Chemical Characterization of Neutral Extracts Prepared by Treating Pinus radiata Bark with Sodium Bicarbonate

  • A neutral extract (NE), that is soluble in cold water and has excellent antioxidant activity, from Pinus radiata pine bark was prepared by sodium bicarbonate treatment, and its chemical characteristics were investigated. NE was prepared by treating P. radiata bark with 0.8% NaHCO3 aqueous solution with a 5 : 1 liquor-to-bark ratio at boiling temperature for 1 h, resulting in 44% yield and final pH of 6.66. The yield of NE was 11% higher than that of the hot water extract (HWE) due to the increase in the solubility of polyphenols, the main component in the bark, by NaHCO3 treatment. NE was characterized through FT-IR, NMR, and MALDI TOF MS analyses. The results indicated that NE is mostly composed of proanthocyanidins (PAs) consisting of procyanidin (PC) units. The acetylated neutral extract (Ac-NE) had weight average molecular weight (${\bar{M}}w$) of 5,300 Da. The Ac-NE had wide molecular weight distribution and its polydispersity (${\bar{M}}w/{\bar{M}}n$) was 6 times higher than that of pure PA. The antioxidant activity of NE was determined by 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay and showed that NE had comparable antioxidant activity with pure PA.

Antimicrobial Activity and Acute Toxicity of Natural Rutin (천연 Rutin의 항균효과와 급성독성에 미치는 영향)

  • As part of our search for less toxic antimicrobial agents from natural resources, antimicrobial activity of rutin isolated from Sophora japonica was tested in vitro against four kinds of gram positive bacteria and four kinds of gram negative bacteria by serial broth dilution method. Among eight kinds of bacteria tested, the antimicrobial activity of rutin was the most potent against Mycobacterium smegmatis showing MIC of $375\;{\mu}g/ml$. The acute toxicity of rutin was examined in mice and rats. and $LD_{50}$ value administered intraperitoneally in mice was 650 mg/kg(Confidence limit: 894-1,473 mg/kg). There were no significant changes in serum biochemical values and histopathological changes as compared with those of control after intraperitoneal administration with rutin in rats.

Nonstoichimetry (x) of Nuclear Materials UO2+x and $(Ce_yU_{1-y})O_{2+x}$ at High Temperatures as a Function of Oxygen Activity$(a_o_2)$

  • The oxygen nonstoichiometry (x) of UO$_{2+x}$ and $(Ce_yU_{1-y})O_{2+x}$ (y=0.05, 0.25) has been measured as a function of oxygen activity (a02) at 100$0^{\circ}C$ by a solid state coulometri titration technique. The results for UO$_{2+x}$ are in a good agreement with literature data and the ao2-dependence of the nonstoichimetry has been well explained with (2:2:2) cluster model. The equilibrium oxygen activity of $(Ce_yU_{1-y})O_{2+x}$ increases with Ce-content (y) for given nonstoichiometry (x), which is ascribed to the fact that Ce is present as Ce$^{3+}\; and\; Ce^{4+}$, and, therefore, the addition of Ce reduces the oxidation capacity of UO$_{2+x}$. From the oxygen activity dependence of x in $(Ce_yU_{1-y})O_{2+x}$ the defect structure of $(Ce_yU_{1-y})O_{2+x}$ is discussed.

Hypocholesterolemic Effects of Soybean Lecithin in Cholesterol-Fed Rats

  • The present study was performed to elucidate the hypocholesterolemic action of lecithin on the diet-induced hypercholesterolemia in rats. Male Sprague-Dawley rats (n=24) were fed lecithin-free (control) diet or diets containing $2\%\;or\;5\%$ lecithin for 4 weeks. Hypercholesterolemia was induced by adding $1\%$ cholesterol and $0.5\%$ cholic acid to all diets. No difference was found in food intake and body weight gain among groups. The lecithin treated groups showed significant improvement in the plasma levels of total cholesterol and LDL-cholesterol (p<0.05) compared to the control group, while the plasma triacylglyceride was not significantly affected 1he atherogenic index and HDL-cholesterol level were decreased in the lecithin groups. The diets with $2\%\;or\;5\%$ lecithin significantly decreased the activity of cholestetyl ester transfer protein (CETP) by $14\%\;or\;17\%$, respectively. Also, lecithin diets increased the activity of lecithin: cholesterol acyltransferase (LCAT). These results suggest that lecithin accounts for the hypocholesterolemic effect due to the decreased CETP activity and increased LCAT activity.

Effect of Occlusion upon Rat Skin on the Activities of Cutaneous Oxygen Radical Metabolizing Enzymes in Rats

  • To evaluate the effect of occlusive skin on the activity of cutaneous oxygen free radical metabolizing enzymes in rats, the dorsal skin was covered with closed glass chamber shaped petri dish, 46 mm in diameter and 10 mm in height and sealed by an adhesive. Five day-occluded group showed more increased activity of xanthine oxidase (XO) than that of control, and the activity of five day-occluded group was higher than that of ten day-occluded group. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx) were significantly higher in ten day-occluded group than in control or five day-occluded group. All the more, five day-occluded group showed the decreasing tendency of SOD and GPx activities compared to those of control. On the other hand, the cerrous perhydroxide deposits were observed in the intercellular space of the stratum basale in five day-occluded group under the electronic microscope using a cytochemistry method. Futhermore, the degree of cerrous perhydroxide reaction was lower in ten day-occluded group than in five day-occluded group. In conclusion, the increased XO activity and the decreased SOD and GPx activities are likely to responsible far the accumulation of $H_2O_2$ in five day-occluded group.

Cytotoxic Triterpenoids from the Fruiting Bodies of Ganoderma lucidum

  • Twelve triterpenoids (1 - 12) were isolated from $CHCl_3$-soluble fraction of fruiting bodies of Ganoderma lucidum. Extensive spectroscopic and chemical studies established the structures of these compounds as butyl lucidenate P (1), butyl lucidenate $E_2$ (2), butyl lucidenate $D_2$ (3), butyl lucidenate Q (4), ganoderiol F (5), methyl ganoderate H (6), methyl ganoderate J (7), lucidumol B (8), ganodermanondiol (9), methyl lucidenate N (10), methyl lucidenate A (11) and butyl lucidenate N (12). All of the compounds were examined for their cytotoxic activity against HL-60, HeLa, and MCF-7 cancer cell lines. Among them, compounds 4 and 8 showed cytotoxic activity with $IC_{50}$ values of 6.6 and 1.6 ${\mu}M$ against HL-60, respectively. In addition, compound 8 also showed cytotoxic activity with $IC_{50}$ values of 2.0 ${\mu}M$ against HeLa cancer cell line, other compounds were moderate or inactive.

Cytotoxic and Mutagenic Effects of Cinnamomum cassia Bark-Derived Materials

  • The cytotoxic activities of Cinnamomum cassia (Blume) bark-derived materials toward six human HeLa epithelioid cervix, A549 lung, SK-OV-3 ovarian, SK-MEL-2 melanoma, XF-498 central nerve system, and HCT-15 colon tumor cell lines were evaluated by using sulforhodamine B assay and compared to those of the anticancer agents, cisplatin and mitomycin C. The biologically active constituent of the Cinnamomum bark was characterized as trans­cinnamaldehyde by spectroscopic analysis. The cytotoxic activity of cinnamaldehyde against HeLa, SK-MEL-2, and HCT -15 cell lines was comparable to that of cisplatin and mitomycin C. The compound showed lower activity against A549, SK-OV-3, and XF-498 cell lines than the anticancer agents. Eugenol exhibited moderate activity against SK-OV­3, XF-498, and HCT-15 tumor cells, and trans-cinnamic acid, cinnamyl alcohol, $\alpha-pinene,\;and\;\beta-pinene$ showed little or no activity against model tumor cells. Cinnamaldehyde was not mutagenic against four strains (TA 98, TA 100, TA 1535, and TA 1537) of Salmonella typhimurium (Castel and Chalm). These results indicate at least one pharmacological action of C. cassia.

Characterization of a Bioflocculant Produced by an Isolate, Bacillus megaterium G31

  • A bacterial strain capable of producing a novel bioflocculant was isolated from a biofilm sample and identified as Bacillus megaterium G31. The highest biopolymer yield was achieved when the organism was cultivated in a medium containing acetate as the sole carbon source and ($NH_4)_2HPO_4$as the nitrogen source. In kaolin suspension, the flocculating activity was highest at 170 mg I$^{-1}$ and decreased at the higher bioflocculant concentrations. The crude bioflocculant produced by the organism was purified by ethanol precipitation and gel permeation chromatography. The FTIR spectrum of the purified bioflocculant revealed that the bioflocculant might be a heterogeneous polysaccharide composed of hexosamines and neutral sugars. The analysis of sugar components of the bioflocculant using high performance anion-exchange chromatography showed that the sugar constituents of the bioflocculant were glucosamine, fucose, galactosamine, galactose, glucose, mannose in approximate molar ratio of 4 : 1 : 6 : 3 : 8 : 19. Its flocculating activity was stimulated by various cations. The bioflocculant was thermo-stable and retained 64% of its original activity after heating at $100^{\circ}C$ for 50 min.

Purification and Characterization of an Antibacterial Substance from Aerococcus urinaeequi Strain HS36

  • A bacterial strain inhibiting the growth of Vibrio anguillarum, the causative agent of vibriosis, was isolated from fish intestines. The isolated strain HS36 was identified as Aerococcus urinaeequi based on the characteristics of the genus according to Bergey's Manual of Systematic Bacteriology and by 16S rRNA sequencing. The growth rate and antibacterial activity of strain HS36 in shaking culture were higher than those in static culture, while the optimal pH and temperature for antibacterial activity were 7.0 and 30℃, respectively. The active antibacterial substance was purified from a culture broth of A. urinaeequi HS36 by Sephadex G-75 gel chromatography, Sephadex G-25 gel chromatography, and reverse-phase high-performance liquid chromatography. Its molecular weight, as estimated by Tricine SDS-polyacrylamide gel electrophoresis, was approximately 1,000 Da. The antibacterial substance produced by strain HS36 was stable after incubation for 1 h at 100℃. Although its antibacterial activity was optimal at pH 6-8, activity was retained at a pH range from 2 to 11. The purified antibacterial substance was inactivated by proteinase K, papain, and β-amylase treatment. The newly purified antibacterial substance, classified as a class II bacteriocin, inhibited the growth of Klebsiella pneumoniae, Salmonella enterica, and Vibrio alginolyticus.

사과 겹무늬썩음병균에 대한 Bacillus sp. SS279의 항진균활성과 생물학적 방제

  • From apple skin, we isolated a bacterial strain which strongly inhibited the growth of apple white rot fungus, Botryosphaeria dothidea. The isolated strain, designated as SS279, was identified to be the genus Bacillus. The antifungal activity of Bacillus sp. SS279 was found in the culture filtrate. The production of antifungal substances occurred during logarithmic phase and was the highest when cultures reached the stationary growth phase. The optimum ranges of temperature and pH for its production were 25-30$\circ$C and 4.5-9.0, respectively. The culture filtrate of Bacillus sp. SS279 exhibited a strong inhibitory effect on the spore germination and germ tube elongation of B. dothidea. Autoclaved culture filtrate of Bacillus sp. SS279 showed only a slight decrease in antifungal activity, indicating that the Bacillus sp. SS279 produce heat-stable antifungal substances. In in vivo bioassay, Bacillus sp. SS279 also showed antagonistic activity against apple white rot caused by B. dothidea.
