Growth inhibition of Perilla frutescens var. acuta extract (차조기 Perilla frutescens var. acuta.의 항균활성)

  • Objectives : This study was performed to investigate the effects of Perilla frutescens var. acuta extracts on growth inhibition of Streptococcus pyogenes KCTC 3208. Methods : Measured growth inhibition of Streptococcus pyogenes KCTC 3208 by concentration of the Perilla frutescens var. acuta. extract and collection periods of the Perilla frutescens var. acuta. and picking region of the Perilla frutescens var. acuta. Results : The Perilla frutescens var. acuta. extract is effective on retarding growth of Streptococcus pyogenes KCTC 3208. At 3% the growth of Streptococcus pyogenes KCTC 3208 was suddenly retarded. The stem of Perilla frutescens var. acuta. extract in a sunny place on June is more effective then its leaf. Conclusions : These results suggest that Perilla frutescens var. acuta. extract is effective in growth inhibition of Streptococcus pyogenes KCTC 3208.

  • The optimal conditions of sterilization for Streptococcus pyogenes treated with disinfectant and antibiotics were investigated. The survivors of Streptococcus pyogenes had no effect at the concentration of 0.2% hydrogen peroxide but decreased abruptly when the concentration increased from 0.4% to 1.0% Minimum inhibitory concentration values of 0.78, 0.39 and 3.125 $\mu\textrm{g}$/$m\ell$ for erythromycin, tetracycline and cephalexin, respectively, were obtained for Streptococcus pyogenes when incubated at 37 $^{\circ}C$ for 24 hrs. Tetracycline and cephalexin showed bactericidal effect against Streptococcus pyogenes, whereas erythromycin did bacteriostatic effect.

  • Studies were made on the optimal conditions of sterilization for Streptococcus pyogenes treated with heat. The results were as follows: The optimal temperature on growth of Streptococcus pyogenes was 37$^{\circ}C$ and mean generation time was 20 minutes in the logarithmic growth phase. The suspension of Streptococcus pyogenes, adjusted to pH 6-9 and treated with heat at 5$0^{\circ}C$, showed logarithmic death rate. Specific death rate constant(k) values at pH 6-9 were 0.1448, 0.1194. 0.1273 and 0.1707 minute$^{-1}$ , respectively.

  • Streptococcus pyogenes 는 지역 사회 폐렴의 드문 원인균으로 독감, 홍역, 만성 폐쇄성 폐질환등의 합병증으로 대부분 발생하고 진행 경과가 빠르며, 고령 및 패혈증을 동반한 경우 특히 사망률이 높다고 알려져 있다. 저자들은 만성 폐쇄성 폐질환 환자에서 균혈증을 동반한 Streptococcus pyogenes 폐렴 1 예를 보고한다.

  • A 5-year-old boy was admitted to the hospital with a high fever and abnormal gait. Magnetic resonance imaging showed extensive swelling of the quadriceps and adductor muscles around the right hip and anterior thigh. A duplex scan demonstrated a thrombus from the external iliac vein to below the popliteal vein. The blood culture revealed Streptococcus pyogenes. The patient was immediately treated with antibiotics and anticoagulants. Follow-up sonography demonstrated complete disappearance of the venous thrombosis. We report a case of streptococcal myositis complicated by deep vein thrombosis.

Rapid Development of Brain Abscess Caused by Streptococcus Pyogenes Following Penetrating Skull Injury via the Ethomoidal Sinus and Lamina Cribrosa

  • Objective : Streptococcus pyogenes is a beta-hemolytic bacterium that belongs to Lancefield serogroup A, also known as group A streptococci (GAS). There have been five reported case in terms of PubMed-based search but no reported case of brain abscess caused by Streptococcus pyogenes as a result of penetrating skull injury. We present a patient who suffered from penetrating skull injury that resulted in a brain abscess caused by Streptococcus pyogenes. Methods : The patient was a 12-year-old boy who fell down from his bicycle while cycling and ran into a tree. A wooden stick penetrated his skin below the right lower eyelid and advanced to the cranium. He lost consciousness on the fifth day of the incident and his body temperature was measured as $40^{\circ}C$. While being admitted to our hospital, a cranial computed tomography revealed a frontal cystic mass with a perilesional hypodense zone of edema. There was no capsule formation around the lesion after intravenous contrast injection. Paranasal CT showed a bone defect located between the ethmoidal sinus and lamina cribrosa. Results : Bifrontal craniotomy was performed. The abscess located at the left frontal lobe was drained and the bone defect was repaired. Conclusion : Any penetrating lesion showing a connection between the lamina cribrosa and ethmoidal sinus may result in brain abscess caused by Streptococcus pyogenes. These patients should be treated urgently to repair the defect and drain the abscess with appropriate antibiotic therapy started due to the fulminant course of the brain abscess caused by this microorganism.

Antibacterial and therapeutic effects of a combination of Sophora flavescens and Glycyrrhiza uralensis Fischer ethanol extracts on mice infected with Streptococcus pyogenes

  • This study evaluated the antibacterial effects of a mixture of Sophorae radix and Glycyrrhiza uralensis Fischer (1 : 1) ethanol extracts (SGE) on mice infected with Streptococcus (S.) pyogenes. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration of SGE necessary for antibacterial effects against S. pyogenes were $20{\mu}g/mL$. Based on the time-kill curves for S. pyogenes, SGE was effective at $4{\times}$ MIC after 16 h. On Day 12 after challenge, the survival rate of mice treated with 2.0 mg/kg SGE was 60%. In conclusion, SGE had potent in vitro and in vivo antibacterial activities against S. pyogenes.

  • Estrous and non-estrous rabbits were inoculated with E. coli or Streptococcus pyogenes, and the mixture of the two organisms, and bacterial count and histopathological studies of uterine horns were made to observe the effects of estrogen on the resistance of the uterus to bacterial infection. The results obtained were summarized as followings; 1. Four hours after inoculation of bactoria into uterine horn, the number of organisms was significantly lower in estrous rabbits than in non-estrous regardless of the kind of organisms inoculated. 2. The highest reduction rate of the organisms among the three bacterial inoculation groups was found in estrous rabbits inoculated with E. coli, and the lowest reduction rate was with Streptococcus pyogenes. 3. Histopathological changes of uterine horns induced five days after bacterial inoculation were observed. In estrous rabbits, a mild inflammatory reaction was found in Streptococcus pyogenes group, but a slight inflammatory reaction and only a negligible inflammatory reaction were observed in mixed bacteria group, and in E. coli group respectively. In non-estrous group, however, a marked inflammatory reaction was observed in Streptococcus pyogenes group, a moderate inflammatory reaction and a slight inflammatory reaction were observed in the mixed bacterial group and E. coli group, respectively.

Isolation Frequency and Antimicrobial Susceptibility of Streptococcus spp. from Clinical Specimens (임상검체에서 분리된 사슬알균종의 분리빈도와 항균제 감수성)

  • From the total 116,429 clinical specimens submitted to "C" hospital from January 2005 to December 2006, 2,195 strains of streptococci were isolated. Twenty four species of Streptococcus were identified with 0.1~19.8% isolation frequencies, of which S. pneumoniae was 19.8%, S. agalactiae 16.2%, S. anginosus 9.8%, S. constellatus 5.0%, S. oralis 3.9%, S. mitis 3.3%, S. pyogenes 2.7%, S. salivarius subsp. salivarius 2.2%, S. sanguinis 1.9%. For S. pneumoniae, clinical specimens showing over 9.0% isolation rate were 82.8% in sputum, 9.2% in blood, and for Streptococcus species other than S. pneumoniae, 18.0% in sputum, 16.0% in urine and 9.7% in blood. The antimicrobial agents that showed over 90.0% susceptibility were cefotaxime, gatilfloxacin, imipenem, levofloxacin, linezolid, moxifloxacin, rifampin and sporfloxacin in S. pneumoniae, ampicillin, cefotaxime, cetriaxone, levofloxacin, linezolid, penicillin, quinupristin/dalfopristin and vancomycin in S. agalactiae, chloramphenicol, clindamycin, levofloxacin and vancomycin in S. anginosus, levofloxacin, vancomycin in S. constellatus subsp. constellatus, vancomycin in S. oralis, vancomycin in S. mitis, chloramphenicol, clindamycin, levofloxacin, quinupristin/dalfopristin and vancomycin in S. pyogenes, chloramphenicol, levofloxacin and vancomycin in S. salivarius subsp. salivarius, chloramphenicol, levofloxacin and vancomycin in S. bovis II, chloramphenicol, levofloxacin, quinupristin/dalfopristin and vancomycin in S. dysgalactiae subsp. dysgalactiae, levofloxacin, chloramphenicol and vancomycin in the whole group of 10 Streptococcus spp. other than Streptococcus pneumoniae.

Pulsed Field Gel Electrophoresis Profile of Erythromycin-Clindamycin Resistant Streptococcus pyogenes Isolated in Korea (국내분리 Erythromycin-Clindamycin 내성 Streptococcus pyogenes에 대한 Pulsed Field Gel Electrophoresis 양상 분석)

  • Ninety two strains of Streptococcus pyogenes were isolated from patients with pharyngitis, scarlet fever, skin infection, and invasive streptococcal infections in Seoul, Korea from January to December, 1998. All isolates were epidemiologically characterized by T protein serotype, and serum opacity factor (OF) detection to phenotypes. To analyze the genetic relationship, fifty two isolates including 32 erythromycin-clindamycin (Em-Cm) resistant strains, 20 antimicrobial susceptible strains were attempted to the pulsed-field gel electrophoresis (PFGE). T protein serotype showed 16 kinds in distribution including T12 and T4. Among the total isolates, 40 strains (43.5%) belonged to the T12 serotype and twenty strains (21.7%) to T4 serotype. On the other hand, when infection aspect of S. pyogenes isolates were analysed by T serotype distribution, T12 type was predominant for pharyngitidis which contributed to 21 strains (53%) and for skin infection isolates which contributed to 11 strains (28%), respectively. In case of T4 type, it was the most predominant pharyngitidis isolates which contributed to 8 strains (40%). In T serotype distribution of Em-Cm resistant strains, 27 strains (84%) of the thirty two showed T12 serotype. In minimum inhibitory concentration (MIC) values of Em-Cm resistance isolates, thirty two isolates showed resistant to erythromycin 27 strains (84%), had high MIC of >$128\;{\mu}g/ml$. And also to clindamycin, twenty two strains (69%) had high MIC of >$128\;{\mu}g/ml$. When OF detection of Em-Cm resistance of S. pyogenes isolates were analyzed by T serotype distribution, T12 serotype isolates revealed that all of the isolates except one strain were OF negative. In PFGE profile analysis to Em-Cm resistance isolates, of the twenty seven, Em-Cm resistance of T12 serotype isolates, 26 strains showed identical PFGE profile and all of these isolates revealed that OF negative. Eighty four percent of Em-Cm resistance S. pyogenes isolates had identical phenotype and PFGE profile. These results strongly suggested that the Em-Cm resistant S. pyogenes isolates from Seoul area showed close genetic correlation and PFGE could be available tool for molecular epidemiology.
