Acute promyelocytic leukemia (APL) is a myeloid leukemia caused by over-expression of fusion protein, PML/RAR$({\alpha})$, which was the result of chromosomal translocation and induces the blockage of differentiation of affected promyelocytes. Pharmacological dose of retinoic acid induces the activation of and subsequent degradation of PML/RAR$({\alpha})$ fusion protein, and then APL cells undergo through the normal differentiation pathway. Arsenic trioxide has proved effective in causing remission of acute promyelocytic leukemia by inducing apoptosis of this tumor cells, whereas the heterogeneity of cellular susceptibility to this cytotoxic agent limited its usage on more types of tumors in clinic. This work showed that arsenic trioxide could induce apoptosis of a panel of acute promyelocytic leukemic cell lines, all-trans-retinoic acid (ATRA) sensitive NB4 cells and ATRA resistant UF-1 cell. They were investigated with regard to the correlation between the inherent or intrinsic cellular level of GSH and the apoptotic susceptibility of the cells to arsenic trioxide. We manifested, in two cell types, the inherently existed difference in intracellular GSH level reactive to the arsenic trioxide, and a positive correlation between the GSH level and their apoptotic sensitivity to arsenic trioxide. And it showed that arsenic trioxide could differentiate promyelocytic cancer cells to the cells possessed of dendritic cell surface markers. Unravelling the cause of the different susceptibility between leukemic cells and proving that promyelocyte could be differentiated to dendritic cells by arsenic trioxide will help not only to understand the mechanism underlying the complete remission of acute promyelocytic leukemia induced by arsenic trioxide, but also to expand its clinical usage.
An understanding of the structure and function of mammalian spermatozoa requires the iso-lation of these components. In this study, frozen-thawed bovine spermatozoa were treated by physical treatments (vortexing, 26 gauge needle, strained 26 gauge needles and freezing-thawing) or chemical treatments (trypsin, dithiothreitol, sodium dodecylsulfate and $\beta$-mercaptoethanoJ) to yield free heads and tails. The most effective treatment was repeated pumping of sperm suspension through a strained 26 gauge needle conneted to a syringe. Spermatozoa by this treatment were mainly broken at the junction of the head and the tail, resulting in 90-100% yields. Also, sperm head surface did not modify during strained 26 gauge needle treatment when either spermatozoa or sperm heads were incubated in 250${\mu}\textrm{g}$/ml of FITC-UEA 1 for 1 h at room temperature to detect the modification of sperm surface components. Other physical treatments were less efficient for the breakdown of spermatozoa. The effects of chemical treatments on bovine spermatozoa are not noticeable. Dissected sperm heads and tails should be fractional leading to nearly pure components by sucrose gradient centrifugation at 1,000 rpm for 15 min. The result suggest that the established method may be useful for the biochemical study of spermatozoal components, and the understanding of oocyte activation mechanism either by spermatozoal components during fertilization or microinjection of isolated components.
Crack-free dried gel monoliths of the composition $Li_2O1{\cdot}7Al_2O_3{\cdot}8.6SiO_2$ have been prepared as a precursor of transparent glass-ceramic by the hydrolysis and polycondensation of mixed metal alkoxides in solutions containing N,N-dimethylformamide as the drying control chemical additive, alcohols, and water. It was investigated that activation energy for gelation according to the variation of water concentration ranged from 13 to 14 kcal/mol. Only when the amount of water for gelation was 3 times higher than the stoichiometric amount, monolithic dry gels were successfully prepared after drying at $70{\sim}75^{\circ}C$ and at a rate of 0.1~0.3%/h. The specific surface area, the pore volume, the average pore diameters of dried gel at $180^{\circ}C$ were about $239.40m^2/g$, 0.001~0.03 mL/g, and $145.62{\AA}$, respectively. It showed that the dried monolithic gel had a porous body. The DTA curve had the first exothermic peak around $800^{\circ}C$ and the 2nd peak around $980^{\circ}C$, which may correspond to crystallization of the gel.
This aim of this study was to examine the immunomodulatory activities of crude polysaccharides from Perilla frutescens Britton var. acuta Kudo (PCP) in mouse bone marrow-derived dendritic cells (BMDC) and splenocytes. The immunomodulatory activity was determined by cell viability, nitric oxide (NO) production, cell surface marker expression (CD 80/86 and MHC class I/II), and cytokine production in BMDC, and cell viability, and cytokine production in splenocytes. Cell proliferation and cytokine production (tumor necrosis factor; TNF-${\alpha}$, interleukin (IL)-6, IL-$1{\beta}$, and IL-12) tested in BMDC were significantly increased by PCP treatment. Additionally, the cell surface markers (CD 80/86, MHC class I/II) were highly increased by PCP treatment. For cytokine production in splenocytes, PCP treatment significantly increased the production of Th 1 cytokines [IL-2 and interferon (IFN)-${\gamma}$], but not Th 2 cytokines (IL-4). Therefore, PCP can induce immune cell activation and is a potential candidate for the development of nutraceuticals to boost the immune system.

Objective: This study was to compare the characteristics between parthenogenetic mES (P-mES) cells and in vitro fertilization mES cells. Materials and Methods: Mouse oocytes were recovered from superovulated 4 wks hybrid F1 (C57BL/6xCBA/N) female mice. For parthenogenetic activation, oocytes were treated with 7% ethanol for 5 min and $5{\mu}g$/ml cytochalasin-B for 4 h. For IVF, oocytes were inseminated with epididymal sperm of hybrid F1 male mice ($1{times}10^6/ml$). IVF and parthenogenetic embryos were cultured in M16 medium for 4 days. Cell number count of blastocysts in those two groups was taken by differential labelling using propidium iodide (red) and bisbenzimide (blue). To establish ES cells, b1astocysts in IVF and parthenogenetic groups were treated by immunosurgery and recovered inner cell mass (ICM) cells were cultured in LIF added ES culture medium. To identify ES cells, the surface markers alkaline phosphatase, SSEA-1, 3,4 and Oct4 staining were examined in rep1ated ICM colonies. Chromosome numbers in P-mES and mES were checked. Also, in vitro differentiation potential of P-mES and mES was examined. Results: Although the cleavage rate (${\geq}$2-cell) was not different between IVF (76.3%) and parthenogenetic group (67.0%), in vitro development rate was significantly low in parthenogenetic group (24.0%) than IVF group (68.4%) (p<0.05). Cell number count of ICM and total cell in parthenogenetic b1astocysts ($9.6{\pm}3.1,\;35.1{\pm}5.2$) were signficantly lower than those of IVF blastocysts ($19.5{\pm}4.7,\;63.2{\pm}13.0$) (p<0.05). Through the serial treatment procedure such as immunosurgery, plating of ICM and colony formation, two ICM colonies in IVF group (mES, 10.0%) and three ICM colonies (P-mES, 42.9%) in parthenogenetic group were able to culture for extended duration (25 and 20 passages, respectively). Using surface markers, alkaline phosphatase, SSEA-l and Oct4 in P-mES and mES colony were positively stained. The number of chromosome was normal in ES colony from two groups. Also, in vitro neural and cardiac cell differentiation derived from mES or P-mES cells was confirmed. Conclusion: This study suggested that P-mES cells can be successfully established and that those cell lines have similar characteristics to mES cells.
Sodium borohydride, $NaBH_4$, shows a number of advantages as hydrogen source for portable proton exchange membrane fuel cells (PEMFCs). Properties of $NaBH_4$ hydrolysis reaction using activated carbon supported Co-B/C, Co-P-B/C catalyst were studied. BET surface area of catalyst, yield of hydrogen, effect of $NaBH_4$ concentration and durability of catalyst were measured. The BET surface area of carbon supported catalyst was over $500m^2/g$ and this value was 2~3 times higher than that of unsupported catalyst. Hydrogen generation of activated carbon supported catalyst was more stable than that of unsupported catalyst. The activation energy of Co-P-B/C catalyst was 59.4 kJ/mol in 20 wt% $NaBH_4$ and 14% lower than that of Co-P-B/FeCrAlloy catalyst. Catalyst loss on activated carbon supported catalyst was reduced to about 1/3~1/2 compared with unsupported catalyst, therefore durability was improved by supporting catalyst on activated carbon.
The electrical properties and defect states in ZnO substrates were studied during high-energy electron beam irradiations. 1 MeV and 2 MeV electron-beam with dose of $1{\times}10^{16}$ electrons/$cm^2$ were irradiated on Zn-surface of the sample. In the sample irradiated by 1 MeV, the leakage current was increased by electron-beam induced surface defects, while the enhancement of on/off property and the decrease of leakage current appeared in the 2 MeV irradiated sample. From the deep level transient spectroscopy measurements for these samples, it showed that the defect states with the activation energies of $E_c$-0.33 eV and $E_v$+0.8 eV are generated during the high energy electron-beam irradiation. Especially, it considered that the $E_c$-0.33 eV state related with O-vacancy affects to their electrical properties.
This study was performed to investigate the influence of fuel/oxygen ratio (F/O= 3.2, 3.0, 2.8) on the oxidation behavior of two kinds of (20wt%NiCr claded $\textrm{Cr}_{3}\textrm{C}_{2}$, and 7wt%NiCr mixed $\textrm{Cr}_{3}\textrm{C}_{2}$) composite powder with different manufacturing method. The results show that the oxidation behavior between the 20wt% NiCr claded $\textrm{Cr}_{3}\textrm{C}_{2}$ and 7wt% NiCr mixed $\textrm{Cr}_{3}\textrm{C}_{2}$ coating was widely different. The surface morphology of the coating composed of 7wt% NiCr mixed $\textrm{Cr}_{3}\textrm{C}_{2}$ was changed to porous with F/O ratio by the aggressive evolution of gas phases($\textrm{CO}_2$, CO and $\textrm{CrO}_3$) and the oxide cluster composed of Ni and Cr were grown after oxidation at $1000^{\circ}C$ for 50 hours. But the surface morphology of the coating composed of 20wt% NiCr claded $\textrm{Cr}_{3}\textrm{C}_{2}$ was not changed to porous after oxidation at $1000^{\circ}C$ for 50 hours. Therefore, the reason for high oxidation rate is due to activation of $\textrm{Cr}_{3}\textrm{C}_{2}$ to oxidation by entrapped oxygen gases within coating layer, and to closely relate with the decomposition of $\textrm{Cr}_{3}\textrm{C}_{2}$ to $\textrm{Cr}_{7}\textrm{C}_{3}$ phase. Accordingly, On the evidence of these results, the study about the oxidation behavoir of the HVOF sprayed $\textrm{Cr}_{3}\textrm{C}_{2}$ coating depending on hydrogen flow rate must be done.
Conventional nickel-based catalyst processes used for dry reforming reactions have high activation temperatures and problems such as carbon deposition and metal sintering on the active sites of the catalyst surface. In this study, the characteristics of butane dry reforming reaction were investigated by using DBD plasma combined with catalytic process and compared with existing catalyst alone process. The physical and chemical properties of the catalysts were investigated using a surface area & pore size analyzer, XRD, SEM and TEM. Using $10%Ni/{\gamma}-Al_2O_3$ at $580^{\circ}C$, in the case of the catalyst+plasma process, the conversion of carbon dioxide and butane were improved by about 30% than catalyst alone process. When the catalyst+plasma process, the conversion of carbon dioxide and butane and the hydrogen production concentration are enhanced by the influence of various active species generated by the plasma. In addition, it was found that the particle size of the catalyst is decreased by the plasma in the reaction process, and the degree of dispersion of the catalyst is increased to improve the efficiency.

This study aimed to identify the effect of the push-up plus exercise with sling on the muscle activity and lung function in adults with forward head posture over a period of 8 weeks. A total of 16 adults with forward head posture were recruited. The participants were divided into two groups : control (n-=8) and experimental (n=8). From July 6 until Aug 28, 2015, the participants were instructed to perform the knee push-up plus exercise and knee push-up plus using sling exercise three times a week to begin with and progressing to three sets of push-ups on each occasion after 8 weeks. The participants in both groups showed an increase in their craniovertebral angle (both groups p<0.001) and a decrease in their crania rotation angle (control p<0.05, experimental p<0.001). The participants in both groups showed a decrease in their trapezius activity (both groups p<0.001), an increase in their serratus anterior and lower trapezius muscle activity (both groups p<0.001), (and an increase in their) lung function (control group p<0.01, experimental group p<0.001). These findings show that push-up plus exercise improves posture and increases activation of lung function, while the push-up plus exercise in the sling is more effective in improving the forward head posture. The push-up plus exercise in the sling was more effective in improving the forward head posture, but there was no significant difference in the lung function between the two groups. Therefore, the push-up plus exercise in the sling is recommended as an exercise that is effective for the improvement of the posture and lung function of persons with forward head posture.
