모델시스템에서 기름과 당이 분리대두단백 두부의 특성에 미치는 영향 (Effects of Oil and Sugar on SPI-Tofu Characteristics Under Model System)

  • Effect of addition of oil , sucrose, dextrin and oil-sucrose (1 : 1 w/w) mixture on SPI tofu was investigated. The characteristics measured were yield , water holding capacity , textural and organoleptic properties. THe SPI tofufwas prepared by coagulation of soyprotein isolate (SP) suspensino by CaCl$_2$ , CaSo$_4$ an dGDL , followed by compression . Addition of oil to SPI increased the tofu yield and water holding capacity, particulary for those tofu coagulated by CaCl$_2$. Eventhough dextrin addition decreased the yield, it showed the most improving effect on water holding capacity. The tofu prepared by CaSO$_4$coagulant resulted highest in yield and water holding capacity. Hardness was found to be decreased as the oil, sucrose and dextrin added more and adhesiveness, cohesiveness and guminess were also affected. The sensory evaluation showed the SPI tofu prepared by CaSO$_4$ and 10% addition of oil and sucrose mixture to be realtively high in hardness , elasticity and uniformity of the texture.

효소분해가 감쥬스의 이화학적 특성에 미치는 영향 (Effect on Enzymatic Hydrolysis on the Physicochemical Properties of Persimmon Juice)

  • 마쇄한 감 puree에 2종류의 상업적 다른 탄수화물 분해효소인 Viscozyme과 Celluclast를 처리하여 얻은 감쥬스의 추출수율, 점도, 색, 적정산도, 당에 미치는 영향을 조사하였다. Arabinase, cellulase, xylanase, hemicellulase와 ${\beta}-glucanase$의 효소활성을 가진 Viscozyme으로 $50^{\circ}C$에서 10분간 가수분해할 때 추출수율, L값, 환원당은 현저히 증가하였고 점도는 감소하였다. 감쥬스의 추출수율은 Viscozyme으로 처리시 42.8%에서 80%로 증가하는 반면 Celluclast는 73.3%로 증가하였다. 한편 관능적 특성중 감맛과 감내, 주홍색과 주황색은 Viscozyme으로 60분간 가수분해한 쥬스에서 크게 향상되었다.

열처리 조건이 포도즙의 착즙 수율 및 품질에 미치는 영향 (Effects of Heat Treatment on Yield and Quality of Grape Juice)

  • 충북 영동 지역에서 수확된 포도(Campbell Early)의 가열조건에 따른 착즙수율, 맛 색도의 변화를 조사한 결과 $60^{\circ}C$ 이하의 온도에서 가열 처리한 후 포도를 착즙한 경우에는 착즙수율이 60% 이하로 낮았으나, $80{\cim}90^{\circ}C$에서 $20{\cim}30$분간 가열하고 착즙한 경우에는 수율이 75% 이상이 되었고, 맛과 색깔도 가장 좋게 나타났다. 반면 $100^{\circ}C$에서 30분 가열한 경우에는 이취가 발생하였고, 포도즙의 색깔도 적색이 아닌 암갈색 계열로 바뀌었다. 또한 당도와 산도가 각기 다른 포도를 $80^{\circ}C$에서 30분간 가열한 후 착즙하였을 때 포도의 당도가 $14^{\circ}Brix$ 이상이면서 당도와 산도의 비(당도/산도)가 20 이상인 포도즙이 가장 맛이 좋게 평가되었다.

  • The present studies were carried out to obtain some basic informations of the breeding of tobacco varieties. Genetically divergent 8 varieties, 3 flue-cured, 2 burley and 3 sun-cured tobaccos, were used in half diallel cross. In order to analyze the heterosis, combining abilities, modes of inheritance and correlations for some agronomic and chemical characters, 8 parents and 28 $F_1$ were tested. The results obtained were summarized as follows: The percentages of heterosis for stem diameter, internode length and total sugar content in $F_1$ hybrid were 3.6%, 3.1% and 10.6%, whereas these for days to flower, total alkaloids and leaves per plant were -6.3%, -6.9% and -5.0%, respectively. Yield had significant positive genotypic correlations with plant height, days to flower and leaf length, but negative with internode length and total sugar content. It also had significant prositive phenotypic correlations with plant height, days to flower, leaves per plant, leaf length, leaf width and leaf shape index (Leaf length/leaf width). General (GCA) and specific combining abilities (SCA) for all characters of $F_1$ hybrid were significant. The effects of GCA were positive on yield, plant height, stem diameter, leaves per plant and days to flower of Burley 21. And those were positive on yield, leaf shape index and plant height, but negative on leaves per plant and total nitrogen of Hicks. The effects of SCA for yield and leaves per plant were greater than those of others on the combinations of Coker 139 and Burley type, respectively.

오탄당과 육탄당의 혼합용액에서 Pichia stipitis에 의한 에탄올 발효 (Ethanol Fermentation by Pichia Stipitis in a Mixture of Pentoses and Hexoses)

  • Pichia stiJitis CBS5776를 이용하여 6탄당과 5 탄당을 함유한 혼합당 배지에서의 에탄올 발효 특성 을 알아 보기 위한 실험을 수행하였다. 탄소원으로 사용된 당 중 에탄올 수율변에셔는 포도당, man­n nose, galactose, xylose 순으로 5탄당보다는 6탄당 이 우수한 기절로 판명되었다. 포도당의 에탄올 수 율이 O.376g ethanol/g glucose로 제 일 높았으며, 다음으로 mannose의 에탄올 수율은 O.326g etha­nnol/g mannose이였다. Spent sulfite liquor와 같은 조성을 가진 혼합당 함유 배지에서 6탄당인 mannose, 포도당이 먼저 이용된 후 galactose와 5탄당인 xylose가 이용됨을 알 수 있었다. Arabinose가 단일 탄소원으로 존재할 경우, 일부가 균체 성장에 이용되였으나 혼합당 배지 에셔는 전혀 이용되지 못하였다. 발효 결과 최대 에 탄올 농도는 $12.2g/\ell$ 이었고, 에탄올 생산 수율은 0.332g ethanol/g sugar이었다.

Production of Mannitol Using Leuconostoc mesenteroides NRRL B-1149

  • A process for the production of mannitol from fructose (5% to 25%) using Leuconostoc mesenteroides NRRL B-1149 was investigated. Fermentations were carried out In batch or fed-batch fermentations without aeration at 28$\^{C}$, pH 5.0. When 5% fructose was used In batch culture fermentation, the yield of mannitol was 78% of that expected theoretically. When the frurtose concentration was Increased to 10%, the yield dropped to 59.6% of the theoretical value. However, In the fed-batch culture, using 10% fructose, the yield was 81.9% of the theoretical value. In a 15% fruttose fed-batch culture, with 5% fructose being added initially and the other 10% fructose being added as a continuous supply the final yield was 83.7% of the theoretical yield. When 20% fructose was used In the same manner, the yield was 89.5% of theoretical yield.

Milk Yield and Its Fat Content as Affected by Dietary Factors: A-Rewiew

  • Milk yield and its composition is governed by level of nutrition and the composition of diet. Higher concentrate input improves milk yield, whereas its input at moderate levels improves yield of milk fat. High level of dietary protein improves dry matter intake and milk production, however, CP content above 14% has less advantage. Milk yield is enhanced by the feeding of cottonseed and soyabean meal, whereas milk fat increases by the supplementation of cottonseed. Dietary fat increases energy intake, production of milk and milk fat. Quality and quantity of feeds consumed affect fermentation patterns in rumen. Among the rumen metabolites, volatile fatty acids (VFA) content and propionate proportion have been related positively with milk yield, whereas proportion of acetate and butyrate have been related positively with milk fat content. Dietary carbohydrates through the source of sugar, starch, roughage and fibre affect VFA concentration in rumen. Therefore, concentration of volatile fatty acids could be altered to the advantage of consumer through judicious manipulation of diet.

Optimization of Culture Conditions for D-Ribose Production by Transketolase-Deficient Bacillus subtilis JY1

  • D-Ribose is a five-carbon sugar used for the commercial synthesis of riboflavin, antiviral agents, and flavor enhancers. Batch fermentations with transketolase-deficient B. subtilis JY1 were carried out to optimize the production of D-ribose from xylose. The best results for the fermentation were obtained with a temperature of $37^{\circ}C$ and an initial pH of 7.0. Among various sugars and sugar alcohols tested, glucose and sucrose were found to be the most effective for both cell growth and D-ribose production. The addition of 15 g/l xylose and 15 g/l glucose improved the fermentation performance, presumably due to the adequate supply of ATP in the xylose metabolism from D-xylulose to D-xylulose-5-phosphate. A batch culture in a 3.7-1 jar fermentor with 14.9 g/l xylose and 13.1 g/l glucose resulted in 10.1 g/l D-ribose concentration with a yield of 0.62 g D-ribose/g sugar consumed, and 0.25 g/l-h of productivity. Furthermore, the sugar utilization profile, indicating the simultaneous consumption of xylose and glucose, and respiratory parameters for the glucose and sucrose media suggested that the transketolase-deficient B. subtilis JY1 lost the glucose-specific enzyme II of the phosphoenolpyruvate transferase system.

맛과 향이 좋은 촉성재배용 딸기 '대왕' 품종의 육성 (Breeding of 'Daewang' Strawberry for Forcing Culture with Good Taste and Fragrance)

  • '대왕' 품종은 농촌진흥청 국립원예특작과학원에서 2010년도 육성한 촉성재배용 신품종이다. 맛과 향이 우수하면서 경도가 강한 품종을 육성하기 위해 2006년 고당도 품종인 '매향'과 고당도 품종인 '원교3111호'를 교잡하여 육성하였다. '대왕' 품종의 주요 특성을 보면 초형이 직립형이고 초세는 매우 강하다. 화방당 꽃 수는 12-15개 정도로 적고 휴면타파를 위한 저온요구시간은 50-100시간 정도로 촉성재배용으로 적합한 품종이다. 과형은 원추형이고 과색은 연적색이며 평균과중은 16-17g 내외의 중대과형 품종이다. 과실의 당도는 $11.1^{\circ}Brix$, 산도는 0.39%로 당산비가 적절하여 식미가 좋으며 특히 과육 내 육질이 치밀하여 씹는 맛이 우수하다. 또한 경도가 $18.2g{\cdot}mm^{-2}$로 아키히메의 $10.3g{\cdot}mm^{-2}$ 보다 $7.9g{\cdot}mm^{-2}$이 더 높아 봄철 늦게까지 수확이 가능하다. 그리고 상품과율은 대조품종인 '아키히메'보다 높았으나 전체적인 수량은 유의적인 차이를 보이지 않았다. 그러나 탄저병과 고온에 다소 약한 경향이 있다.

단수수 착즙액으로부터 에탄올 생산을 위한 반응표면분석법을 이용한 효모 발효조건 최적화 (Optimization of Fermentation Conditions for the Ethanol Production from Sweet Sorghum Juice by Saccharomyces cerevisiae using Response Surface Methodolgy)

  • Optimization of initial total sugar concentration of sweet sorghum juice, aeration time and aeration rate on ethanol production was performed by response surface methodology (RSM). The optimum conditions for ethanol production from concentrated sweet sorghum juice were determined as follows: initial total sugar concentration, 21.2 Brix; aeration time, 7.66h; aeration rate, 1.22 vvm. At the optimum conditions, the maximum ethanol yield was predicted to be 91.65% by model prediction. Similarly, 92.98% of ethanol yield was obtained by verification experiment using optimum conditions after 48 h of fermentation. This result was in agreement with the model prediction.
