Taxonomy of Ulva causing blooms from Jeju Island, Korea with new species, U. pseudo-ohnoi sp. nov. (Ulvales, Chlorophyta)

  • Several species classified to the genus Ulva are primarily responsible for causing green tides all over the world. For almost two decades, green tides have been resulted in numerous ecological problems along the eastern coast of Jeju Island, Korea. In order to characterize the species of Ulva responsible for causing the massive blooms on Jeju Island, we conducted DNA barcoding of tufA and rbcL sequences on 183 specimens of Ulva from eight sites on Jeju Island. The concatenated analysis identified five bloom-forming species: U. australis, U. lactuca, U. laetevirens, U. ohnoi and a novel species, U. pseudo-ohnoi sp. nov. Among them, U. australis, U. lactuca, and U. laetevirens caused to the blooms coming mainly from the substratum. U. ohnoi and U. pseudo-ohnoi sp. nov. were causative the free-floating blooms. Four species, except U. australis, are characterized by marginal teeth. A novel species, U. pseudo-ohnoi sp. nov., is clearly diverged from the U. lactuca, U. laetevirens, and U. ohnoi clade in the concatenated maximum likelihood analysis. Accurate species delimitation will contribute to a management of massive Ulva blooms based on this more comprehensive knowledge.

혐기성 생물막 반응기의 기질 농도 변화에 따른 생물막 부착 특성 관찰

  • 수영 하수처리장의 소화조에서 농축조로 보내지는 혐기성 슬러지를 탈기된 증류수와 1:1 로 희석한 후 연속식 반응기에 주입한 후 합성폐수의 농도를 변화시켜 공급한 결과, 담체 표면적당 생물막 부착량은 23 일 이후에 그 양이 일정해지는 경향을 나타냄을 알 수 있었다. SEM 사진에 의해서는 미생물 형태를 명확히 파악할 수 없고 특히 메탄 형성 미생물의 존재를 규명하기 어려워 형광현미경을 이용하였다. 형광현미경에 의한 담체에 부착된 미생물의 관찰 결과 메탄생성균을 확인할 수 있었고, 시간이 증가됨에 따라 일정하게 증가됨을 볼 수 있었고, 23 일 이후엔 농도 변화엔 큰 영향이 없이 일정함을 알 수 있었다.

Susceptibility of Nereocystis luetkeana (Laminariales, Ochrophyta) and Eualaria fistulosa (Laminariales, Ochrophyta) spores to sedimentation

  • The establishment of algal spores plays an essential role in adult kelp distribution and abundance patterns. Sedimentation is a key variable regulating algal spore settlement and success, possibly controlling species-specific dominance $in$ $situ$. Laboratory experiments were used to determine spore attachment and survival rates of two Alaskan canopy-forming kelps, $Nereocystis$ $luetkeana$ (K. Mertens) Postels & Ruprecht and $Eualaria$ $fistulosa$ (Postels & Ruprecht) M. J. Wynne, to various types of sediment loading. Spore attachment for both species was significantly and similarly affected by three sediment treatments: suspended particles; settled sediment covering the substratum; and smothering of attached spores by settling sediment. Spore attachment decreased by approximately 90% at 420 mg sediment $L^{-1}$, the highest sediment load tested here, under all three treatments for both species. These results suggest that increases in sedimentation may constrain the success of the spore stages, but sediment does not seem to be a likely factor explaining species-specific distribution patterns. However, while sedimentation affected spores of both species similarly, timing of spore release in relation to times of maximum sediment load in the water might differ for different species, possibly explaining kelp species distribution patterns.

  • The science of toxicology is the understanding of the mechanisms by which exogenous agents produce deleterious effects in biological systems. The actions of chemicals such as drugs are ultimately exerted at the cellular and gene levels. Over the past decade. several in vitro alternative methods such as cultured cells for assessing the toxicity of various xenobiotics have been proposed to reduce the use of animals. In this workshop three advanced methods will be presented. These methods are novel important models for toxicologic studies. Dr. Tabuchis group has establishcd two immortalized gastric surface mucosa cell lines from the pminary cultore of gastric fundic mucosal cells of adult transgenic mice harboring a temperature sensitive simian virus 40 large T-anugen gene. As the immortalized cell lines of various tissues possess unique characteristics to maintain their normal functions for several months, these cell lines are extremely useful for not only toxicity testing but also pharmacological screening in new drug development. Professor Funatsu have studied the formation of spherical multicelluar aggregates of adult rat hepatocytes(spheroid) having tissue like structure. The sphcroid shown thre is a prototype module of an artificial liver support system. Thus, the urea synthesis activity of the artificial liver was maintained at least to days in 100% rat blood plasma. Dr. Takezawa and his coworkers have developed a novel culture system of multicellular spheroids considered 〃organoids〃 by utilizing a thermo-responsive polymer as a substratum of anchorage dependent cells. His final goal is to reconstitute the organoids of various normal organs, e.g., liver, skin etc. and also abnormal deseased organs such as tumor.

  • The development of routine techniques for the isolation and in vitro maintenance of conducting airway epithelial cells in a differentiated state provides an ideal model to study the factors involved in the regulation of the expression of mucocilicary differentiation. Several key factors and conditions have been identified. These factors and conditions include the use of biphasic culture technique to achieve mucociliary differentiation and the use of such stimulators, the thickness of collagen gel substratum, the calcium level, and vitamin A, and such inhibitors, the growth factors EGF and insulin, and steroid hormones, for mucous cell differentiation. Using the defined culture medium, the life cycle of the mucous cell population in vitro was investigated. It was demonstrated that the majority of the mucous cell population in primary cultures is not involved in DNA replication. However, the mucous cell type is capable of self-renewal in culture and this reproduction is vitamin A dependent. furthermore, differentiation from non-mucous cell type to mucous cell type can be demonstrated by adding back a positive regulator such as vitamin A to the “starved” culture. Cell kinetics data suggest that vitamin A-dependent mucous cell differentiation in culture is a DNA replication-independent process and the process is inhibited by TGF-${\beta}$1.

Temporal and spatial variation in the distribution of life history phases of Chondrus crispus (Gigartinales, Rhodophyta)

  • Thirty populations of Chondrus crispus Stackhouse from Nova Scotia were collected during the years 1993 to 2011. Taken from estuaries, wave exposed open coasts, high intertidal rock pools and shallow subtidal habitats, the populations were evaluated for relative abundance of tetrasporophytic and gametophytic life history phases. Over 2,800 thalli were characterized using the resorcinol-acetal test to distinguish the kappa- and lambda-carrageenan containing fronds of gametophytes and tetrasporophytes, respectively. These populations had $77{\pm}5%$ gametophytes (mean ${\pm}95%$ confidence interval), with most populations having gametophyte : sporophyte ratios ranging from 2 : 1 to 9 : 1. No population had a dominance of tetrasporophytes, although two populations had 1 : 1 ratios. A meta-analysis of our data along with previously published accounts showed no significant changes in gametophyte dominance with respect to hypothesized gradients of wave exposure, salinity, or water depth. Significant changes occurred in ratios at five sites where replicate sampling occurred in different years. We conclude that C. crispus in Maritime Canada has a natural ratio of 3 : 1 or greater in stable conditions, and that lower ratios represent recovery from disturbance in which bare substratum is created that is subsequently colonized by carpospores from remaining gametophytic thalli.

Scattering of torsional surface waves in a three layered model structure

  • In this article, a comparative study has been made to investigate the scattering behaviour of three layered structure model on torsional surface wave. For such model intermediate layer is taken as fiber reinforced composite, resting over a dry sandy Gibson substratum and underlying by different anelastic media. We consider two distinct mediums for topmost layer. In the first case, topmost layer has been taken as fluid saturated homogeneous porous layer, while in the second case the fluid saturated porous layer has been replaced by a transversely isotropic layer. Simple form expression for the secular equation of torsional surface wave has been worked out in both the cases by executing specific boundary conditions, which comprises Whittaker's function and its derivative, for imminent result that have been elaborated asymptotically. Some special cases have been constituted which are in excellent compliance with recorded literatures. For the sake of comparative study, numerical estimation and graphical illustration have been accomplished to identify the effects of the width ratio of the layers, Biot's gravity parameter, sandy parameter, porosity parameter and other heterogeneity parameters corresponding to the layers and half spaces, horizontal compressive and tensile initial stress on the phase velocity of torsional surface wave.

Long-term Environmental Changes: Interpretations from a Marine Benthic Ecologist's Perspective (II) -Eutrophication and Substratum Properties

  • Chemical oxygen demand (COD), phytoplankton cell number and chlorophyll-a concentration (Chl-a), sediment mean grain size and ignition loss were studied to determine their temporal trends in the study area. Historical data of COD, cell number and Chl-a were gathered from the late 1960s or early 1980s to 1997, and trends in temporal domain were obtained from a simple regression. Sediments for grain size and ignition loss (as organic contents in sediments) were sampled from the Chokchon macrotidal flat bimonthly from September 1990 to November 1996, and were analyzed using the decomposition method of time series analysis. In general, the first three data showed increasing trends based on regression analysis. The trends of sediment grain size fluctuated in a neutral pathway while those of ignition loss yielded no increasing pattern. In contrast with the suggestions from Ahn and Choi (1998) who reported a coarsening variation in sediment grain size to be a cause of the directional and remarkable changes of macrofaunal communities in this area, we could not find such a corresponding variation pattern from our samples. In diagnosing eutrophication, a paradoxical phenomenon was encountered between the trends in water column (COD, cell number and Chl-a) and sediment (ignition loss) data. In this paper, we inferred the possible processes that produce the discrepancy. Some explanations and biological responses to eutrophication were predicted and discussed.

Early Stage of Algal Succession on Artificial Reefs at Muronohana, Ikata, Japan

  • Different substrata fixed as the roof (for shadow) on artificial iron reef had been developed as a tool for valuable fishery resources. The experiment was set up on a sandy bottom substratum at 8m depth in Muronohana, Ikata, Japan. Within one month of placement of the plates, diatoms dominated the experimental plates with a coverage of $100\%$ approximately. Enteromorpha intestinalis and Colpomenia sinuosa dominated within three months after the placement. E. intestinalis coverage on substrata was estimated $7\%$ on the plate shaped iron bar, $12-14\%$ on concrete plates, $18\%$ on the plate fixed pebble, $61\%$ on the plate to accumulated wood, and 80-100% on the steel materials plate. Whereas after four months of placement, C. sinuosa coverage on plates became $1-36\%$ on different plates, and $100\%$ on the plate to accumulated wood. The differences in E. intestinalis and C. sinuosa colonization on the different substrata were probably due to variations in their surface roughness influencing the settlement of zoospores, and thus gametophyte. development. After five months of placement, the above two species slowly disappeared.

Inference Models for Tidal Flat Elevation and Sediment Grain Size: A Preliminary Approach on Tidal Flat Macrobenthic Community

  • A vertical transect with 4 km length was established for the macrofaunal survey on the Chokchon macrotidal flat in Kyeonggi Bay, Incheon, Korea, 1994. Tidal elevation (m) and sediment mean grain size $(\phi)$ were inversely predicted by the transfer functions from the faunal assemblages. Three methods: weighted average using optimum value (WA), tolerance weighted version of the weighted average (WAT) and maximum likelihood calibration (MLC) were employed. Estimates of tidal elevation and mean grain size obtained by using the three different methods showed positively corresponding trends with the observations. The estimates of MLC were found to have the minimum value of sum of squares due to errors (SSE). When applied to the previous data $(1990\sim1992)$, each of three inference models exhibited high predictive power. This result implied there are visible relationships between species composition and faunas' critical environmental factors. Although a potential significance of the two major abiotic factors was re-affirmed, a weak tendency of biological interaction was detected from faunal distribution patterns across the flat. In comparison to the spatial and temporal patterns of the estimates, it was suggested that sediment characteristics were the primary factors regulating the distribution of macrofaunal assemblages, rather than tidal elevation, and the species composition may be sensitively determined by minute changes in substratum properties on a tidal flat.
