells. The fed-batch fermentations were carried out in a 5 L scale. Both chains of the Fab were successfully expressed upon methanol induction. The absorbance ($OD_{600}$) of the broth can reach 350~500 at the end of fed-batch phase. After the induction, the expression level of the recombinant Fab (soluble) reached 420~458 mg/L. The recombinant Fab fragment was purified from the crude culture supernatant by ion exchange chromatography and the purity of the recombinant Fab fragment was over 95%. The affinity activities of the crude fermentation supernatant and the purified Fab were analyzed by indirect ELISA, which showed that the purified recombinant Fab fragment had high affinity activity with hepatitis B surface antigen.
Here, we show that an endophytic bacterial strain, Enterobacter asburiae B1 exhibits the ability to elicit ISR in cucumber, tobacco and Arabidopsis thaliana. This indicates that strain B1 has a widespread ability to elicit ISR on various host plants. In this study, E. asburiae strain B1 did not show antifungal activity against tested major fungal pathogens, Colletotrichum orbiculare, Botrytis cinerea, Phytophthora capsici, Rhizoctonia solani, and Fusarium oxysporum. Moreover, the siderophore production by E. asburiae strain B1 was observed under in vitro condition. In greenhouse experiments, the root treatment of strain B1 significantly reduced disease severity of cucumber anthracnose caused by fungal pathogen C. orbiculare compared to nontreated control plants. By root treatment of strain B1 more than 50% disease control against anthracnose on cucumber was observed in all greenhouse experiments. Simultaneously, under the greenhouse condition, the soil drench of strain B1 and a chemical inducer benzothiadiazole (BTH) to tobacco plants induced GUS activity which is linked with activation of PR promoter gene. Furthermore, in Arabidopsis thaliana plants the soil drench of strain B1 induced the defense gene expression of PR1 and PDF1.2 related to salicylic acid and jasmonic acid/ethylene signaling pathways, respectively. In this study, for the main focus on root colonization by strain B1 associated with defense responses, bacterial cells of strain B1 was tagged with the gfp gene encoding the green fluorescent protein in order to determine the colonization pattern of strain B1 in cucumber. The gfp-tagged B1 cells were found on root surface and internal colonization in root, stem, and leaf. In addition to this, the scanning electron microscopy observation showed that E. asburiae strain B1 was able to colonized cucumber root surface.

Since Anton van Leeuwenhoek first observed a surface-associated multicellular structure of bacterial cells in the 17th century, it has been shown to exhibit an ability to form a biofilm by numerous bacterial species. The biofilm formation is composed of distinct developmental stages, which include an attachment/adhesion of a single cell, a proliferation toward monolayered coverage, a propagation to aggregated microcolony, a maturation to 3-dimensional structure, and subsequently a local degradation. Investigation to identify the essential factors for bacterial biofilm formation has been performed via classical genetic approaches as well as recently developed technologies. The initial stage requires bacterial motility provided by a flagellum, and outermembrane components for surface signal interaction. Type IV-pilus and autoaggregation factors, e.g., type I-fimbriae or Ag43, are necessary to reach the stages of monolayer and micro colony. The mature biofilm is equipped with extracellular polymeric matrix and internal water-filled channels. This complex architecture can be achieved by differential expressions of several hundred genes, among which the most studied are the genes encoding exopolysaccharide biosyntheses and quorum-sensing regulatory components. The status of our knowledge for the biofilms found in humans and natural ecosystems is discussed in this minireview.
To evaluate the effect of flip angle on flow rate measurements obtained with phase contrast MRI according to the flip angle degree in ascending aorta and velocity encoding (VENC) was (150 m/s). 1.5T MRI in patients 17 (female: 8, male: 9, mean age $57.9{\pm}15.4$) as a target by applying a non-breath holding techniques to flip angle VENC (150 cm/s) in each of the ascending aorta was measured by changing $20^{\circ}$, $30^{\circ}$ and $40^{\circ}$. Blood was obtained a peak velocity, average velocity, net forward volume, net forward volume/body surface area. Ascending aorta from average velocity (AV) measured the average value of the flip angle $20^{\circ}$ (9.87 cm/s), $30^{\circ}$ (9.6 cm/s) and $40^{\circ}$ (10.05 cm/s). As we face an aging society, the demand for personal mobility for disabled and aged people is increasing. In fact, as of 2017, the number of electric wheelchair in the country continues to increase to 90,000. However, people with disabilities and seniors are more likely to have accidents while driving, because their judgment and coordination are inferior to normal people. One of the causes of the accident is the interference of personal vehicle steering control due to unbalanced road surface conditions. In this paper, we introduce a encoder type semantic segmentation algorithm that can recognize road conditions at high speed to prevent such accidents. To this end, more than 1,500 training data and 150 test data including road surface damage were newly secured. With the data, we proposed a deep neural network composed of encoder stages, unlike the Auto-encoding type consisting of encoder and decoder stages. Compared to the conventional method, this deep neural network has a 4.45% increase in mean accuracy, a 59.2% decrease in parameters, and an 11.9% increase in computation speed. It is expected that safe personal transportation will be come soon by utilizing such high speed algorithm.
