Skin and soft tissue infections caused by methicillin-resistant Staphylococcus aureus (MRSA) can occur especially in community populations such as military training camps. We investigated antimicrobial resistance patterns and molecular epidemiological characteristics of MRSA isolated from nasal swabs in healthy army trainees. From January 2018 to March 2018, one MRSA strain was isolated from nasal swab and hand of healthy army trainees. mecA gene detection, SCCmec and mec complex typing were performed to analyze the antimicrobial resistance patterns and molecular epidemiological characteristics of MRSA isolates. As a result, SCCmec and mec complex type of MRSA isolate from military trainees was not-typeable (n=1). In conclusion, not-typeable subtype of MRSA isolate from military trainees need to be confirmed by continuous follow-up study to determine whether there is a different genotype or a new subtype of genotype present in the Republic of Korea.
Interfacial evaluation and hydrophobicity of Ni-nanopowder/epoxy composites were investigated for self-sensing and actuation. Contact resistance and resistivity were measured using gradient micro-specimens. The actuation of the composites in the electromagnetic field was studied with three wave functions, i.e., sine, triangle and square functions. Due tothe presence of hydrophobic domains on the heterogeneous surface, the static contact angle of Ni-nanopowder/epoxy nanocomposite wasabout $100^{\circ}$, which was rather lower than that for super-hydrophobicity. The dynamic contact angle showed the similar trend of static contact angle. Ni-nanopowder/epoxy composite was responded wellfor both self-sensing and actuation in electromagnetic field due to the intrinsic metal property of Ni-nanopowder. Displacement of the actuator of Ni-nanopowder/epoxy composite was evaluated to obtain the maximum and the optimum performance using laser displacement sensor as functions of the wave type, frequency, and voltage. Actuation of Ni-nanopowder/epoxy composites also increased as functions of applied frequency and voltage. Actuated strain increased more rapidly at sine wave with increasing voltage compared to those of triangle or rectangular waves.
The trichomonad species Tritrichomonas foetus and Pentatrichomonas hominis were recently detected in the feces of dogs with diarrhea. However, little information is available on the prevalence and pathogenicity of these parasites in the canine population. Therefore, the aim of this study was to determine the prevalence and molecular characterization of trichomonads infecting pet dogs in Anhui and Zhejiang provinces, east China. In total, 315 pet dogs, with or without diarrhea, from 7 pet hospitals were included in this epidemiological survey. Microscopy and PCR detected P. hominis in 19.7% (62/315) and 31.4% (99/315) of fecal samples, respectively. T. foetus infection was detected in 0% (0/315) of samples with microscopy and in 0.6% (2/315) with PCR. The prevalence of P. hominis was significantly higher in young dogs (${\leq}12months$) than in adult dogs (>12 months), and was significantly higher in diarrheic dogs (50.6%) than in non-diarrheic dogs (24.3%; P<0.05). Infection with T. foetus did not correlate with any risk factors evaluated in this study. A sequence analysis of the P. hominis PCR products showed minor allelic variations between our sequences and those of P. hominis strains from other hosts in different parts of the world. Type CC1 was the most common strain in dogs in east China. The internal transcribed spacer 1 (ITS1)-5.8S rRNA gene sequences from the 2 T. foetus isolates detected in this study displayed 100% identity and were homologous to the sequences of other strains isolated from domestic cats in other countries.
Plants expressing Agrobacterium sp. strain CP4 5-enolpyruvylshikimate-3-phosphate synthase (CP4 EPSPS) are known to be resistant to glyphosate, a potent herbicide that inhibits the activity of the endogenous plant EPSPS. In order to develop herbicide-resistant rice, we prepared transgenic rice plants with CP4 EPSPS gene under the control of CaMV 35S promoter for over-expression. A recombinant plasmid was transformed into rice via Agrobacterium-mediated transformation. A large number of transgenic rice plants were obtained with glyphosate and most of the transformants showed fertile. The integration and expression of CP4 EPSPS gene from regenerated plants was analyzed by Southern and northern blot analysis. The transgenic rice plants had CP4 EPSPS enzyme activity levels more than 15-fold higher than the wild-type plants. EPSPS enzyme activity of transgenic rice plants was also identified by strip-test method. Field trial of transgenic rice plants further confirmed that they can be selectively survived at 100% by spay of glyphosate (Roundup$^{(R)}$) at a regular dose used for conventional rice weed control.
Potato virus X (PVX-KO) showing mild mosaic and stunting symptoms on potato (Solanum tuberosum) in Kangwon area has been isolated and characterized. EM observation of the purified virus particles showed flexuous rod shape of about 520 nm in length. The coat protein (CP) of the virus had a molecular weight of 31 kDa in SDS-PAGE analysis, and the viral RNA was approximately 6.4 kb in size in denatured agarose gel electro-phoresis. In gel-immunodiffusion tests, it reacted strongly with an antiserum to common PVX from BIOREABAAG (USA). A rabbit antiserum was produced using purified virus and used for routine PVX detection by ELISA. Cultivated potatoes in Kangwon and other areas were frequently infected with PVX-KO. Both Datura stramonium and Nicotiana tabaccum cultivars developed necrotic local lesions 5 days after inoculation, and systemic mosaic symptoms with vein clearing 2 weeks after inoculation. All the features agree with the description of other PVX strains. To confirm and determine PVX strains, reverse transcription-polymerase chain reaction experiment was conducted using specific primers for viral CP. Amplified DNA fragments were cloned and sequenced. Results showed nucleotide sequence homologies of about 88 to 99% to other PVX strains. Based on CP amino acid sequence deduced from nucleotide sequences and host range studies PVX-KO is considered a member of the type X subgroup of PVX.
Alternanthera mosaic virus (AltMV) is a member of the genus Potexvirus which has been known for less than twenty years, and has been detected in Australasia, Europe, North and South America, and Asia. The natural host range to date includes species in at least twenty-four taxonomically diverse plant families, with species in at least four other families known to be infected experimentally. AltMV has been shown to differ from Potato virus X (PVX), the type member of the genus Potexvirus, in a number of ways, including the subcellular localization of the Triple Gene Block 3 (TGB3) protein and apparent absence of interactions between TGB3 and TGB2. Differences between AltMV variants have allowed identification of viral determinants of pathogenicity, and identification of residues involved in interactions with host proteins. Infectious clones of AltMV differing significantly in symptom severity and efficiency of RNA silencing suppression have been produced, suitable either for high level protein expression (with efficient RNA silencing suppression) or for Virus-Induced Gene Silencing (VIGS; with weaker RNA silencing suppression), demonstrating a range of utility not available with most other plant viral vectors. The difference in silencing suppression efficiency was shown to be due to a single amino acid residue substitution in TGB1, and to differences in subcellular localization of TGB1 to the nucleus and nucleolus. The current state of knowledge of AltMV biology, including host range, strain differentiation, host interactions, and utility as a plant viral vector for both protein expression and VIGS are summarized.

The characteristics of Brown gas was experimentally studied in view of efficiency and flame propagation. For this study, the Brown gas stack with 7 cells was manufactured following the Brown gas related patents and reports. All measuring equipments were re-tested and calibrated by Korea Laboratory Accreditation Scheme (KOLAS) certified laboratories. Since the amount of produced gas is most crucial in determining the efficiency, we adopted two gas collecting methods such as bottle trap method and wet gas meter method. The energy efficiency of our own fabricated stack was measured to be 75%, which is comparable to general alkaline water electrolysis efficiency. In order to analyze the flame propagation characteristics of Brown gas, we measured the flame propagation pressure, velocity, and shape by using strain type pressure sensor, optical sensor, and high speed camera in conjunction with Schliren system, respectively. From the experimental results, it was found that the flame propagation behavior of Brown gas was almost the same as that of hydrogen and oxygen mixture gas in 2:1 molar ratio. Moreover, from the high speed camera analysis, we concluded that Brown gas flame exhibits explosion behavior as does mixture gas ($H_{2}:O_{2}=2:1$).
This study is to clarify the comparative relationship and mechanical anisotropy of granite distributed in the Nam-weon on the subject of weathered rock mass sea water surroundings. Artificial weathering test is defined as a test, which controls the weathering rate and agents by controlling the weathering rate and agents by artificial environmental of salt water. Increased weathering degree is large indicated by weathering salt water, such as apparent specific gravity, absorption, porosity, uniaxial compression strength, P-wave velocity, slake durability, shore hardness, indirect tensile strength(brazilian test) and cohesion were measured. As the Weathering salt water proceeds, cracks develope increasingly. A number the cracks affect the rock deformation. Therefore, stress-strain curve of weathered salt water rock in one confined state are quite differ from weathered fresh water rock those. A reason of their deformation type is the formation of micro-cracks and potential porosity caused by artificial weathering test.
An isolate of Peanut stunt virus (PSV) isolated from black locust tree (Robinia pseudo-acacia L.) showing severe mosaic and malformation symptoms, was designated as PSV-Rp. PSV-Rp was characterized by the tests of host range, physical properties, RNA and coat protein composition and RT-PCR analysis. Nucleotide sequences of the cucumoviruses CP genes were also used for identification and differentiation of PSV-Rp. Six plant species were used in the host range test of PSV-Rp. PSV-Rp could be differentiated from each Cucumovirus strain used as a control by symptoms of the plants. The physical properties of PSV-Rp virus were TIP $65^{\circ}C$, DEP $10^{-3}$, and LIP $2{\sim}3$ days. In dsRNA analysis, PSV-Rp consisted of four dsRNAs, but satellite RNA was not detected. Analysis of the coat proteins by SDS-PAGE showed one major protein band of about 31 kDa. RT-PCR using a part of Cucumovirus RNA3 specific primer amplified ${\sim}950bp$ DNA fragments from the crude sap of virus-infected black locust leaves. RFLP analysis of the RT-PCR product could differential PSV-RP from CMV The nucleotide sequence identity between the PSV-Rp CP and the TAV-P CP genes and the PS-V-RP CP and CMV-Y CP genes were 61.6% and 40.5%, respectively. On the other hand, the nucleotide sequence identity of the PSV-Rp CP gene was $70.9%{\sim}73.4%$ in comparison with those of PSV subgroup I (PSV-ER and PSV-J) and 67.3% with that of PSV subgroup II(PSV-W). Especially, the nucleotide sequence identity of PSV-Rp CP gene and that of PSV-Mi that was proposed recently as the type member of a novel PSV subgroup III was 92.4%.
Recently, prestressed H-Beam bridges with external unbonded Tendons are increasingly built. The mechanical behavior of prestressed steel H-beams is different from that of normal bonded PSC beams in a point of the slip of tendons at deviators and the change of tendon eccentricity that occurs, when service load are applied in external unbonded steel H-beams. The concept of prestressing steel structures has been widely considered, in spite of long and successful history of prestressing concrete members. In the study, The flexural test on prestressed steel H-beams has been performed in the various aspects of prestressed H-beam including the tendon type and profile. The load was plotted against the deflection and the strain respectively in the steel beam and prestressing bars. The value expected with the equation of internal force equilibrium and compatibility between the deflection of the bars and the H-beam was found to correlate well with the measured data.
