Effect of ultrasonification on immuno-stimulatory activities of the extracts from Korean medicinal herbs

  • 물 추출과 초음파를 병행한 모든 조건 중 $60^{\circ}C$에서 초음파 병행 추출물이 보다 높은 추출 수율을 보였고 그 중 복분자가 가장 높은 추출 수율을 보였다. 또한 총 5일간 인간 면역세포인 T-cell의 생육도와 Cytokine 분비량을 확인한 결과도 마찬가지로 물 추출물보다는 초음파를 병행한 추출물을 더 높은 면역 증진 효과를 보였다. 이는 초음파를 통해 발생되는 액체 상호간의 상승작용으로 인하여 생약재중에 함유되어 있는 많은 phenol성 물질 및 식물성분의 증가로 생리활성 물질의 증가를 가져오기 때문이다. 더욱이 $100^{\circ}C$가 아닌 $60^{\circ}C$에서 더 높은 면역 증진활성을 보인 이유는 액체 상태의 과도한 열로 인하여 내부 식물성분의 파괴 및 열 변성을 가져와 이런 결과를 가져온 것으로 생각된다. 이상의 결과로 기존의 $100^{\circ}C$ 물 추출물보다는 $60^{\circ}C$의 비교적 낮은 온도에서 초음파 추출을 통한 생약재의 추출물들이 더 높은 활성을 나타내므로 초음파 추출에 대한 다양한 실험 모델을 통해 더 많은 실험이나 기능성 식품 및 소재 개발에 이용되고자 한다.

인삼사포닌의 흰쥐 LDH-A 유전자와 흰쥐 간세포 재생시 DNA 합성률에 미치는 유도효과 (Inductive Effects of Ginseng Saponins on the Rat LDH A-gene and the Synthetic rate of Hepatocyte DNA in Regenerating Rat Liver Cells)

  • The effects of ginseng saponins, SRbl and G-Rc on the rat liver LDH A-gene transcriptional activity was investigated during prereplicative phase of rat liver after partial hepatectomy. Changes in LDH A-mRNA levels in regenerating rat liver after intraperitoneal administrations of G-Rbl or 'G-Rc were tested by slot blot hybridization methods. The results showed that G-Rbl (1 mg/100g B.W) and G-Rc (1 mg/100g B.W) caused marked increases of LDH A-mRNA contents by respectively 1.9- and 1.5-fold in rat liver at 5-hours after partial hepatectomy Dose dependent elect of G-Rbl and G-Rc (1-25 mg/ 100g B.W) on the LDH A-mRNA levels on regenerating rat liver were also analyzed. The maximal increases of liver LDH A-mRNA levels were observed with the doses of 1 mg for G-Rbl and 5 mg for G-Rc. However, when the administration doses of G-Rbl and G-Rc were increased to 20 mg, G-Rbl caused a marked decrease of LDH A-mRNA level to 61% of those in sham-operated rat liver. In contrast, G-Rc slightly decreased the liver LDH A-mRNA contents by 30% as compared to those of the maximum value but still maintained 22% higher LDH A-mRNA levels then those of sham-operated rate liver. On the basis of these experimental results, we conclude that ginseng saponin, G-Rbl and G-Rc have stimulatory effect at the lower concentration (1 mg/ 100g B.W) and inhibitory effect at the higher concentration (20 mg/ 100g B.W) on the LDH A-gene transcription during regeneration of rat liver. Additionally we also investigated the stimulatory effects of ginsenosides on the protein and DNA sinthetic activities in hepatocyte primary cell cultures isolated from regenerating rat liver. Both of G-Rc and -Re increased the synthetic rates of hepatocytes proteins and DNA at the administration doses of 50 us and 100 $\mu\textrm{g}$/3 ml/dish respectively representing 1.3-1.6 fold increases. From these results we postulate that G-Rc and -Re may have a mitogen ehincer activity for the hepatocyte proliferation during rat liver regeneration period.

생리식염수 세척이 구강 소독제의 타액 내 세균 수의 변화에 미치는 영향 (Effect of saline irrigation used in combination with antimicrobial agents on salivary bacterial counts)

  • Introduction: The aim of the present study was to investigate the effect of mechanical irrigation in combination with mouthwash of antimicrobial agents on salivary bacterial counts. Materials and Methods: This study was performed with a randomized study employing a panel of 40 healthy volunteers (20 males and 20 females) between the age of 26 and 32 years. Volunteers were randomly put in one of four treatment groups. In the first group, 0.2 mL of non-stimulatory saliva was collected from every subjective person. Then, saliva was collected after rinsing with chlorhexidine (CHX) for 1 minute. In the second group, non-stimulatory saliva was collected, and then saliva was collected after rinsing with CHX and irrigation with saline. In the third and fourth groups, the same procedures as the first and second groups were performed with povidone iodine (PVI) instead of CHX. All of these samples were cultured for 48 hours aerobically. The reduction rates of colony-forming units (CFU) were calculated for each group. The reduction rate between each group was tested statistically using student t-test. Results: Using CHX in combination with saline irrigation showed a significant decrease of the salivary bacterial CFU when compared with only using CHX.(P<0.01) And using PVI with saline irrigation showed a little decrease of the CFU when compared with only using PVI, but there was no statistical significance.(P>0.01) Conclusion: It was concluded that the CHX or PVI used with saline irrigation made the salivary bacterial counts reduced more than when CHX or PVI was used alone as an oral antiseptic agent.

A Protective Effect for Panax ginseng in the Rat Stomach

  • The effect of ginseng on gastric ulcer and gastric acid secretion was investigated in pylorus-ligated rats. Methods: Sprague-Dawley strain rats were used after 24 hours fast. Pylorus-ligation was performed under light ether anaesthesia, then gastric mucosal damage was evoked in conscious pylorus-ligated rats by the administration of subcutaneous (s.c.) indomethancin (20mg/kg), s.c. histamine (150mg/kg) or by pylorus-ligation (Shay ulcer). Ginseng was given by intragastric (i.g.) or intraperitoneal (i.p.) route simultaneously with the ulcerogens. Rats were killed after 3h (indomethacin) and histamine models) or after 18h (Shay ulcer), when the gastric secretory responses, the number and severity of gastric mucosal lesions and mucosal mucus content deetermined. the effect of i.p. ginseng on basal gastric acid secretion and on gastric acide secretion in indomethacin (20mg/kg, s.c.)-treated rats was also investigated in urethane anesthetized rats. Gastric acid secretion was measured by flushing of the gastric lumen with saline every 15min through an oesophageal cannula. Results: In conscious pylorus-ligated rats, i.g. ginseng(12.5-50mg/$m\ell$; 50-200mg/kg) protected against gastric mucosal lesions evoked by s.c. indomethacin or s.c. histanmine in the d3-h pylorus-lighted rat, withoutmodifying gastric acid secretory responses. Ginseng given i.p. (150 or 200mg/kg) did not reduce the gastric lesions produced by histamine or by ligating the pylorus (Shay ulcer) Ginseng given orally in 50mg/$m\ell$ (200mg/kg) increased gastric mucus secretion in saline- and indomethacin-treated conscious pylorus-ligated rats. In anaesthetized rats ginseng (50 or 200mg/kg) did not modify basal gastric acid secretion or gastric acid secretion in the indomethacin-treated rats. Conclusions: ginseng given orally exerts gastroprotective effects in the rat stomach. Such anti-ulcer effect does not involve changes in gastric acid secretory responses. In addition, ginseng possesses stimulatory effect on gastric mucus secretion, which could be one mechanism by which the compound exerts its antiulcer effect. Our data are in favor for a beneficial effect for topically applied ginseng on the gastric mucosa.

The effect of PAHs on the regualtion of CYP1 gene in ZR-75-1 and MCF7 human breast cancer cells

  • Recent industrial society has human widely exposed to PAHs that are coming from the incomplete combustion of organic material as widespread environmental contaminants. Biological activities of PAHs are not known although PAHs are considered as carcinogens. The mechanism of action of PAHs has been studied extensively, however it is not clear how PAHs turn on CYPlAl in human breast cancer, Our laboratory have been studied the effect of PAHs in the human breast cancer cells, MCF-7. In this study, we examined the ZR-75-1, human breast cancer cells, as a new system to evaluate bioactivity of PAHs and to compare the PAHs action with that of MCF-7 cells. ZR-75-1 human breast cancer cell line is responsible to estrogen and progesterone. We have been able to establish long term culture system of this cells then used for the study to the effect of 13 different PAHs and environmental samples. We demonstrate that PAHs induced the CYP1A1 promoter and 7-ethoxyresorufin O-deethylase (EROD) activity in a concentration-dependent manner. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1A1 mRNA. Some of PAHs showed stronger stimulatory effect on CYP1 gene expression than TCDD Apparently, ZR-75-1 cells have Aryl hydrocarbon receptors (AhR), therefore it would be a good experimental tool to study the cross-talk between PAHs and steroid actions.

자하차(紫河車)약침이 제2형 콜라겐 유발 관절염에 미치는 영향 (Effect of Hominis placenta Herbal Acupuncture on Type II Collagen-induced Arthritis in Mice)

  • Objectives : We investigated the effects of Hominis placenta Herbal acupuncture on rheumatoid arthritis. Methods : We investigated the Type II collagen-induced arthritis test, vascular permeability test, phenyl benzoquinone-induced writhing test, hot plate test, carbon clearance test, determination of prostaglandin $E_2$ content and cyclooxygenase-2 activity test. Results : Hominis placenta Herbal acupuncture suppressed Type II collagen-induced arthritis in DBA/1J mice and vascular permeability with 10.2% inhibition in ICR mice. We see the phenyl benzoquinone-induced writhing test and the hot plate test in the analgesic activity test. In the phenyl benzoquinone-induced writhing test, Hominis placenta Herbal acupuncture showed that it increased its analgesic effect more than the control(test?) by 85.5%. In the hot plate test, it was also shown that the analgesic effect increased more than the control(test) by 32.1%. In the carbon clearance test, the immuno-stimulatory effect showed more than the control(test) by 8.0%. The formation of prostaglandin $E_2$ was also reduced more than the control(test) by 10.1%. Finally, cyclooxygenase-2 activity was inhibitedwith 36.3%. Conclusions : Hominis placenta Herbal acupuncture may be useful for the treatment of rheumatoid arthritis after injection.

Antifilarial potential of the root extracts of Mirabilis jalapa Linn. (Nyctaginaceae) on cattle filarial parasite Setaria cervi

  • Effect of aqueous and alcoholic extracts of the roots of Mirabilis jalapa Linn. Four Oclock plant, on the spontaneous movements of both the whole worm and the nerve-muscle (n.m.) preparation of Setaria cervi and on the survival of microfilariae in vitro was studied. Alcoholic extract caused the inhibition of spontaneous movements of the whole worm and the n.m. preparation of S. cervi, whereas aqueous extract caused inhibition of spontaneous movements of the n.m. preparation. The initial stimulatory effect was not observed by aqueous and alcoholic extracts on n.m. preparation while effect of alcoholic extract on the whole worm was characterized by an increase in the amplitude of contractions followed by reversible paralysis. The concentrations required to inhibit the movements of the whole worm and n.m. preparation for alcoholic extract of root were $270\;{\mu}g/mL$ and $40\;{\mu}g/mL$, respectively whereas an aqueous extract caused inhibition of n.m. preparation at $30\;{\mu}g/mL$ suggesting a cuticular permeability barrier. Alcoholic extract of the roots of M. jalapa caused concentration related effect on the survival of microfilariae of S. cervi. The $LC_{50}$ and $LC_{90}$ for alcoholic extract as observed after 6 hrs. were found to be 10 ng/mL and 18 ng/mL., respectively.

산화질소가 인간 치주인대세포의 증식과 분화에 미치는 영향 (Effects of nitric oxide on the proliferation and differentiation of human periodontal ligament cells)

  • 교정적 치아이동과 밀접한 관련이 있는 치주인대세포에서, 산화질소가 세포의 증식과 분화에 미치는 영향을 알아보고자 하였다. 인간 치주인대세포를 분주한 후에 산화질소의 donor인 SNP의 농도에 따라 실험군을 구분하고, 세포 활성 염기성 인산분해효소 활성, Western blot 분석을 통한 osteonectin과 bone sialoprotein의 발현 정도를 측정하였다. 0.2 mM 이하의 저농도 SNP를 처리한 실험군에서 대조군과 비교하여 세포 활성, 염기성 인산분해 효소의 활성, 그리고 osteonectin과 bone sialoprotein의 발현이 유의하게 증가하였다. 그러나 1 mM 이상의 고농도 SNP를 처리한 실험군에서는 오히려 감소하였다. 산화질소는 인간 치주인대세포에서 저농도는 세포의 증식과 분화에 촉진 효과를, 고농도는 억제 효과를 보이는 biphasic effect를 갖는다.

경골 파혈산동탕(破血散疼湯)이 골절 생쥐의 골 유합에 미치는 영향 (Effect of Pahyeolsandong-tang (Poxiesanteng-tang) in Tibia Fracture-induced Mice)

  • Objectives The main purpose of this study was to evaluate the bone healing effect of Pahyeolsandong-tang (PHT)(Poxiesanteng-tang) extract in tibia fracture-induced mice. Methods PHT was extracted using a solution of 35% ethanol in 60℃ for 8 hours. Mice were randomly divided into 4 groups (normal, control, PHT 50 and PHT 100). Mice of experimental groups were medicated with PHT 50 or 100 mg/kg for 7 to 21 days. To clarify the effect of bone fracture healing, relative messenger RNA (mRNA) expressions of osteocalcin (OCN), runt-related transcription factor 2 (Runx2), osterix (OSX), Sox9, collagen type II alpha 1 chain (Col2a1), receptor activator of nuclear factor kappa-B ligand (RANKL), osteoprotegerin (OPG) were examined. Results In in vitro experiment, relative mRNA expression of OCN, Runx2, Col2a1 was significantly increased in PHT treated group to compare with control differentiation group. In in vivo experiment, relative mRNA expression of OCN, Runx2, OSX, Sox9, Col2a1, RANKL, OPG was significantly increased in PHT treated group. Conclusions This study showed that PHT accelerates bone fracture healing through the activation of osteoclasts and osteoblasts. It was showed that PHT significantly promotes osteoblasts differentiation by osteoblast differentiation markers such as OCN, Runx2, Col1a2. Also it was investigated that PHT had stimulatory effect on osteoblasts function through enhancing OCN, Runx2, OSX, Sox9, Col2a1 and, osteoclasts function through enhancing RANKL and OPG markers. PHT effectively promotes bone fracture healing process through activation of osteoblasts and osteoclasts.

오가피(五加皮) (Acanthopanacis cortex)추출물(抽出物)을 첨가한 발효김치의 급여가 생쥐의 면역작용(免疫作用)에 미치는 영향(影響) (Effect of Immune Function on the fermentation of Kimchi Intake to append Acanthopanacis cortex Extract in Balb/c Mice)

  • This experimental study was carried out to evaluate the effects of Kimchi intake of Acanthopanacis cortex extract (APCE) supplementation on cytokine-induction and immune response in mice. To study in experiments using male Balb/c mice fed Kimchi and Kimchi of APCE supplementation (addition of 2% of total Kimchi weight) containing fed experimental diet during 2 weeks. Experimental mice were fed control diet or diet containing freeze-dried Kimchi at the level of 5%(w/w) or 5% freeze-dried Kimchi with 2% APCE supplementation. The main ingredient of Kimchi was Korean cabbage and fermentation was carried out at $4^{\circ}C$ for three weeks. Freeze-dried 2% APCE supplementation was added to Kimchi at the begining of fermentation. In order to investigate the effect of Kimchi intake of APCE supplementation (5%Kimchi-2%APCE), the following was performed; body weight, food intake, hematological parameter, serum level of mouse interleukin-4 (mlL-4) and mouse interferon-$\gamma$ (mIFN-$\gamma$ ), and, the percentage of CD3+/CD4+, CD3+/CD8+, B220+ in splenic cells. The results of final body weight, and food diet intake of two Kimchi groups were lower than those of the control group (not supplemented experimental diet). The hematology change obtained from the level of WBC (white blood cell) and platelet were not affected by feeding different dietary regiments, but the level of RBC (red blood cells) HB (hemoglobin), and spleen weight of two Kimchi groups were increased significantly than those of the control group. The serum level of IL-4 and IFN-$\gamma$ of two Kimchi groups were increased significantly than those of the control group, also enhanced the percentages of the CD3+/CD4+ and CD3+/CD8+ by 5% freeze-dried Kimchi, and 5%Kimchi-2%APCE group were 43.9 and 65.2%, and 96.0 and 208% than those of the control group, respectively. From these results, it can be concluded that Kimchi itself has an immuno-stimulatory effect and Kimchi contaning 2% APCE supplementation has the more pronounced effect in vivo system.
