Effect of Delayed Inoculation After Wounding on the Development of Anthracnose Disease Caused by Colletotrichum acutatum on Chili Pepper Fruit

  • Detached chili pepper fruits were inoculated with the conidial suspension of Colletotrichum acutatum JC-24 simultaneously (simultaneous inoculation, SI) and at delayed time (delayed inoculation, DI) after wounding with (delayed wound inoculation, DWI) or without additional wounding (delayed non-wound inoculation, DNI) at the inoculation time. Disease severity was significantly lowered by DNI, compared to SI. By DNI, the disease reduction rates were proportional with the length of delayed time, and greater at the high temperature range (18, 23 and $28^{\circ}$) than at the low temperature ($13^{\circ}$) tested. DWI was also effective in reducing the disease severity especially at 18oC; however, its effectiveness was lower than for DNI. In light microscopy, parenchyma cells at the wounding sites were modified structurally, initially forming new cell walls crossing cytoplasm, enlarged with multiple periclinal cell divisions, and finally layered like wound periderms. In DWI, the above structural modifications occurred, showing the restriction of the fungal invasion by the cell walls in enlarged modified cells, while no definite cellular modifications were found with proliferation of fungal hyphae in SI. Sclerenchyma-like cells with thickened cell walls were proliferated around the wounding sites, which were partially dissolved by DWI, probably leading to some disease development. All of these results suggest that the decline of the anthracnose disease in pepper fruit by the delayed inoculations may be derived from the structural modifications related to the healing processes of the previous wound inflicted on the tissues.

  • The purpose of this study is to dimensionally examine the factors of policy changes by applying the ACMS model to KEPCO's structural reorganization privatization policies and study its implications. As a result of the analysis, among the external variables that affect as a financial source or restriction to advocates, all areas except for the difference in opinion of the people as well as policy decision and effects of other subordinate systems were shown to be in an advantageous condition acting as a financial source to advocates of structural reorganization privatization. On the other hand, advocates affected by the external variables share a certain belief system in which the advocates of privatization share the justification of profitability while the advocates of collectivization share the justification of publicness. In these flows, the announcement of the privatization plan of public enterprises of the committee of planning and budgeting acts as a matter of contact which opens the windows for policy changes, and this window goes through ignition, explosion, and fixation which causes fierce interactions among advocates of both sides. Eventually, in accordance with the contents of excluding electricity(KEPCO) from the list for privatization the window for policy changes has closed and things have changed to a transitional KEPCO privatization policy. Based on this, the first implication is that non-official participants such as the people, social organizations, labor unions, etc. influence policy changes more than they did before. Secondly, in the process of policy changes, there is an order of boundary even in the fierce interactions between each advocate. Thirdly, specific variables can play various roles in complex policy phenomenon. Fourthly, a logical analysis using the ACMS model is possible in Korea as well. Fifthly, as a result of analyzing the change process of the KEPCO structural reorganization privatization policy through the ACMS model, not all matched with the main research results implied by the ACMS model.

Structural health monitoring of high-speed railway tracks using diffuse ultrasonic wave-based condition contrast: theory and validation

  • Despite proven effectiveness and accuracy in laboratories, the existing damage assessment based on guided ultrasonic waves (GUWs) or acoustic emission (AE) confronts challenges when extended to real-world structural health monitoring (SHM) for railway tracks. Central to the concerns are the extremely complex signal appearance due to highly dispersive and multimodal wave features, restriction on transducer installations, and severe contaminations of ambient noise. It remains a critical yet unsolved problem along with recent attempts to implement SHM in bourgeoning high-speed railway (HSR). By leveraging authors' continued endeavours, an SHM framework, based on actively generated diffuse ultrasonic waves (DUWs) and a benchmark-free condition contrast algorithm, has been developed and deployed via an all-in-one SHM system. Miniaturized lead zirconate titanate (PZT) wafers are utilized to generate and acquire DUWs in long-range railway tracks. Fatigue cracks in the tracks show unique contact behaviours under different conditions of external loads and further disturb DUW propagation. By contrast DUW propagation traits, fatigue cracks in railway tracks can be characterised quantitatively and the holistic health status of the tracks can be evaluated in a real-time manner. Compared with GUW- or AE-based methods, the DUW-driven inspection philosophy exhibits immunity to ambient noise and measurement uncertainty, less dependence on baseline signals, use of significantly reduced number of transducers, and high robustness in atrocious engineering conditions. Conformance tests are performed on HSR tracks, in which the evolution of fatigue damage is monitored continuously and quantitatively, demonstrating effectiveness, adaptability, reliability and robustness of DUW-driven SHM towards HSR applications.

  • The necessity of the Toi-Maru has increased greatly in order to enter to the building or to move conveniently in the building after the mid-Joseon period, when the combination of On-dol Ma-ru began to be universally used. According to this process, the 3-Dori type upper structure is thought to have undergone the typological changed by securing a more stable space of Toi-Maru In cases where a relatively large girder was used, it is often the case that the space is divided using a pillar to place a Toi-Maru on the front. However, it is difficult to find a case where the width of the Toi-Maru exceeds 1meter due to its structural limit. The type that made additional Toi-girder to overcome the spatial constraint of this structure and replaced the method of former typology after Mid-Jpseon period. In case of 5-Dori type, Toi-Maru installed in between high post and Toi post in early period. In these type, the width of Toi-Maru was subordinated by the location of milled Dori of roof structure and sometimes it became too excessive. 5-Dori type with Pyon-ju occurred to overcome these restriction between the width of Toi-Maru and the location of milled Dori of roof structure. Not a few remains of Mid-Jpseon period keeping the evidences of changing process of roof structure provoked by Toi-Maru installation. The consistent process of Toi-Maru installation and structural adjustment will be focused to consider the attempt to spatial requirement and architectural reaction in Mid-Joseon perid.

A Study on the Effect of Wind Load to an Articulated type Container Crane by Fluid-Structural Coupled Field Analysis (유동-구조 연성해석기법을 이용한 풍하중이 관절형 컨테이너 크레인에 미치는 영향에 관한 연구)

  • This study was carried out to the effect of wind load on the structural stability of an articulated type container crane according to the wind direction assuming that 75m/s wind velocity is applied on a container crane using FSI(fluid-structural interaction). To consider fluid phenomenon around the container crane, the wind load was derived by the computation fluid dynamic, and it applied to the FSI which can guarantee an accuracy and a reliability in the design stage for wind resistant structural stability to minimize the damage due to high wind load applied in a container crane with a 'ㄱ' type articulated boom which used in the total height restriction region. Following from this, the reaction force on the each support of a container crane was suggested. ANSYS ICEM CFD 10.0 and ANSYS CFX 10.0 used for computation fluid dynamic, and the ANSYS Workbench 11.0 was used for the fluid-structural interaction.

  • For the purpose of optimum design of reinforced concrete structures, pre-determined section database of column and beam are constructed and arranged in order of the resisting capacity. Then, regression equations representing the relation between section number and section resisting capacity are derived. In advance, effective optimization algorithms which search optimized solution quickly using direct search method from these database are proposed. In practice, from the fact that engineers conduct member design close to capacity optimization rather than cost optimization, both capacity and cost optimization using proposed algorithms are performed, and the review for the obtained results are followed. Moreover, the investigation for the applicability and effectiveness of the Introduced design procedure is conducted through correlation study for example structures. Because of no restriction in constructing objective functions with very simple optimization processes and fast convergence, the introduced method can effectively be used in the preliminary design stage. Especially, selected solutions from database are directly applicable in practice because these sections already satisfy all the requirements in design codes and practical restrictions.

Soild-state reaction in Ti/Ni multilayers

  • Ti/Ni multilayered films (MLF) are ideal for neutron optics particularly in neutron guides and focusing devices. This system also possesses the tendency of amorphization through a solid-state reaction (SSR). This behaviors are closely related to the electronic structures and both magneto-optical (MO) and optical properties of metals depend strongly on their electron energy structures. Mutual inter-diffusion of the Tin and Ni atoms in the MLF caused by a low temperature annealing should decrease the thickness of pure Ni, as well as change the chemical and atomic order in the reactive zone. The application of the MO spectroscopy to the study of SSR in the MLF allows us to obtain an additional information on the changes in the atomic and chemical orders in the interface region. The optical one has no restriction on the magnetic state of the constituent sublayers. Therefore, the changes in magnetic, MO and optical properties of the Ti/Ni MLF due to SSR can be expected. To the best of our knowledge, the MO and optical spectroscopies were not used for this purpose. SSR has been studied in the series of the Ti/Ni MLFs with bilayer periods of 0.65-22.2nm and constant ratio of the Ti to Ni sublayers thickness by using MO and optical spectroscopies as well as an x-ray diffraction. The experimental MO and optical spectra are compared with the computer-simulated spectra, assuming various interface models. The relative changes in the x-ray diffraction spectra and MO properties of the Ti/Ni MLF caused by annealing are bigger for the multilayers with "thick" sublayers, or the SSR with the formation of amorphous alloy takes place mainly in the Ti/Ni multilayers with "thick" sublayers, while in the nominal threshold thickness of the Ni-sublayer for the observation of the equatorial Kerr effect in the as-deposited and annealed Ti/Ni MLFs of about 3.0 and 4.5nm thick is explained by the formation of amorphous alloy during the deposition or the formation of the nonmagnetic alloyed regions between pure components as a result of the SSR. For the case of Ti/Ni MLF the MO approach is more sensitive for the determination of the thickness of the reacted zone, while x-ray diffraction is more useful for structural analyses.structural analyses.

  • Lately construction industry tends to prefer remodeling of existing buildings rather than new construction or reconstruction of buildings due to strengthening of several restriction related to real estates along with prolonged depression. And also, remodeling of building costs less and creates less wastes compared to reconstruction and so it is more profitable in financial and environmental view. However remodeling is process of creating new environment with existing building. Therefor remodeling must follow the procedure realizing problem and fix the problem based on through investigation on existing building and users requirement must be faithfully reflected. Specially in case of apartment houses, since vagueness on ownership and management authority on common parts exists. Hereupon, in this study we are to present the procedure of analyzing apartment house remodeling method through user requirement by approaching to several considerable factors in user request side.

  • The CFT columns used in thin-walled steel tubes can be more economical, because it was expected the increase of strength by restriction for the local buckling of steel tubes. The purpose of this paper is to review feasibility of existing design formula and verify the applicability limit of width-to-thickness ratio for increasing the strength of rectangular CFT columns. As the main parameters of experiments, width-to-thickness ratios of steel tube, height of rectangular concrete columns, and concrete filled or not. The strength of concrete are selected to 90MPa. From the test results, the confinement effect of steel tube on the compressive strength of infilled concrete is remarkably appeared in the thin-walled rectangular steel tube columns infilled wih high strength concrete. By the non-linear analysis, the axial strength from experiment result was given higher than analysis result for all CFT stub columns.

In Silico Structural and Functional Annotation of Hypothetical Proteins of Vibrio cholerae O139

  • In developing countries threat of cholera is a significant health concern whenever water purification and sewage disposal systems are inadequate. Vibrio cholerae is one of the responsible bacteria involved in cholera disease. The complete genome sequence of V. cholerae deciphers the presence of various genes and hypothetical proteins whose function are not yet understood. Hence analyzing and annotating the structure and function of hypothetical proteins is important for understanding the V. cholerae. V. cholerae O139 is the most common and pathogenic bacterial strain among various V. cholerae strains. In this study sequence of six hypothetical proteins of V. cholerae O139 has been annotated from NCBI. Various computational tools and databases have been used to determine domain family, protein-protein interaction, solubility of protein, ligand binding sites etc. The three dimensional structure of two proteins were modeled and their ligand binding sites were identified. We have found domains and families of only one protein. The analysis revealed that these proteins might have antibiotic resistance activity, DNA breaking-rejoining activity, integrase enzyme activity, restriction endonuclease, etc. Structural prediction of these proteins and detection of binding sites from this study would indicate a potential target aiding docking studies for therapeutic designing against cholera.
