Opuntia ficus-indica has traditionally been used in prevention and treatment of various diseases such as diabetes mellitus. The current study was performed to determine whether Opuntia ficus-indica is associated with diabetes. Diabetic rat models were induced with streptozotocin (STZ). This study divided rats into 1 day (short-term) and 4 consecutive weeks (long-terms) of daily administration. These groups were subdivided into four groups each other for assessment of blood glucose level as follows: Group 1, untreated rats given distilled water; Group 2, untreated rats given Opuntia ficus-indica; Group 3, STZ-induced diabetic rats given distilled water; Group 4, STZ-induced diabetic rats given Opuntia ficus-indica. Blood glucose level was measured for one day and four weeks. In addition, serum markers of alanine aminotransferase (ALT), aspartate transaminase (AST), cholesterol, and creatinine were determined, and total protein triglycerides were measured at four weeks. Blood glucose level was highest in both groups (Group 3 and Group 4) at 30 minutes and two weeks and gradually decreased in a time-dependent manner. The difference in blood glucose among the four groups was significant (p < 0.05). Additionally, the levels of ALT, AST and triglycerides were significantly decreased by Opuntia ficus-indica.
Protective effects of cheonggukjang fermented with 20% red ginseng (RC) were observed in streptozotocin (STZ)-induced diabetic rats by measuring levels of blood glucose, serum lipid profiles, and hepatic reactive oxygen species generating and scavenging enzymc activities. RC diet was prepared by mixing with AIN-76 diet at the final concentration 2%, and it was fed to STZ-induced diabetic rats for 3 weeks. The RC dict was significantly improved body weight, feed efficiency ratio, levels of serum glucose, and serum and hepatic lipids in diabetes. The significantly elevated O type activity of xanthine oxidase in diabetes was also greatly decreased by the RC diet. The treatment of RC showed the improved hepatic glutathione s-transferase activities in the diabetic animals. The present study indicates that cheonggukjang fermented with red ginseng could ameliorate STZ-induccd diabetic symptoms such as aggravated blood glucose levels, serum lipid profiles, and even the conditions of oxidative stress.

Objectives : For evaluation of preventive and anti-diabetic activities of Cinnamomi ramulus(CR) herbal acupuncture on pancreatic islet damage in streptozotocin(STZ)-induced diabetic rat. Methods : CR herbal acupuncture was performed at Bisu($BL_{20}$) for 3 weeks subcutaneously starting1 week before STZ i.p. injection. SD rats were divided into four groups(n=10 for each group); 1) NC group, non-treated normal control group, 2) STZ group, STZ administered control group, 3) CR125 group, CR(125mg/kg) + STZ administered group, and 4) CR250 group, CR(250mg/kg) + STZ administered group. Results : Both of CR250 and CR125 groups showed increase in insulin secretion and decrease in the level of serum triglyceride and non-esterified fatty acid in a dose-dependent manner compared to the STZ group. Only CR250 group showed decrease in the levels of glucose and total cholesterol compared to the STZ group. CR herbal acupuncture prevents $\beta$-cell damage of pancreatic islet, showing round figure on the sections of the pancreas. In the pancreatic cells, expressions of iNOS, JNK-2, P-JNK-1/2 and ERK-1/2 were decreased compared to the STZ group. CR herbal acupuncture solution did not show any cytotoxicity by MTS assay and inhibited expressions of iNOS and COX-2 in the STZ-induced diabetic rats. Conclusions : Therefore, we suggest that CR herbal acupuncture may act as a prophylactic as well as a therapeutic modality for diabetes mellitus.
Objectives : The object of this study was to observe the effects of Gamioryung-san (GOS), which consists of 22 types of herbs, on streptozotocin (STZ)-induced diabetic nephropathy rats. Methods : Three different dosages of GOS were orally administered once a day for 28 days from 3 weeks after STZ treatment. Six groups, each of 8 rats per group were used. Changes on the body weights, blood glucose levels, serum BUN and creatinine levels, urine volumes, and UAER were observed with changes on the kidney malondialdehyde contents and glutathione, dismutase and catalase contents. In addition, histopathology of kidney, pancreas, thymus and spleen were observed. The results were compared with antioxidant silymarin 100 mg/kg, of which the effects on STZ -induced diabetes and related complications are already confirmed. Results : As a result of treatment of GOS 800, 400 or 200 mg/kg for 28 days, STZ-induced decreases of body weights, hyperglycemia, atrophic changes of pancreatic islets with decreases of insulin-immunoreactive cells and decreases of glucagon -immunoreactive cells were inhibited dose-dependently. Increases in kidney weight, serum BUN and creatinine levels, urine volumes, UAER, vasodilated atrophic glomerulus and abnormal tubules were inhibited dose-dependently. Also increases of kidney MDA contents and decreases of GSH contents, SOD and CAT activities, decreases of thymus and spleen weights, and atrophic changes at histopathological observation were also inhibited. The effects of GOS 400 mg/kg showed similar effects to silymarin 100 mg/kg. Conclusions : These results suggest that 400 mg/kg of GOS retarded the STZ-induced diabetic nephropathies as similarly to silymarin 100 mg/kg, through modulations of oxidative stress and immune systems.
This study was conducted to investigate the effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts on blood hemoglobin, $HbA_{1c}$, levels and biomarkers in the streptozotocin (STZ)-induced diabetic rats. Male Wistar rats divided into normal and diabetic groups. The diabetic groups subdivided into the control group (DM) and Araliaceae water extracts supplemented groups: Aralia elata (AE), Acanthopanacis cortex (AC) and Ulmus davidiana (UD). The extracts were supplemented in diet base on 11.42 g of raw Araliaceae/kg diet for 7 weeks. The diabetes was induced by injecting 572 (55 mg/kg B.W., i.p.) once 2 weeks before sacrifying. Relative weights of liver were significantly lowered in the DM group compared to the normal group, whereas those of kidney and heart were significantly increased in the DM group. Supplementation of the Araliaceae water extracts improved reduced liver weights in STZ-induced diabetic rats. Blood glucose level was significantly higher in the DM group than in the normal group, whereas insulin contents were significantly lowered in the DM groups. However, these parameters were normalized in the An, AC and UD supplemented groups, respectively. Blood hemoglobin and $HbA_{1c}$ levels were significantly higher in the DM group than in the normal group. When all of Araliaceae water extracts were supplemented to the diabetic rats lowered hemoglobin and $HbA_{1c}$ levels. Red blood cell, white blood cell and Lymphocyte were significantly higher in the DM group than in the normal group. The supplementation of Araliaceae family water extracts significantly lowered these parameters compared to the DM group. MCV, MCH contents were declined in the DM group, while the supplementation of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts elevated of these contents in STZ-induced diabetic rats. Accordingly, these results indicate that Aralia elata, Aeanthopanacis corex and Ulmus davidiana water extracts would seem to improve the blood biomarkers in STZ-induced diabetic rats.
Nutritional concentrations by chemical analyses of mushroom fermented milk were protein 2.87%, fat 0.09%, carbohydrates 6.0%, dietary fiber 0.3%, lactose 2.01%, sucrose 1.23%, calcium 95.9 mg/100 g and iron 0.08 mg/100 g. The present study was undertaken to investigate the hypoglycemic effects of the equal volume of either water (streptozotocin (STZ)rontrol rats), mushrooms water-extract (STZ-extrart fed rats), mushroom fermented milk product (STZ-mushroom yogurt fed rats) or mushroom fermented milk supernatant (STZ-supernatant fed rats) (10%, v/w), in STZ-induced diabetic rats for 3 week period. The mushroom fermented milk given to the STZ-diabetic rats decreased the blood glucose significantly and increased the blood insulin, compared with the STZ-control rats. The supernatant and mushroom water extract also slightly retarded the development of hyperglycemia in the STZ-diabetic rats. Taken together the results, the mushroom yogurt may have a potential for the hypoglycemic effect in the STZ-diabetic rats.
The hypoglycemic effects of butanol(BuOH) fraction of Polygonatum odoratum with vitamin E in streptozotocin(STZ)-induced diabetic rats were investigated. Sprague-Dawley rats weighing 200-230g were devided into five groups, and four groups induced diabetes mellitus by the STZ injection(45mg/kg b.w.) into the tail vein : Normal, diabetic-control, and three diabetic experimental groups(p. odoratum group, P. od-vit. E group and Vit. E group). All groups were fed on a AIN-76 diet, and the experimental groups were orally administered with the BuOH fraction of Polygonatum odoratum(500mg/kg b.w.) and vitamin E(10mg/kg b.w.) for 14 days. The body weight, diet intake and organ weights were monitored. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol and aspartate and alanine aminotransferase activities were analyzed. The levels of glycogen in liver and muscle, cholesterol in liver were determined. The malondialdehyde(MDA) levels in liver, kidney and lung were assayed. The body weight loss was seen in P. odoratum group, P. od-vit. E group, Vit. E group and diabetic control group, while the loss in P. odoratum group was much less than that in the diabetic control group. The plasma glucose levels were significantly lowered in P. odoratum group compared to diabetic control group. The plasma insulin levels were noticeably higher in P. odoratum and Vit. E groups. The rats in P. odoratum and P. od-vit. E group showed higher liver glycogen levels than in the diabetic control group. The MDA levels in liver, kidney and lung were also significantly reduced in P. od-vit. E and Vit. E groups compared to the diabetic control group. The results suggest that the administration of BuOH fraction of Polygonatum odoratum along with vitamin E reduced blood glucose levels and peroxidative tissue damage in STZ-induced diabetic rats, showing the possibility of preventive and therapeutic use of the wild edible plant to the diabetes mellitus. The investigation assessed the influence of Compositae plants consumption on the protein profile in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Sprague-Dawley rats by injection of STZ (45 mg/kg body weight) into tail vein. The rats were randomly assigned to five groups: normal and STZ-control fed an AIN-93 diet, and groups whose diets were supplemented with 10% Compositae powder containing Artemisia iwayomogi (A. iwayomogi), Atractylodes lancea (A. lancea) or Taraxacum mongolicum (T. mongolicum). To observe the effects of Compositae plants in the animal model, the levels of protein in liver, kidney, lung, pancreas, and brain were determined after 4 weeks. The level of protein in kidney increased significantly in rats receiving the A. iwayomogi- and T. mongolicum-supplemented diet compared to the STZ-control group. The level of protein in lung was increased significantly in the A. iwayomogi-supplemented group. Blood glucose level correlated well with brain protein level but did not correlate with other protein levels. Also, blood glucose correlated inversely with kidney, lung and brain protein levels. It is suggested that supplementation with A. iwayomogi in diabetic rats leads elevates protein in kidney and lung.
The purpose of this study was to investigate the effects of a butanol fraction of fraction of Alisma canaliculatum All. Braun et Bouche (Ac), and of selenium (Se), on plasma gllucose and lipid levee in streptozotocin (STD-induced diabetic rats. Male Sprague-Dawley rats, fed the AIN-93 recommended diet, were divided into five groups: a non-diabetic control group (no STZ treatment), and four 572-induced diabetic groups which consisted of a diabetic-control group, an Ac-treated group, an Ac-Se treated group, and a Se-treated group. Diabetes was induced in the rats by an injection of STZ into the tail vein at a dose of 45 mg/kg body weight. The butanol (BuOH) fraction of Ac was orally administered at a rate of 400 mg/kg body weight for 21 days to both the Ac and Ac-Se groups. The supplementation of selenium in the Se and Ac-Se groups was achieved by adding (freshly, every day) 2 mg of Se as Na$_2$SeO$_3$ per kg of feed. The rats'body weights and hematocrit (Hct) levels were measured, along with plasma levels of glucose, insulin, cholesterol, HDL-cholesterol, triglyceride (TG), and free fatty acids (FFA). Aminotransferase activities were also analyzed. The non-diabetic rats gained weight, while the diabetic rats lost weight - except in the Ac-Se group, which maintained their initial weight. The blood glucose levels of the Ac group and the Se group were significantly lower than for the diabetic-control group. The plasma triglyceride levels were lowered when both Ac and Se were administered to diabetic rats. The concentrations of plasma FFA in the Ac-Se group were significantly lower compared with the diabetic-control group. Plasma cholesterol levels and alanine aminotransferase activity in the Ac, Ac-Se, and Se groups were significantly lower when compared with the diabetic-control group. Aspartate aminotransferase activity was significantly lower in the Se group compared to the other diabetic groups. These data show that treatment with a butanol fraction of Ac in combination with Se has no synergistic effect. Plasma glucose levels tended to be low when Se was administered to diabetic rats. Supplementation of Se in diabetic rats did not elicit a significant increase in plasma insulin levels or result in hypolipemic effects.
The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.
