  • Objectives : Camellia sinensis (Theaceae) possesses a various beneficial effects such as free radical-scavenging, inactivation of urokinase in cancer cell proliferation, antibacterial, and hypotensive. Dental caries is one of the most common oral infectious disease in a human. Oral microorganisms play a significant role in the etiology of dental caries. An aberration to this ecology due to dietary habits, improper oral hygiene or systemic factors lead to an increase in cariogenic microorganisms. Cariogenic microorganisms like Streptococcus mutans and Streptococcus sobrinus encourage the accumulation and adherence of plaque biofilm by metabolizing sucrose into glucans. The purpose of this study was to investigate the antimicrobial activity of phenolic compounds of Camellia sinensis and R-carvone, monoterpenes, is can be found naturally in numerous essential oils, on Streptococcus mutans and Streptococcus sobrinus .Methods : The antimicrobial activity of these compounds was determined by the broth microdilution method and checkerboard dilution assay to investigate the potential synergistic effects of each five compounds of Camellia sinensis (C. sinensis) and R-carvone.Results : C. sinensis-isolated compounds and R-carvone were determined with MIC of more than 1,000 ㎍/mL. However, the combination test showed significant synergism against S. mutans and S. sobrinus, implicated in the lowered MICs.Conclusions : These results suggest that combinatory application of phenolic five compounds (theophyllin, l-theanine, epicatechin, epicatechin gallate, and caffeine) from C. sinensis and R-carvone has a potential synergistic effect and thus may be useful as a mouthrinse in helping control cariogenic microorganism.

충치균 (Streptococcus mutans)에 대한 다시마 추출물의 항균활성 (Antibacterial Activity of Sea-mustard, Laminaria japonica Extracts on the Cariogenic Bacteria, Streptococcus mutans)

  • 우리 나라 연안에서 채취한 해조류 27종의 $80\%$ ethanol 추출물이 Streptococcus mutans KCTC 3300의 증식에 미치는 영향을 검색하고, 다시마의 추출용매별 항균성과, $80\%$ ethanol 추출물을 용매분획하였을 때 획분별 MIC를 측정하였다. S. mutans KCTC 3300에 대한 해조류 추출물의 항균활성은 다시마가 가장 강하였고, 갈파래, 청각, 감태, 곰피 및 미역도 비교적 높은 활성을 나타내었으며 홍조류보다 갈조류와 녹조류에 속하는 종이 대체적으로 강하였다 다시마의 항균활성은 추출용매에 따라 차이가 있었으나 용매별 가용성 획분의 량과 항균성간에는 뚜렷한 관련이 나타나지 않았다. 5. mutans KCTC 3300과 S. sobrinus KCTC 3307에 대한 해조류의 $80\%$ ethanol 추출물의 MIC는 해조류의 종뿐만 아니라 동일한 해조류 추출물의 경우에도 균종에 따라 차이가 있었다. 다시마의 $80\%$ ethanol 추출물을 유기용매를 사용하여 순차 분획하였을 때 항균활성 물질의 대부분은 ether 획 분에 이행하여 이 획분의 MIC는 105$\mu$g/mL이었고, 수용성 획분은 약 5,500$\mu$g/mL의 농도에서도 균의 증식을 저지하지 못하였다.

광중합기를 이용한 광학적 치아우식활성도 검사법 (DEVELOPMENT OF OPTICAL CARIES ACTIVITY TEST USING DENTAL CURING LIGHT)

  • 레이저와 플라스마 광, 할로겐 광 중합기를 이용하여 각 개인의 우식 활성토를 측정할 수 있는지를 규명하기 위해 7-8세 아동 45명을 대상으로 치아의 순면과 협면에 각 광원을 조사하고 특수 필터를 사용하여 초기 치아우식증으로 판별된 치아의 개수를 측정하여 기존의 우식활성도 측정방법인 dDfFtT rate와 mutans streptococci 측정법, Lactobacilli 측정법과 비교, 평가하고 그 특이도, 민감도, 예측력을 평가하였다. 1. 아르곤 레이저, 플라스마 광, 할로겐 광 조사시 육안검사시 보다 유의하게 많은 초기우식 병소가 관찰되었다(p<0.05). 2. 아르곤 레이저, 플라스마 광, 할로겐 광을 이용한 우식활성도 측정법은 dDfFtT rate와 양의 상관관계 (${\gamma}=0.42,\;0.41,\;0.39$)를 보였다(p<0.01). 3. 기존의 우식활성도 측정방법을 기준으로 하여 평가한 결과 특이도와 민감도, 예측력은 모두 레이저가 가장 높았으며 플라스마 광, 할로겐 광 순으로 나타났다. 이상의 결과를 종합하여 볼 때 아르곤 레이저와 플라스마 광, 할로겐 광을 이용한 형광법은 모두 양호한 진단학적 지표를 보여줌으로써 향후 광학적 우식활성검사법으로 활용 가능성이 높을 것으로 사료된다.

Effect of the Fish Oil Fortified Chitosan Edible Film on Microbiological, Chemical Composition and Sensory Properties of Göbek Kashar Cheese during Ripening Time

  • Objective of the present study is to investigate the effect of coated edible films with chitosan solutions enriched with essential oil (EO) on the chemical, microbial and sensory properties of Kashar cheese during ripening time. Generally, no differences were found in total aerobic mesophilic bacteria, streptococci and lactoccocci counts among cheeses but these microorganism counts increased during 60 and 90 d storage especially in C1 (uncoated sample) as compared with coated samples. Antimicrobial effectiveness of the films against moulds was measured on 30, 60, and 90 d of storage. In addition of fish EO into chitosan edible films samples were showed to affect significantly decreased the moulds (p<0.05) as 1.15 Log CFU/g in C4 (with fish oil (1% w/v) fortified chitosan film) on the 90th d, while in C1 as 3.89 Log CFU/g on the 90th d of ripening. Compared to other cheese samples, C2 (coated with chitosan film) and C4 coated cheese samples revealed higher levels of water-soluble nitrogen and ripening index at the end of storage. C2 coated cheese samples were preferred more by the panellists while C4 coated cheese samples received the lowest scores.

  • Xylitol is a five-carbon sugar alcohol that reduces the incidence of caries by inhibiting the growth of oral streptococci, including Streptococcus mutans. Since xylitol is transported via the fructose phosphotransferase system, we hypothesized that it could also affect the growth of other oral bacteria strains. We tested the effects of xylitol against non-periodontopathogenic oral bacteria frequently found in healthy subjects as well as periodontopathogens including Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia. With 5% xylitol, Streptococcus vestibularis and Gemella morbillorum showed marked growth inhibition. With 10% xylitol, all of the tested periodontopathogens and Actinomyces naeslundii showed marked growth inhibition, whereas the growth inhibition of Neisseria mucosa, Neisseria sicca and Veillonella parvula was mild only. Xylitol is a widely used sweetener and the concentration used in our experiment is easily achieved in the oral cavity. If xylitol reduces the growth of periodontopathogens more preferentially, it could also reduce the prevalence of these pathogens and have clinical utility in the prevention or treatment of periodontal disease.

Characterization and antimicrobial efficacy of Portland cement impregnated with silver nanoparticles

  • PURPOSE. This study investigated the effects of silver nanoparticle (SN) loading into hydraulic calcium silicate-based Portland cement on its mechanical, antibacterial behavior and biocompatibility as a novel dental bone substitute. MATERIALS AND METHODS. Chemically reduced colloidal SN were combined with Portland cement (PC) by the concentrations of 0 (control), 1.0, 3.0, and 5.0 wt%. The physico-mechanical properties of silver-Portland cement nanocomposites (SPNC) were investigated through X-ray diffraction (XRD), setting time, compressive strength, solubility, and silver ion elution. Antimicrobial properties of SPNC were tested by agar diffusion against Streptococcus mutans and Streptococcus sobrinus. Cytotoxic evaluation for human gingival fibroblast (HGF) was performed by MTS assay. RESULTS. XRD certified that SN was successfully impregnated in PC. SPNC at above 3.0 wt% significantly reduced both initial and final setting times compared to control PC. No statistical differences of the compressive strength values were detected after SN loadings, and solubility rates of SPNC were below 3.0%, which are acceptable by ADA guidelines. Ag ion elutions from SPNC were confirmed with dose-dependence on the concentrations of SN added. SPNC of 5.0 wt% inhibited the growth of Streptococci, whereas no antimicrobial activity was shown in control PC. SPNC revealed no cytotoxic effects to HGF following ISO 10993 (cell viability > 70%). CONCLUSION. Addition of SN promoted the antibacterial activity and favored the bio-mechanical properties of PC; thus, SPNC could be a candidate for the futuristic dental biomaterial. For clinical warrant, further studies including the inhibitory mechanism, in vivo and long-term researches are still required.

구강악안면 근막간극감염에 관한 임상적 고찰 (A CLINICAL STUDY ON ORAL & MAXILLOFACIAL FASCIAL SPACE ABCESS)

  • Disturbances of the interrelationship among the host, environment, microorganism will cause the infection clinically. Infection can be classified into bacterial, viral, fungal origin, Bacterial infection is most common due to dental caries, periodontal disease. These infections have the potential to spread via the fascial spaces in the head and neck region. We have undertaken clinical studies on infections in the oral and maxillofacial regions by analyzing 78 hospitalized patients in the Dept. of Oral and Maxillofacial Surgery, Dong-A University Hospital from 1994 to 1997. The results were as fellows; l. Odontogenic infections were most common with the incidence of 84.6%. 2. Considering the number of involved space, single space was 83.3%, double or more space was 16.6%. The most common fascial space involved was submandibular space and followed by buccal space, 3. The most causative organism isolated from the pus cultures was streptococci group 35.4%. 4. Antibiotics were administrated in all cases, and surgical incision and drainage was performed in 87.2%. 5. Combined administration of penicillin and aminoglycoside was most common in 34.6%.. 6. 7 cases were diagnosed as Ludwig's angina and tracheostomy was done in 2 cases of them.

폐슬러지 감량화 및 재활용을 위한 오존 처리시 하수슬러지내의 미생물 군집구조의 변화 (Change of Microbial Community on Ozonation of Sewage Sludge to Reduce Excess Sludge Production)

  • The change of the microbial community structure in excess sludge of different sewage treatment plants by ozone treatment was investigated by quinone profiles. The resulting ozone dosage ranged from 0.1 to $0.4gO_3/gTS$. In terms of overall sludge reduction, more than 50% reduction of the total sludge mass could be achieved by ozone treatment at $0.4gO_3/gTS$. Quinone concentration and type in sludge of different treatment plants were remarkably decreases with increasing ozone dose. Ubiquinones(UQs)-8, -10 and MK-8 were still remained in the ozonized sludge at $0.4gO_3/gTS$. The results of this study showed that the remaining microorganisms belong to UQs-8, -10 and MK-8 were difficult to destruct cell membrane or wall by ozonation. Fecal Streptococci and Salmonella were not detected at ozone dose of $0.2gO_3/gTS$, but Fecal Coliform was not detected at ozone dose of $0.4gO_3/gTS$.

넙치의 세균성(細菌性) 질병(疾病) (Bacterial diseases of flounder, Paralichthys olivaceus)

  • Flounder culture has been developed mainly in the western parts of japan, and, to date, following six bacterial diseases have been reported. Bacterial white enteritis occurs in 16 to 30-day-old flounder larvae and often causes mass mortality in seed production. Bacterium named Vibrio sp. INFL invades and multiplies in the mucosae of posterier part of intestine, and causes desquamative enteritis. Gliding bacterial disease occurs mostly in juvenile stage and in spring to summer. Diseased signs are partial discoloration and erosion of skin and fins. Histologically, epidermis are removed, and the causative bacterium, Flexibacter maritimus, multiplies on the surface of demis and invades into the muscular tissue. Vibriosis caused by Vibrio anguillarum and related organisum is one of the well-known diseases among marine fish. Outbreaks of the disease in flounder culture are relatively few, but mass mortalities in fingerlings due to the disease were reported. An outbreak of nocardiosis in the autumn of 1984 has been reported, but since then the disease scarcely occurred. The disease is characterized by formation of abscesses under the skin and white nodes in the gill, heart, spleen and kidney. Streptococcicosis occurs frequently in recent years. Beta-hemolytic streptococcus is the causative bacterium, which possesses the same biochemical and serological characteristics as $\beta$-streptococci isolated from some marine and freshwater fish, and is seemed to related to Streptococcus iniae. Edwardsiellosis is the disease that causes most damage in flounder culture in Japan. Characteristic symptoms are swelling of abdomen and intestinal protrusion from the anus due to accumulation of ascites. Edwardsiella tarda, a well-known pathogen of freshwater fish, is the causative bacterium of the disease.

Macrolide계 항균제 내성 출현과 소아에서의 임상적 적용 (Emergence of macrolide resistance and clinical use of macrolide antimicrobials in children)

  • Macrolide antimicrobial agents including erythromycin, roxithromycin, clarithromycin, and azithromycin are commonly used in the treatment of respiratory tract infections in children. Newer macrolides that have structural modifications of older drug erythromycin show improved change in the spectrum of activity, dosing, and administration. However, recent studies reported that increasing use of macrolide antibiotics is the main force driving the development of macrolide resistance in streptococci. In particular, azithromycin use is more likely to select for macrolide resistance with Streptococcus pneumoniae than is clarithromycin use, a possible reflection of its much longer half life. Recently, erythromycin resistance rates of S. pneumoniae and Streptococcus pyogenes are rapidly increasing in Korea. Two main mechanisms of acquired macrolide resistance have been described, altered binding site on the bacterial ribosome encoded by the ermB gene and active macrolide efflux pump encoded by the mef gene. Relationship between the susceptibility of S. pneumoniae and the response to macrolides has been shown in studies of acute otitis media, but less clear in cases of pneumonia. This article reviews the spectrum of activity, pharmacokinetic properties, mechanisms of action and resistance, and clinical implication of resistance on the treatment of respiratory tract infections in children.
