어류에 다량 함유된 EPA와 DHA에 대한 관심이 높아지고 있다. 하지만 어유의 이용에 있어 가장 장애가 되는 요소는 비린내 및 산패취이며 이를 제거하기 위한 연구를 필요로 하고 있다. 본 논문에서는 초임계 이산화탄소를 추출용매로 하여 참치 안구유로부터 제거된 휘발성 이취성분을 동정하고 초임계 이산화탄소 추출 전과후의 이취성분을 비교해 보았다. 또한 추출 조건인 온도와 압력 조건을 달리 함으로써 휘발성 성분의 제거에 있어서의 최적조건을 찾아보았다. 추출 전 참치 안구유에서 99종의 휘발성 성분을 동정할 수 있었던 반면, $40^{\circ}C$, 200bar조건에서의 추출 후의 분석결과에서는 14개 이취성분을 동정할 수 있었으며, 어유 이취 유발성분인 heptanal과 octanal이 제거되었다. $60^{\circ}C$, 80-200 bar 영역에서는 휘발성 성분의 제거율이 높게 나타났다. 초임계 이산화탄소 추출에 의한 참치유의 휘발성 성분의 제거는 기존의 방법에 비교하여 효율성과 안정성이 우수하여 생물 및 식품산업분야에 다양하게 이용될 것으로 전망된다.
유기용매에 의한 잔류농약 추출방법을 대체할 신속, 간편, 값싼 방법의 개발을 목표로, 어류조직에 5종의 유기인계 농약 표준용액을 첨가하여, 추출온도와 압력별, 이산화탄소 유속별, 보조용매 종류별, 어류 시료량별, 어류 종류별로 초임계 이산화탄소를 이용하여 첨가회수율을 측정하였다. 조피볼락에 유기인계 농약 표준용액을 첨가한 후 흡수제로 celite를 혼합하여 초임계 이산화탄소로 추출하였을 때 첨가회수율은 추출온도와 추출압력이 낮을수록 증가하였고, 이산화탄소 유속이 높을수록 감소하였다. 보조용매 사용에 따른 회수율의 증가효과는 크지 않았으며, 사용한 보조용매로 methanol, acetone, methylene chloride 중 methylene chloride 10%와 더불어 추출하였을 때 첨가회수율이 가장 높았다. 어류 시료량이 적을수록 첨가회수율이 증가하였다. 조피볼락, 방어, 옥돔, 정어리, 고등어에서의 첨가회수율은 각각 $66.7{\sim}86.3%$, $56.2{\sim}79.2%$, $57.6{\sim}77.8%$, $84.2{\sim}96.3%$, $74.6{\sim}83.6%$로 지방함량이 높은 시료는 지방함량이 낮은 시료보다 첨가회수율이 높았다.
300bar와 313K에서 초임계 이산화탄소와 에틸아세테이트와 메탄올을 보조용매로 사용하여 주목나무로부터 taxol과 baccatin III를 추출하는 실험을 한 결과 추출되어지는 taxol의 추출량은 보조용매로 사용된 에틸아세테이트의 농도에는 영향을 받지 않았으며 3wt%의 메탄올을 사용했을 때 가장 많은 양이 추출되었고 taxol의 선택성은 보조용매로 에틸아세테이트를 사용했을 때 0.117mass%로 가장 높게 나타났다. Baccatin III는 보조용매로 에틸아세테이트를 사용하면 유기용매 추출량과 거의 같은 양이 추출되어지며 이 때의 선택성은 0.7wt% 에틸아세테이트에서 1.245wt%, 3.0wt% 에틸아세테이트에서 1.115wt%로 유기용매 추출에 비해 19배까지 크게 증가하였으며 3wt%의 메탄올을 사용했을 때에는 약 6.4배 증가하였다. 여기서 에틸아세테이트는 baccatin III를 추출하는데 있어 수율과 선택성면에서 가장 효과적인 보조용매라는 것을 알 수 있다. 또한 시료를 n-hexane으로 처리하면 비극성 물질은 상당히 제거되나 그 과정에서 많은 양의 taxol 과 baccatin III가 손실될 뿐만 아니라 선택성도 증가하지 않으므로 공정상 불필요하며 보조용매를 사용한 초임계유체 추출이 주목나무로부터 특정성분을 추출하는데 아주 효과적이라는 것을 알 수있다.
보조용매가 포함되지 않은 초임계이산화탄소를 이용하여 추출한 결과 온도 및 압력, 유속 등을 변경시켜도 genistein은 추출되지 않았다. 유기용매로 soaking한 결과를 보면 1mL ethanol로 soaking한 경우, 7.8%의 추출수율을 보였으며 함량도 약 7.8%정도를 나타내었다. Methanol로 soaking한 경우 더 낮은 수율과 함량을 보였다. 보조용매가 포함되지 않은 초임계이산화탄소로 추출한 결과 추출이 되지 않아서 보조용매를 이용하였다. 동일한 온도 압력 하에서 추출한 결과를 보면, ethanol이 methanol보다 효과가 우수하였다. 보조용매를 이용할 때 온도의 영향은 ethanol을 보조 용매로 사용한 경우, 압력 300bar일 때에 75$^{\circ}C$에서 56.4%의 최대수율을 나타내었고 함량은 55$^{\circ}C$일 때가 가장 높았다. 온도 35$^{\circ}C$조건에서 9%의 ethanol을 보조용매로 사용하여 압력의 영향을 조사하였다. 그 결과 300bar에서 genistein은 48.1%의 추출 수율을 얻을 수 있었다. 함량은 200bar일 때가 가장 높았지만 300bar와 비교해서 큰 차이가 없었다. 또한 ethanol을 보조용매로 사용할 때 초임계이산화탄소 유량에 대한 영향은 유량이 증가할수록 추출량도 증가하는 경향을 보였으며 함량도 약간 증가하였다. 온도 35$^{\circ}C$, 압력 275bar에서 ethanol 농도의 영향을 조사하였다. ethanol의 농도가 증가할수록 추출량도 증가하였으나 19%이후부터는 증가율이 둔화되었다. 보조용매를 첨가한 초임계유체를 이용하여 genistein을 추출하였을 때에 ethyl ether를 사용하여 추출했을 때와 비교해서 35$^{\circ}C$, 300bar, 30분간의 추출에서 genistein은 71% 추출수율과 31.5%의 함량을 나타내었다.
동결 건조된 미역 가공 부산물로부터 색소 및 지방산의 추출 및 조건은 시료 30g, 압력 $10.3\~17.2MPa$, 유량 15mL/min와 온도 $30\~45^{\circ}C$에서 추출 실험을 실시하였다. 지방산의 경우 초임계 처리하지 않은 것에 비해 불포화 지방산의 함량이 높게 나타났고, 온도 $40^{\circ}C$의 조건에서 보조용매 (entrainer)로 에탄올을 첨가하여 압력의 변화에 따른 회수율을 실험한 결과 12.4MPa에서 최대 $92\%$의 회수율을 보였다. 그리고 압력이 증가하면서 초임계 이산화탄소에 대한 용질의 용해도가 감소하는 retrograde 현상이 나타났다 시료입자의 크기를 $500\~1,000{\mu}\;m$와 12.4MPa에서 온도의 변화를 주어 실험한 결과 입자의 크기 $500{\mu}m$가 $1000{\mu}m$에 비해 색소의 추출량이 1.8배정도 많았다 최적 조건 (압력 12.4MPa,온도 $35^{\circ}C$, 입자크기 $500{\mu}m$)에서 추출시간에 따른 chlorophyll a 함량을 비교한 결과 에탄올을 각각 3mL/min, 5mL/min 첨가하였을 때 모두 20분까지 함량이 증가한 후 감소하였으며, 최적 추출시간은 30분까지로 판단되었다. 유기용매 추출과 초임계 추출을 비교한 결과 유기용매로 48시간 추출시 chlorophyll a 함량이 150mg/L였지만 초임계로 1시간 추출시 최대 172mg/L로 약 $20\%$ 증가하였다.
The oil in mackerel muscle was extracted using an environment friendly solvent, supercritical carbon dioxide (SC-$CO_2$) at a semibatch flow extraction process and an n-hexane. The SC-$CO_2$ was maintained at a temperature of $45^{\circ}C$ under pressures ranging from 15 to 25 MPa. The flow rate of $CO_2$ (27 g/min) was constant during the entire 2 h extraction period. The fatty acid composition of the oil was analyzed using gas chromatography (GC). Significant concentrations of eicosapentaenoic acid (EPA) acid and docosahexaenoic acid (DHA) acid were present in the SC-$CO_2$ extracted oil. The oil extracted using SC-$CO_2$ exhibited increased stability compared with n-haxane extracted oil. Particles of mackerel oil together with the biodegradable polymer, polyethylene glycol (PEG) were formed using a gas saturated solution process (PGSS) with SC-$CO_2$ in a thermostatted stirred vessel. Different temperatures ($45-55^{\circ}C$), pressures (15-25 MPa) and a nozzle size $400{\mu}m$ were used for PGSS with a 1 h reaction time. The stability of mackerel oil in the particles did not changed significantly.
Many studies on plant extracts have been reported for the treatment of candidiasis caused by Candida albicans, a representative fungal infection. This study demonstrates the synergistic antifungal activity of the combination of Phellodendri Cortex and Magnoliae Cortex, previously reported to have antifungal efficacy. Considering the antifungal efficacy and the separation of the active constituents, berberine and magnolol, hot water extraction and carbon dioxide supercritical extraction were selected for Phellodendri Cortex and Magnoliae Cortex, respectively. A combination of 0.55 g/L hot water extract of Phellodendri Cortex and 0.59 g/L carbon dioxide supercritical extract of Magnoliae Cortex showed synergistic antifungal activity. The synergistic antifungal activity of 160 μM berberine and 100 μM magnolol, which are representative antifungal compounds of Phellodendri Cortex and Magnoliae Cortex, respectively, contributes to the synergistic antifungal effect of their extracts. The additive decrease in cellular ergosterol level and the increased antifungal efficacy by extracellular ergosterol suggest that disruption of the biological function of ergosterol in the cell membrane is not responsible for the synergistic antifungal activity of berberine and magnolol. Synergistic cellular release of chromosomal DNA upon mixing berberine and magnolol indicates that disruption of the cellular structure is responsible for the synergistic antifungal effect of berberine and magnolol.
In this study, gemcitabine (Gem)-Poly(L-lactic acid) (PLLA) conjugates were synthesized through an amide linkage reaction. Then, the microparticles of Gem-PLLA/PLLA blends containing gemcitabine were prepared using a supercritical fluid process, called aerosol solvent extraction system (ASES). Gemcitabine-loaded Gem-PLLA/PLLA microparticles obtained from the ASES process showed a spherical shape. The amount of gemcitabine released after 30 day incubation in a phosphate buffer solution of pH 7.4 was about 90% of the total amount of gemcitabine present in the product.
To develop mil fat fractions with desirable physico-chemical properties, anhydrous milk fat (AMF) was fractionated by one- and two-stage extractions using supercritical $CO_2$(SC-$CO_2$). Tow-stage extraction of AMF was performed by first producing tow fractions, an extract and a residue at 4$0^{\circ}C$/241bar, which were subsequently used as the feed for an extraction at 6$0^{\circ}C$/241bar and 4$0^{\circ}C$/345bar, and separated into five and four fractions, respectively, based one extraction time. These fractions were quantified and analyzed for fatty acids and physico-chemical properties. SHort-chain (C4~C8) fatty acids in extract fractions from an extract were 200~150% compared with those of the original AF. Long-chain (C14~C18) fatty acids in extract fractions from a residue were 118~141%. The ratio of unsaturated fatty acids in the residue fraction was 131%. Melting point ranged from 22 to 43$^{\circ}C$, iodine value 21.8 to 36.9, and saponification value 255 to 221 in the extract and residue fractions. SC-$CO_2$ fractionation of AMF by two-stage extraction offers the possibility of developing ractions with discrete fatty acid compositions and physico-chemical properties such as melting point, iodine value and saponification value.
초임계 이산화탄소 및 유기용매를 이용하여 동결 건조된 멸치 시료로부터 오일을 추출하였으며, 시료는 700 ${\mu}m$로 균질화 시켜 사용하였다. 다양한 압력 (15~25 MPa) 및 온도 ($40{\sim}60^{\circ}C$) 조건에서 실험을 수행하였으며, 22 g/min의 일정한 유량을 추출기로 유입시켰다. 추출된 오일의 지방산 조성을 확인하기 위하여 가스 크로마토그래피를 이용하여 분석한 결과 추출된 멸치 오일에는 myristic acid, palmitic acid, stearic acid, palmitoleic acid, eicosapentaenoic acid (EPA), 그리고 docosahexaenoic acid (DHA)가 주요 지방산으로 확인되었다. 추출된 오일 내 EPA 및 DHA와 같은 고도불포화지방산(PUFAs)함량을 비교하였을 때, 유기용매 추출 오일보다 초임계 이산화탄소를 이용하여 추출한 오일의 고도 불포화 지방산 함량이 더 높았다. 산화도 비교 실험인 산가 및 과산화물가 값을 측정한 결과, 유기용매 추출 오일보다 초임계 이산화탄소를 이용한 추출 오일의 산화도가 유의적으로 낮았으며, 산화되는 속도 또한 느리게 진행되는 것을 확인할 수 있었다.
