The Usefulness of Selected Physicochemical Indices, Cell Membrane Integrity and Sperm Chromatin Structure in Assessments of Boar Semen Sensitivity

  • The present work describes experiments undertaken to evaluate the usefulness of selected physicochemical indices of semen, cell membrane integrity and sperm chromatin structure for the assessment of boar semen sensitivity to processes connected with pre-insemination procedures. The experiments were carried out on 30 boars: including 15 regarded as providers of sensitive semen and 15 regarded as providers of semen that is little sensitive to laboratory processing. The selection of boars for both groups was based on sperm morphology analyses, assuming secondary morphological change incidence in spermatozoa as the criterion. Two ejaculates were manually collected from each boar at an interval of 3 to 4 months. The following analyses were carried out for each ejaculate: sperm motility assessment, sperm pH measurement, sperm morphology assessment, sperm chromatin structure evaluation and cell membrane integrity assessment. The analyses were performed three times. Semen storage did not cause an increase in the incidence of secondary morphological changes in the group of boars considered to provide sperm of low sensitivity. On the other hand, with continued storage there was a marked increase in the incidence of spermatozoa with secondary morphological changes in the group of boars regarded as producing more sensitive semen. Ejaculates of group I boars evaluated directly after collection had an approximately 6% smaller share of spermatozoa with undamaged cell membranes than the ejaculates of boars in group II ($p{\leq}0.05$). In the process of time the percentage of spermatozoa with undamaged cell membranes decreased. The sperm of group I boars was characterised with a lower sperm motility than the semen of group II boars. After 1 hour of storing diluted semen, the sperm motility of boars producing highly sensitive semen was already 4% lower ($p{\leq}0.05$), and after 24 hours of storage it was 6.33% lower than that of the boars that produced semen with a low sensitivity. Factors that confirm the accuracy of insemination male selection can include a low rate of sperm motility decrease during the storage of diluted semen, low and contained incidence of secondary morphological changes in spermatozoa during semen storage and a high frequency of spermatozoa with undamaged cell membranes.

  • In this study, an attempt is made to apply the concept of fuzzy-bayesian theory to the integrity assessment of structure system, and uncertainty states are represented in terms of fuzzy sets which define several linguistic variables such as "very good", "good", "average", "poor", "very poor", etc. Especially, the concept of fuzzy conditional probability aids to derive a new reliability analysis which includes the subjective assessment of engineers without introducing any additional correction factors. The fuzzy concept are also used as reliability indexes for the condition assessment based on the proposed models, the proposed fuzzy theory-based approach with the results of PEM and AFOSM are applied to retaining wall.

Measurement of vibration and stress for APR-1400 reactor internals

  • The U.S. Nuclear Regulatory Commission, Regulatory Guide 1.20 needs to perform a comprehensive vibration assessment program for reactor internals during preoperational and startup testing for nuclear power plants and extended power uprate. Although the measurement program is one of the core programs, it is rarely carried out except for a first-of-a-kind or a unique design. This article describes measurement results of vibration and stress for the comprehensive vibration assessment program for an APR-1400 reactor internals. The measurement was performed at an upper guide structure during the pre-core hot functional test of Shin Kori unit 4 reactor internals because the Shin Kori unit 3 and 4 are the first construction project for the APR-1400, and the upper guide structure assembly was to design change compared with the valid prototype. We confirmed that all measured results are within the test acceptance criteria. It means that the structural integrity of the APR-1400 reactor internals was secured for the flow-induced vibration.

  • In the present study, the influence of car body structures to the noise and vibration characteristics has been sought. The numerical modal analysis for the body-in-white is employed to predict the vibratory response of structure, and then followed by the experimental modal testing to confirm the validity of the model. Using the results of numerical simulations with the designated modal parameters, the optimal structural configuration has been deduced. Special interests have been paid to the sensitivity of sound quality to the structural integrity. Since the structural integrity has a close relationship to the structure-born noise, the substantially low frequency range, which is far below the frequency range almost barely sensible by human auditory organ but still quite influential to overall impression, is especially examined. The subjective assessment agrees with the objective evaluation by means of traditional sound measures as well as psychoacoustic metrics.

INTEGRITY ANALYSIS OF AN UPPER GUIDE STRUCTURE FLANGE

  • The integrity assessment of reactor vessel internals should be conducted in the design process to secure the safety of nuclear power plants. Various loads such as self-weight, seismic load, flow-induced load, and preload are applied to the internals. Therefore, the American Society of Mechanical Engineers (ASME) Code, Section III, defines the stress limit for reactor vessel internals. The present study focused on structural response analyses of the upper guide structure upper flange. The distributions of the stress intensity in the flange body were analyzed under various design load cases during normal operation. The allowable stress intensities along the expected sections of stress concentration were derived from the results of the finite element analysis for evaluating the structural integrity of the flange design. Furthermore, seismic analyses of the upper flange were performed to identify dynamic behavior with respect to the seismic and impact input. The mode superposition and full transient methods were used to perform time-history analyses, and the displacement at the lower end of the flange was obtained. The effect of the damping ratio on the response of the flange was also evaluated, and the acceleration was obtained. The results of elastic and seismic analyses in this study will be used as basic information to judge whether a flange design meets the acceptance criteria.

Simplified Numerical Load-transfer Finite Element Modelling of Tunnelling Effects on Piles

  • Tunnelling in urban environments is very common nowadays as large cities are expanding and transportation demands require the use of the underground space for creating extra capacity. Inevitably, any such new construction may have significant effects on existing nearby infrastructure and therefore relevant assessment of structural integrity and soil-structure interaction is required. Foundation piles can be rather sensitive to nearby tunnel construction and therefore their response needs to be evaluated carefully. Although detailed three-dimensional continuum finite element analysis can provide a wealth of information about this behaviour of piles, such analyses are generally very computationally demanding and may require a number of material and other model parameters to be properly calibrated. Therefore, relevant simplified approaches are used to provide a practical way for such an assessment. This paper presents a simple method where the pile is modelled with beam finite elements, pile-soil interaction is modelled with soil springs and tunnelling-induced displacements are introduced as an input boundary condition at the end of the soil springs. The performance of this approach is assessed through some examples of applications.

Monothiolglycerol이 동결 융해 후 미니돼지 정자의 활성산소 억제, 미토콘드리아 활성 그리고 DNA Integrity에 미치는 영향 (Effect of Monothioglycerol on ROS Inhibition, Mitochondrial Activity, and DNA Integrity in Frozen-thawed Miniature Pig Sperm)

  • Cryopreservation and in vitro fertilization (IVF) protocols are important in genetic studies and applications to transgenic animals. Various studies about boar sperm cryopreservation have been studied for a long time. Those were about the use of extenders, the choice of sugars, the cooling and warming rates. The factors that influence the boar sperm are the dramatic changes in temperatures, osmotic and toxic stresses, and reactive oxygen species (ROS) generation. Among these factors, ROS generation is the main damage to DNA which is a principal genetic material and the most important for the practical applications. So we wondered whether ROS generation could be reduced. In previous study, monothioglycerol (MTG) was essential for the culture of embryo stem cells. Therefore we added MTG in the freezing extender based on lactose-egg yolk (LEY) with trehalose. For the assessment of the frozen-thawed sperm, we focused onmotility, membrane integrity and DNA damage. First, we used a computer-aided sperm analysis system for overall conditions of sperm such as motility and viability. Then we performed the sperm chromatin structure assay for DNA integrity and hypo-osmotic swelling test for membrane integrity. And our result showed the existence of MTG in the freezing extender caused less damage to DNA and higher motility in frozen-thawed boar sperm. Also we checked a relative antioxidant activity of MTG in modified Modena B extender. We concluded that this reagent can activate sperm mitochondria at MTG $0.2{\mu}M$, contribute to sperm motility and DNA integrity but there was no significant difference on membrane integrity. Also antioxidant activity of MTG in modified Modena B extender was proved.

A potential role for fatty acid biosynthesis genes during molting and cuticle formation in Caenorhabditis elegans

  • Caenorhabditis elegans undergoes a developmental molting process that involves a coordinated interplay among diverse intracellular pathways. Here, we investigated the functions of two fatty acid biosynthesis genes; pod-2, encoding acetyl-CoA carboxylase, and fasn-1, encoding fatty acid synthase, in the C. elegans molting process. Although both the pod-2 and fasn-1 genes were expressed at constant levels throughout C. elegans development, knockdown of the proteins encoded by these genes using RNA interference produced severe defects in triglyceride production, molting, and reproduction that were coupled to suppression of NAS-37, a metalloprotease. An assessment of the structure and integrity of the cuticle using a COL-19::GFP marker and Hoechst 33258 staining showed that downregulation of either pod-2 or fasn-1 impaired cuticle formation and disrupted the integrity of the cuticle and the hypodermal membrane.

VIBRATION AND STRESS ANALYSIS OF A UGS ASSEMBLY FOR THE APR1400 RVI CVAP

  • The most important component of a nuclear power plant is its nuclear reactor. Studies on the integrity of reactors have become an important part regarding the safety of a nuclear power plant. The US Nuclear Regulatory Commission Regulatory Guide (NRC RG) 1.20 presents a Comprehensive Vibration Assessment Program (CVAP) to be used to verify the structural integrity of the Reactor Vessel Internals (RVI) for flow-induced vibration prior to commercial operation. However, there are few published studies related to the RVI CVAP. We classified the Advanced Power Reactor 1400 (APR1400) RVI CVAP as a non-prototype category-2 reactor as part of an independent validation of its design. The aim of this paper is to present the results of structural response analyses of the Upper Guide Structure (UGS) assembly of the APR1400 reactor. These results show that the UGS and the Inner Barrel Assembly (IBA) meet the specified integrity levels of the design acceptance criteria. The vibration and stress analysis results in this paper will be used as basic information to select measurement locations of the vibration and stress for the APR1400 RVI CVAP.

  • A series of vibration assessment programs has been performed for Yonggwang Nuclear Power Plant Unit 4 (YGN 4) in order to verify the structural integrity of the reactor internals for flow induced vibration prior to its commercial operation. The structural analysis was done to provide the basis for measurement and the theoretical evidence for the structural integrity of the reactor internals. The actual flow induced hydraulic loads and reactor internals vibration response data were measured and recorded during pre-core hot functional testing of the plant. Then, the measured data have been reduced and analyzed, and compared with the analysis results such as the frequency contents, stresses, strains and displacements. It is concluded that the structural analysis methodology performed for vibration response of the reactor internals due to the flow induced vibration is appropriately conservative, and also that the structural integrity of YGN 4 reactor internals to flow induced vibration is acceptable for long term operation.
