Expressed sequence tags (ESTs) analysis of Acanthamoeba healyi

  • Randomly selected 435 clones from Acanthamoeba healyi cDNA library were sequenced and a total of 387 expressed sequence tags (ESTs) had been generated. Based on the results of BLAST search, 130 clones (34.4%) were identified as the genes enconding surface Proteins , enzymes for DNA, energy Production or other metabolism, kinases and phosphatases, protease, proteins for signal transduction, structural and cytoskeletal proteins, cell cycle related proteins, transcription factors, transcription and translational machineries, and transporter proteins. Most of the genes (88.5%) are newly identified in the genus Acanthamoeba. Although 15 clones matched the genes of Acanthamoeba located in the public databases, twelve clones were actin gene which was the most frequently expressed gene in this study. These ESTs of Acanthamoeba would give valuable information to study the organism as a model system for biological investigations such as cytoskeleton or cell movement, signal transduction, transcriptional and translational regulations. These results would also provide clues to elucidate factors for pathogenesis in human granulomatous amoebic encephalitis or keratitis by Acanthamoeba.

Identification of Streptomyces sp. Producing New Polyene Antibiotics and In Vivo Antimicrobial Activity of Tetrin C Against Phytopathogenic Fungi

  • A Streptomyces sp. isolated from a soil sample collected in Taejeon, Korea has previously been found to produce two new polyene antibiotics. The two new antibiotics were named "16-methyloxazolomycin (antibacterial)" and "tetrin C (antifungal)", and their chemical structures are presented elsewhere [10, 11]. In the current study, chemotaxonomy, numerical taxonomy, and ISP methods were all employed for the taxonomic study. The spore chains were spirales and the spore surface was smooth. The spore mass was a gray series and no melanin pigment was produced. On the basis of the morphological and physiological properties, the microorganism was identified to be Streptomyces erumpens, belonging to the gray series of category IV, as defined by Bergey′s Manual. Tetrin C at the concentration of 20 ${\mu}g$/ml demonstrated a potent in vivo (pot test) preventive effect against rice blast, rice sheath blight, cucumber gray mold, wheat powdery mildew, and barley leaf rust.

  • Most of the roads are paved with impermeable materials such as asphalt concrete and cement concrete, and in the event of heavy rainfall, rainwater directly flows into river through a drainage hole on the pavement surface. This large quantity of rainwater directly spilled into the river frequently leads to the flooding of urban streams, damaging lowlands and the lower reaches of a river. In recent years there has been a great deal of ongoing research concerning water permeability and drainage in pavements. Accordingly, in this research, a porous polymer concrete was developed for permeable pavement by using unsaturated polyester resin as a binder, recycled aggregate as coarse aggregate, fly ash and blast furnace slag as filler, and its physical and mechanical properties were investigated. Also, 3 types of permeable polymer block by optimum mix design were developed and rainfall runoff reduction effects by permeability pavement using permeable polymer block were analyzed based on hydraulic experimental model. The infiltration volume, infiltration ratio, runoff initial time and runoff volume in permeability pavement with permeable polymer block of $300{\times}300{\times}80$ mm were evaluated for 50, 100 and 200mm/hr rainfall intensity.

Determination of the Protecting Periods of Frost Damage at Early Age in Cold Weather Concreting (한중콘크리트의 초기 동해 방지를 위한 초기 양생기간의 산정)

  • Protections from the frost damage at early ages are one of the serious problems to be considered in cold weather concreting. Frost damage at early ages brings about the harmful influences on the concrete structures such as surface cracks and declination of strength development. Therefore, in this paper, protecting periods of frost damage at early ages according to the standard specifications provided in KCI(Korean Concrete Institute) are suggested by appling logistic curve, which evaluates the strength development of concrete with maturity. W/B, kinds of cement and curing temperatures are selected as test parameters. According to the results, the estimation of strength development by logistic curve has a good agreement between calculated values and measured values. As W/B and compressive strength for protecting from frost damages at early ages increase, it is prolonged. It shows that the protecting periods of FAC(Fly Ash Cement) and BSC(Blast-furnace Slag Cement) concrete are longer than those of OPC(Ordinary Portland Cement) concrete. The protecting peridos from frost damage at early age by JASS are somewhat shorter than those by this paper.

  • In this study, as the resistance of the carbonation for high volume admixture incorporating concrete, coating effect of using refined cooking oil in the surface of high volume admixture incorporating concrete has been tested. The following results could be made as the conclusion. For the fresh concrete, the slump and air content has been identified as satisfying the target range. For the hardened concrete, comparing with specimen of Plain, specimen with coating showed better long age compressive strength. For the carbonation speed, the specimen of FA30 showed highest speed and the specimen of BS60 showed higher speed than specimen of Plain. For all the specimens coated with RCO, as the decrease of capillary pores inside the concrete, the carbonation speed has been obviously decreased and with even better effect than using PEP coating. It could be identified that specimens with coating by RCO showed good effect on refrain the speed of carbonation.

  • This study aims to examine the effects of thickness of the concrete members and curing temperature on the properties of low heat concrete through heat of hydration analysis. Type of the members that was analyzed in the experiment is ternary mixture of ordinary portland cement, blast-furnace slag incorporating ratio(20%) and fly ash incorporating ratio(30%), which formed a mat foundation. Thicknesses of the concrete members were 1, 2 and 3(m) and three levels of curing temperatures were 10, 20 and 30(℃). They were applied to analyze the effects on the temperature and thermal cracking index. As a result, for temperature history, temperature difference between the central area and the surface tended to decrease as the thickness of the concrete members get thinner. For the temperature cracking index, on the other hand, the risk of cracking tended to decrease as the curing temperature gets higher and as the thickness gets thinner.

  • In this paper, we describe a flow meter system for pulverized coal developed for the pulverizer-burner system of a boiler or blast furnace, which uses the density and the velocity of the granule flow. The granule flow density is measured by a sensor that detects the capacitance from the electrode on the surface of the piping system. The velocity of granule flow can be calculated using the distance between two pairs of built-in sensors in the flow direction, the time obtained from the sampling cycle using the correlation method between two waveforms of the sensors. The flow rate is calculated from the density and velocity of the granule flow. The reliability and accuracy of the flow meter system has been verified by comparing the data with the weight measured from a load-cell.

  • Air deck charge blasting method which has been generally used in a surface mine and large scale developing site is one of the improved techniques with blasting effectiveness. Many studies and experiments have been tried to investigate the characteristics of pressure distribution in a blasting hole and increase the effectiveness of air deck charge blasting method. In this study, changes of pressure occurred in sections of air deck installed in various ways was computed and also changes of pressure with the location and length of air deck was analyzed, using numerical analysis program. Basically, all the numerical analysis was 2-Dimensional analysis and equation of status of explosives was JWL-EOS. Only to evaluate the variations of pressure in blast hole, it was assumed that rock mass is homogeneous but rock mass has different density and intensity.

Investigation of the Properties of Sand Tubules, a Biomineralization Product, and their Microbial Community

  • Sand tubules, made up of sand grains cemented by microbe-induced calcium carbonate precipitation, have been found in China's Ningxia Province. Sand tubules grow like a tree's roots about 40-60 cm below the surface. The properties of sand tubules and their bacterial community were examined. X-Ray diffraction analysis revealed that the sand tubules were associated with crystalline calcite. Scanning electron microscopy showed that the crystalline solid had a lamellar structure and lacked the presence of cells, suggesting that no bacteria acted as nucleation sites, nor that the crystalline solid was formed by the aggregation of bacteria. Denaturing gradient gel electrophoresis analysis showed 11 of the 12 detectable bands were uncultured bacteria by BLAST analysis in the GenBank database, and the rest were closely related to Paenibacillus sp. (100% identity). By cultivation techniques, the only strain isolated from the sand tubule was suggested to be related to Paenibacillus sp.; no archaea were found. Furthermore, Paenibacillus sp. was demonstrated to induce calcium carbonate precipitation in vitro.

Influence of Cement Type on the Diffusion Characteristics of Chloride Ion in Concrete (콘크리트의 염소이온 확산특성에 미치는 시멘트 종류의 영향)

  • To predict service life of concrete structures exposed to chloride attack, surface chloride concentration, diffusion coefficient of chloride ion, and chloride corrosion threshold value in concrete, are used as important factors. of these, as the diffusion coefficient of chloride ion for concrete is strongly influenced by concrete quality and environmental conditions of structures and may significantly change the service life of structures, it is considered as the most important factor for service life prediction. The qualitative factors affecting the penetration and diffusion of chloride ion into concrete are water-binder(W/B) ratio, age, cement type and constituents, chloride ion concentration of given environment, wet and dry conditions, etc. In this paper the influence of cement type on the diffusion characteristics of chloride ion in concrete was investigated through the chloride ion diffusion test. For this purpose, the diffusion characteristics in concrete with cement type such as ordinary portland cement(OPC), binary blended cement(BBC), and ternary blended cement(TBC) were estimated for the concrete with W/B ratios of 32% and 38%, respectively. It was observed from the test that the difussion characteristics of BBC containing OPC and ground granulated blast-furnace slag was found to be most excellent of the cement type used in this study.
