Selection of KYC 3270, a Cellulolytic Myxobacteria of Sorangium cellulosum, against Several Phytopathogens and a Potential Biocontrol Agent against Gray Mold in Stored Fruit

  • During 2002-2008 in Korea, 455 extracts from myxobacteria consisting of 318 cellulolytic and 137 bacteriolytic myxobacteria were isolated, which were then screened for antifungal activity against the phytopathogens Botrytis cinerea, Colletotrichum acutatum, Penicillium sp., Pyricularia grisea, and Phytophthora capsici. 204 isolates had antifungal activity, causing both a clear zone due to blocked spore germination and inhibition of mycelial growth; most (199) were from cellulolytic (Sorangium cellulosum) and only five were from bacteriolytic myxobacteria. B. cinerea, the best controlled among the five tested pathogens, had a unique group of antifungal isolates of myxobacterial extracts compared to the other pathogens' groups. Among seventy-nine bioactive myxobacteria, four isolates, KYC 3130, KYC 3247, KYC 3248 and KYC 3270, were selected and all were cellulolytic. Liquid culture filtrates of these four myxobacteria were applied to tomato, cherry tomato, strawberry, and kiwi fruits 5 h before inoculation with gray mold conidia; then the treated fruits were placed in an airtight container and the experiment was repeated six to eight times. Incidence (%) of gray mold on fruit of the infected control treatment was 84-98%, whereas it was only 5-21% after the KYC 3270 treatment. After KYC 3270 treatment of the four fruits, mold control was 79-95%, which was highest among the filtrates and statistically the same as treatment with fludioxonil, a registered chemical against gray mold of stored fruits.

Effects of Water-soluble and Water-insoluble Fractions of Kimchi on the Alteration of Plasma Lipids and Fibrinolytic Activity in Middle-aged Healthy Subjects

  • Freeze-dried kimchi fractions, water-soluble (WSK) versus water-insoluble (WISK), on their effects on plasma lipids and for their fibrinolytic activities were studied. Nineteen middle-aged healthy subjects were assigned to WSK, WISK, or placebo during 6 weeks of kimchi supplementation. Subjects in the WSK and WISK groups were supplemented with 3 grams of kimchi pills (equivalent to 60 grams of fresh kimchi), while the placebo group was on their normal diets. LDL/HDL and atherogenic index decreased in the subjects who received kimchi pills, but not in those in the placebo group. The levels of plasma triglyceride were significantly decreased in the WSK group compared with those in the WISK group (p<0.05). However, there was no difference in the levels of total cholesterol and HDL between the two groups. Fibrinolytic activity of WISK was significantly higher than that of WSK.

Luteolin Inhibits Extracellular Signal-Regulated Kinase Pathway Through Protease-Activated Receptors (-2 and -4) and Their Agonist Activity

  • Luteolin is a major flavonoid of Lonicera japonica and has anti-inflammatory effect. The activation of proteinase-activated receptor (PAR)-2 and -4 by trypsin appears to play a role in inflammation, In the present study, we examined the inhibitory effects of luteolin on activation of trypsin-induced human leukemic mast cells (HMC-1). HMC-1 cells were stimulated with trypsin, PAR-2 and PAR-4 agonist, in the presence or absence of luteolin. The level of TNF-${\alpha}$ secretion was measured by enzyme-linked immunosorbent assay (ELISA). The expression of tryptase and phosphorylated-extracellular signal-regulated kinase (ERK) were assessed by Westem blot analysis. Moreover, trypsin activity was measured by the substrate Bz-DL-Arg-p-nitroanilide (BAPNA). TNF-${\alpha}$ secretion and Tryptase expression in trypsin-stimulated HMC-1 cells were markedly inhibited by pretreatment of luteolin. Furthermore, the pretreatment of luteolin resulted in the reduction of ERK phosphorylation and trypsin activity. These results suggest that luteolin might has the inhibitory effects on the PAR-2 and -4-dependent inflammation.

Characterization of Calcium-Activated Bifunctional Peptidase of the Psychrotrophic Bacillus cereus

  • The protease purified from Bacillus cereus JH108 has the function of leucine specific endopeptidase. When measured by hydrolysis of synthetic substrate (N-succinyl-Ala-Ala-Pro-Leu-p-nitroanilide), the enzyme activity exhibited optimal activity at pH 9.0, $60^{\circ}C$. The endopeptidase activity was stimulated by $Ca^{++},\;Co^{++},\;Mn^{++},\;Mg^{++},\;and\;Ni^{++}$, and was inhibited by metal chelating agents such as EDTA, 1,10-phenanthroline, and EGTA. Addition of serine protease inhibitor, PMSF, resulted in the elimination of the activity. The endopeptidase activity was fully recovered from the inhibition of EDTA by the addition of 1 mM $Ca^{++}$, and was partially restored by $Co^{++}\;and\;Mn^{++}$, indicating that the enzyme was stabilized and activated by divalent cations and has a serine residue at the active site. Addition of $Ca^{++}$ increased the pH and heat stability of endopeptidase activity. These results show that endopeptidase requires calcium ions for activity and/or stability. A Lineweaver-Burk plot analysis indicated that the $K_m$ value of endopeptidase is 0.315 mM and $V_{max}$ is 0.222 ) is $0.222\;{\mu}mol$ of N-succinyl-Ala-Ala-Pro-Leu-p-nitroanilide per min. Bestatin was shown to act as a competitive inhibitor to the endopeptidase activity.

Effect of Supplements $Mn^{2+}$, $Cu^{2+}$, and Aromatic Compounds and Penicillium decumbens on Lignocellulosic Enzyme Activity and Productivity of Catathelasma ventricosum

  • This is the first report on using Catathelasma ventricosum for production of fruiting body and lignocellulosic enzymes. To improve the laccase activity and productivity of mushroom, the substrate was added with different supplements (eight aromatic compounds, $Mn^{2+}$, and $Cu^{2+}$). Based on the results, all these supplements can improve the laccase activity and productivity of C. ventricosum, and it seems that there is a critical value of laccase activity that affects the productivity of C. ventricosum. In addition, when Penicillium decumbens was inoculated into the substrate that had been cultivated with C. ventricosum for 20 days, the highest values of laccase activity, FPA activity, and productivity of C. ventricosum were obtained. Moreover, the laccase activity showed a positive correlation with the productivity of C. ventricosum. Finally, the effect of $Mn^{2+}$, $Cu^{2+}$, and P. decumbens on laccase activity was investigated by response surface methodology (RSM).

Antibacterial Activity of Puer Tea Extract on Pathogenic Microorganisms

  • We have analyzed 11 different commercial brands provided by Daboo Culture and Art Center. This study was carried out to determine the antibacterial activity of water extract from puer tea ingredients against Staphylococcus aureus (S. aureus) and Listeria monocytogenes (L. monocytogenes). Antibacterial activity of the water extracts was shown against S. aureus and L. monocytogenes by paper disc method. In case of S. aureus, antibacterial activity of the extracts appeared as clear inhibitory zone. This antibacterial activity was greater along with pH and showed the highest effect at pH 10. In case of L. monocytogenes, antibacterial activity of the extract appeared as clear inhibitory zone. Some Puer tea appeared to be more powerful in antibacterial activity against S. aureus and L. monocytogenes. The polyphenol compounds were 3.46~8.68%, depending upon samples. In conclusion, puer tea is considered as a good choice for well-being food.

A Potent Anti-Complementary Acylated Sterol Glucoside from Orostachys japonicus

  • In order to isolate substances that inhibit the hemolytic activity of human serum against eryth-rocytes, we have evaluated whole plants of the Orostachys japonicus species with regard to its anti-complement activity, and have identified its active principles following activity-guided isolation. A methanol extract of the O. japonicus, as well as its n-hexane soluble fraction, exhibited significant anti-complement activity on the complement system, which was expressed as total hemolytic activity. A bioassay-guided chromatographic separation of the constituents resulted in the isolation of three known compounds 1-3 from the active n-hexane fraction. The structure of these compounds were analyzed, and they were identified as hydroxyhopanone (1), $\beta-sitosteryl-3-O-\beta-D-glucopyranosyl-6'-O-palmitate$ (2), and $\beta-sitosteryl-3-O-\beta-D-glucopyranoside$ (3), respectively. Of these compounds, compound 2 exhibited potent anti-complement activity $(IC_{50}=1.0\pm0.1{\mu}M)$ on the classical pathway of the complement, as compared to tiliroside $(IC_{50}=76.5\pm1.1{\mu}M)$, which was used as a positive control. However, compounds 1 and 3 exhibited no activity in this system.

Studies on the Fibrinolytic Effect of Germinated Grain Seeds

  • In this study, seven grain seeds(sorghum maize, buckwheat, soy bean, mung bean, red bean, and barngrass) and germinated seven grain seeds were examined the fibrinolytic activity through fibrin plate assay and SDS-PAGE. The results obtained were as follows : 1. In the fibrin plate assay, the extracts of maize, barngrass, sorghum and buckwheat showed fibrinolytic activity. Especially, Maize of them showed fibrinolytic activity that was almost similar to plasmin, fibrinolytic enzyme used as a positive control. 2. In the SDS-PAGE of seven grain seeds, fibrinolytic activity was remarkably shown in mung bean and red bean. 3. In the fibrin plate assay of germinated grain seeds, buckwheat(5 mm), buckwheat(10 mm) and soy bean(10 mm) showed a level of fibrinolytic activity that was about 0.3 fold than 1.0 unit of plasmin, Also maize(10 mm) of them showed a level of fibrinolytic activity that was about 0.5 fold than 1.0 unit of plasmin. As a result, maize of grain seeds was found that it has a strong fibrinolytic activity.

  • In vitro ${\beta}$-lactamase inhibitory activity of 6-triazole exomethylenepenam compounds (1, 2, 3, 4, 5 and 6) was compared with clavulanic acid, sulbactam and tazobactam. The inhibitory activity of 3, 4 and 5 was stronger than those of sulbactam, clavulanic acid and tazobactam against Type IV enzymes. And, inhibitory activity of 3 and 4 was stronger than those of sulbactam, clavulanic acid and tazobactam against Type III enzymes. The in vitro antimicrobial activity of 3, 4 and 5 combined with ampicillin was better than those with sulbactam and with cefoperazone was compared with the sulbactam against ${\beta}$-lactamase producing 27 strains. The synergistic activity of (Z)-form compounds (3 and 5) was better than that of (E)-form compound (4) and sulfone compound (5) was better than sulfide compound (3).

The Association between Health-related Quality of Life and Depression on Activity Restriction in Osteoarthritis: A Cross-sectional Study

  • Purpose: The purpose of this study is to provide basic evidence on the need to approach osteoarthritis patients through a psychological factors-considering rehabilitation program by understanding how activity restrictions in osteoarthritis affect health-related quality of life and depression. Methods: This study assessed 3,761 osteoarthritis patients from the Korea National Health and Nutrition Examination Survey. The subjects were divided into two categories: with and without activity restriction. Results: The prevalence of osteoarthritis in women was higher than that of men (men: 19.7%; women: 80.3%), and high BMI increased the prevalence of osteoarthritis. The EQ-5D index of subjects with activity restriction was 0.84±0.18 (points), while in those without activity restriction was 0.93±0.12, and the diagnosis of depression was 15.8%, 8.2%. There was a statistically significant difference in the odds ratio for each item in the EQ-5D. Moreover, the odds ratio for depression with activity restriction was 2.098 compared with no activity restriction. Conclusion: Activity restriction of osteoarthritis patients significantly decreases the health-related quality of life and increase the probability of depression. Therefore, early diagnosis of depression symptoms to prevent deterioration of symptoms in patients with osteoarthritis and to increase compliance with rehabilitation treatment, and to provide arbitration, including treatment that can alleviate depression.
