Influence of Gluconeogenic Phosphoenolpyruvate Carboxykinase (PCK) Expression on Succinic Acid Fermentation in Escherichia coli Under High Bicarbonate Condition

  • The effects of amplifying the gluconeogenic phosphoenolpyruvate carboxykinase of Escherichia coli ($pck_{Ec}$) on succinic acid production in E. coli were examined under anaerobic condition. No significant increase in succinic acid production was observed in E. coli overexpressing the $pck_{Ec}$ gene without supplementing $NaHCO_{3}$ or $MgCO_{3}$. On the other hand, succinic acid production was enhanced as the $NaHCO_{3}$ concentration was increased. When 20 g/l of $NaHCO_{3}$ was added, succinic acid production in recombinant E. coli overexpressing PCK was 2.2-fold higher than that observed in the wild-type strain. It was concluded that the gluconeogenic $pck_{Ec}$ overexpression enabled E. coli to enhance succinic acid production only under the high bicarbonate supplementation condition.

  • In this study, succinic acid production using fumaric acid fermentation broth was investigated. We tried to produce fumaric aicd from glucose by Rhizopus oryzae and then convert fumaric acid fermentation broth into succinic acid by Enterococcus faecalis RKY1. Conversion ratio of succinic acid was more than 0.90 g/g-fumaric acid. Furthermore, we optimized conditions of conversion from fermentation broth. As a result, when fumaric acid fermentation broth for succinic acid production was employed, we could decrease the amount used of glycerol and yeast extract.

  • The effects of culture conditions on succinic acid production and its possible scale-up have been studied. Mannheimia succiniciproducens LPK7, engineered for enhanced production of succinic acid and reduced by-product secretion, was used for the experiments. Mannheimia succiniciproducens LPK7 is a knock-out strain of wild type deficient in the ldhA, pflB, and pta-ackA genes, and is derived from Mannheimia succiniciproducens MBEL55E. Process optimization of factors including optimal temperature, pH, carbon source, and nitrogen source was performed to enhance the production of succinic acid in flasks. To observe scale-up effects, batch fermentation was carried out at various working volumes. At a working volume of 7.0 l, the final succinic acid concentration and yield were 15.4g/l and 0.86g/g. This result shows similar amount of succinic acid obtained in lab-scale fermentation, and it is possible to scale up to larger fermentors without major problems.

Effect of redox potential on the production of succinic acid by metabolically engineered Escherichia coli

  • Recombinant Escherichia coli NZN111 ($F-{\Delta}pfl::Cam$ ldhA::Kan) harboring pTrcML, a plasmid containing the E. coli malic enzyme gene, produced considerable amount of malic acid along with the desired product succinic acid. This seemed to be due to the unmatched redox states between glucose and succinic acid. Therefore, a more reduced carbon substrate sorbitol was examined for the possibility of matching the potential during succinic acid production. When NZN111 (pTrcML) was cultured in LB medium containing 20 g/L sorbitol under $CO_2$ atmosphere, 10 g/L of succinic acid was produced. The apparent yield of succinic acid was 1.1 g succinic acid per g sorbitol, which is 85% of the maximum theoretical yield.

  • An internal membrane bioreactor system was employed for continuous succinic ac id production from glucose in order to prove its performance and practicality. Succinic acid-producing Anaerobiospirillum succiniciproducens required more $CO_2$ for the proper growth and succinic acid production in cell recycled continuous culture than in batch culture. The maximum productivity obtained in cell recycled continuous culture was about 3.3 g/L-h which was ca. 3.3 times higher than that obtained in batch culture.

  • Modulation of the catabolic PEP-pathway of Anaerobiospirillum succiniciproducens was tried using some enzymatic inhibitors such as gases and chemicals in order to enhance succinic acid production. 10$\%$ CO increased the succinic acid/acetic acid (S/A) ratio but inhibited growth as well as production of succinic and acetic acid. Hydrogen gas also increased the S/A ratio and inhibited the synthesis of pyruvate: ferredoxin oxidoreductase when used in mixture with $CO_2$, Catabolic repression by acetic, lactic and formic acid was not recognized and other modulators such as glyoxylate, pyruvate derivatives, arsenic salt, phosphate and sulfate were shown not to be effective. Magesium carbonate was shown effective for repressing acetate production. Palmitic acid, myristic acid and phenylalanine did not affect acetate production but carprylic acid completely inhibited growth.

  • Statistical medium optimization has been carried out for the production of succinic acid in anaerobic fermentations of Actinobacillus succinogenes. Succinic acid utilized as a precursor of many industrially important chemicals is a fourcarbon dicarboxylic acid, biosynthesized as one of the fermentation products of anaerobic metabolism by A. succinogenes. Through OFAT (one factor at a time) experiments, corn steep liquor (CSL), a very cheap agricultural byproduct, was found to have significant effects on enhanced production of succinic acid, when supplemented along with yeast extract. Hence, using these factors including glucose as a carbon/energy source, interactive effects were investigated through $2^n$ full factorial design (FFD) experiments, showing that the concentration of each component (i.e., glucose, yeast extract and CSL) should be higher. Further statistical experiments were conducted along the steepest ascent path, followed by response surface method (RSM) in order to find out optimal concentrations of each constituent. Consequently, optimized concentrations of glucose, yeast extract and CSL were observed to be 180 g/L, 15.08 g/L and 20.75 g/L respectively (10 g/L of $NaHCO_3$ and 100 g/L of $MgCO_3$ to be supplemented as bicarbonate suppliers), with the estimated production level of succinic acid to be 92.9 g/L (about 3.5 fold higher productivity as compared to the initial medium). Notably, the RSM-estimated production level was almost similar to the amount of succinic acid (92.9 g/L vs. 89.1 g/L) produced through the actual fermentation process performed using the statistically optimized production medium.

Succinic Acid Production by Continuous Fermentation Process Using Mannheimia succiniciproducens LPK7

  • To achieve a higher succinic acid productivity and evaluate the industrial applicability, this study used Mannheimia succiniciproducens LPK7 (knock-out: ldhA, pflB, pta-ackA), which was recently designed to enhance the productivity of succinic acid and reduce by-product secretion. Anaerobic continuous fermentation of Mannheimia succiniciproducens LPK7 was carried out at different glucose feed concentrations and dilution rates. After extensive fermentation experiments, a succinic acid yield and productivity of 0.38 mol/mol and 1.77 g/l/h, respectively, were achieved with a glucose feed concentration of 18.0 g/l and $0.2\;h^{-1}$ dilution rate. A similar amount of succinic acid production was also produced in batch culture experiments. Therefore, these optimal conditions can be industrially applied for the continuous production of succinic acid. To examine the quantitative balance of the metabolism, a flux distribution analysis was also performed using the metabolic network model of glycolysis and the pentose phosphate pathway.

Biological conversion of biomass to succinic acid

  • Batch cultivations of Anaerobiospirillum succiniciproducens have been systematically studied for the economical production of succinic acid from wood hydrolysate with corn steep liquor(CSL) as a nitrogen source. CSL was found to be an alternative complex nitrogen source for A. succiniciproducens when glucose and wood hydrolysate were used as carbon sources. Compared with polypeptone and/or yeast extract, CSL had similar effects on fermentation performance such as succinic acid yield and a ratio of succinic acid to acetic acid in the fermentation of wood hydrolysate as well as glucose. This means that succinic acid can be produced more economically from wood hydrolysate and CSL than relatively expensive carbon and nitrogen sources. Besides its low cost, the alternative medium served as a green technology for succinic acid production because it gives a net-zero effect on global warming.

숙신산 생합성을 위한 Enterococcus sp. RKY1의 분리와 특성 (Isolation and Characterization of the Enterococcus sp. RKY1 for Biosynthesis of Succinic Acid.)

  • 재생가능한 대체원료로서 가치있는 탄소 4개의 디카르복실산인 숙신산은 현재 석유화학공정에 의해 상업적으로 생산되고 있으나, 최근 생물화학공정에 의한 숙신산 대량생산 공정개발을 위한 연구가 집중되고 있다. 본 연구에서는 전자공여체로 포도당과 전자수용체로서 푸마르산을 이용하여 숙신산으로의 생물전환을 연구하였다. 먼저 고수율로 푸마르산을 숙신산으로 생물전환하는 통성박테리아 Enterococus sp. RKY1 KCTC 8890P를 분리하여 그 특성을 조사하였다. 초기 푸마르산 농도 40, 70, 100g/L으로 회분식 배양한 결과 푸마르산의 농도가 증가함에 따라 숙신산 생성량이 증가하였다. 초기 푸마르산 농도 70 g/L에서 평균비생산속도, 평균부피생산속도및 몰수율은각각 0.64 g/g.h, 4.87 g/L.h, 96.5%이었고 최대 숙신산 농도는 초기 푸마르산 농도 100 g/L에서 발효 20시간에 89%의 몰 수율로 88.9 g/L가 얻어졌다.
