In vitro 계에서의 Streptozotocin 분해 및 세포내 흡수

  • Since streptozotocin has been known to be chemically stable only under acidic condition(pH 4), the spontaneous degradation and cellular uptake of streprozotocin in neutral incubation medium was determined by chemical assay. Streptozotocin levels in both phosphate buffered saline (pH 7.4) and Krebs-Henseleit buffer (pH 7.4) decreased with a half life of about 2 hours. The presence of erythrocytes or pepatocytes under the same buffers did not affect the streptozotocin degradation rate at all. However, streptozotocin levels in plasma isolated from rats decreased rapidly compared to those in neutral buffers.

Effect of Psyllium Seed Husk on the Postprandial Glucose Control and Insulin Secretion Dynamics

  • This study was to investigate the effect of psyllium seed husk (PSYL) on postprandial glucose control and insulin secretion dynamics in Sprague-Dawley rats. In experiment 1, the rise in postprandial serum glucose was monitored during a 240-min period using a maltose loading test In normal rats given 16.6 mg/l00 g B.W./ml of PSYL orally, all the blood glucose levels during the 240-min period did not show statistically significant differences from the corresponding levels in normal rats given water. However, in streptozotocin-induced diabetic rats given the same amount of PSYL, the blood glucose level at 30 min was significantly lower than that in diabetic rats given water, and the peak time of the rise in the postprandial glucose was delayed In experiment 2, the normal (N) and diabetic (Db) rats were given PSYL (25 mg/l00 g B.W./ml/day) orally for 5 days. Blood samples were collected in order to measure the s-glucose and s-insulin levels. The final s-glucose level at day 5 in Db-PSYL was significantly lower than that in the corresponding control rats (Db-CONT) and the final s-insulin level in Db-PSYL was significantly greater than that in Db-CONT. In vitro 40-min pancreas perfusion was performed at day 5 in order to examine the insulin secretion dynamics. Results showed that the amounts of insulin secreted during the first phase (11-20 min) and the second phase (21-40 min) in the Db-PSYL were significantly greater than those in Db-CONT. Therefore, it is concluded that psyllium seed husk could be beneficial for controlling postprandial glucose levels in the stretozotocin-induced diabetic rats, and it may be partially mediated by insulin secretion dynamics.

Effects of Coriandrum sativum L. on Lipid Contents in Streptozotocin-Induced Diabetic Rats (식이내 고수(Coriandrum sativum L.)의 첨가가 당뇨성 흰쥐의 혈장과 간의 지질함량에 미치는 영향)

  • This study was designed to investigate the effects of Coriandrum sativum L. on lipid metabolism in nondiaetic and stretozotocin-induced diabetic.Thirty two mael Sprague-Dawley rats weighting 108$\pm$13 g were divided into four groups as follow: control (C), leaf(L), seed (S) and root (R) groups. For four weeks of experimental periods, the C group were fed basal diet and the L, S and R groups were fed Coriandru sativum L. diets containing 5% dried leaf, seed and root of Coriandrum sativum L, respectively. diet intakes, weight gain and food efficiency were not significantly different among the four groups. The liver and heart weights of L, s and R groups were significantly different amount the control group. The levels of plasma insulin was significantly increased but the plasma glucose levels was not decreased in all Coriandrum group. The levels of plasma lipids was not significantly different among the four groups. the levels of plasma total cholesterol was significantly decreased in R groups. The levels of plasma and liver TBARS values were significantly decreased in R groups. The levels of plasma vitamin E were tendency to decrease in all coriandum groups. These results indicate feeding diet with coriander did not show the hypolipidemic effect but show the antioxidative effect.
