This study was conducted to provide scientific information for establishing efficient and systematic measures for nature conservation and management in Manjang-gul lava tube. The number of vascular plants was 204 taxa (82 families, 176 genera, 144 species, 1 subspecies, 26 varieties and 1 forma). Of those, pteridophyte were 5 families, 12 genera, 14 species and 1 variety and gymnosperm were 4 families, 6 genera, 5 species and 1 variety. Seventy three families, 149 genera, 157 species, 1 subspecies, 24 varieties and 1 forma in angiosperm composed of 62 families, 123 genera, 137 species, 1 subspecies, 17 varieties and 1 forma in dicotyledon and 11 families, 26 genera, 20 species and 7 varieties in monocotyledon. The number of naturalized plants was 9 families, 17 genera, 18 species and 3 varieties, totaling 21 taxa.
The Formosan sika deer (Cervus nippon taiouanus) is an endemic subspecies in Taiwan. The original wild deer has been extinct since the late 1960s. The largest captive population is located at the Taipei Zoo. Except for infectious disease outbreaks, no systemic medical research has been reported for this subspecies. This study was conducted to analyze the medical status of the captive Formosan sika deer population, including the hematological and serum chemistry characteristics. To accomplish this, medical records for 34 Formosan sika deer from January 2003 to January 2014 were acquired and analyzed. The most common illness and cause of death was trauma, followed by gastrointestinal and respiratory disease, respectively. The hematologic and serum chemical values of healthy adults were quite different from those of sika deer (Cervus nippon yesoensis). This study provides a closer medical understanding of this subspecies and the results will facilitate its management.
The plant pathogenic bacterial genus Pectobacteirum consists of heterogeneous strains. The P. carotovorum species is a complex strain showing divergent characteristics, and a new subspecies named P. carotovorum subsp. brasiliensis has been identified recently. In this paper, we re-identified the P. carotovorum subsp. brasiliensis isolates from those classified under the subspecies carotovorum and newly isolated P. carotovorum subsp. brasiliensis strains. All isolates were able to produce plant cell-wall degrading enzymes such as pectate lyase, polygalacturonase, cellulase and protease. We used genetic and biochemical methods to examine the diversity of P. carotovorum subsp. brasiliensis isolates, and found genetic diversity within the brasiliensis subsp. isolates in Korea. The restriction fragment length polymorphism analysis based on the recA gene revealed a unique pattern for the brasiliensis subspecies. The Korean brasiliensis subsp. isolates were divided into four clades based on pulsed-field gel electrophoresis. However, correlations between clades and isolated hosts or year could not be found, suggesting that diverse brasiliensis subsp. isolates existed.

Paratuberculosis caused by Mycobacterium avium subspecies paratuberculosis (Mpt) is a chronic infectious enteric disease with deleterious impact on the performance in ruminants. In Korea, ELISA has been introduced to detect antibodies to Mpt in individual cattle. However, comparison study with ELISA has not been studied until now. In total, a panel of 899 serum samples obtained from dairy cattle was analyzed with two commercial ELISAs for Mpt to assess the performance. Two ELISAs employed in this study were both licensed worldwide. Two ELISAs applied onto same serum samples showed the moderate agreement (kappa value = 0.60). There was non-significant McNemar test (p = 0.0614) between two ELISA results indicating that each proportion detected by two kits did not differ. In addition, the percent agreement between two ELISA results was turned out to be 96.8% which interpreted excellent reproducibility. It was shown from this study that two ELISAs revealed moderate kappa agreement performance. The implication raised is that when ELISAs as diagnostics are used to detect Mpt in individual cattle, positive reaction by either ELISA should be interpreted as serologically Mpt positive due to presumed low sensitivity of ELISAs and their test agreement being less than 100%.
Susceptibility to chronic wasting disease (CWD) in cervid species has been associated with polymorphisms in the prion protein gene (PRNP). The single nucleotide polymorphisms (SNPs) were found in the PRNP of the Korean subspecies of Chinese water deer via analyses of the DNA sequences obtained from 34 individual deer. Two SNPs were detected at codons 77 and 100. One SNP at codon 77 encoding Glycine was determined to be a silent mutation but the other SNP detected at codon 100 induced an amino acid change, from Asparagine to Serine. The prion protein (PrP) amino acid sequence of the water deer showed 98.8-99.2% homology with those of American elk, white-tailed deer and mule deer. The PrP of the water deer contained amino acid residues closely related with CWD-susceptibility. This study is the first to describe genetic variations in the PRNP of the Korean subspecies of Chinese water deer.
Mycobacterium avium subspecies paratuberculosis (MAP) causes Johne's disease (JD) in ruminants. This is the first large scale report to estimate the herd-level prevalence of antibodies against MAP by using an ELISA to detect antibodies in bulk tank milk (BTM) samples. The samples were collected from January 2011 to November 2011, from 636 herds of the dairy farms in the Gyeonggi and Chungbuk areas of Korea. The overall apparent prevalence of MAP antibody-positive herds was 8.5%, and regional prevalence were 32/440 (7.3%) and 22/196 (11.2%) of dairy farms in the Gyeonggi and Chungbuk areas, respectively. The results did not differ significantly by region. While we have determined the prevalence rate of MAP in the Gyenoggi and Chungbuk areas in this study, there is a continuing need for well-designed studies to calculate the prevalence of MAP in dairy herds based on culture and molecular findings.
The ground beetles or carabids are essentially predaceous feeding on a wide variety of insects including forest pests, slugs and land snails containing those injurious to livestock or veterinary, thus many are predominantly beneficial and serve as natural enemies. In the present investigation, some morphometric multivariate analysis were done for 9 different populations in 5 subspecies of D. (C.) jankowskii, which are one of the most common ground beetles in Korea As the results, when the comparison was conducted between intraspecific groups regardless of subspecies in external morphological characters, the average group membership revealed 97.46% correct assignment For intersubspecific comparisons alone 96.3% were correctly classified. Between the groups of ssp. jankowskii an average of 100% individuals were classified in their known group. Thus the predict group membership was highly significant(P<0.001), exceeding so-called 'the 75% rule'. Whereas, the average group membership using the male genitalic characters represented less than the 75% assignment, except only in ssp. quelpartianus (85.6%). The population from Is. Chindo were described here as a new subspecies for qualitative as well as the resultant quantitative differences. Therefore, a total of 7 subspecies are represented in Korea. Among them, the nominate subspecies, ssp. jankowskii(sensu lato), revealed distinct intrasubspecific differences between different geographic populations. These differences can be as notable as intersubspecific variation which need substantial revision of the previous intuitional concepts on the infraspecific level.
The parasporal crystals of Bacillus thuringinsis subspecies kurstaki, dendrolimus, finitimus, aizawai and israelensis were compared by polyacrylamide electrophoresis, amino acid composition and immunological analysis. In the subspecies of kurstaki, dendrolimus, finitimus and aizawai, the molecular weights of main subusnits of crystal solubilized by alkaline solution were 1.3${\times}$105 and 6.5${\times}$104 while those of subsp. israelensis were 4${\times}$104 and 1,4${\times}$104. The degradation of lepidopteran toxic subspecies crystals by silkworm midgut protease showed 6.0-6.4${\times}$104 molecular weight and the pattern of degradation of subsp. israelensis crystals was similar to that of alkaline solution treatment. In the amino acid composition, aspartic acid in subsp. israelensis and glutiamic acid in the other four subspecies were the most abundant. The immunological characteristics of the crystals revealed that the antibody produced against the alkali-solubilized crystal protein of subsp. israelensis reacted with only its antigen, but the crystal antigens from the other four lepidopteran toxic subspecies did cross-react with each other as well as with their own homologous antisera.
Morphometric and genetic analyses on six populations of the two subspecies of Cobitis koreensis were performed to investigate the geographic variation and to clarify their taxonomic status. No significant differences between subspecies were found. Puan population of C. k. pumilus tends to be smaller in numbers of cross bars and dorsal spots, than those of C. k. koreensis populations with no statistical significance. Average degree of genic variation among six populations of the two subspecies was A=1.5, P =37.3%, HD=0.053, and HG=0.097. Marginal populations of Samchog (C. k. koreensis) and Puan(C. k. pumilus) were least variable: A= 1.3, P=23.8%, HD=0.0l0, HG=0.043, and A= 1.2, P=19.0%, HD=0.029, HG=0.078 respectively.The difference between HD and HG in all populations may indicate a patchness distribution between demes within each population. Genetic similarities between the two subspecies were, on the average, S=0.894, and this value correspond to the average genetic similarities among five populations of C. k. koreensis (5=0.899). Chongup population of C. k. koreensis and Puan population of C. k. pumilus was most similar genetically (5 =0.931). The results of morphometric and genetic analysis in this study indicate that C. k. pumilus is not a valid subspecies and should be considered as a population of C. koreensis.
Morphological and ecological comparions are made between specimens of Cobitis koreensis KIM and the geographically restricted population of the Cobitid fish which is found along the Begchon stream, Puangun, Cholla-bugdo province over a 12-month period of 1982 and 1983. The Begchon population is tentatively identified as Cobitis koreensis, but the color patterns and two morphometric proportions differ widely fromthose reported previously for this species. In addition to these character, it is easily distinguished from C. koreensis by the smaller body size, by having the larger egg size and the less egg number in female during the breeding season. As the differentiation of this population is primarily due to geographically isolation, it is supposed that Begchon cobitid population may be an undescribed distinct subspecies of K. koreensis or above the conventional subspecies level.
