Bioassay-guided fractionation of methanol extract of Reynoutria sachalinensis flower using DPPH assay has led to the isolation of three anthraquinones and three flavonoids. Their structures were identified as emodin (1), emodin-8-O-$\beta$-D-glucopyranoside (2), physcion-8-O-$\beta$-Dglucopyranoside (3), quercetin-3-O-$\alpha$-L-arabinofuranoside (4), quercetin-3-O-$\beta$-D-galactopyranoside (5), and quercetin-3-O-$\beta$-D-glucuronopyranoside (6) by comparing their physicochemical and spectral data with those published in literatures. All isolated compounds were evaluated for antioxidant activities with free radical 1, 1-diphenyl-2-picrylhydrazyl (DPPH) scavenging, superoxide radical scavenging and $Cu^{2+}$-mediated low density lipoprotein (LDL) oxidation assay. The results demonstrated that three flavonoids, 4, 5, and 6 had remarkable antioxidant activities with the $IC_{50}$ values of 64.3, 54.7, and 46.2${\mu}M$ (DPPH scavenging), the $IC_{50}$ values of 6.0, 6.7, and $4.4{\mu}M$ (superoxide radical scavenging) and the $IC_{50}$ values of 3.8, 3.2, and 5.4${\mu}M$ against LDL oxidation, respectively.
Objectives : This studies were carried out to evaluate the effects on anti-oxidation activities in different parts as trunk bark and root bark of Ulmus devidiana Planchon. var. japonica Nakai and Hemipteleae davidii (Hance) Planchon. Methods : For evaluation of antioxidative effects, scavenging activity on superoxide anion radical and DPPH radical were measured. Also, inhibitory activity on LDL oxidation and linoleic acid peroxidation were measured in each samples. In the in vivo test, inhibitory activity on TBARS production, GSH contents in rat liver were measured. SOD, Catalase, GSH-px and ALDH activity were analysed in ethanol extracts of Ulmi Radicis Cortex. Result : 1. Scavenging activity on superoxide anion radical was higher in water extract than in ethanol extract even in low concentration of 50ppm as over 90%. 2. There was no difference between water extract and ethanol extract in the scavenging activity on DPPH radical but, Ulmi Raclicis Cortex and Ulmi Trunci Cortex showed high effect even in low concentration of 10ppm. 3. GSH reduction was prevented and antioxidative activity such as Mn-SOD, GSH-px in the rat liver recovered in the treatment of ethanol extracts of Ulmi Radicis Cortex. Conclusion : Ulmi Radicis Cortex recorded as Ulmi Trunci Cortex in official regulation book. But, it was known that Ulmi Radicis Cortex was more effective than Ulmi Trunci Cortex in most physical activities.
Hovenia dulcis Thunb fruits were successively extracted with hot water, water, methanol, ethyl acetate, and chloroform. The crude extracts were investigated for potential antioxidant by measuring scavenging against DPPH free radicals, reducing power, superoxide radicals, and protection of protein damage and cultured cells from a lethal dose of hydrogen peroxide ($H_2O_2$). In all chemical assays used, the hot water extract of H. dulcis fruits, which contained $61.14{\pm}2.57$ (Tannic acid mg/g extract, n=3) of total phenolic compounds contents exhibited highest activity in in vitro models of DPPH free radical scavenging activity, reducing power assay, superoxide radical scavenging activity and protection of protein damage. In addition, the hot water extract protected cultured RAW 264.7 macrophages from a lethal dose of $H_2O_2$ and reduced reactive oxygen species level in RAW 264.7 cells.
As a part of an ongoing search for natural plants with antioxidant compounds by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH), a total extract of the aerial parts of Bidens frondosa L. (Compositae) was found to show potent antioxidant activity. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of five compounds, quercetin-3-O-${\beta}$-D-glucopyranoside (1), luteolin-7-O-${\beta}$-D-glucopyranoside (2), 7,8,3',4'-tertrahydroxy-flavanone (3), okanin-4-O-${\beta}$-D-glucopyranoside (4), and okanin (5). Their structures were elucidated by spectroscopic studies. Compounds 3-5 were isolated for the first time from this plant. Among them, compounds 3 and 5 showed the significant radical scavenging effects on DPPH, and compounds 3 and 5 also showed the potent riboflavin and xanthine originated superoxide quenching activities.
This study investigated the antioxidant effects of complex extracts from pine needles (PN), green tea (GT), and sea buckthorn leaves (SL). Measurement of total polyphenol and flavonoid content, DPPH radical scavenging activity (DPPH), ABTS+ radical scavenging activity (ABTS), and reducing power (RP), showed that SL extract (SE) had the highest values. The superoxide radical scavenging activity (SSA) and nitrite scavenging activity (NSA) were the highest in the GT extract (GE). In most experiments, the PN extract (PE) showed higher activity in complex extracts than in single extracts. The combination of GT and SL led to a higher activity than that exhibited by GT (DPPH and ABTS at 300 ppm, RP at 100 and 500 ppm) and SL (SSA at 100 ppm and NSA at 300 ppm) alone. These results suggest that the combination of PE, GE, and SE may be a useful functional food material in the food industry.

To examine the free radical scavenging activities from buckwheat, Suwon No. 5 was extracted with various solvents. The seed of Suwon No. 5 had a high contents of carbohydrate, but the vegetable of that had a 31.4% of protein, 28.6% of ash and 25.7% carbohydrate as moisture free basis. Phenols contents of vegetable extracts had higher than those of seed extracts. Acetone extract of vegetable showed the greatest electron donating ability. Butanol and acetone extracts of vegetable showed high inhibitory effect on lipid peroxidation. Acetone extract of vegetable has also excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effects of extracts on xanthine oxidase were measured. Acetone extract had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was 2.2$\mu\textrm{g}$.
An antioxidant peptide derived from Pinctada fucata meat using an Alcalase2.4L enzymatic hydrolysis method (named AOP) and identified by LC-TOF-MS has promising clinical potential for generating cosmetic products that protect skin from sunshine. To date, there have been few published studies investigating the structure-activity relationship in these peptides. To prepare antioxidant peptides better and improve their stability, the design and expression of an antioxidant peptide from Pinctada fucata (named DSAOP) was studied. The peptide contains a common precursor of an expression vector containing an ${\alpha}$-helix tandemly linked according to the BamHI restriction sites. The DNA fragments encoding DSAOP were synthesized and subcloned into the expression vector pET-30a (+), and the peptide was expressed mostly as soluble protein in recombinant Escherichia coli. Meanwhile, the DPPH radical scavenging activity, superoxide radical scavenging activity, and hydroxyl radical scavenging activity of DSAOP $IC_{50}$ values were $0.136{\pm}0.006$, $0.625{\pm}0.025$, and $0.306{\pm}0.015mg/ml$, respectively, with 2-fold higher DPPH radical scavenging activity compared with chemosynthesized AOP (p < 0.05), as well as higher superoxide radical scavenging activity compared with natural AOP (p < 0.05). This preparation method was at the international advanced level. Furthermore, pilot-scale production results showed that DSAOP was expressed successfully in fermenter cultures, which indicated that the design strategy and expression methods would be useful for obtaining substantial amounts of stable peptides at low costs. These results showed that DSAOP produced with recombinant Escherichia coli could be useful in cosmetic skin care products, health foods, and pharmaceuticals.

This study focused on the evaluation of the antioxidative effects of lotus seeds from golden colored flowers. The lotus seeds were extracted with ethanol and then fractionated into 4 fractions, ethyl acetate (EtOAc), n-butanol, methylene chloride, and n-hexane. The comparison of antioxidative activities of the extract and fractions from the lotus seeds was carried out using an in vitro radical scavenging model and the total phenol content was analyzed. Of the tested extracts and fractions, the EtOAc fraction of the lotus seeds showed the strongest 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity with 96.24% at a concentration of $100{\mu}g/mL$. In addition, the hydroxyl radical scavenging activity of the lotus seed EtOAc fraction was also increased in a concentration dependent manner with the concentrations tested ranging from 5 to $100{\mu}g/mL$. Moreover, the EtOAc fraction showed the highest scavenging activity for nitric oxide and superoxide anion radicals. In particular, of all the extracts and fractions, the EtOAc fraction showed highest contents of total phenols. These results indicate that lotus seeds have potential as an antioxidative agent against oxidative stress involving reactive oxygen species. Furthermore, the EtOAc fraction of lotus seeds includes promising oxidative stress-protective compounds.
The EtOAc-soluble fraction from a methanolic extract of Hovenia dulcis Thunb. exhibited neuroprotective activity against glutamate-induced neurotoxicity in mouse hippocampal HT22 cells. The neuroprotective activity-guided isolation resulted in 8 phenolic compounds (1-8), such as vanillic acid (1), ferulic acid (2), 3,5-dihydroxystilbene (3), (+)-aromadendrin (4), methyl vanillate (5), (-)-catechin (6), 2,3,4-trihydrobenzoic acid (7), and (+)-afzelechin (8). Among these, compounds 6 and 8 had a neuroprotective effect on the glutamate-induced neurotoxicity in HT22 cells. Furthermore, compound 6 had a DPPH free radical scavenging effect with an $IC_{50}$ value of $57.7{\mu}M$, and a superoxide anion radical scavenging effect with an $IC_{50}$ value of $8.0{\mu}M$. Both compounds 6 and 8 had ABTS cation radical scavenging effects with $IC_{50}$ values of $7.8{\mu}M\;and\;23.7${\mu}M$, respectively. These results suggest that compounds 6 and 8 could be neuroprotectants owing to their free radical scavenging activities.

The purpose of this study is to investigate the antioxidative activity and digestive enzyme inhibition of grape seed extract (GSE). The GSE was tested for its effect on various antioxidative potentials (scavenging activities of DPPH radical, superoxide anion radical and hydroxyl radical) and inhibitory effect of various digestive enzymes (trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\beta$-glucosidase and lipase). DPPH radical scavenging activity ($SC_{50}$, 50% scavenging concentration) of GSE was 4.76${\pm}$0.27 ppm while those of positive controls (EGCG and vitamin C) were 2.22${\pm}$0.12 ppm and 9.50${\pm}$0.72 ppm, respectively. $SC_{50}$ value of GSE against superoxide anion radical and hydroxyl radical were 3.82${\pm}$0.07 ppm and 803.23${\pm}$27.16 ppm, respectively. In addition, $IC_{50}$ values of GSE against trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\alpha$-glucosidase and lipase were 2.17${\pm}$0.59 ppm, 7.46${\pm}$1.25 ppm, 18.25${\pm}$3.54 ppm, 12.30${\pm}$1.12 ppm, and 653.23${\pm}$79.34 ppm, respectively. These results suggest that GSE may be useful for the prevention or treatment of obesity.
