HPLC에 의한 식육조직중의 잔류 설파제 동시 분석 (Simultaneous Determination of Residual Sulfonamides in Meat Tissues by High Performance Liquid Chromatography)

  • Four sulfoanmides ; sulfamerazine ; sulfamethazine, sulfathiazole and sulfadimethoxine from muscle, kindney, liver and heart tissues of pork and chicken by LC. Residual sulfonamides were extracted with dichloromethane and determined on a Sperisorb ODS-1 column(250mm$\times$4.6mm id) with acetonitrile/water/acetic acid (30/70/0.3 v/v) as a mobile phase at 260nm. Recoveries from 4 tissues of pork and chicken samples fortified with 50 and 100 ppb were 71.2~87.2% and 73.7~89.6%, respectively. The detection limit was 0.03 $\mu\textrm{g}$/g in each drug. Sulfamethazine in 5 samples of pork. And sulfadimethoxine in 5 samples and sulfamethazine in 3 samples were also detected from 41 samples of chicken. The order of residue levels of sulfonamides in tissues was kidney>liver>heart>muscle, respectively. The residue levels of sulfonamides from kidney and liver were 0.03~0.15 $\mu\textrm{g}$/g in porks and 0.03~0.10 $\mu\textrm{g}$/g in chickens.

  • A multiresidual analysis was performed to determine 12 sulfonamides(sulfacetamide, sulfadiazine, sulfisomidine, sulfathiazole, sulfapyridine, sulfamerazine, sulfamethazine, sulfamonomethoxine, sulfisoxazole, sulfamethoxazole, sulfaquinoxaline, and sulfadimethoxine) in beef and pork simultaneously. The multiresidual analysis for the sulfonamides currently used was able to analyze 5 kinds of sulfonamides at the same time. The method of this 12 sulfonamides multiresidual analysis in this study was matrix solid-phase dispersion(MSPD) by high performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS). The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM: Ethylacetate(3:1), DCM:EA(9:1). Also, samples (84 beef and 205 pork samples) which were positive by EEC-4 plate test from 2001 to 2003 were tested to investigate the kinds of sulfonamides using HPLC. The results from the study were as follows; 1. The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM:Ethylacetate(3:1), DCM:EA(9:1). The method of extraction solvent with DCM:ethyl acetate(9:1) was the most excellent(87.7∼99.3%) in separation and reappearance. 2. In the LC/MS analysis. of sulfonamides, signal to noise ratio was showed relatively high in the positive mode and special ion in the quality analysis was determined via [M+H]$\^$+/ and m/z 156. A spectrum of sulfonamides was showed from all 12 sulfonamides. 3. The samples positive by the EEC-4 plate, a screening test method, were categorized by sulfonamides through Charm II and confirmed the kinds of sulfonamides through HPLC. 1) Among 84 beef samples positive by EEC-4 plate, 20 samples were positive by Charm II and identified as 7 sulfamethazine, 9 sulfadimethoxine, 1 sulfamonomethoxine and 3 unknown status. 2) Among 205 pork samples positive by EEC-4 plate, 42 samples were positive by Charm II and identified as 19 sulfamethazine, 1 sulfadimethoxine, 4 sulfamonomethoxine and 5 unknown status.

  • 가금질병(家禽疾病)의 예방(豫防) 및 치료(治療) 그리고 성장(成長) 및 비육촉진(肥育促進)의 목적(目的)으로 급여(扱與)한 sulfonamides의 계육(鷄肉) 및 계란내(鷄卵內) 잔류(殘留)할 가능성(可能性)이 주목(注目)됨으로 그 이행잔류상(移行殘留狀)을 구명(究明)함으로써 식육내(食肉內)의 허용직(許容直) 설족(設足), 투약법(投樂法)의 개선(改善) 그리고 식품위생산(食品衛生上)의 공해(公害)를 방지(防止)할 수 있을 것으로 생각되어 실시(實施)한 실험결과(實驗結果)를 요약(要約)하면 다음과같다. 1. Bacillus subtilis 균(菌)은 Sulfonamides에 대(對)하여 감수성(減受性)이 예민(銳敏)하여 0.05ppm까지 검출(檢出)할 수 있었다. 2. 혈액내(血液內) 잔류상(殘留狀)은 1회(回) 투약후(投藥後)에는 48시간(時間)까지 검출(檢出)할 수 있었고, 3회(回) 투약(投藥)한 구(區)에서는 3구(區) 모두 최종투약후(最終投藥後) 60시간(時間)까지 검출(檢出)할 수 있었다. 3. 각(各) 장기조직내(臟器組織內)에서는 3구(區) 다같이 최종투약후(最終投藥後) 5일(日)까지는 검출(檢出)할 수 있었으나 10일(日)에서는 담즙(膽汁)을 제외(除外)하고는 모두 검출(檢出)할 수 없었다. 4. 계란(鷄卵)의 난백(卵白)에서는 최종투약후(最終投藥後) 3구(區) 모두 5일(日)까지는 검출(檢出)이 가능(可能)하였으나 난황(卵黃)에서는 3구(區) 모두 5일(日)에서 흔적(痕迹)만 남기고 검출(檢出)할 수 없었다. 5. 담즙내(膽汁內)의 잔류(殘留) 유무(有無)는 각(各) 장기조직(臟器組織) 및 계란(鷄卵)의 식용(食用) 여부(與否)를 판정(判定)하는데 좋은 측도(測度)가 될 수 있을 것이다.

  • This study was carried out to explore the most sensitive and useful method for simultaneous determination of five sulfa drugs(sulfamethazine, sulfamerazine, sulfamonomethoxine, sulfadimethoxine, sulfaquinoxaline) in livestock productions(pork muscle, bovine muscle, chicken muscle, milk ) by HPLC with UV detector and reverse phase column. The results obtained were as follows:1. For mobile phase acetonitrile-0.01M ammonium acetate (23:77) showed more applicable sensitivity and retention times than acetonitrile-1% acetic acid(23:77). Thus acetonitrile-0.01M ammonium acetate(23:77) selected and applied to the modification test, from which it was found pH 6.75 was the most adequate. 2. Optimal wavelength of UV for SMT(sulfamethazine), SMR(sulfamerazine), SMM(sulfamonomethoxine), SD(sulfadimethoxine), and SQ(sulfaquinoxaline) were 266, 266, 265, 269 and 250nm, respectively, and that for simultaneous application it was 263nm. 3. The average recovery rate by extractant(chloroform, dichloromethane, chlorform+dich-loromethane) in the classic method was not significantly different(p>0.05) but that by chloroform higher than the others. Thus chloroform was found to be adequate as extractant in this classic method. 4. The average recovery rate was 86.5% by the MSPD(matrix solid phase disperse) method, which was significantly higher than that by the classic method(p<0.05). Also the recovery rates by method were significantly different(p<0.05) in accordance with sample and type of drug. The MSPD method was much superior to classic method on clean-up.

Sulfadimethoxine(SDM) 투여(投與) 가토(家兎)의 난소(卵巢), 갑상선(甲狀腺) 및 뇌하수체(雷下垂體)에 관한 병리학적(病理學的) 연구(硏究) (Pathological Studies on Ovaries, Thyroid Glands and Hypophyses of Rabbits Following Administration of Sulfadimethoxine)

  • In order to investigate the effects and acting mechanism on ovaries, thyroid glands and hypophyses of rabbits in short term administration of sulfadimethoxine (SDM) as medical dose, a total of 90 virgin albino rabbits (mean body weight, 1,362g) were selected at random and alloted to two groups. Rabbits in one group served as controls and the others were administered SDM of 50 mg/kg/day for 5 weeks, and then reared without medication for 4 weeks. Pathological changes of the three organs were observed each week for 9 weeks and the results obtained were summarized as follows: 1. Mean body weights of both groups manifested slow increasing tendency but mean hypophysis weights fluctuated throughout the experimental term. Mean ovary weights of experiments were decreased significantly from the 3rd to 6th week but mean thyroid weights of experiments were increased significantly from the 1st to 6th week compared with those of controls. 2. Many ovarian follicles of each developing stage showed follicular atresia accompanying atrophy or necrosis of oocytes and of disintegrated follicular cells. Theca interna cells and sudanophilic interstitial cells showed atrophy and diminished sudanophilic granules and also liquor folliculi were diminished. These changes icreased from the 1st week, remaining so for 5 weeks and returned to normal status in the 8th or 9th week. 3. The thyroid gland showed a typical hyperplastic goiter. Hypertrophic and hyperplastic epithelia follicular manifested cuboidal or columnar form showing tiny or small vacuoles in cytoplasm. The follicles showed atrophy and decreasing colloidal materials. Necrotic and regenerative changes were also present. The interfollicular vessels showed congestion and hemorrhage. These changes increased from the 1st week, remaining so for 5 weeks and returned to normal status in the 9th week. 4. The rates of differential cell counts of hypophyses revealed increase of basophils (gonadotrophs and thyrotrophs) and decrease of chromophobes. Basophils which had diminished granules stainable with HE, PAS and AF revealed hypertrophy, hyperplasia, and increasing of tiny or small vacuoles in cytoplasm. These changes increased from the 1st week, remaining so for 5 weeks and returned to normal status in the 8th or 9th week. As summarized above histologically, administration of SDM led thyrotrophs and gonadotrophs of pituitary glands to hyperactivity but revealed retrogressive and compensatory changes with functional disturbance in ovaries and thyroid glands. These changes were transitional and attributed to direct actions of the drugs on the ovaries and thyroid glands.

Dimethylformamide 수용액중에서의 Sulfa제-Cu 착화합물의 안정도정수 (Stability Constants of Copper Sulfa-Drug Complexes in Dimethylformamide Aqueous Solution)

  • Acid dissociation constants of sulfamethoxypyridazine, sulfadimethoxine, sulfamerazine, sulfathiazole and sulfadiazine, and stability constants for formation of copper chelate were calculated from their titration curves in 80% dimethylformamide with ionic strength 0.1 at $25{\deg}$ It was found that the acid dissociation constants (pKa) of sulfa-drugs were in the range of 9.33 - 10.05, and the stability constants (log $k_{1}$, $k_{2})$ of their copper chelates were in the range of 9.33-9.71.
