TRANSIENT SIMULATION OF SOLID PARTICLE DISTRIBUTION WITH VARIOUS DESIGN PARAMETERS OF THE BAFFLE IN A STIRRED TANK (배플 형상에 따른 교반기 내부 고체입자 분포의 비정상상태 해석)

  • In the present study, numerical simulations were performed in a stirred solid/liquid system by using Eulerian multi-phase model. The transient flow field of liquid and distribution of solid particles were predicted in the stirred tank with pitched paddle impeller and baffles. The Frozen rotor method is adopted to consider the rotating motion of the impeller. The effects of number and width of baffles on the mixing time and the quality of solid suspension in the stirred tank are presented numerically. The result shows that the mixing time decreases as the width and number of baffles increase. The present numerical methodology can be applied to optimizing mixing condition of industrial mixer.

  • Stirred tank is widely used in various industries for mixing operations and chemical reactions for single- or multi-phase fluid systems. For designing agitator of high performance, quantity data of internal flow characteristics influenced by mixing performance are definitely confirmed but quantity analysis about the transient flow characteristics of complicate structure is recognized as difficult problem in the present. In this study, two models of commercial CFD code Fluent 6.3 used to propose suitable for the tank analysis. Agitation of Stirred tank is analyzed using a mixed model and the flow in the stirred tank is analyzed using a standard k-${\varepsilon}$ model. Multiple reference frame(MRF) and Sliding mesh(SM), the analysis techniques were used For compare a result of CFD with a visualization experiment result, to grasp internal flow and mixing characteristic in stirred tank and to present fundamental analysis method.

Simultaneous analysis of concentration and flow fields in a stirred tank using large eddy simulation (대형 와 모사를 사용한 혼합 탱크 내의 농도장과 유동장의 동시 해석)

  • Transport of a scalar quantity, such as chemical concentration or temperature, is important in many engineering applications and environmental flows. Here we report on results obtained from the large eddy simulations of flow and concentration fields inside the tank performed using a spectral multi-domain technique. The computations were driven by specifying the impeller-induced flow at the blade tip radius (Yoon et al. $^{(1)}$). This study focused on the concentration development at different molecular diffusivities in a stirred tank operated under turbulent conditions. The main objective of the work presented here is to study the large-scale mixing structure at different molecular diffusivities in a stirred tank by using the large eddy simulation. The time sequence of concentration and flow fields shows the flow dependency of the concentration development. The presence of spatial inhomogenieties is detailed by observing the time variation of local concentration at different positions.

  • Stirred tanks are widely used in various industries for mixing operations and chemical reactions for single- or multi-phase fluid systems. In this study, a numerical study was conducted to predict the mixing characteristics in a simple stirred tank. The flow in the model stirred tank was calculated utilizing the multiple reference frame (MRF) and the sliding mesh (SM) capabilities of a commercial CFD code (Fluent 6.2). The results of the flow simulation were analyzed in terms of the mixing efficiency, and the applicability of MRF and SM methods was also discussed.

Analysis of Fluid Flows in a Stirred Tank Using Computational Fluid Dynamics (전산유체역학을 이용한 교반탱크 내 유체흐름 해석)

  • The flow patterns in a stirred tank, 1m in diameter and 1 m in height, were studied using CFX, a commercial computational fluid dynamics program, with the impeller rotation speed, the impeller blade angle and the tank-bottom shape varied and the baffles included or excluded. A vortex was observed in the center of the tank in the absence of the baffles, and the intensity of the vortex increased with increasing the rotation speed. The vortex was considerably reduced in the presence of the baffles. An increase in the blade angle increased the vertical flow and decreased the vortex intensity. The flow in the corners of the tank bottom turned smoother as the tank bottom was varied in shape from flat to round.

Bioreactor Cultures of Lithospermum erythrorhizon for Shikonin Production with In Situ Extraction (동시 추출을 겸한 생물반응기에서 Lithospermum erythrorhizon 배양에 의한 shikonin 생산)

  • Plant cell cultures of Lithospermum erythrorhizon were performed in stirred tank and packed-bed reactors with in situ extraction by n-hexadecane. The specific shikonin production and volumetric shikonin productivity of stirred tank reactor reached 1.5 mg shikoninlg cell and 400$\mu g$ shikonin/(L.day), respectively. In packed-bed reactor with calcium alginate-immobilized cells specific shikonin production and volumetric productivity reached 2.0 mg shikoninlg cell and 2857$\mu g$ shikonin/(L.day), which were 1.3 and 7.1 times higher than those of stirred tank reactor, respectively. The higher shikonin production and productivity of packed-bed reactor seemed to be due to high cell loading capacity of calcium alginate immobilized cells in packed-bed reactor and improved cell-cell contact.

Optimization of $\beta$-Galactosidase Production in Stirred Tank Bioreactor Using Kluyveromyces lactis NRRL Y-8279

  • This paper investigates the production and optimization of $\beta$-galactosidase enzyme using synthetic medium by Kluyveromyces lactis NRRL Y-8279 in stirred tank bioreactor. Response surface methodology was used to investigate the effects of fermentation parameters on $\beta$-galactosidase enzyme production. Maximum specific enzyme activity of 4,622.7 U/g was obtained at the optimum levels of process variables (aeration rate 2.21 vvm, agitation speed 173.4 rpm, initial sugar concentration 33.8 g/L, incubation time 24.0 hr). The optimum temperature and pH of the $\beta$-galactosidase enzyme produced under optimized conditions were $37^{\circ}C$ and pH 7.0, respectively. The enzyme was stable over a pH range of 6.0-7.5 and a temperature range of $25-37^{\circ}C$. The $K_m$ and $V_{max}$ values for O-nitrophenol-$\beta$-D-galactopyranoside (ONPG) were 1.20 mM and $1,000\;{\mu}mol/min{\cdot}mg$ protein, respectively. The response surface methodology was found to be useful in optimizing and determining the interactions among process variables in $\beta$-galactosidase enzyme production. Hence, this study fulfills the lack of using mathematical and statistical techniques in optimizing the $\beta$-galactosidase enzyme production in stirred tank bioreactor.
