Relationship between Seed Size and Seed Vigor in Soybean. (콩의 종실크기와 종자활성 간의 관계)

  • This experiment was carried out to investigate the difference of seed vigor according to seed size, and the physiological aspects and physico-chemical phenomena related with seed vigor in soybean cultivars. Portion of seed coat and radicle to entire seed was much higher in cultivars with small seed than with large. Seed coat rate ranged 7.9% to 9.9%, and radicle 2.5% to 3.3% in small seed group, Whereas in large seed, seed coat rate did 5.5-6.4% and radicle, 1.5 to 2.1%. After accelerated aging treatment, there are significant difference in germination ability between seed size. The germination rate after aging ranged 47 to 80% in cultivars with small seed, but in large seed, only 14 to 24%. After seed was carried out dehydration in incubator at 25 after soaking for 6 hours, the moisture content of seed in drying for 12 hour was 25.5% in small seed, while it was 51% in large seed. Electrical conductivity, leaching soluble nitrogen and sugar content were higher in large seed cultivars. Besides, cotyledon damage after soaking was occurred frequently in large seed cultivars, and seed vigor within same cultivars was higher in small seed than large.

Characteristics of Soycurd-forming Lactic Acid Bacteria that Produce Gammaaminobutyric Acid (GABA) from Kimchi (김치로부터 GABA를 생산하는 커드 형성 젖산균의 특성)

  • Lactobacillus sakei 383, which showed the highest GABA content in fermented soycurd, survived in artificial gastric fluid (pH 3.0) up to 3 h, and the survival rate was 88%. L. sakei 383 was tolerant to bile juice during incubation in MRS broth with 0.3% oxgall, and the survival rate was 99%. The survival ratio of L. sakei 383 was high in media containing less than 6% NaCl. L. sakei 383 produced an antibacterial substance against various pathogens, including Listeria monocytogenes, Bacillus cereus, Staphylococcus aureus, Escherichia coli, and Salmonella typhi. The quality characteristics of soycurd fermented with L. sakei 383 were measured during the fermentation period. The viable cell number reached a peak ($10^{11}CFU/ml$) 36 h after fermentation and then slowly decreased. According to the fermentation time of L. sakei 383, the acidity of soycurd increased and the pH decreased until 12 h, and they were maintained thereafter. The moisture, crude ash, crude protein, crude fat, and crude fiber content was 94.88, 0.22, 2.38, 1.16, and 0.03%, respectively. The content of total and reducing sugar was comparatively higher in the soycurd fermented with L. sakei 383 than in nonfermented soycurd. The essential and nonessential amino acid content was 11.2 and 38.65 mg/100 g.

Quality characteristics of tangerine peel Soksungjang prepared from different koji strains (균주를 달리하여 제조한 귤피 속성장의 특성)

  • This study was performed to provide information for improving the quality characteristics of tangerine peel Soksungjang. Tangerine peel Soksungjangs were prepared with 10% tangerine peel powder by addition of different koji such as Rhizopus oligosporus (RS, RTS) and Aspergillus oryzae (AS, ATS). As a result, the moisture content was not changed and crude protein content decreased during fermentation in all samples. The pH of RS (Soksungjang manufactured with Rhizopus oligosporus koji) and RTS (Soksungjang manufactured with Rhizopus oligosporus koji and 10% tangerine peel powder) was not changed, whereas pH of AS (Soksungjang manufactured with Aspergillus oryzae) and ATS (Soksungjang manufactured with Aspergillus oryzae koji and 10% tangerine peel powder) decreased rapidly after 10 days of fermentation. The acidity values of all the samples increased during fermentation. The major free sugars were glucose, fructose, and maltose; while, the major organic acids were citric acid, succinic acid, acetic acid, and lactic acid in all the samples. The amino nitrogen content and protease activity were highest in the AS and ATS. Conversely, the reducing sugar content and ${\alpha}$-amylase activity were highest in the RS and RTS. The major free amino acids in all samples were glutamic acid, lysine, leucine, and arginine. Sixty kinds of volatile flavor compounds were identified from all samples; the majority of which were 21 alcohols, 14 hydrocarbons, 5 esters, and others identifies as acids, ketones, and aldehydes.

Effects of Electron Beam Irradiation and High-Pressure Treatment with Citrus Peel Extract on the Microbiological, Chemical and Sensory Qualities of Marinated Chicken Breast Meat

  • Chicken breast meat samples were injected with marinade solution (salt, sugar, phosphate, monosodium glutamate, and nucleic acid) with or without 2% citrus peel extract (CPE), and then a subset were irradiated with a 1 or 2 kGy electron beam (EB) and/or subjected to high-pressure (HP) at 300 or 400 MPa. The initial total aerobic bacterial (TAB) count of the control sample was 4.57 log CFU/g and reached 7.17 log CFU/g after 3 days of storage at $4^{\circ}C$. The 2 kGy EB reduced the TAB count to 4.61 log CFU/g after 7 days. The 400 MPa HP treatment was also effective in reducing the TAB count, but the effect was slightly less than that noted with the 2 kGy EB. The CPE, in combination with the EB and HP, decreased the TAB count by 1.71 and 1.32 log CFU/g at the initial stage and further decreased the count during storage. The 2 kGy EB and the HP (300 and 400 MPa) increased the thiobarbituric acid-reactive substances value, whereas the CPE did not show an antioxidative effect. The EB and HP caused no difference in the sensory qualities. In contrast, the CPE decreased all sensory qualities tested. Sensory panelists commented that the samples with CPE were not in the "rejection" category but were "unfamiliar" for chicken breast meat. In conclusion, the EB was more effective than HP in improving the microbial quality of marinated chicken breast meat. The use of CPE in the marinade solution may synergistically increase the shelf life; however, it is necessary to develop an appropriate formulation to ensure that the sensory qualities are maintained.

Influence of Repeated Application of Chlorotharonil and Cellulose on the Bacterial Population in Soil Suspension Culture (토양현탁액중(土壤懸濁液中)에서 Cellulose와 살균제(殺菌濟) Chlorotharonil연용(連用)이 세균(細菌)의 밀도변화(密度變化)에 미치는 영향(影響))

  • This study was conducted to find out the influence of single and/or repeatedly combined application of cellulose and chlorotharonil(TPN) on bacterial population and dissipation of these chemicals by microorganism in the soil suspension culture. 1. The number of total bacteria and Gram-negative bacteria were rapidly increased for 1st week, after the time, decreased in the single application but maintained in repeatedly combined application for 5th week and that were higher at combined application of TPN and cellulose than at single application of these. 2. TPN-degrading bacteria were gradually increased by the repeated application of TPN, and elevated the increasing extents of those with the repeatedly combined application of cellulose with TPN. 3. Cellulose-degrading bacteria showed the peak at 1st week, and was increased in the repeated application of cellulose, but the bacteria was decreased in the repeatedly application of TPN with cellulose. 4. The dissipation of TPN were faster at the combined application of both than at the single application of TPN, wheras that was delayed by the repeatedly combined application. 5. The content of reducing sugar was rapidly increased at 1st week and after 4th week except for the TPN single application, that was higher in the combind application than the cellulose single, but there was no difference between them.

Quality Evaluation and Residual Pesticides of Lettuce during Growth after Transplanting (정식(定植) 후 양상추의 품질평가 및 잔류농약 분석)

  • Lettuce (Lactuca Sativa L.) is the most popular salad vegetable crop The purpose of this study was to analyze the quality of lettuce, and residual pesticides in this vegetable, during growth after transplanting. The eatable weight was 40% of the total weight at the commencement of transplanting, but had doubted at harvest after transplanting. As lettuce grew after transplanting, the pH, the sugar level, and the moisture content increased slightly, and the color tended towards yellow rather than greenThe SPDA chlorophyll measurement was 16.84 at 42 day post-ransplantation, and increased to 26.44 at 67 day after transplanting. The contents of vitamin C were 2.5mg/100g wet weight at 60 d after transplantation, but decreased to 2.2mg/100g wet weight 67 day post-transplantation. Mineral (Ca, Fe, K, Mg, and Na) levels rose after transplantation, and attained their highest levels 60 day post-transplantation. The lettuce crop was analyzed for residues of each of 48 pesticides nt 54 after transplantation, and the only such chemical detected was chlorthalonil(0.03 0.25mg/kg wet weight).

Effect of Suboptimal Nutritional Status on Mineral Uptake and Carbohydrate Metabolism in Tomato Plants

  • A suitable supply of mineral elements into shoot via a root system from growth media makes plants favorable growth and yield. The shortage or surplus of minerals directly affects overall physiological reactions to plants and, especially, strongly influences carbohydrate metabolism as a primary response. We have studied mineral uptake and synthesis and translocation of soluble carbohydrates in N, P or K-deficient tomato plants, and examined the interaction between soluble carbohydrates and mineral elements. Four-weeks-old tomato plants were grown in a hydroponic growth container adjusted with suboptimal N ($0.5mmol\;L^{-1}\;Ca(NO_3)2{\cdot}4H_2O$ and $0.5mmol\;L^{-1}\;KNO_3$), P ($0.05mmol\;L^{-1}\;KH_2PO_4$), and K ($0.5mmol\;L^{-1}\;KNO_3$) for 30 days. The deficiency of specific mineral element led to a significant decrease in its concentration and affected the concentration of other elements with increasing treatment period. The appearance of the reduction, however, differed slightly between elements. The ratios of N uptake of each treatment to that in NPK sufficient tomato shoots were 4 (N deficient), 50 (P deficient), and 50% (K deficient). The P uptake ratios were 21 (N deficient), 19 (P deficient), and 28% (K deficient) and K uptake ratios were 11 (N deficient), 46 (P deficient), and 7% (K deficient). The deficiency of mineral elements also influenced on carbohydrate metabolism; soluble sugar and starch was substantially enhanced, especially in N or K deficiency. In conclusion, mineral deficiency leads to an adverse carbohydrate metabolism such as immoderate accumulation and restricted translocation as well as reduced mineral uptake and thus results in the reduced plant growth.

Effects of Saline Irrigation Water on Crop Growth in Strawberry and Red Radish (딸기 및 적환무의 관개용수 염도수준에 따른 생육영향 분석)

  • Since the salinity of irrigation water is a critical constraint to the production of certain vegetable crops, salinity was considered as one of the most important factors of irrigation water. The purpose of this study were to monitor and assess the effects of saline irrigation water on strawberry and red radish growth in protected cultivation. One control and three treatments, which were differentiated according to the level of salinity in irrigated water, were employed for each vegetable to assess the effects of the irrigation with saline water. Monitoring has shown that using irrigation water with salinity above a certain level causes excessive accumulation of sodium (Na+) in both strawberry and red radish. Increased Na+ content was analyzed to be able to decrease the sugar content in strawberry. In addition, the salinity higher than the threshold level of irrigation water was found to reduce the growth and yield of strawberry and red radish. This study could contribute to suggest criteria for safe use of saline water in protected cultivation, although long-term monitoring is needed to get more representative results.

Complete Biotransformation of Protopanaxatriol-Type Ginsenosides in Panax ginseng Leaf Extract to Aglycon Protopanaxatriol by β-Glycosidases from Dictyoglomus turgidum and Pyrococcus furiosus

  • Aglycon protopanaxatriol (APPT) has valuable pharmacological effects such as memory enhancement and tumor inhibition. ${\beta}$-Glycosidase from the hyperthermophilic bacterium Dictyoglomus turgidum (DT-bgl) hydrolyzes the glucose residues linked to APPT, but not other glycoside residues. ${\beta}$-Glycosidase from the hyperthermophilic bacterium Pyrococcus furiosus (PF-bgl) hydrolyzes the outer sugar at C-6 but not the inner glucose at C-6 or the glucose at C-20. Thus, the combined use of DT-bgl and PF-bgl is expected to increase the biotransformation of PPT-type ginsenosides to APPT. We optimized the ratio of PF-bgl to DT-bgl, the concentrations of substrate and enzyme, and the reaction time to increase the biotransformation of ginsenoside Re and PPT-type ginsenosides in Panax ginseng leaf extract to APPT. DT-bgl combined with PF-bgl converted 1.0 mg/ml PPT-type ginsenosides in ginseng leaf extract to 0.58 mg/ml APPT without other ginsenosides, with a molar conversion of 100%. We achieved the complete biotransformation of ginsenoside Re and PPT-type ginsenosides in ginseng leaf extract to APPT by the combined use of two ${\beta}$-glycosidases, suggesting that discarded ginseng leaves can be used as a source of the valuable ginsenoside APPT. To the best of our knowledge, this is the first quantitative production of APPT using ginsenoside Re, and we report the highest concentration and productivity of APPT from ginseng extract to date.

Changes of Free Sugars, Non-Volatile Organic Acids and Fatty Acids in Flue-cured Leaf Tobacco during Aging (황색종 잎담배의 후숙과정 중 유리당, 비휘발성 유기산 및 지방산의 변화)

  • This study was carried out to investigate the changes of free sugars, non-volatile organic and fatty acids in flue-cured leaf tobacco during aging. The threshed leaf tobacco(B1O and C1L) produced in 2002 crop year was aged for 21 month in warehouse of Oc-Cheon Leaf Tobacco Processing Factory. The leaf tobacco were sampled at three month intervals for analysis of free sugars, non-volatile organic and fatty acids. The major free sugars of flue-cured were glucose and fructose regardless of tobacco grades. After aging period of 21 months, the contents of glucose and fructose showed a tendency to slightly decrease, and there was no appreciable change in the contents of sucrose in B1O and C1L grades. The major compound of non-volatile organic acid and fatty acid were malic, oxalic and citric, linolenic, linoleic and palmitic acid regardless of tobacco grade. After aging period of 21 months, the contents of malic, malonic, oxalic and fumaric acid showed a tendency to slightly decrease, whereas succinic acid was slightly increased, and maleic acid was not changed in B1O and C1L grades. The decreases in fatty acid contents in B1O and C1L grade tobacco loaves after 21 month aging were 16.5% and 9.8%, respectively. The decreases in linolenic acid contents in two grades were the highest, showing 22.1% and 12.0% reduction after 21 month aging.
