Single-stranded DNA binding protein (SSB) is well-characterized as having a helix-destabilizing activity. The helix-destabilizing capability of SSB has been re-examined in this study. The results of restriction endonuclease protection assays and titration experiments suggest that the stimulatory effect of SSB on strand exchange acts by melting out the secondary structure which is inaccessible to RecA protein binding; however, SSB is excluded from regions of secondary structure present in native single-stranded DNA. Complexes of SSB and RecA protein are required for eliminating the secondary structure barriers under optimal conditions for strand exchange.
Whether there exists a sympathetic neural regulation on the aquaporin (AQP) channels in the kidney was examined. Male Sprague-Dawley rats were used. They were renal nerve denervated by stripping the nervous and connective tissues passing along the renal artery and vein, and painting these vessels with 10% phenol solution through a midline abdominal incision. Three days later, the expression of AQP1-4 proteins in the denervated kidneys was determined. The content of norepinephrine was found significantly decreased following the denervation. Accordingly, the expression of AQP2 proteins was markedly decreased. The expression of AQP3 and AQP4 was also slightly but significantly decreased, while that of AQP1 was not. Neither the basal nor the AVP-stimulated accumulation of cAMP was significantly affected in the denervated kidney. It is suggested that the sympathetic nervous system has a tonic stimulatory effect on AQP channels in the kidney.
The effects of Ginseng saponin on respiration and $Na^+$, $K^+$ content of rat cerebral cortex slices were investigated to determine the action of Ginseng saponin on brain cortex at cellular level. There are many reports for the study of Ginseng on central stimulatory action in experimental animals. The electrical stimulation of slices of cortex causes a loss of potassium. And the respiration is needed to maintain a supply of energy for active cation transport. The reduction in $Qo_2$ is a consequence of primary cessation of active cation transport. Ginseng saponin stimulated respiration which was depressed by Morphine. But there was no significant change of electrolyte. It is suggested that the Ginseng saponin act rather on metabolic process than neural excitatory mechanism in vitro.

${\beta}$-Glucans are naturally occurring polysaccharides that are produced by bacteria, yeast, fungi, and many plants. Although their pharmacological activities, such as immunomodulatory, anti-infective and anti-cancer effects, have been well studied, it is still unclear how ${\beta}$-glucans exert their activities. However, recent studies on the ${\beta}$-glucans receptors shed some light on their mechanism of action. Since ${\beta}$-glucans have large molecular weights, they must bind surface receptors to activate immune cells. In this review, we summarize the immunopharmacological activities and the potential receptors of ${\beta}$-glucans in immune cells.
Dexamethasone (Dex) was shown to inhibit the differentiation, maturation, and antigen-presenting function of dendritic cells (DC) when added during DC generation or maturation stages. Here, we examined the direct effects of Dex on MHC-restricted antigen processing. Macrophages were incubated with microencapsulated ovalbumin (OVA) in the presence of different concentrations of Dex for 2 h, and the efficacy of OVA peptide presentation was evaluated using OVA-specific CD8 and CD4 T cells. Dex inhibited both class I- and class II-restricted presentation of OVA to T cells; this inhibitory effect on antigen presentation was much more potent in immature macrophages than in mature macrophages. The presentation of the exogenously added OVA peptide SIINFEKL was not blocked by Dex. In addition, short-term treatment of macrophages with Dex had no discernible effects on the phagocytic activity, total expression levels of MHC molecules or co-stimulatory molecules. These results demonstrate that Dex inhibits intracellular processing events of phagocytosed antigens in macrophages.
The possible effects of garlic (Aliium sativurn L.) extract on growth and aflatoxin production by Aspergillus parasiticus R- 716 were investigated. Various solvent extracts of garlic strongly inhibited growth and sporulation by Aspergillus parasiticus R-716, and effective solvents used for extraction of garlic were chloroform, benzene, and water-chloroform. The growth and aflatoxin production decreased with the increase in extract concentration, and extract equivalent 1.5g of raw garlic weight in 25ml SLS medium completely inhibited, and at a level of 1.25g garlic, total aflatoxin was reduced 64% (472 ${\mu}g$/25ml) of that produced in the control (1, 352 ${\mu}g$/25 ml). During cultivation inhibitory rate of growth was reduced from 89.1% to 40% and aflatoxin $B_2$, $G_1$ production increased with the laps of time. Especially garlic extract appeared to have a stimulatory effect on lipid accumulation on the contrary aflatoxin production.
Burkholderia cepacia is an important pathogen that often causes pneumonia in immunocompromised individuals. Here, it was demonstrated that the TLR5 agonist flagellin could locally activate innate immunity. This was characterized by rapid expressions of IL-$1{\beta}$, TNF-${\alpha}$, and iNOS mRNA and a delay in the expression of IL-10 mRNA. A significant elevation in the IL-$1{\beta}$, TNF-${\alpha}$, and nitric oxide levels was also noted. In the respiratory tract, flagellin induced neutrophil infiltration into the airways, which was observed by histopathological examination and confirmed by the neutrophil count and level of myeloperoxidase activity. This was concomitant with a high activity of alveolar macrophages that engulfed and killed B. cepacia in vitro. The flagellin mucosal treatment improved the B. cepacia clearance in the mouse lung. Thus, the present findings illustrate the profound stimulatory effect of flagellin on the lung mucosal innate immunity, a response that needs to be exploited therapeutically to prevent the development of respiratory tract infection by B. cepacia.

Keloids are characterized by abnormal proliferation of fibroblasts and overproduction of collagen. Recently, it is reported that transforming growth factor beta (TGFb) and its signaling molecule, SMAD3 are related to the mitogenic effect of fibroblasts and a stimulatory factor for collagen synthesis. Cationic SLN was developed to improve the complex formation of DNA/SLN and enhance the uptake efficiency to cells. SLN was formulated by DC-Chol, DOPE, trimyristin as a solid core and other surfactant. The physical properties of the SLN and the ATS-SLN complex were characterized. (omitted)
포도당을 포함한 생장 배지속에 L-사과산을 첨가하면 Leuconostoc oenos균들의 생장률을 크게 촉진 시켰다. L-사과산은 동시에 C-젖산탈수소 효소(D-LDH)의 합성과 활성도를 촉진시켜 포도당에서 D-젖산의 생산을 빠르게 하였다. L-사과산 존재 하에서 이렇게 빨라진 포도당의 이용이 leuconostoc 균들의 생장률을 촉진시키는 것 같다.
백수오의 국내 재배활성화와 함께 농가소득을 높이기 위한 기능성 식품소재로서의 우수성을 산업적으로 응용하고자 백수오 추출물의 면역증진 활성을 평가하였다. 낮은 온도에서 액체상태의 추출물을 효과적으로 농축하고 정제하기 위하여 비열처리 공정인 한외거르기를 통해 분자량(MWCO)이 30 kDa인 여과막을 이용하여 고분자 분획물(CWUF>30) 및 저분자 분획물(CWUF<30)을 얻었으며 이를 큰포식세포 활성화를 통한 면역 증진 효과를 알아보기 위해 RAW 264.7 세포를 이용하여 면역활성의 지표가 되는 산화질소(II) 생성량 및 사이토카인인 TNF-${\alpha}$, 인터루킨-6의 생성량을 측정한 결과, 정상대조군(CON)에 비해 CWUF>30을 처리하였을 때 농도 의존적으로 유의하게 증가하였다. 세포실험을 통해 면역증강활성이 높은 CWUF>30은 정상동물모델에서 면역기관인 비장 및 흉선의 무게를 증가시켰고 비장세포 유래 림프구 증식도 유의하게 증가시켰다. 이는 CWUF>30의 투여가 비장세포를 증식시키는 mitogen 활성이 있음을 보여주며 외부의 항원에 노출 시 항원에 대한 면역반응을 유도하는 면역세포의 수를 증가시켜 항원에 대한 효과적인 방어에 도움을 줄 것으로 보여진다. 또한 CWUF>30을 투여한 마우스 비장세포(splenocyte)의 Yac-1 세포 살해 정도를 측정한 결과, 200 mg/kg BW의 농도로 CWUF>30을 투여한 마우스의 비장세포는 정상대조군(CON)에 비하여 effector 세포와 target 세포의 비율이 10:1에서 유의적으로 증가하였으며 이는 양성대조군(PC)과 유사한 높은 활성을 보이는 것을 확인 할 수 있었다. 이와 같은 결과를 통해 백수오 고분자 분획물(CWUF>30)이 강력한 면역활성 증진효능을 갖고 있으며 각종 바이러스 등 외부 항원들에 대응하여 초기 면역세포를 자극하고 면역매개물질을 생성함으로써 인체의 비특이적 면역반응을 증가시키는데 중요한 역할을 할 수 있을 것으로 기대된다.
