Application of a solid-phase fluorescence immunoassay to determine streptomycin residues in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major)

  • Parallux (IDEXX Laboratories, USA), a solid-phase fluorescence immunoassay (SPFIA) developed for antibiotics residue detection in milk, was applied for analysis of antibiotics in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major). Fishes were dipped in streptomycin 20 g/ton water, the recommended therapeutic dose, for 3 consecutive days. Muscle was sampled after 1st, 2nd, 3rd, 4th and 5th day drug treatment. The concentration of streptomycin in muscle, determined by SPFIA, was compared with that of internal standard (200 ppb as streptomycin). The absorbance ratio of sample to internal standard (S/C) was employed as an index to determine the muscle residues in fishes. The standard solutions were added to muscle samples to give final concentrations in muscle of 0.2 and 0.5 mg/ml to investigate the recovery rate. The recovery rates of all spiked samples were > 84% of the spiked value. Streptomycin was detected in muscles of fishes treated after the 1st day of withdrawal period. The present study showed that the SPFIA can be easily adopted in predicting muscle tissue residues for streptomycin in farmed fishes.

  • 우리나라의 주요(主要) 포도품종(葡萄品種)을 공시(供試)하여 SM처리(處理)에 의(依)한 단위결과(單爲結果) 유기(誘起) 방법9方法)을 조사(調査)하였던 바 다음과 같이 결과(結果)를 얻었다. 1. Schuyler, Delaware, 대평(大平) Dela 및 다노렛드는 SM처리(處理)에 의(依)하여 무핵화(無核化) 재배(栽培)가 가능(可能)하였다. 또한 SM 200ppm에 GA 25ppm을 가용(加用) 처리(處理)하므로서 단위결과(單爲結果) 유기(誘起)나 착립(着粒)의 면(面)에서 안정성(安定性)을 높일 수 있었다. 2. SM처리(處理)에 의한 무핵과(無核果) 유기(誘起) 가능시기(可能時期)는 Schuyler는 만개(滿開) 5~10일전(日前), Delaware는 만개(滿開) 10~18일전(日前), 대평(大平) Dela는 만개(滿開) 7일전(日前), 다노렛드는 만개(滿開) 0~12일전(日前) 처리(處理)가 좋았다. 3. Campbell Early는 SM 200ppm과 초산(醋酸)칼슘 0.05M이나 BA 150ppm을 첨가(添加)하여 처리(處理)할 때 무핵율(無核率)이나 착립율(着粒率)을 높일 수 있으나 아직 실용화(實用化)할 수 있는 정도(程度)는 아니었다. 4. 거봉(巨峰)과 Pione의 경우는 SM처리(處理)에 의하여 용이(容易)하게 단위결과(單爲結果)는 유기(誘起)할 수 있었으나 착립수(着粒數)나 과립중(果粒重)이 수세(樹勢), 환경(環境) 등(等)에 따라 안정성(安定性)이 없었다. 5. Campbell Early와 다노렛드 품종(品種)에서 SM 처리(處理)는 화분(花粉)의 발아력(發芽力)을 현저(顯著)히 감퇴(減退)시켰다.

The Suppression of Maturational Competence by Streptomycin during In vitro Maturation of Goat Follicular Oocytes

  • Antibiotics are common additives in culture media during in vitro embryo development, but their effects on oocyte maturation in vitro have not been tested. The effects of penicillin, streptomycin and gentamicin on the maturational competence and subsequent development potential of goat follicular oocytes were examined after parthenogenetic activation in vitro. Maturation rates at 24 h after in vitro maturation, and parthenogenetic development at 48 h after activation, were evaluated by observing the protruding first polar body and the 4 cell stage cleavage, respectively. When streptomycin was present in the maturation medium, the percentages of matured oocytes 24 h after activation were significantly (p<0.01) lower than those from the other groups (42.5-45.7% vs. 69.1-73.8%). Penicillin and gentamicin treatment did not affect the maturation rates or the percentages reaching the 4 cell stage 48 h after activation. There was no significant difference in cleavage rates among the different antibiotic treatments 48 h after activation. Therefore, streptomycin suppresses the in vitro maturation of immature goat oocytes, but does not influence their subsequent development.

우유(牛乳)의 잔류항생물질(殘溜抗生物質)에 관(關)한 연구(硏究) (Studies on Antibiotic Residues in Cow Milk)

  • The antibiotics are usually used for the medical treatments or the other purpose for cow and almost of them are transfered to milk. Moreover a serious problems on public health are caused by misuse or overuse of antibiotics. These studies were conducted to determine the rise and fall of antibiotics in the raw milk which is milked from the cow treated with antibiotics. The number of samples examined were 500 which were taken from 36 cows, and the antibiotics in milk were detected by cylinder plate assay method. The results were as follows: 1) When penicillin, chloramphenycol and streptomycin were infused into a udder, they were detected up to the period of 84 hours, 36 hours and 60 hours after injection in the milk from the same udder, however in the milk from the other udder streptomycin was not detected but penicillin and chloramphenycol were detected up to 72 hours and 24 hours after injection. When streptomycin and penicillin were injected intramusculary, streptomycin was not detected in the milk but penicillin was detected up to 60 hours after injcetion. 2) When penicillin, streptomycin and chloramphenycol were infused into the udder, the lesser amount of milk was produced, the more antibiotics were detected in the milk. 3) It is necessary policy for the public health to inhibit to be sold the milk up to 3 days or more after the injection on antibiotics or to add food blue to mastitis drugs.

Pseudomonas syringae pv. syringae에 의한 애호박 세균점무늬병 방제를 위한 약제 선발 (Selection of the Antibacterial Agents for Control Against Pseudomonas syringae pv. syringae causing Leaf Spot Disease on Green Pumpkin (Cucurbita moschata))

  • 공주와 논산 육묘장에서 Pseudomonas syringae pv. syringae에 의한 세균점무늬병이 애호박에 심각한 피해를 주고 있지만, 점무늬병을 방제하기 위한 방제법이 개발되어 있지 않다. 종자처리와 방제약제를 선발하기 위하여 살균제 6종을 선택하여 병원균의 생장억제효과, 유묘에서 병 방제효과, 종자처리에 의한 병원균 및 병 방제효과를 각각 조사하였다. King's B배지에서 병원균 생장억제효과는 oxytetracycline 170 ppm, oxytetracycline 15 ppm + Streptomycin sulfate 188 ppm, oxolinic acid 200 ppm, streptomycin 200 ppm 처리 순으로 효과가 높게 확인되었다. Oxytetracycline 1.5% + streptomycin sulfate 18.8% WP와 oxytetracycline 17% WP의 유묘처리 시 방제가는 각각 71.4%, 49.4%이었다. Oxytetracycline 1.5% + streptomycin sulfate 18.8% WP를 인위적으로 감염된 종자에 처리하였을 때, 병원균의 생장을 완전히 억제하였으며, 유묘 발아실험에서 96% 방제효과를 나타내었다. Oxytetracycline 17% WP 2,000배 처리구에서 인위 감염 종자로부터 80 cfu/g의 병원균이 검출되었으며, 유묘발아실험에서 90%의 방제효과를 보였다. Streptomycin 20% WP의 2,000배액을 종자 처리하였을 때 병원균이 280 cfu/g으로 검출되었으며, 유묘발아실험에서 60% 방제효과를 나타내었다. 이러한 결과 oxytetracycline 1.5% + streptomycin sulfate 18.8% WP가 Pseudomonas syringae pv. syringae 에 의한 세균점무늬병의 방제와 종자처리를 위한 최적의 살균제로 판단된다.

  • Fecal samples from Kim-Hae farm animals were examined for the frequency of gram-negative enteric organisms resistant to tetracycline, streptomycin, or penicillin. The propertions of antibiotic resistant to total organisms in fecal specimens of poultry, swine and cow were as follows: 95%, 92%, 70% for tetracycline, 100%, 27%, 9% for streptomycin, 18%, 1%, 1% for penicillin, respectively. The bacteria had multiresistance to antibiotics. These strains had more than one plasmid. From the transformation study, it was concluded that the resistance to streptomycin was attributed to one of these plasmids.

유우유방원(乳牛乳房源) Staphylococcus aureus 의 각종 화학요법제(化學療法劑)에 대한 감수성시험(感受性試驗) (Sensitivity of Staphylococcus aureus Isolated From Bovine Udders to Various Chemotherapeutic Agents)

  • The minimum inhibitory concentration (MIC) of five chemotherapeutic agents (penicillin, streptomycin, tetracycline, oxytetracycline and furazolidone) was measured for 126 strains of Staphylococcus aureus isolated from the udder of dairy cattle. The results obtained were as follows: 1. The MIC of penicillin, streptomycin, tetracycline, oxytetracycline and furazolidone ranged from 0.03 to 32 ug/ml, 0.06 to 128 ug/ml, 0.06 to 128 ug/ml, 1.0 to 512 ug/ml, and 0.06 to 32 ug/ml, respectively. The most frequent MIC of the above drugs were; penicillin 0.5ug/ml, streptomycin 1.0ug/ml, tetracycline 0.5ug/ml, oxytetracycline 4.0ug/ml, and furazolidone 2.0ug/ml. 2. The number of strains resistant to penicillin. streptomycin, tetracycline and oxytetracycline were 89(70.6), 9(7.1%), 10(7.9%), and 26(20.6%), respectively. Twenty-eight (29.2%) strains showed multiple resistance to more than two antibiotics tested.

Polymorphism in Intrinsic Antibiotic Resistance of Azospirillum Isolates from Ornamental Plants

  • The polymorphism of Azospirillum isolates from ornamental rhizosphere and two reference strains were examined with respect to intrinsic antibiotic resistance (IAR) profile. All the isolates showed different intrinsic resistances to different antibiotics viz., tetracycline, kanamycin, nalidixic acid, streptomycin, ampicillin, spectinomycin and chloramphenicol. All the strains demonstrated susceptibility to high concentration of all antibiotics used in the present experiment. In addition to these general patterns, we also obseved the multiple antibiotic resistances of Azospirillum strains. The Azospirillum sp. OAD-11 was resistant to tetracycline, streptomycin and ampicillin, and Azospirillum sp. OAD-57 was resistant to tetracycline and streptomycin. Conversely, Azospirillum sp. OAD-9 possessed the dual susceptibility to tetracycline and spectinomycin, whereas Azospirillum sp. OAD-37 was dual susceptible to streptomycin and kanamycin. Such multiple antibiotic resistant/susceptible traits could be useful for the identification of the strains in field experiments or in molecular genetic transfer experiments.

몇가지 항생제의 느타리버섯 Pseudomonas tolaasii에 의한 세균성갈색무늬병 방제 효과 (Effect of some antibiotics on control of bacterial brown blotch of oyster mushroom caused by Pseudomonas tolaasii)

  • 느타리버섯 세균성갈색무늬병 방제 약제를 선발하기 위하여 33종의 항생제를 공시하여 P. tolaasii에 대한 항균활성을 조사하고 선발된 4종의 항생제를 이용하여 방제효과를 검정한 결과는 다음과 같다. Kanamycin등 19개의 항생제가 병원균 P. tolaasii에 대하여 항균활성을 나타내었으며, 그 중에서 tetracycline, streptomycin, kanamycin과 kasugamycin이 강한 항균활성을 나타내었다. Tetracycline과 kanamycin은 10ppm에서도 세균생장을 완전히 억제시켰고 Kasugamycin은 100ppm, streptomycin은 200ppm 이상의 농도에서 병원세균에 항균활성을 보였다. Tetracycline, kanamycin, kasugamycin, streptomycin을 이병균상에 처리한 결과 각각 72.9, 71.2, 68.1 그리고 62.7%의 방제가를 보였다. tetracycline 처리구의 느타리버섯 생산량은 무처리구보다 31.8% 이상의 증수효과를 보였다. 항생제 처리가 느타리버섯 균사생장에 미치는 영향을 조사한 결과 kanamycin을 처리한 배지에서는 항생제 처리의 영향을 전혀 받지 않았으며 오히려 균사생장을 촉진하는 것으로 밝혀졌다. Streptomycin을 처리한 배지에서는 항생제 무처리 배지에서와 동등한 수준의 균사생장을 보였다. Tetracyclin의 경우는 무처리에 비해 약 10에서 20%의 균사 생장 억제를 보였으나 Kasugamycin 100ppm 이상의 농도에서는 약 40%의 균사생장 억제를 보였다.

가금유래 주요병원성세균의 분리와 분리균주에 대한 약제감수성조사 (Isolation of the Pathogenic Bacteria from Chicken and Antimicrobial Drug Sensitivity of the Strain Isolated)

  • A total of 1503 specimens were submitted to the Poultry Disease Diagnostic Service Laboratory during the year 1966 and 1978. The most frequently diagnosed diseases in order of prevalence were avian mycoplasmosis, staphylococcosis, colibacillosis, salmonellosis and pullorum disease, the percentages of the conditions being 24.6%, 20.0%, 18.0%, 12.6% and 6.4%, respectively, The drug resistance of pathogenic mirnoorganisms isolated during the year 1978 from chicken with colicabacillosis, staphylococcosis or salmonellosis were investigated by the use of disc diffusion technique, the results being as follow. 1) Drug resistance of 63 strains of Escherichia coli More than 95% of the strains tested were sensitive to colistin and gentamicin. The percentages of strains sensitive to kanamycin, chloramphenicol, ampicillin and nitrofurantoin were 66.7%, 60.3%, 60.3% and 47.6%, respectively. Majority of the strains were highly resistant to streptomycin and tetracyline. All the strains were resisistant to bacitracin lincomycin, oleandomycin, penicillin and erythromycin. All the strains tested were resistant to more than two among 10 drugs in common use such as penicillin, erythromycin, streptomycin, tetracycline, neomycin, chloramphenicol, kanamycin, ampicillin and gentamicin, and 27 different resistance patterns were noted. The most frequent multiple resistance pattern was PC, EM, SM and TC (11.1%). 2) Drug resistance of 48 strains of Salmonella More than 95% of the strains tested were sensitive to colistin, gentamicin ana ampicillin. The percentages of st rains sensitive to kanamycin, tetracycline, neomycin and nitrofurantoin were 81,3%, 79%, 72.9%, and 68.0% respectively. None of them was sensitive to streptomycin, oleandomycin, erythromycin, lincomycin and bacitracin. All the strains were resistant to more than one among 7 drugs in common use such as streptomycin, erythromycin, neomycin, tetracycline, kanamycin, ampicillin and gentamicin. The most frequent resistance pattern was SM and EM(66.7%). 3) Drug resistance of 54 strains of Staphylococci All the strains tested were sensitive to gentmaicin, kanamycin and cephalothin. Majority of them were highly sensitive to bacitracin, methicillin, nitrofurantoin and chloramphenicol. The Percentages of strains sensitive to streptomycin, ampicillin, lincomycin and tetracycline were 66.7%, 55.6%, 44.4% and 27.8%, respectively. Among them, 51 strains were resistant to more than one among 11 drugs in common use such as tetracycline, lincomycin, ampicillin, penicillin, streptomycin, erythromycin, neomycin, oleandomycin, chloramphenicol, methicillin and bacitracin, and thirty one different resistance patterns were noted.
