Antibacterial Activity of Lactobacillus casei Isolated from a Fermented Milk Against Pathogenic Enteric Bacteria (발효유(醱酵乳)에서 분리된 Lactobacillus casei의 병원성 장내세균에 대한 항균작용)

  • The present study was undertaken to elucidate the antibacterial spectrum of L. casei phage type $J_1$ strain isolated from a fermented milk product against pathogenic enteric bacteria. Growth inhibitory effects and minimum inhibitory concentration(MIC) of culture supernatants of L. casei grown in MRS broth were measured by both plate culture method and microplate broth dilution technique against Salmonella typhi, Salmonella typhimurium, Shigella flexneri, Shigella dysenteriae, enterpathogenic E. coli, Klebsiella pneumoniae and Pseudomonas aeruginosa. The results are summarized as follows: 1. The MRS broth culture of L. casei gave a similar extent of growth inhibitory effects against S. typhi, S. typhimurium, S. flexneri, S. dysenteriae, E. coli, K. pneumoniae and P. aeruginosa, respectively. 2. The inhibitory effects of L. casei culture were observed either in whole broth culture or in culture supernatant, but neither the bacterial suspension nor the neutralized culture supernatant showed such as antibacterial activities. 3. The MIC titres of the culture supernatants were ${\log_2}5$ to ${\log_2}6$, whereas those of the neutralized culture supernatant dropped markdely to ${\log_2}2$ to ${\log_2}3$. These results indicated that major portion of growth inhibitory effects of MRS broth culture of L. casei against enteric bacterial pathogens was possibly due to the acids produced, and minor portion to other antibacterial substances.

  • In order to increase the production and secretion rate of mouse salivary $\alpha$-amylase from Saccharomyces cerevisiae, various experiments were attempted. A plasmid pCNNinv (AMY) was constructed by the substitution of ADCl promoter and native signal sequence of mouse salivary $\alpha$-amylase cDNA gene with PRBI promoter and yeast invertase leader sequence, which resulted in 25% increase in the production of $\alpha$-amylase in the culture medium. The respiratory deficient transformant carrying pCNNinv (AMY) were obtained by treating yeast cells with ethidium bromide, and the $\alpha$-amylase activities in the culture brothes of the respiratory-deficient transformants were 5-8 times higher than that of parental wild type strain. $\alpha$-Amylase activity was also increased 3 times when the 0.015% (w/v) of 2-mercaptoethanol was added to the culture medium.

Study on Effect of Takrisodokyeum Extract on Antitumoral Activity and Immune Response (탁리소독음(托裏消毒飮)의 항종양(抗腫瘍) 효과(效果) 및 면역조절반응(免疫調節反應)에 관(關)한 연구(硏究))

  • Tish study was carried out to evaluate the possible therapeutic or antitumoral effects of Takrisodokyeum extract against tumor, and immunomodulatory effect. Some kinds of tumor were induced by the typical application of 3-methylcholanthrene (MCA) or by the implantation(s.c) of malignant tumor cells such as leukemia cells(3LL cells) or sarcoma cells(S-I80 and Fas II cells). Treatment of the Takrisodokyeum water-extract(daily 1mg mouse, i.p.) was continued for 7 days prior to tumor induction and after that the treatment was lasted for 15 days. Against squamous cell carcinoma induced by MCA, Takrisodokyeum decreased not only the frequency of tumor production but also the number and the weight of tumors per tumor bearing mice (TBM). Takrisodokyeum also significantly suppressed the development of 3LLcell and S-180 cell by frequency and their size, and some developed tumors were regressed by the continuous treatment of Takrisodokyeum extract into TBM. However, when tumor was induced by FsaII cell-implantation, the growth of implanted cells in mice was delayed by the water extract of Takrisodokyeum until day 7 and then rapid growth ensued. In vitro, treatment of Takrisodokyeum extract had no effect on the growth of some kind of cell lines such as FsaII, A-131 strain but significantly inhibited the proliferation of 3LL, S-180 cells. Takrisodokyeum also stimulated the migrative ability of leucocyte, the MIF and IL 2-production of T lymphocytes, but not IL 6 production of B cells. Takrisodokyeum enhanced Arthus reaction and DTH to sheep erythrocytes, and NK cells activities. These results demonstrated that Takrisodokyeum extract different results according to the type of tumor cells. And these results also suggested that antitumor effect of Takrisodokyeum might be chiefly due to nonspecific enhancement of NK cell activities and cell-mediated immune responses.

Rheology of Concentrated Xanthan Gum Solutions : Steady Shear Flow Behavior

  • Using a strain-controlled rheometer, the steady shear flow properties of aqueous xanthan gum solutions of different concentrations were measured over a wide range of shear rates. In this article, both the shear rate and concentration dependencies of steady shear flow behavior are reported from the experimentally obtained data. The viscous behavior is quantitatively discussed using a well-known power law type flow equation with a special emphasis on its importance in industrial processing and actual usage. In addition, several inelastic-viscoplastic flow models including a yield stress parameter are employed to make a quantitative evaluation of the steady shear flow behavior, and then the applicability of these models is also examined in detail. Finally, the elastic nature is explained with a brief comment on its practical significance. Main results obtained from this study can be summarized as follows: (1) Concentrated xanthan gum solutions exhibit a finite magnitude of yield stress. This may come from the fact that a large number of hydrogen bonds in the helix structure result in a stable configuration that can show a resistance to flow. (2) Concentrated xanthan gum solutions show a marked non-Newtonian shear-thinning behavior which is well described by a power law flow equation and may be interpreted in terms of the conformational status of the polymer molecules under the influence of shear flow. This rheological feature enhances sensory qualities in food, pharmaceutical, and cosmetic products and guarantees a high degree of mix ability, pumpability, and pourability during their processing and/or actual use. (3) The Herschel-Bulkley, Mizrahi-Berk, and Heinz-Casson models are all applicable and have equivalent ability to describe the steady shear flow behavior of concentrated xanthan gum solutions, whereas both the Bingham and Casson models do not give a good applicability. (4) Concentrated xanthan gum solutions exhibit a quite important elastic flow behavior which acts as a significant factor for many industrial applications such as food, pharmaceutical, and cosmetic manufacturing processes.

Effects of Motor Learning Guided Laryngeal Motor Control Therapy for Muscle Misuse Dysphonia (운동학습이론에 기초한 발성운동조절법이 근오용성 발성장애의 음성에 미치는 효과)

  • Muscle misuse dysphonia (MMD) is defined as a behavioral voice disorder resulting from inappropriate contractions of intrinsic and/or extrinsic laryngeal muscles. The purpose of this study was to investigate the effect of motor learning guided laryngeal motor control therapy (MLG-LMCT) which is designed to improve an existing LMT and further the effective voice treatment on people with muscle misuse dysphonia. Forty-six people with MMD (M:F=16:30) participated in this study. The voice samples of the participants were recorded to investigate the effect of MLG-LMCT before and after the voice therapy. Voice samples were analyzed via electro-glotto-graph (EGG). Contact quotient (CQ), speed quotient (SQ), and waveform were reported. In addition, perceptual and acoustical evaluation were conducted to determine the change of voice improvement after treatment. The experimenter massaged the tensioned muscles around the neck. In order to find more proper phonation the experimenter showed the subjects their EGG wave forms as to whether or not they are moving the vocal folds to the appropriate position. Therefore, the EGG wave forms were used as a type of visual feedback. With the wave form, the experimenter helped subjects move the vocal folds and laryngeal muscles to find more proper voice production. The sensory stimuli from the experimenter gradually faded out. A paired dependent t- test revealed that there was significant differences in CQ between pre- and post-therapy. Perceptually, overall, rough, breathy, strain, and transition were significantly reduced. Acoustically, there were significant differences in Fo, jitter, shimmer, and NHR. After using MLG-LMCT, most of the subjects showed improvements in voice quality. The results from this study led us to the following conclusions: Motor learning guided laryngeal motor control therapy (MLG-LMCT) has reduces muscle misuse dysphonia. These results may occur because a visual feedback from EGG wave form can maintain the effect of the muscle tension reduction from laryngeal manual therapy. In case of people with MMD who reduced muscle tension from the therapy (LMT) but, not appropriately manipulating the location of larynx or adducting the vocal folds, MLG-LMCT might be an alternative therapy approach.

A Study on the LPG Explosion Characteristics of Non-uniform Concentration (불균일 농도 LPG의 폭발 특성에 관한 연구)

  • LPG explosion characteristics in non-uniform concentration was investigated with a 270 liter explosion vessel of which the scale is 100 cm${\times}$60 cm${\times}$45 cm. Vented explosion and closed explosion system were used. Experimental parameter were position of ignition source, nozzle diameter and flow rate of gas. Non uniform concentration was controlled by the nozzle diameter and flow rate. Explosion pressure were measured with strain type pressure sensor and the flame behavior was pictured with the video camera. Based on this experimental result, it was found that the flow rate of gas and the duration of gas injection are important factor for mixing the gas in the vessel. And as the increase the non-uniformity of gas concentration, explosion pressure and pressure rise rate Is decrease but the flame resident time in the vessel is increase. Therefore gas explosion to fire transition possibility will increase in non-uniform concentration gas explosion.

Prevalence and Characteristics of Antimicrobial-Resistant Staphylococcus aureus and Methicillin-Resistant Staphylococcus aureus from Retail Meat in Korea

  • This study was to investigate the prevalence and characteristics of antimicrobial-resistant Staphylococcus aureus and methicillin-resistant S. aureus (MRSA) from 4,264 retail meat samples including beef, pork, and chicken in Korea between 2013 and 2018. A broth microdilution antimicrobial susceptibility testing was performed for S. aureus. Molecular typing by multilocus sequence typing (MLST), spa typing, and pulsed-field gel electrophoresis (PFGE), was performed on mecA-positive S. aureus strain. S. aureus was isolated at a rate of 18.2% (777/4,264), of which MRSA comprised 0.7% (29 strains). MLST analysis showed that 11 out of the 29 MRSA isolates were predominantly sequence type (ST) 398 (37.9%). In addition, ST72, ST692, ST188, ST9, and ST630 were identified in the MRSA isolates. The spa typing results were classified into 11 types and showed a high correlation with MLST. The antimicrobial resistance assays revealed that MRSA showed 100% resistance to cefoxitin and penicillin. In addition, resistance to tetracycline (62.1%), clindamycin (55.2%), and erythromycin (55.2%) was relatively high; 27 of the 29 MRSA isolates exhibited multidrug resistance. PFGE analysis of the 18 strains excluding the 11 ST398 strains exhibited a maximum of 100% homology and a minimum of 64.0% homology. Among these, three pairs of isolates showed 100% homology in PFGE; these results were consistent with the MLST and spa typing results. Identification of MRSA at the final consumption stage has potential risks, suggesting that continuous monitoring of retail meat products is required.

Heterologous Expression and Characterization of a Thermostable Exo-β-D-Glucosaminidase from Aspergillus oryzae

  • An exo-β-D-glucosaminidase (AorCsxA) from Aspergillus oryzae FL402 was heterologously expressed and purified. The deduced amino acid sequence indicated that AorCsxA belonged to glycoside hydrolase family 2. AorCsxA digested colloid chitosan into glucosamine but not into chitosan oligosaccharides, demonstrating exo-β-D-glucosaminidase (CsxA) activity. AorCsxA exhibited optimal activity at pH 5.5 and 50℃; however, the enzyme expressed in Pichia pastoris (PpAorCsxA) showed much stronger thermostability at 50℃ than that expressed in Escherichia coli (EcAorCsxA), which may be related to glycosylation. AorCsxA activity was inhibited by EDTA and most of the tested metal ions. A single amino acid mutation (F769W) in AorCsxA significantly enhanced the specific activity and hydrolysis velocity as revealed by comparison of Vmax and kcat values with those of the wild-type enzyme. The three-dimensional structure suggested the tightened pocket at the active site of F769W enabled efficient substrate binding. The AorCsxA gene was heterologously expressed in P. pastoris, and one transformant was found to produce 222 U/ml activity during the high-cell-density fermentation. This AorCsxA-overexpressing P. pastoris strain is feasible for large-scale production of AorCsxA.

Molecular Characterization of Plasmids Encoding CTX-M β-Lactamases and their Associated Addiction Systems Circulating Among Escherichia coli from Retail Chickens, Chicken Farms, and Slaughterhouses in Korea

  • Extended-spectrum β-lactamases (ESBLs), particularly those of the CTX-M types, are the predominant resistance determinants of Escherichia coli that are rapidly spreading worldwide. To determine CTX-M types, E. coli isolates were collected from retail chickens (n = 390) and environmental samples from chicken farms (n = 32) and slaughterhouses (n = 67) in Korea. Fifteen strains harboring blaCTX-M genes were isolated from 358 E. coli isolates. The most common CTX-M type was eight of CTX-M-15, followed by six of CTX-M-1 and one of CTX-M-14. The blaCTX-M genes were identified in the isolates from retail chickens (n = 9), followed by feces, water pipes, floors, and walls. Conjugations confirmed the transferability of the plasmids carrying blaCTX-M genes to the recipient E. coli J53 strain. Furthermore, eight addiction systems carried by the replicons in CTX-M types were confirmed. The dominant system was identified as ccdAB, vagCD, and pndAC in donor strains and transconjugants. The clonal relationship between the two strains carrying blaCTX-M genes indicates that E. coli may transmit from the farm to retail chickens, suggesting a possible public health risk. Our findings demonstrate that the detection of CTX-M types in E. coli isolates is important for tracking ESBL production in animals, and suggest linkage of multiple addiction systems in plasmids bearing blaCTX-M genes.

Isolation and Culture Conditions of Hydrogen Producing Bacterium Enterobacter sp. ES392 (수소생산균 Enterobacter sp. ES392의 분리 및 배양조건)

  • A hydrogen-producing bacterium (strain ES392) was isolated from pond water located in the Dong-Eui University, Busan, Korea. The cell was long-rod type ($1.4\;{\mu}m$) of about ($0.6\;{\mu}m$) in diameter, and not formed flagellum and spore. Phylogenetic analysis based on the 16S rRNA sequence and biochemical studies indicated that ES392 belonged to the genus Enterobacter sp. The optimum pH and temperature for hydrogen production was 7.5 and $35^{\circ}C$, respectively. The optimization of medium compositions which maximize hydrogen production from Enterobacter sp. ES392 was determined. As a result, the maximum hydrogen production was obtained under the conditions of 4% (w/v) sucrose, 0.5% (w/v) yeast extract and 50 mM potassium phosphate buffer (pH 7.5). Under batch culture conditions, the maximal hydrogen production and yield were obtained as 3481 mL/L and 1.33 mol/mol sucrose, respectively.
