Characteristics of electric field in the liquid metal ion source with a suppressor

  • The liquid metal ion sources(LMIS) in FIB system have many advantages of high current density, high brightness and low ion energy spread. Most FIB systems use LMIS because the ion beam spot size of LMIS is smaller than other ion sources. LMIS is basically emitted by an extractor but the new electrode called the suppressor is able to control the emission current. We investigated characteristics LMIS with a suppressor, the function of the suppressor in LMIS, the change of the electric field by the suppressor and the advantages of using the suppressor. The characteristics of the threshold voltage and current-voltage (I-V) were observed under the varying extracting voltage with floated suppressor voltage, and under the varying suppressor voltages with fixed extractor voltage. We also simulated LMIS with the suppressor through CST(Computer Simulation Technology). The emission current increases as the suppressor voltage decreases because the suppressor voltage which restrains the electric field goes down, The threshold voltage increases as the suppressor voltage increases. We can explain characteristics and functions of LMIS with a suppressor using the electric field.

LMIS basically consists of a emitter(needle, anode), a reservoir(gallium) and a extractor(cathode). But several LMIS have new electrode called the suppressor. We investigated characteristics of LMIS with a suppressor. The characteristics of the threshold voltage and current-voltage (I-V) were observed under the varying extracting voltage with floated suppressor voltage, and under the varying suppressor voltages with fixed extractor voltage. We also simulated LMIS with the suppressor through CST(Computer Simulation Technology). We can explain characteristics of LMIS with a suppressor using the electric field.

두경부 편평상피세포암에서 종양억제유전자들의 변이 (Alteration of Multiple Tumor Suppressor Genes in Head and Neck Squamous Cell Carcinoma)

  • Objectives: Head and neck squamous cell carcinoma (HNSCC) is the most common head and neck malignant tumor. The molecular genetic changes involving both oncogenes and tumor suppressor genes are known to be involved in head and neck squamous cell carcinogenesis, but the roles of the known tumor suppressor genes in carcinogenesis are not fully elucidated. The objectives of this study are to demonstrate the genetic alterations including the loss of heterozygosity (LOH) , amplification, and microsatellite instability of known tumor suppressor genes in HNSCC and to evaluate the relationship between genetic alterations of tumor suppressor genes and clinicopathologic features. Materials and Methods: Genetic alterations of 10 micro satellite markers of the 6 known tumor suppressor genes (APC, EXT1, DPC4, p16, FHIT, and PTEN) were analysed by DNA-PCR in paraffin-embedded histologically confirmed HNSCC specimens. Results: The genetic alterations of tumor suppressor genes were found frequently. Among the genetic alterations, LOH was most frequently found one. LOH was found frequently in APC (45.4%), EXT1 (36.4%), DPC4 (54.5%), and p16 (50%), but not found in FHIT. Also, the author found that abnormalities of APC gene was related to cervical lymph node metastasis and recurrence and that abnormalities of EXT1 gene were coexisted with those of APC gene or DPC4 gene. But these coexistences had no correlation with clinical features. Conclusion: These results suggested that APC, EXT1, p16, and DPC4 genes might play important roles and multiple tumor suppressor genes may participate dependently or independently in the carcinogenesis of HNSCC. These results also suggested that APC gene might relate to prognosis.

모세관 컬럼 이온 크로마토그래피를 위한 Conductivity Cell과 Suppressor의 개발 (Development of Conductivity Cell and Suppressor for Capillary Column Ion Chromatography)

  • 적은 양의 시료를 분석할 수 있는 마이크로 컬럼 이온 크로마토그래피용 전도도 셀과 억압컬럼을 개발하였다. 안지름이 작은 모세관 컬럼을 사용하는 경우 이동상의 유속은 보통 $5{\sim}20{\mu}L/min$ 정도 된다. 따라서 이 경우 일반적인 전도도 셀은 사용할 수가 없기에 내부 부피가 작은 모세관 컬럼용 전도도 셀과 억압컬럼을 제작하였다. 전도도 셀은 두 개의 백금 피하주사 바늘을 안지름이 0.010 mm인 튜브 속에 넣어 간격이 $2{\mu}m$ 되게 만들었고, 억압컬럼은 Nafion 튜브를 이용하여 막-억압컬럼 형태로 개발하였다. 이 전도도 셀과 억압컬럼을 이용하여 실제 음이온들(플루오르화물, 아질산염, 질산염, 염소산염)을 분석한 결과 재현성 있고 양호한 크로마토그램을 얻었다.

Naturally occurring reoviruses for human cancer therapy

  • Naturally occurring reoviruses are live replication-proficient viruses that specifically infect human cancer cells while sparing their normal counterpart. Since the discovery of reoviruses in 1950s, they have shown various degrees of safety and efficacy in pre-clinical or clinical applications for human anti-cancer therapeutics. I have recently discovered that cellular tumor suppressor genes are also important in determining reoviral tropism. Carcinogenesis is a multi-step process involving the accumulation of both oncogene and tumor suppressor gene abnormalities. Reoviruses can exploit abnormal cellular tumor suppressor signaling for their oncolytic specificity and efficacy. Many tumor suppressor genes such as p53, ataxia telangiectasia mutated (ATM), and retinoblastoma associated (RB) are known to play important roles in genomic fidelity/maintenance. Thus, a tumor suppressor gene abnormality could affect host genomic integrity and likely disrupt intact antiviral networks due to the accumulation of genetic defects which in turn could result in oncolytic reovirus susceptibility. This review outlines the discovery of oncolytic reovirus strains, recent progresses in elucidating the molecular connection between oncogene/tumor suppressor gene abnormalities and reoviral oncotropism, and their clinical implications. Future directions in the utility of reovirus virotherapy is also proposed in this review. [BMB Reports 2015; 48(8): 454-460]

  • In the firing operation, muzzle jumping phenomenon was investigated, and a method of reducing muzzle jump was suggested. The muzzle jumping force was determined from the torque acting on the rifle due to a recoil force. For the purpose of reducing a muzzle jump, the gas flowing through the M16A1 flash suppressor was considered. A flash suppressor, improved in reducing a muzzle imp, was modeled and tested. The experiment shows that muzzle jump has quantitative relation to shape of a flash suppressor, and can be reduced by the improved modeling of a flash suppressor.

  • Introduction: We investigated frameshift suppressor tRNAs previously reported to use five-base anticodon-codon interactions in order to provide a collection of frameshift suppressor tRNAs to the synthetic biology community and to develop modular frameshift suppressor logic devices for use in synthetic biology applications. Results and Discussion: We adapted eleven previously described frameshift suppressor tRNAs to the BioBrick cloning format, and built three genetic logic circuits to detect frameshift suppression. The three circuits employed three different mechanisms: direct frameshift suppression of reporter gene mutations, frameshift suppression leading to positive feedback via quorum sensing, and enzymatic amplification of frameshift suppression signals. In the course of testing frameshift suppressor logic, we uncovered unexpected behavior in the frameshift suppressor tRNAs. The results led us to posit a four-base binding hypothesis for the frameshift suppressor tRNA interactions with mRNA as an alternative to the published five-base binding model. Conclusion and Prospects: The published five-base anticodon/codon rule explained only 17 of the 58 frameshift suppression experiments we conducted. Our deduced four-base binding rule successfully explained 56 out of our 58 frameshift suppression results. In the process of applying biological knowledge about frameshift suppressor tRNAs to the engineering application of frameshift suppressor logic, we discovered new biological knowledge. This knowledge leads to a redesign of the original engineering application and encourages new ones. Our study reinforces the concept that synthetic biology is often a winding path from science to engineering and back again; scientific investigations spark engineering applications, the implementation of which suggests new scientific investigations.

  • Saccharomjyces cerevisiae에서 KEM1 유전자는 세포분열시 microtubules과 spindle pole body 구조체의 새로운 기능에 관여하는 것으로 알려져 있다. KEM1과 유사하거나 연관이 있는 기능을 갖는 새로운 유전자들을 찾는 목적으로 kem1 돌연변이의 high copy suppressor 유전자, ROK1, 를 찾아냈다. ROK1은 high copy 플라스미드에 클로닝되었을 때 kem1을 suppression 하고, low copy 플라스미드에서는 suppression 하지 않는다. kem1 돌연변이의 benomyl에 대한 민감성과 Kar enhancing 표현서을 동시에 suppression 하는 두 개의 클론을 분리하였으며, 제한요소로 분석했을 때 9.0kb의 insert 를 지닌 동일한 클론이었다. 이 suppressor 유전자 ROK1의 제한지도를 작성하였고, 그 결과 KEM1 이 아닌 다른 유전자인 것으로 나타났다. Suncloning 실험으로 ROK1은 적어도 3.0kb의 기능부위를 갖음을 확인했다.

대장균에서 비천연 아미노산의 위치특이적 삽입을 위한 Amber Suppressor tRNA와 Aminoacyl-tRNA Synthetase의 Amber Suppression 활성측정시스템 개발 (Establishment of an In Vivo Report System for the Evaluation of Amber Suppression Activity in Escherichia coli)

  • 대장균에서 비천연 아미노산을 단백질 생합성시 특정 위치에 삽입하는 방법의 하나로 amber suppressor tRNA와 여기에 비천연 아미노산을 특이적으로 결합할 수 있는 변형된 aminoacyl-tRNA synthetase 쌍을 이용한다. 이러한 기술의 개발을 위해 필요한 여러 요소 중 하나는 이러한 시스템이 대장균에서 얼마나 잘 작동하는지를 확인할 수 있는 in vivo 보고시스템을 설정하는 것이다. 본 논문에서는 $\beta$-galactosidase 유전자의 N-말단에 amber 코돈을 삽입한 보고유전자를 제작하였으며, 이를 대장균(DH10B)의 chromosomal DNA에 삽입하여 DH10B(Tn:lacZam) 균주를 개발하였다. Genomic PCR과 Southern blot 분석을 통하여 lacZ amber 유전자가 대장균의 염색체에 삽입된 것을 확인하였으며, DH10B(Tn:lacZam)은 amber suppression을 유도할 수 있는 벡터가 형질 전환될 경우만 $\beta$-galactosidase 활성을 나타냈다. DH10B(Tn:lacZam)에 효모균의 amber suppressor $tRNA^{Tyr}$와 Tyrosyl-tRNA synthetase 쌍을 동시에 발현하는 벡터를 형질전환하였을 때, amber suppression에 의해서 $\beta$-galactosidase 활성이 나타났다. 하지만 이 활성은 대장균의 amber suppressor $tRNA^{Gln}$를 발현하는 pSupE2를 형질전환하였을 때와 비교 하여 매우 낮은 $\beta$-galactosidase 활성을 나타냈다. 이러한 결과는 DH10B(Tn:lacZam) 균주가 $\beta$-galactosidase 활성을 통하여 정성 및 정량적으로 in vivo amber suppression 활성을 비교 분석할 수 있는 특성을 가졌음을 나타낸다.

비천연 아미노산의 위치특이적 단백질 삽입을 위한 Amino Acyl-tRNA Synthetase 선별시스템 개발 (Establishment of a Selection System for the Site-Specific Incorporation of Unnatural Amino Acids into Protein)

  • 생명체에서 비천연 아미노산을 단백질의 특정 위치에 삽입하는 방법으로 orthogonal suppressor tRNA와 여기에 비천연 아미노산을 특이적으로 결합시킬 수 있는 유전자 변형된 aminoacyl-tRNA synthetase (ARS)가 활용되고 있다. 이 기술개발을 위해서는 돌연변이를 유발한 ARS library로부터 비천연 아미노산만을 특이적으로 결합시킬 수 있는 변형된 ARS를 탐색하기 위한 선별시스템이 필요하다. 본 논문에서는 대장균에서 작용하는 2단계로 구성된 새로운 선별시스템을 개발하였다. 먼저 양성선별 시스템은 27번 잔기를 amber 코돈으로 치환한 Chloramphenicol acetyl transferase 유전자로 구성되어 있으며, 이유전자의 amber suppression에 의해 chloramphenicol 배지에서 생존함에 따라 활성을 나타내는 ARS를 최고 $9.0{\times}10^5$배로 농축할 수 있었다. 반면 음성선별 시스템은 대장균의 Topoisomerase II의 기능을 억제하는 단백질을 암호화하는 control of cell death B (ccdB) 유전자의 N-말단 앞에 3개의 amber 코돈을 삽입하여 제작하였다. 이 음성선별 시스템을 가진 대장균에 orthogonal pair인 Saccharomyces cerevisiae tyrosyl-tRNA synthetase (Scc TyrRS)와 amber suppressor tRNA를 형질전환하면 amber suppression으로 CcdB가 발현되어 대장균의 성장이 억제되는 것을 확인하였으며, 천연 아미노산에 대한 특이성을 가진 ARS를 효과적으로 제거하는 것을 관찰하였다. 따라서, 양성선별 및 음성선별 시스템을 순차적으로 거침으로써 무작위적으로 아미노산에 대한 특이성을 변형시킨 ARS 라이브러리로부터 비천연 아미노산을 suppressor tRNA에 특이적으로 결합하는 유전자 변형 ARS를 탐색하는데 유용하게 사용될 수 있을 것이다.
