Phanerochaete chrysosporium의 액체 배양 및 Lignin Peroxidase 생산 (Submerged Culture of Phanerochaete chrysosporium and Lignin Peroxidase Production)

  • This study characterizes the growth of white rot fungi Phanerochaete chrysosporium IFO 31249) and lignin peroxidase(LiP) activity in different submerged culture media. P. chrysosporium was grown in the form of pellet of various sizes from a spore inoculum under shaking liquid culture condition. While the growth of mycelia was higher under the nitrogen-sufficient culture than under the nitrogen-limited culture, ligninase activity was relatively lower. The lignin peroxidase appeared in nitrogen-limited culture and was suppressed by excess nitrogen. High level(40U/l) of lignin peroxidase activity was obtained in the growth medium containing 1.5mM veratryl alcohol, a secondary metabolite of P. chrysosporium. Lignin peroxidase production was not observed under conditions of nitrogen sufficiency or in balanced media, suggesting that control parameters could increase the activity by manipulating the secondary metabolism.

Antioxidant compounds from the stem bark of Sorbus commixta

  • The MeOH extract of Sorbus commixta (Rosaceae) exhibited strong DPPH radical scavenging activity, and through activity-guided fractionation two antioxidant compounds were isolated and identified as $catechin-7-O-{\beta}-D-xylopyranoside$ (1) and $catechin-7-O-\;{\beta}-D-apiofuranoside$ (2) by physicochemical and spectrometric methods. To evaluate the antioxidant effect of these compounds, some in vitro tests, such as the DPPH radical scavenging activity test, the superoxide radical scavenging activity test and the lipid peroxidation inhibitory activity test, were performed. Compounds 1 and 2 showed stronger activities than both a-tocopherol and butylated hydroxy anisole (BHA) in each assay.

  • The investigation aimed to study the effects of methanol fraction of licorice (FM 100) and glycyrrhizin on prostaglandin synthetase activity, in relation to their analgesic effects. Effects of FM 100 and glycyrrhizin on the activity of prostaglandin synthetase extracted from bull seminal vesicles were examined by the modified method of Takeguchi et al. The analgesic effect of FM 100 was tested in mice by the acetic acid writhing method. FM 100 was administered orally to mice. BSV prostaglandin synthetase activity was inhibited significantly by FM 100 in a dose-dependent manner, whereas the activity was slightly inhibited by glycyrrhizin. Statistically significant analgesic effects were also observed with FM 100. The results suggest that analgesic effect of licorice may be due to the inhibition of prostaglandin synthesis.

Roles of the Hsp90-Calcineurin Pathway in the Antifungal Activity of Honokiol

  • Honokiol, a bioactive compound isolated from the cone and bark of Magnolia officinalis, has been shown to have various activities including inhibition of the growth of Candida albicans. We investigated the roles of the Hsp90-calcineurin pathway in the antifungal activity of honokiol. The pharmacologic tool was employed to evaluate the effects of Hsp90 and calcineurin in the antifungal activity of honokiol. We also evaluated the protective effects of the calcineurin inhibitor cyclosporin A (CsA) on honokiol-induced mitochondrial dysfunction by the fluorescence staining method. The Hsp90 inhibitor potentiated the antifungal activity of honokiol. A C. albicans strain with the calcineurin gene deleted displayed enhanced sensitivity to honokiol. However, co-treatment with calcineurin inhibitor CsA attenuated the cytotoxic activity of honokiol due to the protective effect on mitochondria. Our results provide insight into the action mechanism of honokiol.

다빈도 한약 처방 50종의 아세틸콜린 분해 효소 활성 및 아밀로이드 베타 단백질 응집 억제 효능 비교 연구 (Screening of 50 Korean Herbal formulas with Inhibitory Effects on Acetylcholinesterase Activity and Amyloid-β Aggregation)

  • Acetylcholinesterase (AChE) activation and amyloid-${\beta}$ ($A{\beta}$) aggregation are major biological markers of Alzheimer's disease. In the present study, we evaluated the inhibitory effects of 50 kinds of herbal formulas on AChE activity and $A{\beta}$ aggregation. Among them, Hwanglyeonhaedok-tang, Cheonwangbosim-dan, Makmundong-tang, and Gamisoyo-san had a potent effects on the inhbition of AChE activity. Sosiho-tang, Samsoeum, Cheonsimyeunjaeum, and Bunsimgieum exerted to have the inhibitory activity on $A{\beta}$ aggregation. In addition, these 8 herbal formulas showed the 3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, indicating their antioxidant activities.

Effect of Hydrothermal Treatment on the Antioxidant Activity of Rice Hull Extracts

  • Hydrothermal treatment of rice hull was hydrothermal carried out at 105, 110, $121^{\circ}C$ for 15, 30, 60 min, respectively, using a conventional autoclave. Antioxidant activity of the hydrothermal treated rice hull extract was evaluated by determining total phenol contents (TPC), 1,1-diphenyl-2-picylhydrazyl (DPPH) radical scavenging activity (RSA), reducing power, and ABTS RSA. TPC, DPPH RSA, reducing power, and 2,2-azinobis(3-ethylenebenzothiozoline-6-sulfonic acid) (ABTS) RSA of the extract were significantly increased with increasing treated temperature and time. For example, hydrothermal extracts at $121^{\circ}C$ for 60 min increased the TPC, DPPH RSA, reducing power, and ABTS RSA to 0.840 mg/mL, 64.77%, 1.437, and 92.11%, respectively, while those of the extracts treated at $105^{\circ}C$ for 60 min were 0.508 mg/mL, 51.23%, 0.819, and 45.22%, respectively. The results indicated that hydrothermal treatment of rice hull was very effective to increase phenolic compounds and antioxidant activity of rice hull extract.

Cultivation of Phanerochaete chrysosporium and Lignin Peroxidase Activity

  • Effects of exogenous veratryl alcohol addition on the growth of basidiomycete Phanerochaete chrysosporium ME-446 and the induction of lignin peroxidase activity were investigated in this study. The organism was grown in ligninolytic (low-nitrogen) culture conditions in which extracellular enzymes are produced. Analyses showed that a statistically significant decrease of cell growth was associated with the veratryl alcohol addition. The effect of veratryl alcohol addition on LiP activity was nearly instantaneous and this effect diminished with culture aging. The extent of this effect was different depending on the time of addition, which led to a speculation that there might be some other effector species which played a role in regulation of lignin peroxidase activity.

  • Antimicrobial activity of GL (the aqueous extract from the carpophores of Ganoderma lucidum ($F_RK_{KARST}$) was tested in vitro aginst Gram positive and Gram negative bacteria by serial broth dilution method, and the antimicrobial activity was expressed by minimal inhibitory concentration (MIC). Among fifteeen species of bacteria tested, the natimicrobial activity of GL was of antimicrobial combinations of GL with four kinds of antibiotics (ampicilin, cefazolin, oxytet-racycline and chloramphenicol), the fractional inhibitory concentraction index (FICI) was determined by checkerboard assy for each stain. The antimicrobial combinations of GL with four antibiotics resulted in additive effect in most instances, synergism in two instances, and antagonism in two instances. Synergism was obversed when GL was combined with cefazolin against Bacillus subtilis and Klebsiella oxytoca.

Cytotoxicity and L-Amino Acid Oxidase Activity of Crude Insect Drugs

  • The cytotoxicity of crude insect drugs was measured using HeLa cells originating from human cervix and uterine cancer. using the dye uptake assay in order to find potential anticancer agents. Three kinds of extracts (buffer, methanol and ethylacetate) were prepared from 26 insects and used as raw materials for the activity assay. Among these, the buffer extracts from Tabanus, Mylabris and Huechys showed a potent anticancer activity, and those from Catharsius, Red ant, Scorpion, Tabanus and Vespae Nidus showed a strong L-amino acid oxidase (AAO) activity as well as cytotoxicity. In contrast, buffer extracts from Gryllotalpa orientalis and Apriona germari larvae showed greater/more rapid Hela cell growth than that of other insects.

Psammaplin A, a Natural Bromotyrosine Derivative from a Sponge, Possesses the Antibacterial Activity against Methicillin-resistant Staphylococcus aureus and the DNA Gyrase-inhibitory Activity

  • Psammaplin A, a natural bromotyrosine derivative from an associated form of two sponges (Poecillastra sp. and jaspis sp.) was found to possess the antimicrobial effect on the Gram-positive bacteria, especially on methicillin-resistant Staphylococcus aureus (MRSA). The minimal inhibitory concentration of psammaplin A against twenty one MRSAs ranged from 0.781 to 6.25 ${\mu}g/ml$, which that of ciprofloxacin was 0.391~3.125${\mu}g/ml$. Psammaplin A could not bind to penicillin binding protein, but inhibited the DNA synthesis and the DNA gyrase activity with the respective 50% (DNA synthesis) and 100% (DNA gyrase) inhibitory concentration 2.83 and 100 ${\mu}g/ml$. These results indicate that psammaplin A has a considerable antibacterial activity, although restricted to a somewhat narrow range of bacteria, probably by inhibiting DNA gyrase.
