  • The study was carried out to investigate sensory characteristics of Chongilppong mulberry fruit wine, as well as biological effects of subfraction of MeOH extracts. In the cholesterol-induced hyperlipidemia rats administered with subfraction of mulberry fruit extract, serum triglyceride, GPT and GOT level in serum were generally decreased. Therefore, the above results suggested that mulberry fruit water soluble fraction can help to maintain normal liver functions and to protect hyperlipidemia.

Effects of Anthriscus sylvestris Hoffmann Extract on the Biosynthesis of Collagen in Human Dermal Fibroblasts (사람 섬유아세포에서 전호(Anthriscus sylvestris Hoffmann)추출물이 콜라겐 생합성에 미치는 영향)

  • In this study, we investigated the boosting effects on collagen biosynthesis of $Anthriscus$ $sylvestris$ ethanol extract (ASE) in human dermal fibroblasts. To obtain more effective fraction and subfraction for collagen biosynthesis, standard solvent partition and open column chromatography were performed. The EtOH extract, solvent fractions, and 8 EtOAc subfractions were tested for their collagen synthesis capacity by [$^3H$]Proline-incorporation and ELISA assay. ASE increased 25% of total collagen synthesis and 27% of procollagen biosynthesis. The total collagen biosynthesis was increased by EtOAc fraction and E6 subfraction to 28% and 50% respectively. Type I procollagens were also upregulated by EtOAc fraction and E6 subfraction to 30% and 47%, each. Taken together, our data suggest that potential anti-aging effect of ASE on skin is via increasing collagen biosynthesis and effective subtraction is E6 subfraction of EtOAc fraction.

  • The effects of bioactive fractions from Brachyglottis monroi on biological activity were investigated. this bioactive subfraction 6-5 is the most cytotoxic to P388 murine leukaemia cell lines. A comparison of IC/sub 50/ values of these subfraction in cancer cell lines showed that their susceptibility to these subfractions decreased in the following order; Fr. 6-5 > Fr. 6-3 > Fr. 6-6 > Fr. 6-1 > Fr. 6-2 > Fr. 6-4 by the MTT method. Silica gel flash column chromatography concentrated the cytotoxic activity in subfraction 6-5 which eluted with 30% and 40% ethyl acetate : hexane gave a major bioactive (51 mg, P388 IC/sub 50/ 8,286 ng/mL at 7.5 ㎍/disc).

Screening for Components to Inhibit N-Nitrosodimethylamine Formation from Tomato (토마토로부터 N-nitrosodimethylamine 생성을 억제시키는 유효성분의 검색)

  • A tomato (Lycopersicon esculentum cv. Naomi F1) juice was separated into ascorbate and phenolic portions using a Sep-pak $C_{18}$ cartridge, and its each portion was tested for inhibition of N-nitrosodimethylamine (NDMA) formation. Ascorbate and phenolic portions of tomato juice inhibited NDMA formation by $81.37{\pm}0.25%$ and $72.03{\pm}0.25%$, respectively. The phenolic portion was further fractionated by prep-HPLC and inhibitory effects of NDMA formation by 4 fractions $(1{\sim}4)$ from tomato juice was tested under the different pH conditions (pH 1.2 and 4.2). Fraction 2 inhibited NDMA formation by $50.10{\pm}0.46%$ (pH 1.2) and $64.30{\pm}0.20%$ (pH 4.2), respectively. Fraction 2 was further separated into 4 subfractions $(2a{\sim}2d)$. Subfraction 2b especially inhibited NDMA formation by $70.62{\pm}0.45%$ (pH 1.2) and $75.30{\pm}0.45%$ (pH 4.2). This subfraction was confirmed o-coumaric acid through the analysis of GC-Mass spectrum, $^1H-NMR$ and $^{13}C-NMR$.

Antioxidative Activity of Extract from Bangah Herb (방아 추출물의 항산화 효과)

  • Bangah, one of the herbs grown in Korea, was investigated for its antioxidant activity. The ether extracts of bangah herb was separated into neutral, phenolic, acidic and basic fractions and further separated into subfractions. Antioxidative activities were measured by hydrogen donating activity (HDA), peroxide value (POV), thiobarbituric acid (TBA) value and inhibition activity against lipid peroxidation of rat liver microsomes, The subfraction components were identified by GC/MS and NMR. Phenolic, though being very small in quantity, showed higher antioxidant activity at all assay system by hydrogen donating activity. POV, TBA value and inhibition activity against lipid peroxidation of rat liver microsomes. Five subfractions(P-1, P-2, P-3, P-4 and P-5) were fractionated from phenolic fraction of bangah herbs, and subfraction P-2 among them showed strong antioxidant activity on a level with BHT or gallic acid at each assay system. Four compounds (peak I, peak II, peak III and peak IV) were isolated by gas chromatogram of TMS derivatives of subfraction P-2 and thes compounds were confirmed to be phenolic substance having -OH and COOH group. There subfractions (N-1, N-2 and N-3) were fractionated from neutral fraction of bangah herbs, and subfraction N-2 among them showed highest antioxidant activity and inhibition activity against lipid peroxidation of rat liver microsomes. Subfraction N-2 was indentified to be estragole by H-NMR spectroscopy.

Anxiolytic Activities of Sanguisorbae Radix (지유(Sanguisorbae Radix)의 항 불안 활성)

  • To detect anxiolytic activity from Sanguisorbae Radix we used various animal models of fear or anxiety that are sensitive to known anxiolytic drugs. While diazepam showed significant anxiolytic activities in all five animal models empolyed in this study, $5-HT_3$ antagonist ondansetron and ethylacetate fraction of Sanguisorbae Radix did show anti-anxiety effects in social interaction and two compartment exploration tests. Ethylacetate fraction of Sanguisorbae Radix and 5-HT related drugs like ondansetron and buspirone, however, seem to have merit over diazepam in terms of not causing drowsiness. Among ten subfractions obtained from ethylacetate fraction of Sanguisorbae Radix by silica gel chromatography, subfraction I showed higher anxiolytic activities than subfraction DEF in two animal models, social interaction and two compartment exploration tests. There is growing evidence for the role of 5-HT in the control of anxiety. We hope that new compound(s) will be found from the active fractions of Sanguisorbae Radix as a potential anxiolytic agent in the future.

  • Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl4-induced hepatotoxicity. The hepatoprotective effect of subfractions has been evaluated by analyzing blood and hepatocyte biochemical analyses and biotransformation enzyme analyses. Treatment of BS-5, subfraction has significantly decreased the activities of alanine aminotransferase and aspartate aminotransferase. In addition, the levels of cholesterol and triglyceride in liver have been decreased as compared with that of CCl4 treated rats. The hepatoprotective effect of BS-5, subfraction on the CCl4-induced hepatotoxicity would be mediated of the attenuation of the level of cytochrome P450 and the enhancement of the activity of glutathion S-transferase.

Effect of Tocopherol Supplementation on HDL Subfraction and Lipoprotein Pattern in Rabbits Fed High Polyunsaturated Fatty Acids (Tocopherol 첨가가 고불포화지방 식이를 먹인 토끼의 HDL Subfraction과 Lipoprotein Pattern에 미치는 영향)

  • This study was designed to evaluate the effects of tocopherol supplementation on serum $HDL_{2}$ and $HDL_{3}$ subfraction and lipoprotein pattern of young rabbits fed the different levels of dietary polyunsaturated fatty acids. White male rabbits with an average bo요 weight of 90kg were assigned into 4 groups : LP( Low PUFA) ; HP( High PUFA) ; LPT( Tocopheror added to LP), HPT(Tocopherol added to HP) by feeding safflower oil or coconut oil at loft( w/w )in addition to thew diet with or without tocopherol for 4 weeks. HDL-Chol and HDL subfractions were determined by polyanionic precipitation method, and the relative amount of lipoprotein by electrophoresis. Serum total cholesterol and TG wer slightly lowered in the group of high PUFA diet and its effect was more in the case of tocopherol supplemented. HDL-Chol level was reduced in the high PUFA diet with or without tocopherol, but overall, HDL-Chol level was not influenced by tocopherol addition and also no significant change in lipoprotein pattern when tocopherol added to the HP diet. $HDL_{2}$ level was increased in HPT group but not in LPT group when tocopherol was added to each HP and LP group, respectively. $HDL_{3}$ levels of both LP and HP groups were reduced with tocopherol supplement, but the reduction was less in the high PUFA groups so that $HDL_{2}$ : $HDL_{3}$ ratio of HP was rather close to that of LP, but it was increased when tocopherol supplemented to the HP diet. Therefore, high PUFA diet could improve the anti atherogenic factor when tocopherol supplemented.

Isolation and Characterization of Bio-active Materials from Prickly Lettuce (Lactuca serriola) (가시상추 유래 생리활성물질의 분리 및 특성 규명)

  • The bio-active materials were isolated from prickly lettuce (Lactuca serriola) by using several extraction solvents. The contents of general compounds, vitamins, polyphenols and flavonoids of prickly lettuce were analysed. In addition, nitrite scavenging activity, anti-oxidative activity and anti-microbial activity of methanol extracts were measured. The ethylacetate (EtOAc) fraction of methanol extracts of prickly lettuce showed the best on the nitrite scavenging activity, anti-oxidant activity and anti-microbial activity. And also, the contents of polyphenols and flavonoids were the highest among all fractions. The subfraction 2 (EtOAc:MeOH=3:1) of EtOAc fraction showed the strongest anti-oxidative activity among 5 subfractions. The subfraction 2 was identified as ${\rho}$-coumaric acid and caffeic acid by GC-MS. The anti-microbial activity of each solvent fraction from prickly lettuce was measured. The EtOAc fraction showed the strongest anti-microbial activity. Overall, these results may provide the basic data to understand the biological activities of bio-active materials derived from prickly lettuce.
