This study aimed to isolate and identify L-(+)-lactic acid-producing bacteria from tree barks collected in Thailand and evaluate the potential strain as probiotics. Twelve strains were isolated and characterized phenotypically and genotypically. The strains exhibited a rod-shaped morphology, high-temperature tolerance, and the ability to ferment different sugars into lactic acid. Based on 16S rRNA gene analysis, all strains were identified as belonging to Weizmannia coagulans. Among the isolated strains, BKMTCR2-2 demonstrated exceptional lactic acid production, with 96.41% optical purity, 2.33 g/l of lactic acid production, 1.44 g/g of lactic acid yield (per gram of glucose consumption), and 0.0049 g/l/h of lactic acid productivity. This strain also displayed a wide range of pH tolerance, suggesting suitability for the human gastrointestinal tract and potential probiotic applications. The whole-genome sequence of BKMTCR2-2 was assembled using a hybridization approach that combined long and short reads. The genomic analysis confirmed its identification as W. coagulans and safety assessments revealed its non-pathogenic attribute compared to type strains and commercial probiotic strains. Furthermore, this strain exhibited resilience to acidic and bile conditions, along with the presence of potential probiotic-related genes and metabolic capabilities. These findings suggest that BKMTCR2-2 holds promise as a safe and effective probiotic strain with significant lactic acid production capabilities.

The present paper will give some results of the fatigue behavior of typical axi-symmetric forward extrusion die. The extrusion process is analyzed by rigid-plastic FEM and the deformation analysis of extrusion die is conducted by elasto-plastic FEM. To approach the crack problem LEFM (Linear Elastic Fracture Mechanics) is introduced. Using special element in order to conside the sigularity of stress/ strain in the vicinity of the crack tip, stress intensity factor and the effective stress intensity factor is calculated. Applying proper fatigue crack propagation criterion such as Paris/Erdogan fatigue law and maximum principal criterion to these data, then, the angle and the direction of fatigue crack propagation is simulated. In result, it is proved that the simulated fatigue crack propagates in the zigzag path along the radial direction and fatigue life of the extrusion die is evaluated by using the computed crack growth rate.
Low cycle fatigue characteristics are very important in the development of automobile suspension parts. Fatigue properties using the strain life approach are usually obtained from low cycle fatigue tests. However, low cycle fatigue testing requires a lot of time and cost. In the current study, an attempt to estimate low cycle fatigue properties of high strength steel sheet from tensile test and tensile simulations is performed. In addition, low cycle fatigue testing was conducted to compare the fatigue properties obtained from tensile testing and simulations. In conclusion, the results effectively predict the low cycle fatigue properties. However, some deviations still exist.

A prototype probabilistic approach to thickness design for asphalt pavement surface course was developed using first-order second moment probability model. The tensile strain (load effect) developing at the bottom of surface layer due to the wheel load and the critical strain (resistance) of asphalt concrete were used as random variables for pavement reliability analysis. Based on the parameters for load effect and resistance data collected from reference and field, simulated data were generated by Monte Carlo method for reliability evaluation of the pavement for a typical rural highway. Thickness of pavement surface course was defined in terms of target reliability of the pavement, growth factor of traffic, design life of pavement and resistance of the asphalt concrete to be placed on the pavement. According to these rationales, prototype thickness design chrats were sugested through example studies. From these, similar design charts can be developed for many pavements if appropriate data and target reliability are determined.
This research was designed to illuminate that work domains and family domains are not separated but interacting spheres and to suggest efficient coping strategies than alleviate work/family role conflicts. The subjects of this study were 395 employed professional wives living in Seoul. Korea. Survey methods were questionnaires. Data analysis strategies were percentile, frequency, one-way-anova and multiple classification analysis. The results were as follows; 1)Employed wives experienced time-based conflicts and strain-based role conflicts. This finding supports the scarcity approach that multiple roles inevitably create strain. 2) Individual, family and work-related variables were predictors of work/family role conflicts. This findings indicate that work domains and family domains cannot be regraded as separated spheres. 3) Employed wives tried to meet all the given roles demands not by reducing role responsibilities, but by increasing the efficiency of role performance. This finding reflects the characteristics of employed wives who try to perform successfully all the responsibilities. 4)The negative correlation of coping strategies and work/family role conflicts suggests that strategies can function effectively in alleviating the work/family role conflicts.
A large residual fraction of aliphatic components of diesel prevails in soil, which has adverse effects on the environment. This study identified the most bio-recalcitrant aliphatic residual fraction of diesel through total petroleum-hydrocarbon fractional analysis. For this, the strain Acinetobacter sp. K-6 was isolated, identified, and characterized and investigated its ability to degrade diesel and n-alkanes (C18, C20, and C22). The removal efficiency was analysed after treatment with bacteria and nutrients in various soil microcosms. The fractional analysis of diesel degradation after treatment with the bacterial strains identified C18-C22 hydrocarbons as the most bio-recalcitrant aliphatic fraction of diesel oil. Acinetobacter sp. K-6 degraded 59.2% of diesel oil and 56.4% of C18-C22 hydrocarbons in the contaminated soil. The degradation efficiency was further improved using a combinatorial approach of biostimulation and bioaugmentation, which resulted in 76.7% and 73.7% higher degradation of diesel oil and C18-C22 hydrocarbons, respectively. The findings of this study suggest that the removal of mid-length, non-volatile hydrocarbons is affected by the population of bio-degraders and the nutrients used in the process of remediation. A combinatorial approach, including biostimulation and bioaugmentation, could be used to effectively remove large quantities of aliphatic hydrocarbons persisting for a longer period in the soil.

Creating an artificial strain with a minimal gene set for a specific purpose is every biologist's dream. With the complete genome sequencing of more than 50 microorganisms and extensive functional analyses of their genes, it is possible to design a genetic blueprint for a simple custom-designed microbe with the minimal gene set. Two different approaches are being considered. The first 'top-down' approach is trimming the genome to a minimal gene set by selectively removing genes of an organism thought to be unnecessary based on microbial genomics. The second 'bottom-up' approach is to synthesize the proposed minimal genome from basic chemical building blocks. The 'top-down' approach starting with the genome of a well known microorganism is more technically feasible, whereas the bottom-up approach may not be attainable in the nearest future because of the lack of the complete functional analysis of the genes needed for a life. Here in this study, we used the top-down approach to minimize the E. coli genome to create an artificial organism with 'core' elements for self-sustaining and self-replicating cells by eliminating unnecessary genes. Using several different kinds of sophisticated deletion techniques combined with a p:1age and transposons, we deleted about 19% of the E. coli genome without causing any damages to cellular growth. This smaller E. coli genome will be further reduced to a genome with a minimal gene l;et essential for cell life. This minimized E. coli genome can lead to the construction of many custom-designed strains with myriad practical and commercial applications.

Multiple surface crack distributed randomly along a weld toe influences strongly on the fatigue crack propagation life of welded joint. It is investigated by using statistical approaches based on series of systematic experiments. From the statistical results, initial crack numbers and its locations follow the normal distribution, and the probability of initial crack depths and lengths can be described well by tile Weibull distribution. These characteristics are used to calculate the fatigue crack propagation life, in which the mechanisms of mutual interaction and coalescence of the multiple cracks are considered as well as the Mk-factors obtained from a parametric study on the crack depths and lengths. The automatic calculation is achieved by the NESUSS, where the parameters such as the number, location and size of the cracks are all treated as random variables. The random variables are dealt through the Monte-Carlo simulation with sampling random numbers of 2,000. The simulation results show that the multiple cracks lead to much shorter crack propagation life compared with those in single crack situation. The sum of the simulation and tile fatigue crack initiation life derived by the notch strain approach agrees well with the experiments.
In this paper, rotating bending fatigue tests have been carried out to investigate the growth behabiors of surface fatigue crack initiated from a small artificial surface defect, that might exist in real structures, on 2024-T3 and 6:4 brass. The test results are analysed in the viewpoints of both strength of materials and fracture mechanics, it can be concluded as follows. The effect of a small artificial surface defect upon the fatigue strength is very large. The sensitivity of 2024-T3 on the defect is higher than that of 6:4 brass. The growth behavior of the surface fatigue crack of 2024-T3 is different from that of 6:4 brass. The growth rate of the surface fatigue crack of 2024-T3 is considerably rapid in the early stage of the fatigue life and apt to decrease in the later stage. It was impossible to establish a unifying approach in the analysis of crack growth begabior of 2024-T3 and 6:4 brass using the maximum stress intensity factor because of their dependence on stress level. But if the elastic strain and cyclic total strain intensity factor range were applied to obtain the growth rate of surface fatigue cracks of the materials, the data were found to be nearly coincided.

Rolling stock structures such as bogie frame and car body play an important role for the support of vehicle leading. In general, more than 25 years' durability is needed for them. A lot of study has been carried out for the prediction of the fatigue life of the bogie frame and car body in experimental and theoretical domains. One of the new methods is a probabilistic fatigue lift evaluation. The objective of this paper is to estimate the fatigue lift of the bogie frame of an electric car, which was developed by the Korea Railroad Research Institute (KRRI). We used two approaches. In the first approach probabilistic distribution of S-N curve and limit state function of the equivalent stress of the measured stress spectra are used. In the second approach, limit state function is also used. And load spectra measured by strain gauges are approximated by the two-parameter Weibull distribution. Other probabilistic variables are represented by log-normal and normal distributions. Finally, reliability index and structural integrity of the bogie frame are estimated.
