The purpose of this study was to examined the antioxidant activities by water and 70% ethanol extract from durian (Durio zibethinus.) seed, sarcocarp and peel. Durian extract were studied for reducing sugar content, polyphenol content, superoxide dismutase (SOD) like activity, electron donating ability, nitrite scavenging ability, flavonoid content, hydroxy radical scavenging activity. Reducing sugar content were increased peel > sarcocarp > seed. Total polyphenol, flavonoid content, DPPH radical scavenging ability and SOD like activity were increased seed > peel > sarcocarp. Total polyphenol content was relatively high as $21.90{\pm}0.50mg/g$ in the ethanol extract of the seed. DPPH radical scavenging ability was relatively high as $62.08{\pm}2.63%$ in the water extract of the seed. Nitrite scavenging ability was no significant difference. Hydroxy radical scavenging activity was increased seed > peel > sarcocarp, was relatively high as $58.27{\pm}1.13%$ in the water extract of the seed.

Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical increases with age, the reactive oxygen species(ROS) such as hydrogen peroxide($H_2O_2$), nitric oxide(NO). Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. In this study, we have conducted to investigate the biological activities of Hominis Placenta Herbal Acupuncture by measuring total polyphenol content, DPPH radical scavenging, ABTS radical scavenging, Superoxide dismutase(SOD)-like activity, Nitrite scavenging ability in vitro. The total polyphenol contents of Hominis Placenta Herbal Acupuncture was $24.6m{\ell}/m{\ell}$. Elctron donation ability on DPPH was 49.4%. The 2,2'-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) was 50.01%, similar 10 the DPPH free radical scavenging. The superoxide dismutase (SOD)-like activities of hominis placenta herbal acupuncture was 50.876%. The nitrite scavenging abilities at pH 1.5, pH 3.0, pH 6.0 were 52.8%, 29.4%, 15.4%, respectively; these abilities decreased as pH increased. We conclude that Hominis Placenta Herbal Acupuncture may be useful as potential sources of antioxidant.
To evaluate an effect of liver damage on the dermal oxygen free radical metabolizing enzyme activities, the $CCl_4$ (0.1 ml/ 100 g body wt., 50% $CCl_4$ in olive oil) was intraperitoneally given to the rats every other day for 2 weeks. Based on the histopathological findings, liver weight (%), serum alanine aminotransferase, xanthine oxidase activities and hepatic lipid peroxide contents, the animals were induced severe liver damage. In the present liver damaged animal, all the activities of dermal scavenging enzymes such as superoxide dismutase, catalase and glutathione peroxidase were significantly decreased compared with central. And under the cytochemical electron microscopy the more coarse granules of cerrous perhydroxide were found compared with the control. In conclusion, the $CCl_4$-induced liver damage may influence upon the activities of some dermal oxygen free radical scavenging enzymes.
Spermidine is a ubiquitous polycation that is synthesized from putrescine, which serves as a precursor of spermine. In recent years, spermidine was found to be a polyamine that plays an important role in longevity. Reactive oxygen species (ROS) such as hydroxyl radical, superoxide and hydrogen peroxide have been shown to be involved in various pathogenic processes as well as aging. The direct scavenging effect of spermidine on DPPH radical, $H_2O_2$ and hydroxyl radical, and its protective effect against DNA oxidation related to oxidative stress were evaluated in vitro. It was observed that spermidine exhibits scavenging activities on DPPH radical and H2O2 above 500 ${\mu}M$. Spermidine was especially effective in exerting a scavenging activity on hydroxyl radical. In addition, spermidine at 1000 ${\mu}M$ showed a clear protective effect against DNA oxidation. Furthermore, the expression level of anti-oxidant enzymes such as superoxide dismutase in humam dermal fibroblasts increased in the presence of spermidine compared with blank group. These results suggest that spermidine can be used as an antioxidant to prevent ROS-related diseases including inflammation, cancer and aging.
This study was conducted to compare the antioxidant, anticytotoxic, and anti-inflammatory properties of Euphorbia maculata ethanol extract with those of E. supina ethanol extract. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical and superoxide scavenging activities of E. maculata at $50{\mu}g/mL$ were $38.3{\pm}3.7$ and $21.5{\pm}1.2%$, respectively, whereas those of E. supina at the same concentration were $109.4{\pm}0.9$ and $59.5{\pm}4.8%$, respectively. Oxygen radical absorbance capacities of E. maculata and E. supina at $10{\mu}g/mL$ were $14.70{\pm}0.63$ and $26.17{\pm}1.36nmol/mL$ Trolox, respectively. Cupric reducing antioxidant capacities of E. maculata and E. supina at $10{\mu}g/mL$ were $10.22{\pm}0.97$ and $62.99{\pm}5.28nmol/mL$ Trolox, respectively. Total phenolic contents of E. maculata and E. supina at $50{\mu}g/mL$ were $29.03{\pm}0.14$ and $87.89{\pm}0.20nmol/mL$ gallic acid, respectively. E. maculata and E. supina were reported to prevent supercoiled DNA breakage induced by peroxyl and hydroxyl radicals in a concentration-dependent manner, where protection against the supercoiled DNA breakage provided by E. supina was greater than that provided by E. maculata. E. maculata and E. supina at $100{\mu}g/mL$ inhibited tert-butyl hydroperoxide-induced cytotoxicity in HepG2 cells by $49.4{\pm}4.3$ and $87.3{\pm}4.5%$, respectively. E. maculata and E. supina at $500{\mu}g/mL$ inhibited lipopolysaccharide-induced nitric oxide production in RAW 264.7 cells by $63.1{\pm}7.0$ and $85.2{\pm}1.6%$, respectively. The antioxidant capacities including DPPH radical scavenging, superoxide scavenging, oxygen radical absorbance, and cupric reducing antioxidant activity were found to be highly correlated with total phenolic content (0.896 < r < 0.983, p < 0.01) and anticytotoxic activities (0.915 < r < 0.960, p < 0.01). However, the superoxide scavenging activity was not significantly correlated (r = 0.604, p > 0.05) with the anti-inflammatory activity. Thus, these findings demonstrated that the radical scavenging, anticytotoxic, and anti-inflammatory capacities of E. supina were more potent than those of E. maculata. Further studies are needed to elucidate the properties of polyphenolic constituents in E. supina responsible for these effects and the underlying mechanisms.
In this study, we investigated the free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\cdot}OH$) and superoxide anion (${O_2}^-$)] scavenging activity of MeOH extract and 3 fractions of Citrus junos. Of the tested fractions, the BuOH fraction showed the strongest DPPH scavenging activity, showing the $IC_{50}$ values of 63.4 mg/mL. Therefore, we continuously carried out DPPH, ${\cdot}OH$ and ${O_2}^-$ scavenging activity tests of BuOH fraction of Citrus junos. The BuOH fraction of Citrus junos inhibited DPPH radical to 97.5% at a concentration of 1000 mg/mL and the scavenging activities were increased concentration-dependently. In addition, BuOH fraction from Citrus junos also scavenged ${\cdot}OH$ in a concentration dependent manner from 5 to 1000 mg/mL. Furthermore, BuOH fraction showed about 56% ${O_2}^-$-scavengimg activity at 25 mg/mL concentration but, the scavenging activities were not enhanced in a dose dependent manner. The present results suggest that BuOH fraction of Citrus junos would have the protective potential from oxidative stress induced by free radicals.

The antioxidant activities of ethanolic extracts obtained from medicinal plants (Scutellaria baicalensis Georgi, Acanthopanax sessiliflorum Seeman, Pueraria lobata Ohwi, Portulaca oleracea Linne, Crataegus pinnatifida Bunge var. typica Schneider, Euonymus alatus Apterus, Hovenia dulcis Thunberg, Prunus yedoensis Matsumura, Albizzia julibrissin Durazz., Chrysanthemum indicum Linne) were evaluated for total phenolic content, total flavonoid content, DPPH radicals, nitrites, $Superoxide^-$ radicals, $ABTS^+$ radical scavenging activity, and reducing power. Antioxidant capacities were the highest in Prunus yedoensis Matsumura for DPPH radical scavenging activity ($IC_{50}$$5.39{\mu}g/mL$), reducing power (2.72, $A_{700}$), and nitrite scavenging activity ($IC_{50}$$167.94{\mu}g/mL$). Hovenia dulcis Thunberg and Acanthopanax sessiliflorum Seeman were effective for their nitrite scavenging activities (over 90% at 1 mg/mL). The $Superoxide^-$ radical scavenging activity of Prunus yedoensis Matsumura ($IC_{50}$$43.39{\mu}g/mL$) was stronger than tannic acid ($IC_{50}$$46.51{\mu}g/mL$). Five samples (Prunus yedoensis Matsumura, Acanthopanax sessiliflorum Seeman, Hovenia dulcis Thunberg, Crataegus pinnatifida Bunge var. typica Schneider, Scutellaria baicalensis Georgi) were effective for their $ABTS^+$ radical scavenging activity (more than 90% at 0.5 mg/mL). These results suggest that the ethanolic extracts of Prunus yedoensis Matsumura could be used as a functional ingredient in food products.
The antioxidant activities of water ($H_2O$) and ethanol (EtOH) extracts from hamcho (Salicornia herbacea L.) juice and cake prepared by enzymatic treatments were evaluated by in vitro assays against DPPH, superoxide, and hydroxyl radicals. Among the $H_2O$ and EtOH extracts from five different carbohydrases treated, the EtOH extract from viscozyme-treated hamcho cake had higher yield and phenolic content, and exhibited the strongest radical scavenging activity against DPPH ($IC_{50}=186.91\;{\mu}g/mL$), superoxide ($IC_{50}=87.54\;{\mu}g/mL$), and hydroxyl radicals ($IC_{50}=367.07\;{\mu}g/mL$). Antioxidant assay-guided fractionation and purification of the EtOH extract led to isolation and identification of five phenolic compounds, procatechuic, ferulic and caffeic acids, quercetin, and isorhamnetin. Most of these phenolic compounds exhibited considerable DPPH, superoxide, and hydroxyl radical scavenging activities, and in particular, caffeic and ferulic acids had stronger superoxide and hydroxyl radical scavenging activities than the well-known antioxidant radical scavenger, (+)-catechin (p<0.05). Quercetin and isorhamnetin were the primary compounds responsible for the strong antioxidant activity in the EtOH extract of the viscozyme-treated hamcho cake. Meanwhile, these five phenolic compounds were detected in the EtOH extract of the viscozyme-treated hamcho cake at the following levels (dry base of hamcho); procatechuic acid (1.54 mg%), caffeic acid (6.87 mg%), ferulic acid (8.45 mg%), quercetin (12.63 mg%), and isorhamnetin (6.65 mg%). However, three of these phenolic compounds (procatechuic, caffeic acid, and ferulic acids) were detectable in the $H_2O$ extract of viscozyme-treated hamcho juice. These results suggest that the EtOH extract of viscozyme-treated hamcho cake may be a potential source of natural antioxidants.
The objective of this study was to investigate anticytotoxic and antioxidatative capacities of ethanol extracts from Acer tegmentosum Maxim (A. tegmentosum) stem in vitro. The extract at concentration of 200 ug/mL inhibited 10 and 20 ug/mL arsenic trioxide-induced cytotoxicity of HepG2 cells by 79.3 and 57.5%, respectively. The extract at concentration of 200 ug/mL inhibited 0.2 and 0.5 mM t-BHP-induced cytotoxicity of HepG2 cells by 66.3 and 35.7%, respectively. Antioxidative effects of the extract were examined via measurement of ABTS, superoxide, and peroxyl radical scavenging activities. ABTS radical scavenging activity of the extract was higher than that of ${\alpha}$-tocopherol. Superoxide scavenging activity of the extract was higher than that of catechin. Oxygen radical absorbance capacity of the extract was higher than that of ascorbic acid. Cupric reducing antioxidant capacity of the extract was higher than that of ${\alpha}$-tocopherol. The extract at concentrations of 100 and $500{\mu}g/mL$ inhibited 10 mM t-BHP-induced lipid peroxidation of HepG2 cells by 38.2 and 80.7%, respectively. The extract prevented supercoiled DNA strand breakage induced by hydroxyl or peroxyl radical. Total phenolic and flavonoid contents of the extract at concentration of $100{\mu}g/mL$ were 71.3 nmol/mL gallic acid and 18.8 nmol/mL catechin equivalents, respectively. Thus, strong cytoprotective and antioxidant effects of A. tegmentosum stem extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in polyphenolic contents.

The major objective of this study was to investigate the usability as cultivar for processing of new Citrus hybrid 'Hamilgam'. We investigated various antioxidant activities, such as DPPH radical, ABTS radical, Hydroxyl radical and Superoxide anion radical effect, along with total polyphenol content and total flavonoid content of Citrus hybrid 'Hamilgam', C. unshiu, C. natsudaidai. The total polyphenol content and total flavonoid content were the highest in the Hamilgam. The major flavonoids of Hamilgam and Natsudaidai were naringin and neohesperidin, and those of Unshu were narirutin and hesperidin, as determined by HPLC. Especially, the neohesperidin content of Hamilgam showed much higher (100 times over) than that of Unshu. However, the aglycone form was not found. DPPH and ABTS radical scavenging activities were the highest in Hamilgam peels. Hydroxyl radical scavenging activity was high in the order of Unshu, Hamilgam and Natsudaidai. Superoxide anion radical scavenging of 3 cultivars peels displayed low activities compared to DPPH and ABTS radical scavenging. Based on these results, Hamilgam peel extracts possess antioxidant activities and may thus serve as potential sources of functional food, cosmetic products, etc.
