Impact Evaluation of Rubber Type, Hardness and Induced Prestress Force on the Dynamic Properties of a Damper (감쇠장치의 동적특성에 대한 고무의 종류, 경도 및 프리스트레스력의 영향 평가)

  • The objective of this study is to evaluate the dynamic properties of DUS (damping-up system) composed of the materials with excellent damping capacity, and to compare with those of the conventional hangar bolt. The main parameters are the type and hardness (𝜂H), of rubber and the prestress force (value converted from the compression strain (𝜂R) in the stress-strain relationship of rubber). The dynamic properties were examined from the natural frequency (𝜔n), maximum response acceleration (Am), amplification coefficient (𝛼p), maximum relative displacement (𝚫m), and damping ratio (𝜉D). The test results showed that the Am, 𝛼p, and 𝚫m values of DUS were 46.3%, 46.6% and 62.9% lower, respectively, and the 𝜉D value was 3.89 times higher, when compared to those of the conventional hangar bolt. In particular, the 𝛼p value was 1.3 for DUS, and 2.45 for the conventional hanger bolt, which were similar to those of rigid and flexible components specified in KDS 41 17 00, respectively. Consequently, in the optimal details of DUS, the 𝜂H values of 50 and 45 were required for the NR (natural rubber) and EPDM (ethylene propylene diene monomer), and the 𝜂R value of 5% was also recommended.

Characterization of Protocatechuate 4,5-Dioxygenase Induced from p-Hydroxybenzoate -Cultured Pseudomonas sp. K82

  • Pseudomonas sp. K82 has been reported to be an aniline-assimilating soil bacterium. However, this strain can use not only aniline as a sole carbon and energy source, but can also utilize benzoate, p-hydroxybenzoate, and aniline analogues. The strain accomplishes this metabolic diversity by using dif-ferent aerobic pathways. Pseudomonas sp. K82, when cultured in p-hydroxybenzoate, showed extradiol cleavage activity of protocatechuate. In accordance with those findings, our study attempted the puri-fication of protocatechuate 4,5-dioxygenase (PCD 4,5). However the purified PCD 4,5 was found to be very unstable during purification. After Q-sepharose chromatography was performed, the crude enzyme activity was augmented by a factor of approximately 4.7. From the Q-sepharose fraction which exhibited PCD 4,5 activity, two subunits of PCD4,5 (${\alpha}$ subunit and ${\beta}$ subunit) were identified using the N-terminal amino acid sequences of 15 amino acid residues. These subunits were found to have more than 90% sequence homology with PmdA and PmdB of Comamonas testosteroni. The molecular weight of the native enzyme was estimated to be approximately 54 kDa, suggesting that PCD4,5 exists as a het-erodimer (${\alpha}$$_1$${\beta}$$_1$). PCD 4,5 exhibits stringent substrate specificity for protocatechuate and its optimal activity occurs at pH 9 and 15 $^{\circ}C$. PCR amplification of these two subunits of PCD4,5 revealed that the ${\alpha}$ subunit and ${\beta}$ subunit occurred in tandem. Our results suggest that Pseudomonas sp. K82 induced PCD 4,5 for the purpose of p-hydroxybenzoate degradation.

Genetic Diversity, Pathogenicity, and Fungicide Response of Fusarium oxysporum f. sp. fragariae Isolated from Strawberry Plants in Korea (국내 딸기 시들음병균 Fusarium oxysporum f. sp. fragariae의 유전적 다양성, 병원성과 살균제 반응)

  • Fusarium wilt caused by Fusarium oxysporum f. sp. fragariae (Fof) is the most important diseases of a strawberry field in Korea. We surveyed phylogenetic analysis, pathogenicity test, and fungicide response about Fof isolates isolated from Korea. Twenty-seven isolates of F. oxysporum isolated from strawberry plants were conducted in this study. Specific amplification by Fof specific primer was confirmed in all 26 isolates except Fo080701 isolate. The nuclear ribosomal intergenic spacer region and the translation elongation factor EF-lα gene sequences of isolates revealed three main lineages. Most of all isolates were contained DNA lineage group 1, but 2 and 3 group was shown only one and three isolates, respectively. All isolates were shown in pathogenicity with cv. Seolhyang. The EC50 mean values of prochloraz ranged 0.02-0.1 ㎍/ml except for Fo080701 and effectively inhibited mycelial growth at low concentrations. The EC50 value of metconazole was also 0.04-0.22 ㎍/ml, showing a similar inhibitory effect to that of prochloraz. The EC50 value of pyraclostrobin was 0.23-168.01 ㎍/ml, which was different according to the strain. In the field trial, boscalid+fludioxonil, fluxapyroxad+pyraclostrobin, and prochloraz manganese were selected as the effective fungicides for controlling Fusarium wilt.

Isolation of Aeromonas sobria Containing Hemolysin Gene from Arowana (Scleropages formosus) (Arowana(Scleropages formosus)에서 Hemolysin Gene을 지닌 Aeromonas sobria 분리 및 특성)

  • Arowana (Scleropages formosus) is the most valuable group of ornamental fishes and very much in demand in the ornamental fish trade and commands high price ranging from hundreds to thousands of dollars per fish. In this paper, we described a case of mortality of arowana from a private aquarium in Korea. A bacterial pathogen from fish organs (brain, kidney, liver) was cultured, identified and confirmed using Vitek System 2, API 20E test, multiplex PCR and 16S rRNA gene sequencing. The morphological and biochemical properties of the bacterium isolated from the brain, kidney and liver of the fish were similar to Aeromonas sobria. Positive amplification products using the multiplex PCR assay for detection of A. sobria were obtained from these organs. The 16S rRNA gene of the isolates from fish was identical and exhibited 100% sequence similarity with A. sobria (AY987762.1) strain available from GenBank. This bacterium contained hemolysin gene, a virulence factor that plays an important role in outbreaks of disease and is pathogenic to humans as well as in fish. Although this opportunistic bacterium was isolated from a fish without any external symptoms, this pathogen may act as a reservoir and enhance chances of zoonosis to human such as during handling.
