Investigation of the Microbiological and Biochemical Properties of Kimchi in the Submerged Model System Designed for Fermented Sausages

  • The objective of this study was to investigate the potential of the application of lactic acid bacteria (LAB) from kimchi as a starter culture in the production of fermented sausages. To achieve this, a submerged model medium that contained LAB as part of a complex system of kimchi (0.5, 1.0, 1.0, 3.0, and 5.0%) and lyophilized kimchi powder (0.2 and 0.5%) was fermented for 120 h. During the fermentation period, the growth of total viable organisms and LAB, and the changes in the pH and the titratable acidity, were investigated. The initial LAB counts ranged from 6.4 to 7.7 Log CFU/mL for the kimchi media, and from 6.9 to 6.9 Log CFU/mL for the kimchi powder media. In all the kimchi batches, the LAB increased logarithmically, and the highest LAB counts (around 9 Log CFU/mL) were reached in 24 h. An evident lag phase of the LAB was observed in the kimchi powder samples and reached 8.8 Log CFU/mL in 8 h. The decrease in the pH and the formation of lactic acid were rapid in the kimchi batches, and reached pH values of 3.4-3.5 in 12 h. With these results, the LAB that was integrated with the addition of kimchi or kimchi powder demonstrated its potential utility as a substitute for starter culture.

Phellinus SP.의 액체배양에 의한 세포외 색소물질의 생산 및 특성 (Production and Characteristics of an Extracellular Pigment through the Submerged Cultivation of Phellinus Sp.)

  • An extracellular pigment production of three Phellinus sp. (Phellinus 421, P. linteus and P. hartigil) through submerged cultivation was investigated. The maximum brown pigment from culture broth was obtained from the precipitate by addition of 10% 1M HCI solution. This precipitate showed absorption characteristics with ${\lambda}_{max}$ of 360nm. The maximum production of extracellular pigment obtained at optimum medium and culture condition was 3.54 ($A_{360}$). The precipitate was fractionated by Sephadex G-75 gel chromatography, and the isolated brown pigment contained a large amount of polyphenol and the small amounts of sugar and protein. The brown pigment fraction was stable in temperature range of $30{\sim}60^{\circ}C$, pH range of $4{\sim}6$, sugar addition ranges of $1{\sim}5%$ and salt addition concentration of 3 molarity. Antioxidative activity of the brown pigment by TBA method was better than that of vitamin E (${\alpha}$-tocopherol).

Phellinus baumii으로부터 세포외 다당체 생산의 최적화 (Optimal Conditions of Mycelial Growth and Exopolysaccharide Production in Submerged Culture of Phellinus baumii)

  • 장수상황버섯(Phellinus baumii)의 균사체 성장 및 세포외 다당체(exopolysaccharides) 생산을 위한 액체배양의 최적 배지 및 배양조건에 관한 실험을 수행한 결과, 최적배양 온도와 초기 pH는 각각 3$0^{\circ}C$와 5.0으로 결정되었다. 탄소원의 경우, cellobiose와 maltose가 균사체 성장에는 양호하였으나 저조한 세포외 다당체 생산을 보였으며, 반면에 fructose 와 mannitol 은 상대적으로 높은 수율의 세포외 다당체 생산과 균사체 성장 또한 양호하였다. 최적 질소원으로는 yeast extract가 균사체 성장뿐만 아니라 세포외 다당체 생산에 효과적인 것으로 결정되었다. 최적 배지의 조성은 fructose 20 g/L, yeast extract 20 g/L 및 $CaCl_2$ 0.55 g/L이었으며, 최적 배양조건 하에서 5-L 교반 발효조를 운전한 결과, 최대 균사체 성장(17.43 g/L)과 최대 세포외 다당체 생산(3.6 g/L)을 얻을 수 있었다. 회수된 세포외 다당체는 glycoprotein이었으며 그 조성은 amino acid분석 결과 주로 arginine (14.1%)과 glycin (12.0%)이었으며, cabohydrate의 경우는 mannose (48.7%)와 arabinose (38.4%)이 주성분이었다.

생물반응기 유형에 따른 잔나비불로초(Ganoderma applanatum)의 균사체 및 수용성 다당체 생산 특성 (Production of Mycelia and Water Soluble Polysaccharides from Submerged Culture of Ganoderma applanatum Using Different Types of Bioreactor)

  • 잔나비불로초 배양에 적합한 생물반응기를 선발하기 위하여 4종류의 생물반응기 형태별 균사체 생장특성과 다당체 생산량을 비교하였다. 균사체 생산량은 기포통기형 생물반응기($7\;g/{\ell}$) > 풍선형 생물반응기($6.2\;g/{\ell}$) > 교반형 생물반응기($6\;g/{\ell}$) > 컬럼형 생물반응기($5\;g/{\ell}$) 순으로 나타났으며, 기포통기형 생물반응기에서 가장 높은 세포외 다당체($1\;g/{\ell}$)와 세포 내다당체(2.7%)를 생산하여 기포통기형 생물반응기가 잔나비불로초 균사체 배양에 가장 적합한 것으로 나타났다. 세포외 다당체의 분자량도 생물반응기의 형태별에 따라 다르게 나타났으며, 기포통기형 생물반응기에서 가장 높은 분자량을 나타냈다. 기포통기형 생물반응기에서의 적정한 공기공급율은 0.05부터 0.1 vvm 사이로 나타났다. 특히 잔나비불로초 균사체는 배양하면서 배지의 점도가 높아져 점차 공기 공급량을 높여 주는 것이 좋은 것으로 판단된다.

Composition of Culture Medium and Culture Conditions for In vitro Culture of Rice Panicle

  • The in vitro culture of rice panicles is a culturing technique only panicle without other organs in culture solution containing organic substance, so that would be useful to study how assimilate supply affects grain development and maturation. To find the optimum stage for in vitro culture, rice panicles grown in greenhouse were sampled periodically after anthesis and cultured in nutrient medium. The panicles older than 1 weeks after anthesis had produced normal grains. Grain-filling was apparently dependent upon sucrose concentration (8-12 %) in medium, but not affected by nitrogen concentration supplied with glutamine. As far as rice panicle was supplied with sucrose and N in nutrient medium, grains continued accumulation of dry matter and maturation regardless to light condition. Considerably, grain-filling was improved when panicles were positioned horizontally inside flask, so that each grain was partially submerged to nutrient medium.

표고버섯 균사체의 생산과 당뇨 유발 흰쥐에 대한 혈당강하 효과 (Production of Lentinus edodes Mycelia in Submerged Culture and It's Hypoglycemic Effect in Diabetic Rats)

  • 표고버섯 균사체가 항당뇨 효과에 미치는 영향을 검토하기 위해 당뇨가 유발된 흰쥐에 표고버섯 균사체을 7일간 급여시킨 후 혈장 glucose, total cholesterol, triglyceride 및 ALT, AST 활성을 관찰하였고, 표고버섯 균사체 생장의 최적 배양 조건을 조사하였다. 균사체 생장을 위한 최적의 pH와 온도는 5와 $30^{\circ}C$로 나타났으며, 발효조배양시에는 최대 균사체 생장(8.13g/l)이 진탕배양일 때보다 6일 단축되었다. Hypoglycemic 효과에서 혈장 glucose는 당뇨 유발군에서 표고버섯 균사체 급여군이 대조군보다 23.0%가 유의하게 감소하였으며, 혈장 total cholesterol과 triglyceride는 20.3%와 27.1%로 낮게 나타났다. 또한 혈장 ALT와 AST 활성은 당뇨 대조군에 비해 표고버섯 균사체 급여군이 29.6%와 34.3% 낮은 활성을 보였다.

Production of Polysaccharide by the Edible Mushroom, Grifola frondosa

  • The production of polysaccharide according to various developmental stages(mycelium growth, primordium appearance, and fruiting-body formation) in the edible mushroom Grifola frondosa was studied. The cap of the mature mushroom showed the highest amount of polysacchride. Mycelial growth and polysaccharide synthesis were optimal at pH 5 and $20^{\circ}C$. Polysaccharide synthesis was maximal after 12 days of cultivation, whereas maximum mycelial growth was shown after 18 days. Mannose, cellobiose and starch increased the level of polysaccharide as well as growth in submerged culture. Glucose and sucrose appeared to be good substrates for fruiting of Grifola frondosa.
