A non-inferiority study evaluating a new extended-release preparation of tilmicosin injected subcutaneously vs. ceftiofur administered intramammary, as dry-cow therapy in Holstein Friesian cows

  • Background: A new, extended long-acting tilmicosin (TLAe) preparation was tested against intramammary ceftiofur (CEF) using a non-inferiority trial model during dry-cow therapy (DCT) in a farm with high bovine population density and deficient hygiene application. Objectives: To evaluate the possibility that TLAe administered parenterally can achieve non-inferiority status compared to CEF administered intramammary for DCT. Methods: Cows were randomly assigned to TLAe (20 mg/kg subcutaneous; n = 53) or CEF (CEF-HCl, 125 mg/quarter; n = 38 cows) treatment groups. California mastitis testing, colony-forming unit assessment (CFU/mL), and number of cases positive for Staphylococcus aureus were quantified before DCT and 7 d after calving. A complete cure was defined as no bacteria isolated; partial cure when CFU/mL ranged from 150 to 700, and cure-failure when CFU/mL was above 700. Results: TLAe and CEF had overall cure rates of 57% and 53% (p > 0.05) and S. aureus cure rates of 77.7% and 25%, respectively (p < 0.05). The pathogens detected at DCT and 7 days after calving were S. aureus (62.71% and 35.55%), Staphylococcus spp. (22.03% and 35.55%), Streptococcus uberis (10.16% and 13.33%), and Escherichia coli (5.08% and 15.55%). Non-inferiority and binary logistic regression analyses revealed a lack of difference in overall efficacies of TLAe and CEF. Apart from S. aureus, S. uberis was the predominant pathogen found in both groups. Conclusions: This study is the first successful report of parenteral DCT showing comparable efficacy as CEF, the gold-standard. The extended long-term pharmacokinetic activity of TLAe explains these results.

Evaluation of microbiological, cellular and risk factors associated with subclinical mastitis in female buffaloes

  • Objective: This study aimed to evaluate the microbiological and cellular milk profile for the diagnosis of subclinical mastitis in female buffaloes and to assess risk factors for predisposition of the disease. Methods: Analyses were carried out by standard plate count (SPC), identification of species and antibiotic resistance, somatic cell count (SCC), electrical electrical conductivity of milk (ECM), and lactoferrin content in milk. Teat cups were swabbed to evaluate risk factors, observing hyperkeratosis, milking vacuum pressure and cleanliness of the site. Hence, 30 female buffaloes were randomly selected (15 from a group in early lactation and 15 in late lactation). Results: The most common bacteria in the microbiological examination were Staphylococcus spp., Streptococcus spp. and Corynebacterium sp. In the antibiotic sensitivity test, 10 (58.82%) of the 17 antibiotics tested were sensitive to all isolates, and resistant bacteria were Streptococcus uberis, Streptococcus dysgalactiae, Streptococcus haemolyticus, and Escherichia coli. It was observed that positive samples in the microbiological examination showed total bacterial count between $9.10{\times}10^3$ to $6.94{\times}10^6$ colony forming units/mL, SCC between 42,000 to 4,320,000 cells/mL and ECM ranging from 1.85 to 7.40 mS/cm. It was also found that the teat cups had high microbial counts indicating poor hygiene, and even faults in the cleanliness of the animals' waiting room were observed. It is concluded that values of SCC above 537,000 cells/mL and ECM above 3.0 mS/mL are indications of mammary gland infection for this herd; however, the association of these values with a microbiological analysis is necessary to more accurately evaluate the health status of mammary glands with subclinical mastitis. Conclusion: Through phenotypic characterization of bacteria involved in the samples, the genera Staphylococcus spp., Streptococcus spp., and Corynebacterimum bovis were the most prevalent in this study. Faults in environment and equipment hygienization are factors that are directly associated with mastitis.

수의학강좌 II: 연쇄상구균과 장구균으로 인한 환경성 유방염의 감염위험요인과 통제방법

  • 최근 몇년간 국립수의과학검역원에서 조사한 바에 따르면, 국내 젖소에서 유방염을 일으키는 주된 원인균은 황색포도상구균을 제외하고는 거의 대부분 젖소의 환경으로부터 유래하는 환경성 세균에 의한 것으로 확인된 바 있다. 국내 젖소의 유방염 원인균으로 작용하고 있는 이러한 환경성 세균 중 coagulase-음성 포도상구균 (Coagulase-negative Staphylococci, CNS)과 E. coli 등의 그람 음성균이 가장 많은 비율을 차지하였지만 그 외에 Streptococcus uberis를 비롯한 환경성 연쇄상 구균과 장구균도 상당한 비율을 차지하는 것으로 조사되었으며 이에 대해서는 지난 9월호에서도 상세히 기술한 바 있다. 환경성 연쇄상구균과 장구균은 젖소에서 유선감염 및 임상형 유방염의 중요한 원인이며 환경성 연쇄상구균 중 가장 흔히 발견되는 유방염 원인체는 Streptococcus uberis 로서, 이는 국내에서뿐만 아니라 미국, 유럽, 뉴질랜드, 호주 등에서 수행된 많은 연구결과에서도 마찬가지로 나타났다. 장구균 중 가장 흔히 분리되는 균종은 E. faecium과 E. faecalis로서 검역원에서 지난 몇 년간 조사된 결과에서도 이 두 가지 균 종이 유방염우유에서 분리된 장구균의 90% 이상을 차지하였다. 이러한 환경성 연쇄상구균과 장구균은 사람과 동물의 장관, 분변, 감염된 유방 및 전체적인 목장 환경에서 분리되고 있다. 장구균은 일반적으로 환경성 연쇄상구균/장구균 유방염 감염증 중의 일부일뿐이고 장구균에 의한 유방염이 목장의 주된 문제가 되는 일은 비교적 드물다. 현재, 장구균에 의해 발생하는 유방염은 S. uberis를 통제하기 위해 사용되는 방법과 동일한 방법으로 통제할 수 있다고 여겨진다. 원인체에 상관없이 유방염을 통제하기 위한 기본적인 원칙은 유방염을 일으킬 수 있는 병원체에 유두끝이 노출되는 기회를 줄이거나 감염에 대한 젖소의 저항성을 증가시키는 두 가지 방법이다. 역으로 얘기하면, 병원체에 대한 유두끝 노출을 증가시키거나 감염에 대한 젖소의 저항성을 감소시키는 요인들은 우군 내에서 더 많은 유방염을 발생시킬 가능성이 높기 때문에 위험요인으로 간주된다. 본 고에서는 환경성 연쇄상구균에 의한 유방염과 관련이 있는 것으로 알려진 이러한 여러 가지 위험요인들에 대해 알아보고 그러한 위험요인을 줄일 수 있는 방법에 대해서도 고찰하고자 한다.

Studies on Chronic Mastitis of Dairy Cattle in Kyungnam Province (경남지방(慶南地方)에서 발생(發生)한 젖소의 만성(慢性) 유방염(乳房炎)에 대한 연구(硏究))

  • Some investigations on chronic mastitis in dairy cattle in Kyungnam Province during the year 1982 were conducted with the special reference to the causative agents and their drug resistance. Milk samples from 46 isolated cases of chronic mastitis cattle were investigated bacteriologically and the organisms recovered were examined for their drug susceptibility against the major antibiotics used in this country by the use of disk diffusion susceptibility test. Four major causative agents involved in chronic mastitis were in order of prevalence Staphylococcus aureus (32.6%), Escherichia coli (28.3%), Streptococcus agalactiae (8.7%) and Candida albicans (8.7%). Staph. epidermidis, Streptococcus uberis, Klebsiella pneumoniae and Candida subtropicalis were found to be one of the minor agents. The majority of staphylococcal isolates were highly resistant to the most of antibiotics employed while 8% of them were resistant to gentamicin and 32% to chloramphenicol. The percentages of staphylococcal cultures resistant to penicillin, lincomycin. streptomycin, methicillin, oleandomycin, tetracycline, cephalothin, ampicillin and erythromycin were 100%, 96%, 96%, 92%, 84%, 84%, 80%, 76%, and 64% respectively. Streptococcal isolates were also highly resistant to the majority of the drugs used although 85.7% of them were susceptible to gentamicin. All Escherichia coli isolates were found to be resistant to erythromycin, lincomycin and penicillin while the majority of them were resistant to ampicillin (92.9%), carbenicillin (85.7%), oleandomycin (85.7%), streptomycin(85.7%), kanamycin (78.6%), methicillin (78.6%) and tetracycline (71.4%). The percentages of E. coli cultures resistant to gentamicin, nitrofurantoin, cephalothin and chloramphenicol were 21.4%, 21.4%, 35.7% and 50.0% respectively.

Antibacterial Effects of Propolis Extracts on Pathogenic Bacteria (Propolis 추출물의 병원성 미생물에 대한 항균 효과)

  • The optimal concentration of ethanol to separate a high quantity of propolis was $60\%$ but that for the best flavonoids extraction was $80\%$ We compared the yields of propolis from different countries. In this study we used $60\%$ ethanol concentration as a standard. The yield of propolis was proportional to the contents of flavonoids. Namely, Polish propolis which showed the highest yield with $56\%$ by the extraction with $60\%$ ethanol revealed also the highest flavonoids content with $3.49\%$ among all the samples tested The major constituents of propolis differed from country to country. It has been suggested that the different geographical origin influenced the efficacy and the constituents of propolis. Antibacterial activity of ethanol extracted propolis from different countries was tested against 6 microbial strains of type cultures including Gram-positive (Staphylococcus aureus, Streptococcus uberis, Streptococcus agalactiae) and Gram- negative bacteria (Klebsiella pneumoniae, Proteus vulgaris and E coli) in vitro. Propolis extract showed anti-microbial activity against all the tested bacterial strains. In addition, propolis was sensitive to E coli which was resistant to broad spectrum antibiotics like ampicillin. These results showed that propolis may substituted for commercial antibiotics. The efficiency of anti-microbial activity of the propolis was slightly higher in $80\%$ than $97\%$ ethanol extract.

Purification and Characterization of Lacticin NK34 Produced by Lactococcus lactis NK34 against Bovine Mastitis (Lactococcus lactis NK34에 의해 생산된 소 유방염 원인균에 효과가 있는 lacticin NK34의 정제 및 특성)

  • Lactococcus lactis NK34, isolated from jeotgal (Korean traditional fermented fish), produces bacteriocin against bovine mastitis pathogens such as Staphylococcus aureus 7, S. aureus 8, Staphylococcus chromogenes 10, S. chromogenes 19, Staphylococcus hominis 9, Streptococcus uberis E290, Enterococcus faecium E372, Streptococcus agalactiae ATCC 13813, Pseudonocardia autotrophia KCTC 9455, and Staphylococcus simulans 78. Lacticin NK34 was inactivated by protease XIV but not by protease IX, protease XIII, proteinase K, $\acute{a}$-chymotrypsin, trypsin, and pepsin. Also, lacticin NK34 was stable over a pH range of 2 to 9 for 4 hr and withstood exposure to temperatures of 30-$100^{\circ}C$ for 30 min. Lacticin NK34 showed bactericidal effects against S. simulans 78. This bacteriocin was purified using ammonium sulfate precipitation, ion exchange chromatography, ultrafiltration, and hydrophobic chromatography. Tricin-SDS-PAGE of purified bacteriocin gave the same molecular weight (3.5 kDa) as nisin. The gene encoding this bacteriocin was amplified by PCR using nisin gene-specific primers. It showed similar sequences to this nisin Z gene. These results indicate that lacticin NK34 is a nisin-like bacteriocin, and could be used as an antimicrobial alternative for livestock.

Antimicrobial susceptibility and prevalence of gram-negative bacteria isolated from bovine mastitis (젖소 유방염으로부터 분리한 그람음성균의 분포 및 항생제 감수성)

  • Environmental mastitis has increased particularly in well-managed or low somatic cell countherds that have successfully controled contagious pathogens. Major pathogens of environmental mastitisare Escherichia coli (E. coli) and Streptococcus uberis. The present study was conducted to investigate1,865 quaters of 241 Korean dairy farms from 2001 to 2004. Prevalence of major gram-negative bacteriaisolated from mastitis milk were E. coli (22.7%) and Enterobacter spp. (16.3%) in coliforms and Pseudomoassp. (10.3%) and Serratia spp. (7.9%) in non-coliforms. The results on antibiotic susceptibility by agardifusion test against these pathogens were 86.7% in piperaciliin, 94.6% in cefepime, 85.5% in amikacin,87.7% in gentamicin and so on. In contrast, the susceptibility against ampicillin (41.9%), cephalothin (9.9%),streptomycin (39.9%) and tetracycline (46.7%) appeared to be below 50%. Gram-negative bacteria showed(96.8%). Acording to year, distribution of high $256{\sim}64{\mu}g/ml$ on cephalothin get increased, but the othersare diferent. These findings demonstrate that major gram-negative bacteria were E. coli and Enterobacterspp. isolates, and often encountered the diverse antibiotic resistant patterns.

Efficacy of the AHI Mastitis Detector for the Diagnosis of Bovine Mastitis (전도율측정법(傳導率測定法)(AHI Mastitis Detector)을 이용(利用)한 유방염진단(乳房炎診斷)의 야외적용시험(野外適用試驗))

  • A total of 136 dairy cows were subjected to test for bovine mastitis by AHI mastitis detector, microbiological test (MT) and California mastitis test (CMT). The results obtained were summarlized as follows: MT indicated that the most important causative microorganisms isolated from mastitic milk were Staphylococcus aureus (59.3%), Staphylococcus epidermidis (16.4%), Streptococcus agalactiae (12.4%) and Streoticoccus uberis (4.4%). In Holstein breed, the critical threshold of electrical conductivity values of normal and mastitic milk were found to be less than $6,900{\mu}S$ and above, $7,700{\mu}S$, respectively. Although there was goad agreement (92.0%) between AHI mastitis detector test and CMT for the diagnosis of bovine mastitis, the diagnostic efficiency of AHI mastitis detector (80.0%) was higher than that of CMT (74.0%) when compared with microbiological findings. In addition, handiness and objectivity of AHI mastitis detector for the detection of mastitic milk suggested that this could effectively be used for the diagnosis of both clinical and subclinical bovine mastitis in field.

Medium Optimization for Pediocin SA131 Production by Pediococcus pentosaceus SA131 against Bovine Mastitis Using Response Surface Methodology

  • Pediococcus pentosaceus SA131 was isolated from jeotgal, is the bacteriocin producer against bovine mastitis pathogens, Streptococcus uberis E290, Enterococcus gallinarum E362, and Staphylococcus epidermidis ATCC 12228. The medium composition for pediocin SA131 production by P. pentosaceus SA131 was optimized using response surface methodology. Component of medium was studied as carbon source (glucose, fructose, lactose, glycerol, sucrose, maltose, and mannitol), nitrogen source (beef extract, yeast extract, peptone, malt extract, and tryptone), mineral and surfactant ($MgSO_4$, $KH_2PO_4$, $(NH_4)_2SO_4$, $MnSO_4$, NaCl, sodium acetate, and Tween 80). Through one factor-at-a-time experiment, glucose, fructose, yeast extract, malt extract, NaCl, $MgSO_4$, and Tween 80 were determined as the good ingredient. The effects of major factors for pediocin SA131 production were investigated by two-level fractional factorial designs (FFD). By a $2^4$ FFD, fructose, yeast extract, and $MnSO_4$ were found to be the important factors for the bacteriocin production. Subsequently, a $2^3$ central composite design (CCD) was adopted to derive a statistical model for optimizing the composition of the fermentation medium. The estimated optimum composition for the production of pediocin SA131 by P. pentosaceus SA131 was as follows; 0.13% fructose, 1% glucose, 1.8% yeast extract, 2.58% $MnSO_4$, 0.2% NaCl, and 0.2% Tween 80. The pediocin production under optimized medium was increased to 1,000 AU/mL, compared to the 400 AU/mL in MRS medium.

THE EFFECT OF CELL WALL PROTEINS OF STREPTOCOCCUS SPECIES ON MICROSTRUCTURAL CHANGES OF L929 CELLS (연쇄구균의 세포벽 단백질이 L929 세포의 미세구조 변화에 미치는 영향에 관한 연구)

  • Bacteria are one of the most important causative agents of the pulpal and periapical diseases. Streptococci are one of the most frequently isolated facultative anarerobic bacteria in the infected root canals. Bacterial cell wall components have a direct effect in the pathogenesis of the pulpal and periapical infections. Hyaluronidase produced by bacteria has been implicated in dissemination of the diseases. The purpose of this study was to evaluate the effect of cell wall extract of streptococci on the L929 cells using inverted microscope and the transmission electron microscopy (TEM). Hyaluronidase production of streptococcal strains were investigated to determine the correlation between the severity of cell damage and the activity of enzymes. Bacterial cell wall extracts of S. sanguis, S. mitis and S. uberis isolated from infected root canals and ATCC type strains of S. mutans (ATCC 10449) and E. faecalis (ATCC 19433) were prepared by sonication and confirmed with SDS-PAGE. Silver stain of SDS-PAGE of sonic extract was efficient at $100{\mu}g$/ml concentration of cell wall protein, while Coomasie blue stain was efficient at $100{\mu}g$/ml concentration. Inverted microscope showed that sonic extract-treated L929 cells were round and detached from the substratum while others lost their fibroblastic shapes. Transmission electron microscopic examination revealed that streptococcal extracts induced death of L929 cells. Sonic extracts of streptococci had variable effect on microstructure of L929 cells. significant chromatin condensation was observed in the nucleus of the cells. Disappearance of cell surface microvilli and nuclear fragments with dense chromatin were observed. The cell nucleus had an irregular shape and numerous large vacuoles were seen in the cytoplasm and some breaks of the cell membrane could be seen. Cell organelles were in various stages of destruction and cristae of mitochondria were disoriented or disappeared. Eighteen strains of streptococci did not produce hyaluronidase.
