The urea effect on skin permeation of clonidine was investigated to reduce a log time and to increase a permeability. ICR mouse skin and human skin were used and were assumed to be a two-layer membrane consisted of stratum corneum and viable epidermis. The urea acted as a skin denaturant and humectant in the whole epidermis. Also it enhanced the skin permeability of clonidine about 3.5 times. On the other hand, it enhanced the skin permeability by acting as a humectant in the viable epidermis. But the urea effect on the whole epidermis was shown to be greater than that on the viable epidermis. Therefore, it was found that the effect of urea was greater on the stratum corneum than the viable epidermis. Variation of enhancing effect according to the concentration of urea was not found in the range of 1% to 20%.
In order to reduce the systemic side effects and gastrointestinal irritation after its oral adminitration, ketoprofen was formulated as water-soluble packs. The effects of fatty acids and fatty alcohols on the penetration of ketoprofen through excised rat skins were evaluated. The role of stratum corneum as a protective barrier was also investigated. Fatty acids and fatty alcohols were generally effective in promoting ketoprofen penetration. The flux of ketoprofen through rat skin was maximized when oleic acid or lauryl alcohol was used as an enhancer. As the concentration of fatty acids and fatty alcohols varied from 0% to 10%, the amounts of ketoprofen penetrated were in direct proportion to that of fatty acids but those had no relationship with that of fatty alcohols. The penetration of ketoprofen through stripped skin was enhanced compared to normal skin irrespective of enhancer type, which indicated that the action site of enhancers would be stratum corneum.
The purpose of this study is to establish experimental canine skin barrier disruption model in dog. The study was designed to investigate the predictive properties of acetone damage using as effect parameters transepidermal water loss (TEWL) and stratum corneum (SC) hydration. To compare the structures of SC intercellular lipids in normal and acetone damaged skin, TEM observations were performed. Six clinically normal, male Beagles without dermatological problems were chosen for this study. Acetone damage was performed at 48hrs after clipping. Efficacy measurements for TEWL and skin hydration were performed before ($t_1$) and after ($t_2$) damage in a temperature- and humidity-controlled room ($2{0\sim}22^{\circ}C$, 50-60%). TEWL and SC hydration values were decreased in the acetone damaged model compared with non damaged skin. In TEM observation of acetone damaged sample, the intercellular lipid lamellae exhibit abnormal and incomplete structure compared with those of normal skin. It seems that acetone damage would be one of canine skin epidermal barrier disruption model for the study of canine atopic dermatitis (AD) as well as dry skin in veterinary dermatology research.
The effect of poly[2-methacryloyloxyethyl phosphorylcholine] (pMPC) on the skin permeation property was investigated by performing in vitro skin permeation study of a model drug, nicotinic acid (NA). Effect of pMPC polymer in donor solution on skin permeation rates was evaluated using side-by-side diffusion cells. Also, the structural alterations in the stratum corneum (SC), inter-lamellar bilayer (ILB) and dermis layers in pMPC-treated and -untreated skin sections were investigated with transmission electron microscopy (TEM). The permeation profile of NA without pMPC in donor solution showed biphasic mode: initial $1^{st} phase and 2^{nd}$ hydration phase. The sudden, more than 10-fold increase in flux from the initial steady state (43.5 $\mu g/cm^2$/hr) to the $2^{nd}$ hydration phase (457.3 $\mu g/cm^2$/hr) suggests the disruption of skin barrier function due to extensive hydration. The permeation profile of NA with 3% pMPC in the donor solution showed monophasic pattern: the steady state flux (10.9 $\mu g/cm^2$/hr) without abrupt increase of the flux. The degree of NA permeation rate decreased in a concentration-dependent manner of pMPC. TEM of skin equilibrated with water or 2% pMPC for 12 h showed that corneocytes are still cohesive and epidermis is tightly bound to dermis in 2% pMPC-treated skin, while wider separation between corneocytes and focal dilations in inter-cellular spaces were observed in water-treated skin. This result suggests that pMPC could protect the barrier property of the stratum corneum by preventing the disruption of ILB structure caused by extensive skin hydration during skin permeation study.
In the cosmetics field, facial skins have been classified into four types according to self estimation as well as physiological parameters. The aim of this study is to understand skin condition in the levels and composition of stratum corneum (SC) lipids, and to propose new classification for skin types. We assessed the relationship between the SC lipid composition and the self-estimated skin types or physiological parameters of the skin. Sensitive skin has been of great concern over the last decades, and it should be recognized as a skin type. Therefore, we also investigated the influence of the SC lipid composition on variations of sensitivity evaluated by the Stinging Test. Fifty-five healthy Japanese women aged 22-44 participated in this study. Skin biopsies were taken from facial skin using polyethylene sheet with cyanoacrylate. SC lipids were extracted and separated into individual lipid classes. The combined ceramides and cholesterol were quantified by HPTLC. Free fatty acid was quantified according to the ACS-ACOD method. Instrumental measurements were made at the site around the biopsy. In addition, the Stinging Test and a sensory evaluation questionnaire were administered to all subjects. The generally recognized O-D skin type classification is dependent on the consumer`s subjective assessment with respect to their skin troubles. The product of hydration state and skin surface lipid level showed a significant correlation with self-estimated skin types. The oily-dry skin type classified according to physiological parameters as well as SC lipid levels should be characterized as delicate skin with its barrier function deteriorating in the winter. Two groups of sensitive skin were established. One is the barrier function of the skin is deteriorated, and another is the sebaceous gland activity is in decline.
Objectives This study was conducted to confirm the inhibitory effect of β-glucan on epithelial inflammation induced by atopic dermatitis through Endocannabinoid system (ECS) activity. Methods Six-week-old NC/Nga mice were divided into a control group (Ctrl), atopic dermatitis elicitation group (ADE), and a β-glucan-treated group (β-glucan treatment after atopy dermatitis elicitation, β-GT). After 3 weeks, CB1, CB2, and GPR55 were observed to confirm the regulation of ECS activity, and filaggrin in the stratum corneum and Kallikrein-related peptidase (KLK) 7 in the stratum corneum and protease activated receptor (PAR)-2 were observed to confirm the inhibition of the inflammation, Phosphorylated extracellular signal-related kinase (p-ERK), Phosphorylated mammalian target of rapamycin (p-mTOR), and E-Cadherin were observed to confirm microenvironmental regulation. Results β-GT was significantly increased in CB1, CB2, and GPR55 positive reactions compared to that of the ADE. In positive reaction of the filaggrin in the stratum corneum, β-GT was significantly increased than that of the ADE. For KLK7 positive and PAR2 positive, β-GT was significantly reduced compared to the ADE. The p-ERK-positive and p-mTOR-positive reactions were significantly reduced in β-GT than in ADE. E-cadherin positive reaction was significantly increased in β-GT than in ADE (All p < 0.01). Conclusions It was confirmed that β-glucan has the effect of inhibiting the epithelium induced by atopic dermatitis through the ECS activity.

Glucosylceramides (GluCer) are known to play an important role in both water retention and epidermal permeability barrier function in the mammalian stratum corneum. In this study, we investigated the effects of pineapple fruit extract containing glucosylceramides (PFEG) on the maintenance and recovery of skin barrier function using atopic dermatitis-induced animal models. Five-week-old male Hos:HR-1 mice were divided into four groups fed on standard diet, unsaturated fatty acids-deficient (HR-AD) diet, and HR-AD diet supplemented with 0.01% or 0.1% pineapple-GluCer. Skin barrier function was evaluated by transepidermal water loss (TEWL), dermal moisture content, moisture content of the stratum corneum and wrinkle formation. The control group (HR-AD administration group) showed increased transepidermal water loss (TEWL), while the epidermal moisture content and the moisture content of the stratum corneum slowly decreased. However, in the PFEG groups (with 0.1% or 0.01% glucosylceramide), the TEWL levels were significantly reduced at 2 weeks. The PFEG also helped maintain skin moisturizing function by significantly suppressing the decrease of the epidermal moisture content and the moisture content of the stratum corneum. These results show that the PFEG is effective for maintaining and improving the function of the skin barrier. Therefore, this study suggests that PFEG is a potential candidate material for skin functional foods.

We are made up to the variation of the structure modeling of the stratum corneum by the skin on the hydration. The given skin structure is consist of the control level of the intercellular route and transcellular route, and is confirmed to the variation of the modeling for algorithm. Hydration condition is consisted to modeling by the size, form and combination, that skin impedance is appeared to result the value of measurement by the several layer such as forms of ${\xi}-R-SC-RH$, ${\xi}-R-SL-RH$, ${\xi}-R-SG-RH$, ${\xi}-R-SS-RH$ and ${\xi}-R-SB-RH$. Hydration condition is showed to the alteration difference value of S-Corneum, S-Lucidum, S-Granulosum, S-Spinosum and S-Basale. And, composition condition was constructed with the alteration modeling that was established to separate the division parts for conversion system. We will be possible to progress the improvement effectiveness of the skin and to consist of the continuous hydration control system for functional on the skin. This study aims at compensation of the skin moisture level using skin impedance and SR factor. SR factor is related with the current diffusion into the skin layer. To efficiently analyze the current diffusion on the skin model, we used an electromagnetic simulation program called Ansys 10.0 Emag. We confirmed that the measured value decreases as the electric current gets more diffused to the layer below the horny layer. In order to conduct actual experiments based on the simulated result, we manufactured special electrodes with 24 pins by arranging 0.8mm-diameter electrodes every 0.5mm, in a $3{\times}8$ array. By simultaneously achieving both impedance value and SR value of skin with the manufactured electrodes, we compared the skin moisture level using the existing equipment to the skin moisture level applied using the skin impedance as well as the SR factor developed in this study. The correlation coefficient between the skin moisture level achieved from the existing equipment and the reference value was 0.615 (p<0.01), whereas the correlation coefficient between the skin moisture achieved from the regression equation in this study was 0.677 (p<0.01). Accordingly, it was confirmed that applying SR factor additionally improves the moisture level more precisely.
Transdermal drug delivery offers various advantages over conventional drug delivery systems, such as avoidance gastrointestinal degradation and hepatic first-pass effect. encourages patient compliance. and possible sustained release of drugs. However, transdermal transport of drugs is low permeability of the stratum corneum, the superficial layer of the skin. Many physicochemical and biological factors influencing transdermal transport is described together with the corresponding experimental and clinical results. Phonophoresis is medical treatment with drugs introduced into the skin by ultrasound energy. Enhanced drug penetration is through to result from the biophysical alterations of skin structure by ultrasound waves. The frequency used for phonophoresis is usually from 20 kHz to 15MHz. Phonophoresis can be categorized in to three ranges: low-frequency range(below 1 MHz). therapeutic frequency range(1 to 3MHz), and high-frequency range(above 3 MHz). The depth of penetration of ultrasound into skin is inversely proportional to the frequency. Cavitation may cause mechanical stress. temperature elevation, or enhanced chemical reactivity causing drug transport. One theory is that ultrasound affects the permeation of the stratum corneum lipid structure as the limiting step in permeating through the skin. The range of indications for phonophoresis is wide. Aspecific classification of the range of indications is obtained by classification of pathological conditions. The continuous research is needed for many interesting issucs of phonophoretic transdermal delivory in new future.
