Effects of Araliaceae on Lipid Levels of Plasma and Liver in Streptozotocin Induced Diabetic Rats (두릅나무과 식물이 Streptozotocin으로 유발한 당뇨 흰쥐의 혈장과 간조직 중의 지질농도에 미치는 영향)

  • This study was conducted to investigate the effects of Araliaceae water extracts on lipid concentrations in streptozotocin(STZ)-induced diabetic rats. The Male Wistar rats were divided into normal and diabetic group. The diabetic group was futher subdivided into the control group(DM) and the Araliaceae water extracts supplemented group: Aralia elata(AE), Acanthopanacis cortex(AC) and Ulmus davidiana(UD). The extracts were supplemented with 1.14% of raw Araliaceae/kg diet for 7 weeks. Diabetes was induced by injecting STZ(55 mg/kg B.W., i.p.) once 2 weeks before sacrifying. The net weight gain and feed efficiency ratio were significantly lower in the STZ-induced diabetic group than in the normal group. However, all of the Araliaceae water extracts supplemented groups resulted in an increase of body weight compared to the DM group. The triglyceride, total cholesterol and free cholesterol concentrations in plasma and liver were significantly higher in the DM group than in the normal group. However, the supplementation of Araliaceae water extracts increased plasma HDL cholesterol concentration, while decreased plasma VLDL, LDL-cholesterol concentra-tions in Araliaceae water extracts supplemented group.

Effect of the New Diabetic Formula on Streptozotocin-induced Diabetic Rats (Streptozotocin 당뇨모델에 대한 신당뇨방의 효과)

  • The present study evaluated the beneficial effect of new diabetic formula(NDF) in diabetic rats. Adult Sprague-Dawley rats weighing 250-300 g were used. Diabetes mellitus was induced by intramuscular injections of streptozotocin(STZ, 50㎎/㎏). The extracts of NDF were orally administered at low or high dose two times a day to fasted diabetic rats for 3 weeks. Adminstration of NDF alliviated a significant reduction in weight gain in rats with STZ-induced diabetes. Following acute and repeated treatment, low dose of NDF suppressed the blood glucose concentrations of fasted diabetic rats. Repeated adminstration of NDF for 21 days improved liver and kidney functions in diabetic rats, as indicated by decline of serum alanine aminotransferase(AL T), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine level and kidney weights. The present study showed that NDF exerted antihyperglycemic effects and alliviated liver and renal damages caused by streptozotocin-induced diabetes.

Hypoglycemic effects of nano powder propolis on streptozotocin-induced diabetic rats (Nano powder propolis 투여가 streptozotocin으로 유발된 당뇨병 rat의 혈당에 미치는 영향)

  • To evaluate the hypoglycemic effect of nano powder propolis, streptozotocin (STZ) induced diabetic rats were divided into 2 groups : diabetic control group and nano powder propolis (0.9ml) group. Then the rats were fed with nano-powder-propolis for 4 weeks. After 4 weeks, oral glucose tolerance test (oral GTT) was performed and blood sugar levels after 16 hours fasting, body weights, and blood lipid levels were measured. Finally, pancreas were collected and examined by histopathology and immunohistochemistry. In conclusion, the nano-powder-propolis was effective in the treatment of diabetes due to the reduction of blood sugar level and the regeneration of damaged ${\beta}$-cells observed in streptozotocin-induced diabetic rats.

Effects of Nefopam on Streptozotocin-Induced Diabetic Neuropathic Pain in Rats

  • Background: Nefopam is a centrally acting non-opioid analgesic agent. Its analgesic properties may be related to the inhibitions of monoamine reuptake and the N-methyl-D-aspartate (NMDA) receptor. The antinociceptive effect of nefopam has been shown in animal models of acute and chronic pain and in humans. However, the effect of nefopam on diabetic neuropathic pain is unclear. Therefore, we investigated the preventive effect of nefopam on diabetic neuropathic pain induced by streptozotocin (STZ) in rats. Methods: Pretreatment with nefopam (30 mg/kg) was performed intraperitoneally 30 min prior to an intraperitoneal injection of STZ (60 mg/kg). Mechanical and cold allodynia were tested before, and 1 to 4 weeks after drug administration. Thermal hyperalgesia was also investigated. In addition, the transient receptor potential ankyrin 1 (TRPA1) and TRP melastatin 8 (TRPM8) expression levels in the dorsal root ganglion (DRG) were evaluated. Results: Pretreatment with nefopam significantly inhibited STZ-induced mechanical and cold allodynia, but not thermal hyperalgesia. The STZ injection increased TRPM8, but not TRPA1, expression levels in DRG neurons. Pretreatment with nefopam decreased STZ-induced TRPM8 expression levels in the DRG. Conclusions: These results demonstrate that a nefopam pretreatment has strong antiallodynic effects on STZ-induced diabetic rats, which may be associated with TRPM8 located in the DRG.

Effects of Cynanchum wilfordii Extract on Serum Lipid Components and Enzyme Activities in Hyperlipidemic and Streptozotocin-Induced Diabetic Rats (백하수오 추출액이 고지혈증 및 Streptozotocin 유발 당뇨성 흰쥐의 혈청 지질성분 및 효소활성에 미치는 영향)

  • The purpose of this study was designed to observe the effects of the feeding Cynanchum wilfordii extract on the improvement of the blood glucose, lipid components in the serum of dietary hyperlipidemic and streptozotocin(STZ) -induced diabetic rats(S.D. strain, ♂) fed the experimental diets for 5 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-Cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose in serum were significantly higher in the cholesterol administration groups((group 2(cholesterol+water), 4(cholesterol+Cynanchum WIlfordii 3.5g% extract)) than those in the control group (group1 , basal diet+water). But the concentrations of total cholesterol. atherosclerotic index, LDL, LDL- cholesterol. free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum were remakably lower in the group 4 than those in the group 2. In the STZ(55mg/kg B.W.)-induced diabetic groups((group 3(STZ, IP.)+water), 5(STZ(IP.)+Cynanchum WIlfordii 3.5g% extract? the serum total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose concentrations actions were rather lower in the group 5 than those in the group 3. In the ratio of HDL -cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Cynanchum wilfordii extract administration groups were higher percentage than III the groups 2 and 3. The activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase (ALP) and lactate dehydrogenase(LDH) in serum were rather lower in the Cynanchum wllfordii extract administration groups (group 4,5) than in the cholesterol diet group(group 2) and STZ-induced diabetic group (group 3). From the above research, the physiological activity substances in Cynanchum wllfordii were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic and STZ-induced diabetic rats. And particularly, physiological activity substance in Cynanchum wilfordii was more effective therapeutic regimen for the control of metabolic derangements in adult disease.

The Effects of Alisma canaliculatum Butanol Fraction with Selenium on Glycogen Level, Lipid Metabolism and Lipid Peroxidation in Streptozotocin-Induced Diabetic Rats (택사 Butanol 분획물과 Selenium 보충이 당뇨 흰쥐의 글리코겐 함량, 지질대사 및 지질과산화에 미치는 영향)

  • The purpose of this study was to investigate the effect of butanol (BuOH) fraction of Alisma canaliculatum (Ac) and/or selenium (Se) treatment on glycogen level, lipid metabolism and lipid peroxidation in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were assigned to one of the five groups: normal, STZ-control, and three experimental groups (Ac group, Ac-Se group, and Se group). Diabetes was experimentally induced by intravenous administration of 45 mg/kg of STZ in citrate buffer. The BuOH fraction of Ac (400 mg/kg bw) was orally administered for 3 weeks. The Se group were fed a AIN-93 recommended diet mixed with Na$_2$SeO$_3$ (2 mg/kg diet). The liver glycogen level of Ac and Ac-Se groups were significantly higher, when compared with the STZ-control groups. The muscle glycogen level was not significantly differ among all groups. The levels of liver triglyceride were higher in Ac-Se group than the STZ-control group. Pancreas protein levels were significantly increased in Ac-Se group than STZ-control group. The concentration of liver malondialdehyde (MDA) was significantly decreased in Ac and Se groups and decreased in Ac-Se group. Administration of BuOH fraction of Alisma canaliculatum and selenium supplementation increased the liver glycogen and triglyceride levels, and reduced peroxidative liver damage in STZ induced diabetic rats. These results suggest that treatment with a BuOH fraction of Alisma canaliculatum in combination with selenium has no synergistic antioxidative effect. Selenium supplementation may lead a decrease MDA of liver in diabetic rats.

Effect of Saponin from the Shoot of Aralia elata in Normal Rats and Steptozotocin Induced Diabetic Rats (두릅추출물이 정상쥐 및 당뇨쥐에 미치는 영향)

  • The purpose of this study was to investigate the effect of saponin in the shoot of Aralia elata on serum lipid level and glucose in streptozotocin(STZ) induced diabetic rats. Sprague Dawley normal male rats weighing 70$\pm$5g were randomly assigned to normal(control group), crude saponin(S group), and shoot of Aralia elata(D group). Experimental diets have been fed for 6 weeks. STZ induced diabetic rats were classified to diabetic control(DC group) and crude saponin(DS group). Diabetic rats were experimentally induced by intravenous injection of STZ(65mg/kg of body weight) dissolved in citrate buffer(pH 4.5). DS group has been i.p. injected with crude saponin solved in phosphate buffer(pH 7.0, 10mg/100g body weight) and DC group fed for 10 days. Body weight decreased significantly in crude saponin group. Feed intakes and feed efficiency ratio were not significantly different among C, S, and D group. The crude saponin group has indicated the lowest values of serum total cholesterol, glucose, and triglyceride. However, the values of serum glucose and triglyceride were not significant. Insulin levels among the crude saponin group, the shoot powder group, and the control group were not significantly different. When STZ induced diabetic rats have i.p. injection of crude saponin, the crude saponin has reduced the serum glucose but it is not been significant.

Antidiabetic Effect of Ethanol Extract of Liriope platyphylla in Streptozotocin Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨쥐에서 맥문동 에탄올 추출물의 당뇨개선효과)

  • The ethanol extraction yield of Liriope platyphylla(Lp) was about 30.7% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities effects of Lp in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in Lp treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in Lp treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in Lp treated group. Also the activity of glucokinase(Gk) was increaed in Lp treated group. The content of hepatic glycogen was significantly increaed in Lp treated group. These results indicated that ethanol extract of Lp would have antidiabetic effect in STZ-induced diabetic rats.

Effect of Jaeumyangyung-tang on the Hyperglycemic Mice Induced with Streptozotocin (자음양영탕이 Streptozotocin(STZ)로 유발된 생쥐의 고혈당에 미치는 영향(影響))

  • Objectives : The aim of this study was to investigate the effect of Jaeumyangyung-tang(滋陰養榮場(JY), JA-0.65g/kg;JB-1.3g/kg) on hyperglycemic mice induced with streptozotocin(STZ). Methods : The experiment operated for 6 weeks. The rats were divided into 3 groups : diabetic group(control group), diabetic group treated with JA(0.65g/kg) for 6 weeks, and diabetic group treated with Jb(1.3g/kg) for 6 weeks. Results : In the STZ-induced diabetic group, blood glucose levels significantly increased as well as the loss of body weight. The levels of serum glucose decreased significantly (p<0.05 or p<0.01) in the JA and JB groups compared with the control. According to a tolerance test, intraperitoneal glucose was ameliorated in the JA and JB groups. The blood urea nitrogen (BUN) and creatinine levels slightly decreased. Histologic analyses of the pancreases revealed that the ${\beta}-cells$ on Langerhans' islets were destroyed by STZ, but the ${\beta}-cell$ mass was larger in the JY than in the control mice. Conclusions : These results indicate that JY can exert beneficial effects on diabetes. preservation of in vivo ${\beta}-cell$ function and regeneration of ${\beta}-cell$ dysfunction by STZ.

  • The purpose of this study was designed to observe the effects of the Prunus persica Batsch var. davidiana Max. hot-water extract on the improvement of the glucide and lipid metabolism in the serum of streptozotocin (STZ, 55, mg/kg B.W., I.P. injection)-induced diabetic rats(S.D. strain, male) fed the experimental diets for 5 weeks. Electrolyte(Na, K, Cl) concentration in serum were fairly reduced in the group BSP(basal diet+STZ+Prunus persica $5.0g\%$ extract) than in the STZ(I.P.)-induced diabetic rats group(group BSW, basal diet+STZ(I.P.)+water). Although there was no significant difference among the groups. Concentrations of free fatty acid and lipid peroxide in serum were significantly higher in the STZ-induced diabetic group(group BSW) and STZ+Prunus persica $5.0;g\%$ extract group(group BSP) than those in the control group(group BW, basal diet+water). However, the concentrations of free fatty acid and lipid peroxide in serum were remarkably reduced in the group BSP than those in the group BSW, The activity of creatine phophokinase In serum was significantly lower in the group BSP than in the group BSW However, the activity of LCAT in serum was increased in the group BSP(Prunus persica $5.0\;g\%$ hot-water extract administration group) than in the STZ-induced diabetic group(group BSW). The above results shows that Prunus persica Batsch var. davidiana Max. were effective on the improvement of the glucide and lipid metabolism in serum of streptozotocin-induced diabetic rats.
