A Nudix Hydrolase Protein, Ysa1, Regulates Oxidative Stress Response and Antifungal Drug Susceptibility in Cryptococcus neoformans

  • A nucleoside diphosphate-linked moiety X (Nudix) hydrolase-like gene, YSA1, has been identified as one of the gromwell plant extract-responsive genes in Cryptococcus neoformans. Ysa1 is known to control intracellular concentrations of ADP-ribose or O-acetyl-ADP-ribose, and has diverse biological functions, including the response to oxidative stress in the ascomycete yeast, Saccharomyces cerevisiae. In this study, we characterized the role of YSA1 in the stress response and adaptation of the basidiomycete yeast, C. neoformans. We constructed three independent deletion mutants for YSA1, and analyzed their mutant phenotypes. We found that ysa1 mutants did not show increased sensitivity to reactive oxygen species-producing oxidative damage agents, such as hydrogen peroxide and menadione, but exhibited increased sensitivity to diamide, which is a thiol-specific oxidant. Ysa1 was dispensable for the response to most environmental stresses, such as genotoxic, osmotic, and endoplasmic reticulum stress. In conclusion, modulation of YSA1 may regulate the cellular response and adaptation of C. neoformans to certain oxidative stresses and contribute to the evolution of antifungal drug resistance.

  • Thermal stress of pigs causes decreased feed consumption and weight gain rate, immunosuppression, reproductive disorders, and increased mortality. The concept of the temperature-humidity index (THI) has been widely used to evaluate the degree of thermal stress of pigs. However, use of this concept is strongly restricted for animals living in the enclosed facilities. In this study, Building Energy Simulation (BES) technique was used to realize the energy flow among outside weather conditions, building materials, and animals. Especially, mechanisms of sensible and latent heat generation from pigs according to surrounding air temperature and their weight were designed to accurately evaluate the THI values inside the pig house. The THI values computed by the BES model were compared to those calculated by method of the report (NIAS, 2016), the model of this study predicted the start date of heat stress about 9~76 days earlier compared to the NIAS model. Results of the BES model also showed higher frequencies of the THI above the THI threshold for pigs, indicating that conventional model has a possibility of underestimating the degree of heat stress of pigs.

투석환자의 삶의 질에 미치는 영향요인 분석 (A Study on factors Affecting Quality of Life in Patients on Dialysis)

  • Purpose: This study was done to identify stress, fatigue, and self efficacy in patients on dialysis and identify their effect on quality of life. Method: The participants (n=149) who agreed to participate in the research were selected from among those who were on dialysis treatment at C University Medical Centers in Seoul and Inchon, and at C University Medical Center in Jeju. The data were collected from July, 2001 to March, 2002 using interviews. Data analysis was done with t-test, ANOVA, and multiple stepwise regression using the SAS program. Result: Quality of life was significantly different according to economic status, and type of dialysis. Quality of life for the group with higher economic status was better than for the group with lower economic status, and the group on peritoneal dialysis had higher quality of lift scores than the group on hemodialysis. As for self-efficacy, there were significant difference according to duration of disease, type of dialysis, and duration of dialysis. Stress was significantly different according to marital status. Quality of life was significantly predicted by stress (40.02%) and fatigue (3.85%). Conclusion: To improve quality of life for people on dialysis, their stress should be treated positively, also multi-dimensional nursing interventions to provide emotional support and nursing interventions to diminish fatigue are required.

하지정맥류 환자의 질병관련 지식과 스트레스 및 삶의 질 (Disease-related Knowledge, Stress, and Quality of Life in Patients with Varicose Veins)

  • Purpose: The purpose of this study was to investigate the disease-related knowledge, stress and quality of life for the patients with varicose veins. Methods: Data were collected via questionnaire from 138 patients with varicose veins in C University Hospital in Korea from November in Korea 2012 to August 2013. Results: Knowledge of the symptoms and diagnosis was the lowest. The scores from patients with disease-related knowledge tended to be low for both subjects over the age of 60 and subjects with low education (less than high school). The score for stress over complications was the highest. The level of stress is higher for women, subjects without spouses, subjects with higher severity of disease and patients who had been taking hormones in the past or are still taking hormones. The disease-related quality of life tended to be lower for women and subjects without spouses. 21% of the variance in quality of life was explained by the level of disease-related stress, disease severity and knowledge. Conclusion: Education and the management of nurses to improve disease-related knowledge for patients with varicose veins is highly recommended. Intervention, by reducing stress, is necessary to improve the quality of life for women, subjects without spouses, and subjects with higher severity of the disease.

남자 간호대학생의 성역할 갈등, 학업스트레스 대처, 사회적 지지가 대학생활 적응에 미치는 영향 (The Influence of Gender Role Conflicts, Academic Stress Coping Ability, and Social Support on Adaptations to College Life among Male Nursing Students)

  • 본 연구의 목적은 남자 간호대학생의 성역할 갈등, 학업스트레스 대처, 사회적 지지가 대학생활 적응에 미치는 영향을 파악하기 위함이다. 경기도, 충청도, 경상도 소재의 4년제 간호학과 6곳에 재학 중인 남학생 225명을 대상으로, 2014년 4월 1일부터 6월 4일까지 자가 보고식 설문조사로 시행되었다. 연구결과, 학업스트레스 대처(${\beta}=0.33$, p<.001)가 가장 큰 영향력을 나타내는 변수로, 남자 간호대학생의 대학생활 적응을 약 22%설명하였다. 또한 전공만족도, 사회적지지, 성역할 갈등, 동아리 활동 순으로 대학생활 적응에 영향을 주는 것으로 나타났으며, 이들 전체 변수는 대학생활 적응에 대해 약 33%의 설명력을 나타냈다(F=23.50, p<.001). 그러므로 남자 간호대학생의 학업스트레스 대처능력 강화와 성역할 갈등을 해결하기 위한 프로그램 개발과 교수, 선후배로 구성된 사회적 지지망 구축이 대학생활 적응력 향상을 위해 도움이 될 것으로 사료된다.

성조숙증 아동 어머니의 삶의 질에 영향을 미치는 요인 (Factors Influencing the Quality of Life in Mothers of Children with Precocious Puberty)

  • Purpose: In this study factors affecting quality of life in mothers who have children with precocious puberty were identified. Methods: The participants were 102 mothers of children with precocious puberty. From September 12 to September 30, 2016, data were collected using a structured questionnaire and analyzed using SPSS/WIN 22.0. Results: Regression analysis showed that the model's explanatory power was 45%(F=28.71, p<.001). Role stress (t=-4.83, p<.001), objective burden (t=-2.71, p<.001), and average monthly income (t=4.12, p<.001) were factors affecting quality of life among these mothers. The most influential factor was role stress (${\beta}=-.41$), which showed a negative correlation. Conclusion: Role stress and objective burden were major factors that decreased the quality of life in these. The findings indicate that effective strategy programs for reducing role stress and objective burden are necessary to improve the quality of life in mothers of children with precocious puberty.

생산직 근로자의 직무스트레스와 구강건강 관련 삶의 질의 관련성 (Relationship between job stress and oral health-related quality of life in the blue collar workers)

  • Objectives: The purpose of the study is to investigate the relationship between job stress and oral health-related quality of life in the blue collar workers. Methods: This study has been conducted with subjects of blue-collar workers who work in the industrial section located in Gyeongnam area from August 2013 to September 2013. Korean Occupational Stress Scale(KOSS-SF) and oral health impact profile(OHIP-14) were used as research tools. Data were analyzed by descriptive analysis, t-test, ANOVA, and multiple regression analysis. Results: Overall job stress and oral heal-related quality of life level of subjects were $57.32{\pm}8.03$ and $14.17{\pm}9.27$. The influential factors of oral heal-related quality of life of blue-collar workers were job demand, interpersonal conflict and organizational climate (subcategory of job stress), respectively. Conclusions: It is necessary to improve the better working condition for the oral health-related quality of life due to job stress in the blue collar workers.

Expression of Bacillus subtilis proBA Genes and Reduction of Feedback Inhibition of Proline Synthesis Increases Proline Production and Confers Osmotolerance in Transgenic Arabidopsis

  • Proline accumulation has been shown to correlate with tolerance to drought and salt stresses in plants. We attempt to introduce the wild-type, mutant, and fusion proBA genes derived from Bacillus subtilis into Arabidopsis thaliana under the control of a strong promoter cauliflower mosaic virus 35S (CaMV35S). The transgenic plants produced higher level of free proline than control and the overproduction of proline resulted in the increased tolerance to osmotic stress in transgenic plants. Besides, the mutation in proBA genes, which were proved to lead $\alpha$-glutamyl kinase ($\alpha$-GK) reduces sensitivity to the end-product inhibition and the fusion of proB and proA also result in increasing proline production and confer osmotolerance in transgenic lines.

  • Galactinol and rafinose accumulation in plants is associated with stressful environmental conditions such as cold, heat, or dehydration by the action of galactinols synthase (GolS) in the raffinose family of oligosaccharides biosynthetic pathway from UDP-galactose. Moreover, several reports mentioned that GolS transcription is up regulated by various environmental stresses like cold, heat, dehydration. Therefore, to determine whether MoGolS1 was induced with the abiotic stress we analyzed the expression pattern of the gene under various abiotic stresses like heat, cold, abscisic acid, sucrose and salt concentration in the lemon balm plants grown in standard MS medium. The MoGolS1 gene was 981-bp in length encoding 326 amino acids in its sequence and shared 77 and 76% sequence similarity with Arabidopsis thaliana galactinol synthase4 (AtGolS4) and AtGolS1 genes respectively. The MoGolS1 gene was strongly expressed by the abiotic stress induced by sucrose, ABA or heat shock. It was also expressed in responses to cold, Identification and Functional Characterization of the GALACTINOL SYNTHASgene induction with various stresses may be possible for itscrucial function in abiotic stress tolerance in plants, providing a good engineering target for genetic engineering.
