Stem bark of Cinchona sp. (Rubiaceae) is one of the well known drugs for its therapeutic values in traditional as well as modern medicine. Even though a lot of work has been carried out on quinoline alkaloids of Cinchona, its phenolic constituents received very little attention. In the present study, we evaluated antioxidant properties of C. officinalis stem bark methanolic extract and water extract containing phenolic compounds (total phenolics 21.37, 5.18% w/w respectively in the two extracts) in different in vitro and ex vivo models viz., antiradical activity by DPPH reduction, superoxide radical scavenging activity in riboflavin/light/NBT system, nitric oxide radical scavenging activity in sodium nitroprusside/Greiss reagent system and inhibition of lipid peroxidation induced by iron-ADP-ascorbate in liver homogenate and haemolysis of erythrocytes induced by phenylhydrazine in erythrocyte membrane stabilization study. Both the extracts exhibited very good antioxidant activity in all the models tested. The phenolic compounds including tannins present in the stem bark seem to offer protection from the oxidative damage.
An antioxidant was isolated from Korean mistletoe (Viscum album var. coloratum) by consecutive purification using silica gel, Sephadex LH-20, and RP-HPLC. The active principle was identified as homo-flavoyadorinin-B (3',7-dimethoxyluteolin-4'-O-[apiosyl $(1{\rightarrow}2)$ glucoside]) by spectral analyses. It inhibited 74.6% of hydroxyl radical and 30.6% of superoxide anion radical at 0.01 mM; however, th~compound did not show any scavenging activity against hydrogen peroxide radical. At 0.1 mM, above compound scavenged superoxide anion radical about twice as effective as positive controls, BHT and ${\alpha}-tocopherol$. Radical scavenging activities of homo-flavoyadorinin-B on DPPH, hydroxyl, and hydrogen peroxide radicals were almost same with those of positive controls.
In this study, S. glauca (Bunge) Bunge extract was testified total polyphenol contents, electron donating ability on DPPH, and scavenging ability of superoxide anion radical and hydrogen peroxide. Total polyphenol contents of S. glauca (Bunge) Bunge extract were 7,053.30±915.93 ㎍ GAE/mL. In the fractions, fraction by ethanol was the highest content of 10,973.30±1,000.24 ㎍ GAE/mL. Fractions of ethyl acetate and water were 2,386.70±166.53 ㎍ GAE/mL, 2,413.30±41.63 ㎍ GAE/mL respectively. It was shown that total content of polyphenol according to solvent was significant relation at p<0.05. In the experiment of the electron donating ability, 70% ethanol extract and methylene chloride fraction were -246.15±24.17%, -254.01± 16.54% respectively. In case of ethyl acetate fraction, it was the highest electron donating ability to DPPH radical, 39.06±0.34%, and then water fraction of 36.71±5.55%, ethanol fraction of 29.77±2.57%, gradually. Electron donating abilities revealed significant difference (p<0.05) between the solvents. The superoxide radical scavenging ability of standard material was 0.029±0.0011; 70% ethanol extract of 0.022±0.00052, methylene chloride fraction of 0.027±0.00031, ethyl acetate fraction of 0.024±0.0011, ethanol fraction of 0.021±0.00024 and water fraction of 0.024±0.00019. Hydrogen peroxide scavenging ability of 70% ethanol extract was -0.0029±0.00040 and the others were as follow; methylene chloride fraction (-0.0042±0.00058), ethyl acetate fraction (-0.003± 0.0041), ethanol fraction (-0.0029±0.0015) and water fraction (-0.0028±0.00090).
The objective of this study was to investigate the antioxidant and anti-inflammatory activities of Eugenia caryophyllata Thunb. water and 70% ethanol extracts. The content of total polyphenol was significantly higher in water extract than in 70% ethanol extract. The DPPH radical scavenging activity of water extract was similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. The ABTS radical scavenging activities of water and 70% ethanol extract were similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. SOD-like activity of water extract was higher than that of 70% ethanol extract at a concentration of $1,000{\mu}g/mL$ but lower than that of Vit. C. The DPPH radical scavenging activity, ABTS radical scavenging activity, and SOD-like activity increased as concentrations of water and 70% ethanol extracts increased. Cell cytotoxicity was not observed at all concentrations except at $100{\mu}g/mL$ concentration of water extract. Inhibitory activity on NO production effect of water extract was significantly higher than that of 70% ethanol extract. These results show that E. caryophyllata Thunb. has potent biological activities, and their activities were different depending on extraction solvent.
Ginseng seeds were treated with different autoclaving temperatures and autoclaving times, and extracted with 80% methanol to measure changes in antioxidant activity. The antioxidant activity of ginseng seeds treated by autoclaving was measured by 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, 2,2'-aziono-bis(3-ethylbenzthiazoline)-6-sulfonic acid radical scavenging activity, superoxide dismutase SOD-like activity, ferric reducing antioxidant power (FRAP), and total phenolic compound content. As autoclaving temperature and time were increased, the L lightness value decreased and the redness value tended to increase. Total phenolic compound content was about three times higher in ginseng seeds treated with autoclaving at 130°C than in ginseng seeds that were not treated. DPPH radical scavenging activity and ABTS radical scavenging activity increased as autoclaving temperature and time were increased. In particular, when the concentration was 100 ppm, the ABTS radical scavenging activity was 91.80% in ginseng seeds treated by autoclaving at $130^{\circ}C$, which was the highest antioxidant activity. FRAP and SOD-like antioxidant activity tended to increase significantly as autoclaving temperature and time were increased.

The human skin is constantly exposed to environmental irritants such as ultraviolet, smoke, chemicals. Free radicals and reactive oxygen species (ROS) caused by these environmen tal facts play critical roles in cellular damage. These irritants are in themselves damaging to the skin structure but they also participate the immensely complex inflammatory reaction. The purpose of this study was to investigate the skin cell protective effect of Campsis grandiflora extract on the UVB-irradiated human dermal fibroblasts (HDFs). We tested free radical and superoxide scavenging effect in vitro. C. grandiflora extracts had potent radical scavenging effect by 82% at $100{\mu}g/ml$, respectively. For testing intracellular ROS scavenging activity the cultured HDFs were analyzed by increase in DCF fluorescence upon exposure to UVB 20 $MJ/cm^2$ after treatment of C.grandiflora extracts. The results showed that oxidation of CM-DCFDA was inhibited by C.grandiflora extracts effectively and C.grandiflora extracts has a potent free radical scavenging activity in UVB- irradiated HDFs. In ROS imaging using confocal microscope we visualized DCF fluorescence in HDFs directly. In conclusion, our results suggest that C.grandiflora can be effectively used for the prevention of UV-induced adverse skin reactions such as radical production, and skin cell damage.

Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical increases with age, the reactive oxygen species(ROS) such as hydrogen peroxide($H_2O_2$), nitric oxide(NO). Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. Our reserch objective was to examine the in vitro biological activity of Scolopendrid Pharmacopuncture, including the total poly-phenol content, DPPH radical scavenging, ABTS radical scavenging, Superoxide dismutase(SOD)-like activity, Nitrite scavenging ability. The total poly-phenol contents of Scolopendrid Pharmacopuncture was 35.859mg/L. Elctron donation ability on DPPH was 36.82%. The 2,2'-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) was 84.7%. The superoxide dismutase (SOD)-like activities of Scolopendrid Pharmacopuncture was 44.33%. The nitrite scavenging effects were pH dependent, and were highest at pH 1.5(45.2%) and lowest at pH 6.0(11.3%). We conclude that Scolopendrid Pharmacopuncture may be useful as potential sources of antioxidant.
This study was conducted to develop a functional syrup by using maca extract. Five different levels (0%, 7%, 14%, 21%, 28%) of maca extract was added and the physicochemical properties were investigated. The pH of control syrup was lower than that of syrup added with maca extract. Viscosity increased with increasing amount of maca extract. Hunter L value decreased while b value increased as maca extract level increased. Results of consumer acceptance test revealed that no significant differences in flavor, sweet taste and overall acceptability were observed among samples. Therefore, it is suggested that maca extract can be incorporated into syrup up to 28% without depressing the quality. Total polyphenol content and DPPH radical, superoxide anion radical and hydroxyl radical scavenging tests were conducted in view of estimating the functionality of maca syrup. Results showed that incorporation of maca extract into syrup exhibited higher polyphenol content and DPPH radical and superoxide anion radical scavenging activity than control syrup (p<0.05).
Objective : The purpose of this study was to investigate anti-aging and antioxidant effects of extracts of Nelumbo nucifera leaves (NN-L) using ethanol on skin .Methods : Each part of leaves(NN-L), flowers(NN-F) and stem(NN-S) was extracted with 70% ethanol. We performed radical scavenging assay(DPPH, ABTS+, Superoxide anion radical), elastase inhibition assay, collagenase inhibition assay. NN-L extracts were tested for cell viability(MTT assay), MMP-1 inhibition and MMP-1 protein expression on CCD-986sk cells (human fibroblast line). Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. We measured its free radical scavenging activity, elastase inhibitory activity and expression of MMP-1 (matrix metalloprotease-1) in human fibroblast cells. Among the parts of Nelumbo nucifera, NN-L showed the highest antioxidant activities and in radical scavenging. DPPH, ABTS+ and Superoxide anion radical scavenging activity of NN-L at concentration of 1,000 μg/mL were 91.43%, 99.31% and 73.7% respectively. In vitro elastase and collagenase inhibition effects of NN-L at concentration of 1,000 μg/mL was 42.8% and 55.3% respectively. The ethanol extract of NN-L showed cell viability of 95.4% in 50 μg/mL concentration. In addition, The results from Western blot assay showed that NN-L decreased the expression of MMP-1 protein in a dose-dependent manner (by up to 35.0% at 50 μM).Conclusion : The findings suggest that the NN-L great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

In this study, we investigated the antioxidative effects of AY by measuring 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), and superoxide radical ($O_2{^-}$) scavenging activities in vitro. AY was extracted with ethanol and then partitioned with n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc) and n-butanol (n-BuOH). In the DPPH radical scavenging assay, AY at concentrations of 10 to $100{\mu}g/mL$ dose-dependently increased inhibition of DPPH oxidation, with the EtOAc fraction of AY showing the highest DPPH radical scavenging activity fractions. The $^{\cdot}OH$ radical scavenging activities of the extract and four fractions of AY increased by over 80% at a concentration of $50{\mu}g/mL$. In particular, the IC50 value of the EtOAc fraction was $0.03{\mu}g/mL$, which was the lowest value among all fractions. We also found that the EtOAc fraction of AY was better at $O_2{^-}$ radical scavenging than other fractions. Taken together, these results suggest that AY, especially the EtOAc fraction, can be used as a natural antioxidant against free radicals.
