Physical and Chemical Properties of Coal Fly Ash Ball Substrates, the Salt Accumulation and the Effects of Washing Out Salt with Water (석탄회성형배지(Ash Ball)의 이화학적 특성과 염류집적 및 제거효과)

  • Physical and chemical properties, the salt accumulation and leaching of salt by water of coal fly ash ball (ash ball) were evaluated in comparison with perlite and granule rockwool (rockwool). Bulk density, particle density, solid phase, and porosity of ash ball were 0.93 g.cm$^{-3}$ , 2.29 g.cm$^{-3}$ , 40.6%, 59.4%, respectively. The bulk density of ash ball was greater, while porosity was smaller, than that of perlite and rockwool. Saturation moisture capacity was 52% in ash ball, 71% in perlite, and 90% in rockwool. Water contents after drainage for 1 hr of ash ball, perlite, and rockwool were 21%, 27%, and 80%, respectively. Water content of small granules (3-5 mm) of ash ball was 5% greater than that of large (7-15 mm) grannules. The ash ball was a weak alkali substrate with pH 7.6, but not electric conductivity (EC), of the nutrient solution supplied to ash ball slightly increased. When the absorption of mineral ions to substrates were analyzed, ash ball and RW absorbed mainly PO ̄$_4$. On tomato culture, salt accumulation in ash ball substrate was similar to that in perlite. Most of the salts in the ash balls were removed by submerging the substrate eight times in distilled water. It is concluded that water holding capacity of ash ball substrate was lo as compared to other substrates, but air permeability, and water diffusion was excellent, making control of medium water content easy.

  • The cyclic metabolism of poly(3-hydroxyhutyrate-co-3-hydroxyvalerate) synthesized from glucose by Alcaligenes sp. SH-69 in the presence or absence of new carbon substrate was investigated. In batch culture, the content and weight average molecular weight of the copolymer already stored in the cell decreased rapidly when there was no other carbon source available. After the depletion of carbon source, the amount of high molecular weight copolymer decreased more rapidly than that of low molecular weight copolymer, and as a result, average molecular weight distribution shifted to the lower value. The addition of a mixture of glucose and levulinic acid when the initial carbon substrate, glucose, was nearly depleted supported the continual increase in cell mass and the accumulation of poly(3HB-co-3HV) with high molar fraction of 3HV. However, solvent fractionation of the polymer with acetone revealed the degradation of pre-existing polyhydroxyalkanoale (PHA) in parallel with the synthesis of PHA from new carbon substrate. Even though PHAs obtained from each substrate alone were the copolymer of 3HB and 3HV, it was found that the polymer accumulated in the cells grown by sequential feeding was mainly physical mixture of two poly(3HB-co-3HV) copolymers containing different molar fractions of 3HV.

Cultivation characteristics and yield of Sparassis latifolia depend on the substrate mixture (꽃송이버섯(Sparassis latifolia) 생육배지 조성에 따른 재배특성 및 수량)

  • We investigated the standard cultivation substrate for Sparassis latifolia "Neowul" bred in Jeollabuk-do Agricultural Research and Extension Services. Cultivation characteristics and yield were assessed by varying the kind of sawdust and additives, and the mixing ratio. The cultivation period in larch sawdust was the shortest at 97 days. The yield was excellent (143.6 g). The findings indicated that larch is a tree species appropriate for the cultivation of S. latifolia. When the additives were varied, the yield and productivity (53.1%) were highest (116.6 g) for the wheat bran treatment. Thus, wheat bran would be an additive appropriate for culturing S. latifolia. Concerning the use of different mixing ratios, larch sawdust:beet-pulp:wheat bran ratios of 80:15:5 and 85:10:5 resulted in yields of 114.4 g and 111.4 g, and productivity of 52.5% and 51.8%, respectively. These yield and productivity values were not statistically different. Thus, the standard cultivation substrate for S. latifolia can comprise larch sawdust, beet pulp, and wheat bran at a ratio of 80:15:5 or 85:10:5. The carbon/nitrogen (C/N) ratio assumed to be appropriate for the cultivation of S. latifolia was 184-223. Pinheading would be difficult below a C/N substrate ratio of 105. Thus, the C/N ratio of the media, as well as the pH, would be vital factors affecting pinheading during S. latifolia cultivation.

  • Some experiments on the citric acid production were carried out from soluble starch and molasses as raw materials. When soluble starch was used as substrate for the fermentation of citric acid by the strain M-80 which had assimilating ability of soluble starch in surface culture, the optimal culture media was 120g of soluble starch, 3.0g of (N $H_4$)$_2$S $O_4$, 2.0g of K $H_2$P $O_4$, 0.2g of MgS $O_4$.7$H_2O$, 1.5mg of F $e^{++}$, 1mg of Z $n^{++}$ and 20ml of methanol were added to 1 liter and optimal pH was 5.5. In about 8 days 61.8mg/ml of citric acid was produced. When treated molasses with potassium ferrocyanide was used as substrate for the fermentation of citric acid by the strain of M-315, the optimal condition in surface culture was 250g of molasses, 0.3g of N $H_4$N $O_3$, 0.05g of K $H_2$P $O_4$, 0.01g of MgS $O_4$.7$H_2O$, 0.5g of Potassium ferrocyanide and 30ml of methanol were added to 1.0 liter. On the other hand, the optimal condition in submersed culture was 250g of molasses, 0.3g of N $H_4$N $O_3$, 0.1g of K $H_2$P $O_4$, 0.01g of MgS $O_4$.7$H_2O$, 0.5g of potassium ferrocyanide, and 30m1 of methanol were added to 1.0 liter and optimal pH was all 5.0. After 9 days culture, 69.4mg/ ml, 39.6mg/ml of citric acid were separately produced in surface and submerged culture media.dia.

  • An actinomycetes, KT-10 isolated from ginseng field in Kyongpook, Korea was selected based on its ability to produce a lysosomal cathepsin B inhibitor. The inhibitor purified from the culture supernatant of the isolate KT-10 showed strong inhibitory effects against cathepsin B as well as against papain when the activities were measured using synthetic substrate, ${\alpha}$-N-benzyloxycarbonyl-L-Iysine p-nitrophenyl ester (CLN) or ${\alpha}$-N-benzoyl-D,L-arginine 2-naphthylamide (BANA). The isolate KT-10 was identified as a species of Streptomyces based on its morphological characteristics and chemotaxonomic data. The TAXON program of Ward was used to identify Streptomyces sp. KT-10 as a strain of Streptomyces luteogriseus belong to cluster 18 of the genus Streptomyces with a Willcox probability 0.999388. The cathepsin B inhibitor was presumed to a novel material composed of a polyhydroxylamine.

  • Substrate inhibition of 2,4-dinitrophenol (DNP) degradation was investigated using activated sludge which had been adapted to mineralize DNP. DNP is a metabolic uncoupler, preventing cells from making energy for growth and it has been suggested that pH may be important in mitigating effects of uncouplers. After acclimation of the activated sludge, the effect of pH on toxicity of DNP at high concentration (75 mg/L) was investigated, over a pH range of 5 to 9. DNP inhibition was found to be strongly dependent on mixed liquor pH. The DNP degradation rate was highest in the pH range of 6.95 to 7.84; at pH 5.94 degradation of 75 mg/L DNP was significantly inhibited; at pH < 5.77, DNP degradation was completely inhibited after approximately 30% of the DNP was degraded. By comparison, no significant effect of pH variation in the same range was seen on glucose uptake by the activated sludge culture.

  • A real time optimization algorithm for fed-batch cultures of recombinant yeast to determine the optimal substrate feed rate profile has been developed. Its development involved four key steps: (1) development of reliable adaptive model. (2) development of optimization algorithm. (3) design of on-line model update algorithm to be incorporated into the optimization algorithm and (4) experimental validation. A recombinant Saccharomyces cerevisiae producing human parathyroid hormone (hPTH) was chosen as the model strain. It was found to be very successful in maintaining cell growth and galactose consumption at leigh levels, thus resulting in significant improvements in the productivity (up to 2.1 times) and intact hPTH concentration (up to 1.5 times) compared with the case of an intermittent glucose and galactose, or galactose feeding.

  • The fermentation conditions for the maximal production of the useful polysaccharides(water soluble polysaccharide and cell bound polysaccharide) from marine bacterium Zoogloea sp.(KCCM 10036) were investigated with a 5 L jar fermentor. The maximal production of these polysaccharides was obtained under the conditions of initial pH 7.8, 30$^{\circ}C$, 400 rpm of agitation speed, 2 Wm of aeration rate, 10%(w/v) of inoculum size and 2.5%(w/v) of glucose substrate and 10.38 g/L of total polysaccharide was produced. Apparent viscosity of the culture broth was increased with the production of these polysaccharides and the maximum value was reached to 22,500 cp.

  • We investigated the fermentative production of lactic acid from cheese whey and corn steep liquor as cheap raw materials using Lactobacillus sp. RKY2 to reduce the manufacturing cost of lactic acid. Lactic acid yields were obtained at more than 0.98 g/g from medium containing whey lactose. Lactic aid productivities and yields obtained from whey lactose were slightly higher than those obtained from pure lactose. The final concentration of lactic acid increased with increase, in whey lactose concentration, whereas the lactic acid productivity decreased probably due to substrate inhibition. The fermentation efficiencies were improved by addition of more corn steep liquor to the medium.

  • Streptomyces sp. KISl3 isolated from soil was found to produce low molecular weight thiol protease inhibitors. The protease inhibitor production was closely linked to the cell growth and regulated by growth condition. The inhibitor was purified from the culture broth through butanol extraction, silicagel 60 column chromatography, Sephadex LH-20 gel filtration and preparative HPLC. The inhibitor showed specific inhibitory activity to thiol protease such as papain, picin and bromelain. The mode of inhibition against papain to Hammersten casein as a substrate was non-competitive.
