The preparation of Streptococcus faecalis RSI is used as a medicinal preparation for human intestinal disorders. But the microbe in this preparation is very sensitive to rifampicin. If this preparation is taken with rifampicin, its therapeutic effect can not be expected. To develope rifampicin resistant mutants, the rifampicin sensitive strain S. faecalis RSI was treated with Nmethyl-N'-nitro-N-nitrosoguanidine(MNNG). Twelve strains of the MNNG-induced mutants showed distinct resistance to rifampicin and five mutants were selected for further studies. They also exhibited identical characteristics with the parent S. faecalis RSI when they were tested for lactic acid formation and growth inhibition of E. coli. From in vitro test, it was identified that rifampicin is not inactivated by certain factors of the rifampicin resistant mutants. Conclusively, the rifampicin resistant mutants are efficient strains that have insensitivity against rifampicin and original biochemical characteristics of the parent strain.
Optimum conditions for production of casein phosphopeptides (CPP) from sodium casenate by immobilized cell culture of Streptococcus faecalis var. liquefaciens were investigated. Immobilized cells were made by mixing 60% sodium alginate solution with an equal volume of culture broth at the end of exponential phase and subsequently dropping the mixture into $CaCl_{2}$ solution. Optimum conditions for CPP production by the immobilized cells were the same as those ($50^{\circ}C$, pH 7.0, and 10% substrate concentration) by the crude enzyme solution from the supernatant of culture broth. Optimum loading volume of the immobilized cells into a batch reactor was 30% (w/v). Using a continuous reactor loaded by the immobilized cells under the identified optimal conditions, we were able to produce CPP continuously up to 30 days with a maximum CPP conversion efficiency of 20%.
The broad-host plasmid PAM $\beta_1$ of Streptococcus faecalis DS 5 which codes for erythromycin resistance and lactose utilization was transferred into L. casei M-3 (lac-mutant) by conjugation, but was not transferred by protoplast fusion and protoplast transformation. For conjugal transfer of plasmid PAM $\beta_1$ the method of membrane filter mating was more efficient than that of agar surface mating. The rate of acid production of transconjugant C-1, C-3 was similar to L. casei YIT 9018. The proteolytic activity of transconjugant C-3 was increased 20% higher than that of wild type. Plasmid PAM $\beta_1$ was detected by a11 of the transconjugants. The transconjugants expressed lactose ulitization and erythromycin resistance.
Streptococcus faecalis var. liquefaciens was examined for the presence of plasmid deoxyribonucleic acid. An analysis by agarose gel electrophoresis revealed the presence of at least four plasmids of approxymately 6.8, 5.2, 2.6, and 2.1 Mdal. Two plasmid cured strains were obtained by novobiocin treatment. SKR2, which lost 5.2 mdal plasmid (pSK2) and 2.1 Mdal plasmid(pSK4) was sensitive to lincomycin and erythromycin. However, all cured strains showed identical response as parental strain in sugar fermentation, temperature sensitivity, proteolytic activity, and liquefaction of gelatin. The results imply that pSK2 or pSK4 is associated with antibiotic resistance of Str. faecalis var. liquefaciens.
This study was examined the microbiological adequacy of fish feed treated with high-dose irradiation (5 kGy). 125 flounder (Paralichthys olivaceus) were grouped into 5 and then the fishes were fed the following feeds for 28 days: (1)standard feed; (2)standard feed, inoculated with Edwardsiella tarda ($1{\times}10^8-1{\times}10^9CFU/g$ of feed); (3)standard feed, inoculated with Vibrio anguilarum ($1{\times}10^8-1{\times}10^9CFIT/g$ of feed); (4)standard feed, inoculated with Streptococcus faecalis ($1{\times}10^8-1{\times}10^9CFU/g$ of feed); (5)standard feed, inoculated with Edwardsiella tarda, Vibrio anguillarum and Streptococcus faecalis, and then irradiated the mixed feed to 5 kGy. The flounders feed the mixed diet with Edwardsiella tarda, Wbrio anguillarum or Streptococcus faecalis inoculated feed were showed severe cumulative mortalities of 60, 48 and 52% respectively. The gross and histological changes were observed on the fishes. However, fishs fed with the feed of bacteria inoculation before irradiation demonstrated excellent protection against the bacteria-related disease. The results from experiments with bacteria inoculated feed indicated that the irradiation methods employed were capable of preventing contamination of the fishs with pathogenic bacteria.
This experiment was designed to evaluate effects of freezing and frozen storage on survival of sanitary indicative bacteria in seafoods. Culture of bacteria such as Escherichia coli type I, Citrobacter freundii type I, Klebsiella aerogenes type I and Streptococcus faecalis was inoculated into homogenates of pollack, shrimp, and sardine frozen in a contact plate freezer at $-40^{\circ}C$ and chest freezer at $-20^{\circ}C$, stored at $-20^{\circ}C$, and then survival of the inoculated bacteria was determined over a period of 95 days. Coliform group was highly sensitive to freezing and frozen storage showing survival of about $2\%$ after 95 days of frozen storage at $-20^{\circ}C$, whereas Streptococcus faecalis was relatively resistant with $20\%$ survival rate. The sanitary indicative bacteria count was rapidly decreased in the early stage of frozen storage revealing 90 to $95\%$ loss of coliform group and 40 to $70\%$ loss in case of Streptococcus faecalis after 10 days storage. In determining recovery rate, most probable number (MPN) method gave more reproducible recovery of the tested strain than did the selected agar plate method.

The gamma-radiation sensitivity of three kinds of pathogenic bacteria to fish was investigated. $D_{10}$ values (irradiation dose required to inactivate 90% of the population) of Edwardsiella tarda, Vibrio anguillarum and Streptococcus faecalis were 0.08, 0.10 and 0.44 kGy, respectively. The inactivation factors were 4.50 - 24.30 and 6.75 - 36.45 at radiation doses of 2 and 3 kGy. Raw moist pellets were inoculated with Edwardsiella tarda, Vibrio anguillarum or Streptococcus faecalis. Inoculated feed samples were packaged in air and irradiated at 5 kGy. This dose was effective in controlling the inoculated and general bacteria in fish feed. We consider gamma-radiation to be an effective method to sterilize pathogenic bacteria in fish feed.
The streptococcal plasmid pAM$\beta_{1}$ (erythromycin resistance)was transferred via conjugation from Streptococcus faecalis DS 5 to Lactobacillus casei YIT 9018 by a filter mating method. The transfer frequency depended greatly on the type, pore size and side(front or back) of membrane filter. Water passing through a membrane under reduced pressure induced very tight contact between the cells, increased the transconjugation frequency about 1.3 to 37-fold when Millipore membrane filter (0.45.$\mu$m, front side up) was used.
A simple procedure for rapid isolation of plasmid DNA from lactobacillus species and streptococcus species is described. Lactic acid bacteria were cultured in the TCM broth containing 0.5% glycine and plasmid DNA was isolated from cells treated with mutanolysin by alkaline-detergent lysis method. Good results for releasing and isolating plasmid DNA from lactobacillus species were obtained by treatment of cells with 30$\mu\textrm{g}$ of mutanolysin per ml at 37$^{\circ}C$ for 5 to 10 min. For the streptococcus species, the optimum conditions were slightly different. The procedure could be used for rapid characterization of plasmid DNA in Lactobacillus casei, Lactobacillus acidophilus, Lactobacillus helveticus, Streptococcus lactis, Streptococcus faecalis, Streptococcus faecium, and Streptococcus cremoris strains. Using this procedure, plasmids isolated from $1.5m\ell$ cultures could readily be visualized in agarose gel.
Lactic acid bacteria isolated from Kimchi were tested for inhibitory activity against E. coli, Streptococcus faecalis and Lactobacillus bulgaricus. The inhibitory strains appeared most frequently around the middle stage of fermentation during two weeks of observation, and the majority of them were identified as Pediococcus cerevisiae. The agent(s) responsible for the inhibitory activity of P. cerevisiae was inactivated by heat treatment at moderate temperature, but resistant to proteolytic enzymes. The production of the inhibitory compound(s) decreased when the pH of the medium was maintained at about 7 with phosphate buffer.
